Ketoprofen Associated with Hyaluronic Acid Hydrogel for Temporomandibular Disorder Treatment: An In Vitro Study.

Temporomandibular disorders (TMD) are a public health problem that affects around 12% of the global population. The treatment is based on analgesics, non-steroidal anti-inflammatory, corticosteroids, anticonvulsants, or arthrocentesis associated with hyaluronic acid-based viscosupplementation. However, the use of hyaluronic acid alone in viscosupplementation does not seem to be enough to regulate the intra-articular inflammatory process. So, we propose to develop and evaluate the physicochemical and biological properties in vitro of hyaluronic acid hydrogels (HA) associated with ketoprofen (KE

A comprehensive analytical model for predicting drug absorption in the olfactory region: Application to nose-to-brain delivery.

Nose-to-brain drug delivery offers a promising route for administering pharmaceutical substances directly to the brain. However, this pathway faces significant challenges, such as mucociliary clearance and enzymatic activity in the nasal cavity, which limit drug absorption. Although several formulation strategies exist to enhance drug solubility, diffusion across the airway surface liquid, and protection from enzymatic degradation, there is a lack of tools to systematically evaluate which strategy is best suited for specific molecules. To address this, we developed an analytical model of the o

Enhanced oral delivery of antimony from meglumine antimoniate/beta-cyclodextrin nanoassemblies.

The composition comprising the highly water-soluble drug meglumine antimoniate (MA) and beta-cyclodextrin (beta-CD) was shown previously to enhance the absorption of Sb by oral route and render MA orally active in a murine model of cutaneous leishmaniasis. This unexpected behaviour was attributed, in part, to the fact that the heating of equimolar mixture of MA and beta-CD (first step of preparation of MA/beta-CD composition) induced the depolymerization of MA from high-molecular weight Sb complexes into 1:1 Sb-meglumine complex, resulting in an enhanced oral bioavailability of Sb. Evaluation of a novel micro-sampling device, Mitra™, in comparison to dried blood spots, for analysis of praziquantel in Schistosoma haematobium-infected children in rural Côte d'Ivoire.

Pharmacokinetic (PK) studies with paediatric populations are increasing in importance for drug development. However, conventional PK sampling methods are characterised by invasiveness and low patient adherence, unsuitable for use with sensitive population, such as children. Mitra™ is a novel volumetric absorptive micro-sampling device, which offers an alternative to the dried blood spotting (DBS) technique, a current popular sampling technique within PK studies. We tested Mitra™ for the first time in the framework of a randomised controlled trial in rural Côte d'Ivoire. Thirty-five school-aged

Bone Regeneration Using Hydroxyapatite Sponge Scaffolds with In Vivo Deposited Extracellular Matrix.

There is currently an increased interest in studying the extracellular matrix (ECM) and its potential applications for tissue engineering and regenerative medicine. The ECM plays an important role by providing adhesive substrates to cells during migration, morphogenesis, differentiation, and homeostasis by signaling biochemical and biomechanical cues to cells. In this study, the ECM was incorporated into hydroxyapatite by implanting sponge replica scaffolds in subcutaneous pockets in rats, and the implants were tested for bone regeneration potential. The resulting scaffolds were characterized

Exploring the properties of antituberculosis drugs through QSPR graph models and domination-based topological descriptors.

Tuberculosis (TB) is a global health concern caused by the bacterium Mycobacterium tuberculosis. This infectious disease primarily affects the lungs but can also impact other organs. Effective TB control involves early diagnosis, appropriate treatment with a combination of antibiotics, and public health measures to prevent transmission. However, ongoing challenges include drug-resistant strains and socioeconomic factors influencing its prevalence. Drugs such as isoniazid, pyrazinamide, ethambutol, ethionamide, linezolid, and levofloxacin are approved for the treatment of drug-susceptible tuber

Impact of yeast cell wall incorporation on the mucoadhesion, stability, oral permeability and release profile of alginate/whey protein beads loaded with insulin.

This study aimed to evaluate the impact of incorporating yeast cell wall (YCW) into alginate/whey protein (ALG/WP) particles as a strategy to improve oral insulin delivery. Insulin-loaded particles were produced by an extrusion-gelation process with or without YCW, and their physicochemical, mucoadhesive, and permeability properties were assessed in vitro, ex vivo, and in vivo. The inclusion of YCW increased the viscosity of the polymeric solution, resulting in more cohesive particles and a significant reduction in insulin loss during coating. Encapsulation efficiencies ranged from 65 to 99%.

A novel nanomatrix system consisted of colloidal silica and pH-sensitive polymethylacrylate improves the oral bioavailability of fenofibrate.

A novel solid particle system with a nanomatrix structure and without surfactant for the oral delivery of insoluble drugs was prepared. This used a combination of pH-sensitive polymethylacrylate and nano-porous silica, in order to improve the drug absorption using only pharmaceutical excipients and a relative simple process. The in vitro drug dissolution and in vivo oral bioavailability of this formulation, using fenofibrate as the model drug, were compared with other reference formulations such as a suspension, micronized formulation or self microemulsion drug delivery system (SMEDDS). Binding of plutonium and americium to small molecular weight ligands favors their paracellular transfer across lung epithelium in vitro.

Pulmonary absorption is a critical step in the time-dependent processes governing actinide biodistribution and toxicity following internal contamination by inhalation. The current study was undertaken to investigate on a simple in vitro Calu-3 model the mechanisms involved in the transfer across epithelial cells of plutonium (Pu) and americium (Am) as a function of their intrinsic properties (nature, physicochemical form) or their association with serum proteins, inorganic ligands or synthetic chelating agents. Both the contribution of the transcellular and paracellular routes to the transfer

Combustion Conditions are the Major Drivers of the Variability in Organic Aerosol Chemical Composition and Secondary Organic Aerosol Formation in Emissions from Simulated Wildland Fires.

Wildland-fire emissions exhibit large variability in aerosol physicochemical properties and potential for forming secondary organic aerosol (SOA). Whether this variability is primarily driven by differences in fuel type or combustion conditions remains unresolved. To address this gap, we performed simulated wildland-fire experiments using fuel beds that contained either surface fuels only or a duff layer underneath the surface fuels. We varied the dry mass loading and moisture content of the surface fuels within ranges encountered in prescribed fires in the Southeastern U.S. Less efficient com

Toward Rational Design of Orally Bioavailable PROTACs: Development of a Fully Computational Oral Absorption Score.

Achieving adequate oral absorption for proteolysis-targeting chimeras (PROTACs) remains challenging due to their size, polarity, and conformational complexity. While existing approaches often rely on physicochemical property space or chameleonicity-driven models, a generalizable framework for guiding compound design prior to synthesis is lacking. Here, we introduce comPASS, a fully computational scoring function derived from molecular dynamics-based descriptors that is designed to capture the balance between molecular extension and exposed polarity through normalized radius of gyration and thr

Preparation, Characterization, and In Vivo Evaluation of an Oral Triptolide Nanomatrix System for Rheumatoid Arthritis Therapy.

Background: Triptolide (TP), a principal bioactive component of Tripterygium wilfordii, exhibits potent anti-inflammatory activity. However, its application is still limited due to its poor solubility and systemic toxicity, primarily caused by uncontrolled absorption after oral administration. Our previously established oral nanomatrix system, composed mainly of commercially available nanoporous Sylysia and Eudragit®, can not only enhance the in vitro dissolution of poorly water-soluble drugs, but also modulate their absorption sites in gastrointestinal tract. Methods: We prepared a TP nanomat

The Interplay Between Liver First-Pass Effect and Lymphatic Absorption of Cannabidiol and Its Implications for Cannabidiol Oral Formulations.

For highly lipophilic drugs, passage into the intestinal lymphatic system rather than the portal vein following oral administration may represent a major alternative route of delivery into the general circulation. Increasing intestinal lymphatic transport provides an effective strategy to improve oral bioavailability when hepatic first-pass metabolism is a major rate-limiting step hampering access to the systemic circulation after oral dosing. The transfer of orally administered, highly lipid-soluble drugs to the lymphatic system is mediated by their association with chylomicrons, large intest

Surface modification strategies of oral liposomes: functional design and barrier enhancement.

Oral administration is the most prevalent and preferred clinical route due to its non-invasiveness, high patient compliance, and convenience. However, the oral delivery of many therapeutic drugs is hindered by low bioavailability, attributed to multiple gastrointestinal (GI) barriers including acid degradation, enzymatic hydrolysis, poor epithelial permeability, and first-pass metabolism. Liposomes have emerged as promising oral nanocarriers owing to their biocompatibility, versatile drug-loading capacity, and biomimetic membrane structure. Nevertheless, their poor physicochemical stability an
