The influence of gender, diabetes, and acetaldehyde on the intrinsic contractile properties of isolated rat myocardium.

Diabetes is associated with ventricular dysfunction. Ethanol consumption increases the risk of cardiovascular disease among diabetics. Acetaldehyde (ACA), the main ethanol metabolite, depresses cardiac contraction and contributes to ethanol-induced cardiac dysfunction. This study examined the influence of gender and diabetes on ACA-induced myocardial dysfunction. Adult male and female rats were made diabetic with streptozotocin (55 mg/kg). Left ventricular papillary muscles were isolated and stimulated to contract at 0.5 Hz. The mechanical parameters measured were peak tension development, tim

Effects of liver disease on red blood cell acetaldehyde in alcoholics and non-alcoholics.

High endogenous levels of acetaldehyde were found in the blood (mainly in red blood cells) of patients with liver disease of various etiologies. The presence of severe liver injury increased the blood acetaldehyde response to ethanol consumption and resulted in a more prolonged persistence of high levels after ethanol withdrawal.

Role of acetaldehyde in ethanol-induced increase in the activity of phosphatidate phosphatase in rat liver.

The effect of ethanol on the activity of phosphatidate phosphatase was studied in rat liver using membrane-bound phosphatidate and phosphatidate emulsion as substrate. A single large dose of ethanol (5 g/kg body wt) caused an increase in the enzyme activity measured with membrane-bound phosphatidate after an approximate 2-hr lag period in both cytosolic and microsomal fraction and the increase was approximately 2.2- and 1.8-fold that in control rats at 5 hr in cytosol and microsomes, respectively. A similar time-course of the increase was obtained with phosphatidate emulsion as substrate. Thes

Effect of ethanol, acetaldehyde, and acetate on insulin and glucagon secretion in the perfused rat pancreas.

The effects of varying concentrations of ethanol (1, 10, and 30 mM) and its metabolites (1 mM acetate and 1 and 10 mM acetaldehyde) on insulin and glucagon secretion induced by glucose (11.1 mM) and arginine (20 mM) were studied in isolated perfused pancreas of Sprague-Dawley rats. Ethanol and its metabolites did not significantly modify basal secretion of the two hormones. Ethanol reduced glucose-induced insulin secretion by means of a dose-related effect. Arginine-induced insulin output did not seem to be influenced to any significant degree. Acetate and acetaldehyde significantly inhibited

Effects of acetaldehyde on the membrane resistance in the smooth muscle of the guinea-pig taenia caecum.

The action of acetaldehyde on the membrane resistance of the guinea-pig taenia caecum was investigated with a double sucrose-gap method in different ionic environments. Application of acetaldehyde (10--20 mM) produced hyperpolarization, block of spike discharge and reduction of the membrane resistance. The reduction of the membrane resistance by acetaldehyde was potentiated by increasing the external K+ concentration and by replacement of Cl- with NO3-; it was reduced by lowering the external K+ concentration and by replacement of Cl- with benzenesulfonate. Replacement of NaCl with Tris-Cl, su

Acetaldehyde level in the blood and liver aldehyde dehydrogenase activities in trichloroethylene-treated rats.

The liver NAD+-dependent aldehyde dehydrogenase (AldDH) activity and the acetaldehyde level in the blood during ethanol metabolism after trichloroethylene (trichlene) exposure were studied in rats. Trichlene inhalation caused large elevations in acetaldehyde levels during ethanol metabolism and caused decreases in the activity of the AldDH with a low Km value in mitochondrial and soluble fractions of liver cells. No significant effects were found in the activity of the high Km-enzyme in mitochondrial, soluble and microsomal fractions. Time course of inhibition of the mitochondrial low Km-enzym

Targeting microbiota-generated acetaldehyde to prevent progression of metabolic dysfunction-associated steatotic liver disease.

The progression from metabolic dysfunction-associated steatotic liver disease (MASLD) to steatohepatitis (MASH) entails rapid, often irreversible hepatic injury, underscoring the urgent need for innovative therapeutic strategies. Here, we demonstrate that excessive dietary sugar intake, particularly fructose, exacerbates liver disease progression through microbiota-mediated amplification of endogenous acetaldehyde production. Analysis of over 210,000 participants from the UK Biobank revealed a dose-dependent correlation between sugar consumption and liver-related mortality, accompanied by a mi

Natural Sunlight IR-Driven Highly Efficient Synthesis of Acetaldehyde From Bioethanol Over Cu/Fe2O3.

Using renewable biomass to synthesize valuable chemicals can reduce fossil fuel dependence and achieve carbon neutrality. Here, for the first time an infrared light-driven catalyst, Cu/Fe2O3, was designed to convert bioethanol to valuable acetaldehyde, accompanied by green hydrogen as a by-product, under both indoor IR light and natural sunlight. It achieves an initial acetaldehyde yield of 237 mmol g-1 h-1 under indoor IR irradiation and 205 mmol g-1 h-1 under real sunlight, with exceptional selectivity (97.7%) and nearly stoichiometric H2 byproduct production. Notably, the turnover number an

Biodegradation of ethanol and acetaldehyde by Lactiplantibacillus plantarum YDL-01 from healthy human feces.

Excessive alcohol intake triggers a series of systemic health hazards through the accumulation of acetaldehyde, a major carcinogen that poses a severe threat to human health. Here, a promising probiotic bacterial strain capable of biodegrading both ethanol and acetaldehyde was successfully isolated from healthy human feces and identified as Lactiplantibacillus plantarum YDL-01. In vitro assays demonstrated that an initial concentration of 5.0 g/L of ethanol or acetaldehyde could be completely biodegraded within 72 h or 16 h, respectively, by whole cells or cell-free extracts (CEs) of YDL-01, i

Brain Acetaldehyde Exposure Impacts upon Neonatal Respiratory Plasticity and Ethanol-Related Learning in Rodents.

Prior studies indicate that neonates are very sensitive to ethanol's positive reinforcing effects and to its depressant effects upon breathing. Acetaldehyde (ACD) appears to play a major role in terms of modulating early reinforcing effects of the drug. Yet, there is no pre-existing literature relative to the incidence of this metabolite upon respiratory plasticity. The present study analyzed physiological and behavioral effects of early central administrations of ethanol, acetaldehyde or vehicle. Respiration rates (breaths/min) were registered at post-natal days (PDs) 2 and 4 (post-administra

Acetaldehyde-ethanol interactions on calcium-activated potassium (BK) channels in pituitary tumor (GH3) cells.

BACKGROUND: In the central nervous system ethanol (EtOH) is metabolized to acetaldehyde (ACA) primarily by the oxidative enzyme catalase. Evidence suggests that ACA is responsible for at least some of the effects on the brain that have been attributed to EtOH. Various types of ion channels which are involved in electrical signaling are targets of EtOH like maxi calcium-activated potassium (BK) channels. BK channels exhibit various functions like action potential repolarization, blood pressure regulation, hormone secretion, or transmitter release. In most neuronal and neuroendocrine preparation

Experienced acetaldehyde reaction does not improve treatment response in outpatients treated with supervised disulfiram.

AIM: The major mode of action of disulfiram is assumed to be a psychological deterrence of an acetaldehyde reaction after alcohol consumption. However, it is still unclear whether patients need to experience an acetaldehyde reaction with the help of a "test drink" at the beginning of the therapy to achieve a better efficacy. Therefore, in this study, we test the hypothesis if an experienced acetaldehyde reaction during the therapy with disulfiram predicts better treatment outcome in alcohol-dependent patients. METHOD: We evaluated outcome data of 46 patients treated with supervised disulfiram

Sensitizing potential of acetaldehyde and formaldehyde using a modified cumulative contact enhancement test (CCET).

The contact allergenic activity of acetaldehyde was investigated with a modified cumulative contact enhancement test (CCET) method in guinea pigs. Possible cross-reactivity between acetaldehyde and formaldehyde was also studied. In contrast to the original CCET protocol, we used sham-treated controls and the chemicals were tested with closed epicutaneous application at 1st challenge. The suitability of the method was verified with formaldehyde and the results were comparable with those previously found with the guinea pig maximization test (GPMT). For the 1st time, acetaldehyde was shown to be

Citrullinated and Malondialdehyde-Acetaldehyde-Modified Fibrinogen Activates Macrophages and Promotes Coronary Endothelial Cell Inflammation.

Individuals with rheumatoid arthritis (RA) face increased cardiovascular mortality due to heart failure (HF) complications. Post-translational modifications, such as citrullination (CIT) and malondialdehyde-acetaldehyde (MAA) adduction, are implicated in RA pathogenesis. However, their role in RA-associated HF is not well understood. This study examines the deposition of MAA and CIT in cardiac tissues of RA-HF patients and investigates how MAA and CIT adducts on fibrinogen (FIB-MAA-CIT) drive crosstalk between macrophages and endothelial cells in vitro. We demonstrated elevated MAA and CIT add

Evaluation of Analysis Methods for Formaldehyde, Acetaldehyde, and Furfural from Fast Pyrolysis Bio-oil.

Fast pyrolysis bio-oil (FPBO), a second-generation liquid bioenergy carrier, is currently entering the market. FPBO is produced from biomass through the fast pyrolysis process and contains a large number of constituents, of which a significant part is still unknown. Various analytical methods have been systematically developed and validated for FPBO in the past; however, reliable methods for characterization of acetaldehyde, formaldehyde, and furfural are still lacking. In this work, different analysis methods with (HS-GC/ECD, HPLC, UV/Vis) and without derivatization (GC/MSD, HPLC) for the cha
