The tuberous sclerosis complex model Eker (TSC2+/-) rat exhibits hyperglycemia and hyperketonemia due to decreased glycolysis in the liver.

Tuberous sclerosis complex (TSC) presents as benign tumors that affect the brain, kidneys, lungs and skin. The inactivation of TSC2 gene, through loss of heterozygosity is responsible for tumor development in TSC. Since TSC patients are carriers of heterozygous a TSC2; mutation, to reveal the risk factors which these patients carry prior to tumor development is important. In this experiment, Eker rat which carry a mutation in this TSC2 gene were analyzed for their metabolic changes. Wild-type (TSC2+/+) and heterozygous mutant TSC2 (TSC2+/-) Eker rats were raised for 100 days. As a result, the

L-Cysteine supplementation reduces high-glucose and ketone-induced adhesion of monocytes to endothelial cells by inhibiting ROS.

Type 1 diabetic (T1D) patients are hyperglycemic and also show elevated blood levels of ketone bodies, particularly acetoacetate (AA) and β-hydroxybutyrate (BHB). T1D patients have a greater risk of developing endothelial dysfunction and cardiovascular disease (CVD). Supplementation with cysteine-rich milk proteins has been shown to be beneficial in improving various biomarkers of endothelial dysfunction and CVD. This study examines whether L-cysteine (LC) per se prevents monocyte adhesion to endothelial cells, a critical step in endothelial dysfunction. Human umbilical vein endothelial cells

Mitochondrial 2-methylacetoacetyl-CoA thiolase deficiency: an inborn error of isoleucine and ketone body metabolism.

A review is presented of 22 published cases of verified or probable mitochondrial 2-methylacetoacetyl-CoA thiolase deficiency, a disorder of isoleucine and ketone body metabolism. The clinical expression, characterized by ketoacidosis, vomiting and lethargy, is highly variable. Typical age of onset is between 6 and 24 months. The disorder, which has been observed in several ethnic groups, is apparently inherited as an autosomal, recessive trait. The prognosis is relatively good if acute episodes of ketoacidosis and dehydration are adequately treated. There is abnormal urinary excretion of 2-me

Cerebral blood flow and metabolic rate of oxygen, glucose, lactate, pyruvate, ketone bodies and amino acids.

The cerebral blood flow (CBF) and cerebral metabolic rate (CMR) of oxygen, glucose, lactate, pyruvate, ketone bodies and 24 amino acids were examined in 12 patients with presenile dementia and in seven with normal-pressure hydrocephalus. Both groups of patients showed significantly lower values of CBF and cerebral uptake of oxygen and glucose than 10 healthy subjects examined concurrently. The values decreased roughly in proportion to the degree of clinical deterioration. Furthermore, the patients exhibited a significant release of lactate and pyruvate. A positive correlation was found between

Catabolism of aromatic acids in Trichosporon cutaneum.

Trichosporon cutaneum readily metabolized protocatechuate, homoprotocatechuate, and gentisate, but lacked ring fission dioxygenases for these compounds. Benzoic, salicylic, 2,3-dihydroxybenzoic, and gentisic acids were converted into beta-ketoadipic acid before entry into the Krebs cycle. Benzoic acid gave rise successively to 4-hydroxybenzoic acid, protocatechuic acid, and hydroxyquinol (1,3,4-trihydroxybenzene), which underwent ring fission to maleylacetic acid. Salicylate and 2,3-dihydroxybenzoate were both initially metabolized to give catechol. 2,3-Dihydroxybenzoate was the substrate for

Intravenous infusion of a medium-chain triglyceride-enriched lipid emulsion.

There has been much interest in the parenteral use of medium-chain triglycerides (MCT) as a noncarbohydrate energy source. This study examines the effects of infusion iv into postabsorptive normal subjects with three doses (0.5, 1.0, 1.5 g/kg.15 h) of an emulsion containing 56% MCT and 44% long-chain triglyceride (LCT) and comparing its metabolic effect with infusions of normal saline (NS) and an emulsion containing LCT alone. There were significant increases in serum beta-hydroxybutyrate and acetoacetate 5 and 15 h after the start of the MCT-LCT infusions. Increases were seen only after 15 h

Ketone body metabolism activates the immune response against Staphylococcus aureus infection by fueling the tricarboxylic acid cycle and affecting histone β-hydroxybutyrylation.

Staphylococcus aureus (S. aureus) is a prominent human pathogen that causes persistent inflammation and is notoriously difficult to treat. Fasting is one of host adaptations to infection, and the induction of ketogenesis and ketolysis is a well-described host metabolic adaptation in response to fasting. However, more information about the energy substrates required to meet the immune response to S. aureus infection demands is needed. This study shows that the production of β-hydroxybutyrate (BHB) is enhanced in individuals with S. aureus, and BHB levels correlate with inflammatory cytokines an

Impaired ketogenesis in Leydig Cells drives testicular aging.

Testicular aging commonly leads to testosterone deficiency and impaired spermatogenesis, yet the underlying mechanisms remain elusive. Here, we show that Leydig cells are particularly vulnerable to aging processes in testis. Single-cell RNA sequencing identifies the expression of Hmgcs2, the gene encoding rate-limiting enzyme of ketogenesis, decreases significantly in Leydig cells from aged mice. Additionally, the concentrations of ketone bodies β-hydroxybutyric acid and acetoacetic acid in young testes are substantially higher than that in serum, but significantly diminish in aged testes. Bifidobacterium animalis Probio-M8 improves sarcopenia physical performance by mitigating creatine restrictions imposed by microbial metabolites.

Sarcopenia is a major health challenge due to an aging population. Probiotics may improve muscle function through gut-muscle axis, but their efficacy and mechanisms in treating sarcopenia remain unclear. This study investigated the impact of Bifidobacterium animalis subsp. lactis Probio-M8 (Probio-M8) on old mice and sarcopenia patients. We analyzed 43 subjects, including gut microbiome, fecal metabolome, and serum metabolome, using a multi-omics approach to assess whether Probio-M8 can improve sarcopenia by modulating gut microbial metabolites. Probio-M8 significantly improved muscle function

Blood metabolomic and transcriptomic signatures stratify patient subgroups in multiple sclerosis according to disease severity.

There are no blood-based biomarkers distinguishing patients with relapsing-remitting (RRMS) from secondary progressive multiple sclerosis (SPMS) although evidence supports metabolomic changes according to MS disease severity. Here machine learning analysis of serum metabolomic data stratified patients with RRMS from SPMS with high accuracy and a putative score was developed that stratified MS patient subsets. The top differentially expressed metabolites between SPMS versus patients with RRMS included lipids and fatty acids, metabolites enriched in pathways related to cellular respiration, nota

Unexplained acidosis of malnutrition: a study by ion-exchange chromatography/mass spectrometry.

Keto-acidosis is usually associated with uncontrolled diabetes and typically poses few diagnostic problems when presenting as hyperglycaemia, metabolic acidosis and a high anion gap. An emaciated patient suffering from Duchenne Muscular Dystrophy and volume depletion presented with acidosis of unknown origin. Preliminary investigations appeared to rule out lactic acidosis, diabetic keto-acidosis and acidosis due to base loss. We have previously reported a technique utilizing liquid chromatography coupled to mass spectrometry (LC-MS) which can be used to characterize the underlying aetiology of

Adrenergic mechanism in hyperketonemia in thyrotoxic and starved rats.

To evaluate the possible role of the adrenergic mechanism in hyperketonemia in hyperthyroidism and starvation, the plasma concentrations of FFA, acetoacetate (AcAc), and beta-hydroxybutyrate (BOHB) were measured in thyrotoxic and starved rats for 96 hours. Thyrotoxemia was induced in rats by a daily subcutaneous thyroxine (100 micrograms/kg/day) injection carried out for 7 days. Some of the thyrotoxic and starved rats were administered subcutaneous phentolamine (2 mg/kg, twice daily) or propranolol (0.1 mg/kg, twice daily). Plasma levels of FFA, AcAc, and BOHB were significantly increased in t

Decrease in hepatic mitochondrial redox state by massive infusion of citrate-phosphate-dextrose solution in jaundiced rabbits.

To clarify the mechanism of metabolic derangement by massive blood transfusion to the damaged liver, the changes in the hepatic mitochondrial redox state, as reflected in arterial ketone body ratio (acetoacetate/3-hydroxybutyrate), were studied in jaundiced rabbits by infusion of massive citrate-phosphate-dextrose (CPD) solution. The jaundiced rabbits received common bile duct ligation (BDL group), and the sham group had a simple exploration of the bile duct. CPD solution was infused for 3 h at a rate of 9 ml/kg/h in each group. As metabolic parameters, blood gas, pyruvate, lactate, citrate an

Blood ketone body disappearance rate in diabetics and normals after rapid infusion of dl-3-hydroxybutyrate. Studies before and after diabetic treatment.

Ketone body tolerance has been studied in 26 newly diagnosed diabetics and 9 normal control persons after rapid i.v. infusion of DL-3-hydroxybutyrate. In juvenile diabetics with high initial fasting concentrations of ketone bodies, disappearance rates of acetoacetate and total ketone bodies in blood were low before diabetic regulation. Insulin treatment normalized disappearance rates. In the non-obese maturity onset diabetics, on the other hand, ketone body disappearance rates remained abnormally low after treatment with glibenclamide. In the obese maturity onset diabetics, disappearance rates

Selective one-pot fluorescent probing technique for determination of tolterodine in dosage forms, content uniformity, and human plasma via coumarin synthesis.

Tolterodine tartrate (TTD), the inaugural antimuscarinic drug specifically created to treat overactive bladder syndrome, received FDA approval in 1998. Due to its widespread use in the local population since its approval, there is an urgent demand for the development of a spectrofluorometric method that is cost-effective, highly sensitive, selective, and reproducible. The spectrofluorometric method was developed by derivatization of TTD's phenol via Von Pechmann condensation. TTD reacts with ethyl acetoacetate (EAA) to form a highly fluorescent coumarin compound (Von Pechmann reaction) detecta

Ketone Bodies in Ischemic Stroke: A Review of Neuroprotective Mechanisms and Therapeutic Implications.

Ischemic stroke disrupts cerebral metabolism and initiates a cascade of pathological events, including excitotoxicity, mitochondrial dysfunction, and neuroinflammation. These interconnected processes drive neuronal apoptosis, necrosis, and synaptic loss, ultimately leading to irreversible brain injury, persistent neurological deficits, and high rates of long-term disability and mortality. Ketone bodies, including β-hydroxybutyrate, acetoacetate, and acetone, have demonstrated neuroprotective properties through multiple mechanisms. These include sustaining cerebral energy supply and mitochondri

Effect of consumption of anthocyanin-rich products on NMR lipoprotein subclasses and biomarkers in hypercholesterolemic subjects: a randomized controlled trial (the AppleCOR study).

Our aim was to assess the effect of intake of anthocyanin biofortified red-fleshed apples (RFA) versus that of common white apples (WFA) without anthocyanins on the NMR lipoprotein subfraction profile and other NMR metabolites. Additionally, an aronia infusion (AI) arm, matching the anthocyanin content and profile of the RFA, was included. A 6-week, randomized, parallel study was conducted in hypercholesterolemic subjects (n = 121). Anthocyanin-rich products (RFA and AI) decreased LDLc; ApoB; total, large, and small LDL-P; LDL size; TG/HDL ratio; and large TRL, versus WFA. All treatments signi

Intermittent fasting and ketone bodies.

Intermittent fasting (IF) is a dietary intervention based on time-restricted energy intake. Over the years, IF has been widely investigated as a non-pharmacological approach to increasing life expectancy and promoting brain health. The underlying mechanisms by which IF promotes its beneficial effects are attributed to hormesis, an evolutionary adaptive strategy that regulates cellular responses to stress. These effects include elevated levels of brain-derived neurotrophic factor (BDNF), enhanced neurogenesis and autophagy, increased synaptic plasticity, and improved cognitive functions such as

Integrated Gut Microbiota and Urine Metabolite Analyses of T2DM with NAFLD Rat Model.

Globally 80% type 2 diabetes mellitus (T2DM) patients suffer nonalcoholic fatty liver disease (NAFLD). The interplay of gut microbiota and endogenous metabolic networks has not yet been reported in the setting of T2DM with NAFLD. As such, this study utilized 16S rRNA gene sequencing to assess the changes in intestinal flora and nuclear magnetic resonance spectroscopy (1H NMR) to identify potential metabolites in a T2DM with NAFLD rat model. Spearman correlation analysis was performed to explore the relationship between gut microbiota and metabolites. Results revealed that among T2DM with NAFLD
