PFOA exposure decreases acrosome reaction and increases DNA oxidative damage by impairing the sperm head shaping in mice.

Perfluorooctanoic acid (PFOA) is a reproductive toxicant, but its effects on sperm head shaping and acrosome reaction remain unknown. Adult male Bal b/c mice were given 0, 2 or 10 mg/kg/day of PFOA for 6 weeks by gavage. Elevated sperm head malformations and decreased acrosome reaction were observed in PFOA-exposed mice. Transmission electron microscopy (TEM) revealed that PFOA exposure caused ultrastructural changes of endoplasmic reticulum, acrosomal granules, acrosomal membrane and nuclear membrane at different phases of acrosome biogenesis. Additionally, key proteins participated in acroso

Manchette-acrosome disorders and testicular efficiency decline observed in hypercholesterolemic rabbits are recovered with olive oil enriched diet.

High-fat diet is associated with hypercholesterolemia and seminal alterations in White New Zealand rabbits. We have previously reported disorders in the development of the manchette-acrosome complex during spermiogenesis and decreased testicular efficiency in hypercholesterolemic rabbits. On the other hand, olive oil incorporated into the diet improves cholesterolemia and semen parameters affected in hypercholesterolemic rabbits. In this paper, we report the recovery-with the addition of olive oil to diet-from the sub-cellular mechanisms involved in the shaping of the sperm cell and testicular

Calpain-1 and Cholesterol Regulate the Dynamics of the Lipid Membrane Domains Essential for Suitable Capacitation and Acrosomal Reactions.

Calpain-1 and -2 are calcium-dependent cysteine proteases associated with sperm processes, such as capacitation, the acrosomal reaction, and motility, making them important for the acquisition of fertilizing capacity among spermatozoa. Moreover, their inhibition significantly reduces in vitro fertilization. Guinea pig spermatozoa express only calpain-1, which has been implicated in spectrin cytoskeletal remodeling and NOX2 and NOX4 activation during capacitation. However, little is known about the mechanisms by which calpains participate in capacitation and the acrosomal reaction. The interact

Investigation of endocannabinoid 2-AG and its hydrolases for regulation on sperm motility and acrosome reaction in Kunming mice.

CONTEXT: A key endocannabinoid, 2-arachidonoyl glycerol (2-AG), regulates sperm hyperactivation in humans via alpha/beta hydrolase domain-containing protein 2 (ABHD2)-mediated CatSper calcium channel activation. However, the role of ABHD2 in mice is unclear, whereas monoacylglycerol lipase (MAGL), ABHD-containing protein 6 (ABHD6), and ABHD-containing protein 12 (ABHD12) modulate 2-AG in the mouse central nervous system (CNS). AIMS: This study explored the impact of 2-AG on mouse sperm motility, its regulatory mechanisms, and the involvement of MAGL, ABHD6, and ABHD12. METHODS: 2-AG concentr

Bicarbonate and BSA increase the capacitation pattern and acrosomal exocytosis in boar sperm after 120 min of incubation.

Sperm capacitation is a crucial step towards the acquisition of fertilizing capacity. Despite the attempts to mimic the in vivo situation, there is still a lack of standardization in vitro techniques. Bicarbonate and serum albumin (BSA) are routinely used, although controversial results are reported regarding the optimal concentration of each compound. In addition, whether caffeine is needed on in vitro capacitation media in boar sperm remains to be elucidated. Here, 18 boar commercial artificial insemination doses were used to test different concentrations of bicarbonate (19, 37 or 56 mM) in

ACRBP (Sp32) is involved in priming sperm for the acrosome reaction and the binding of sperm to the zona pellucida in a porcine model.

In boar sperm, we have previously shown that capacitation is associated with the appearance of the p32 tyrosine phosphoprotein complex. The principal tyrosine phosphoprotein involved in this complex is the acrosin-binding protein (ACRBP), which regulates the autoconversion of proacrosin to intermediate forms of acrosin in both boar and mouse sperm. However, the complete biological role of ACRBP has not yet been elucidated. In this study, we tested the hypothesis that tyrosine phophorylation and the presence of the ACRBP in the sperm head are largely necessary to induce capacitation, the acroso

An epididymis-specific secretory protein Clpsl2 critically regulates sperm motility, acrosomal integrity, and male fertility.

The epididymis performs an important role in the maturation of spermatozoa including their acquisition of progressive motility and fertilizing ability. However, the molecular mechanisms that govern these maturational events are still poorly defined. Here we report that Clpsl2, a novel colipase homology, is exclusively expressed in the caupt epididymis and conserved in mammalian. Clpsl2 was secreted into the lumen and covered the acrosome region and principal piece of spermatozoa tail. And during epididymal transit, the binding rate between Clpsl2 protein and the spermatozoa gradually decreased

Contribution of different Ca²⁺ channels to the acrosome reaction-mediated initiation of sperm motility in the newt Cynops pyrrhogaster.

Initiation of sperm motility in urodeles, which is induced by a sperm motility-initiating substance (SMIS) in the sequestered granules on the surface of egg jelly, is mediated by the acrosome reaction (AR), which is triggered by an AR-inducing substance (ARIS) on a sheet-like structure. Details of the unique process of the interaction between egg jelly and sperm in these species is still unclear. The current study showed the fine structure of egg jelly in the newt Cynops pyrrhogaster, a urodele species, revealing that its outer surface was covered by a sheet-like structure of approximately 0.2

Acrosome reaction in the epididymal sperm of Chinchilla lanigera. Effect of progesterone, A23187 and stimulation of a GABAA-like receptor.

Little is known about the reproductive biology of Chinchilla lanigera in captivity. Currently there are no parameters to evaluate the quality of the semen of males that are sold as breeding stock, so they are selected according to their phenotypic characteristics, which cannot guarantee their reproductive efficiency. We worked with sexually mature males; the sperm were collected from the cauda epididymis by puncture. Capacitated sperm were incubated at 37 °C with different concentrations of progesterone. The results showed a biphasic behavior, where the increase in the percentage of the acroso

Assessment of the fertilizing potential of frozen bovine spermatozoa by in vitro induction of acrosome reactions with calcium ionophore (A23187).

The relationship between nonreturn rates of bulls in a commercial artificial insemination program and in vitro induction of acrosome reactions in frozen-thawed spermatozoa by the calcium ionophore, A23187, was investigated. Washed spermatozoa from 3 to 5 ejaculates, collected from each of 23 Holstein bulls, were incubated for 1 h with 1 microM A23187. Acrosome reactions were determined by fluorescence microscopy. The percentage of increase in acrosome reaction in the ionophore-treated compared with control samples was significantly correlated to the 90-d nonreturn rate of the bulls (r = 0.86;

Detection of altered acrosomal physiology of cryopreserved human spermatozoa after sperm residence in the female reproductive tract.

At least some of the spermatozoa that remain motile following cryopreservation have sustained sublethal damage that reduces their functional capacity in vivo. Although it is believed that acrosomal damage is partly responsible for impaired sperm function in vivo, direct evidence for this hypothesis is lacking because spermatozoa have not been collected from the female reproductive tract for evaluation. In the study reported here, cervical mucus was collected from women 24 h after artificial insemination by cervical cup. For both cryopreserved and nonfrozen inseminates, spermatozoa within the c

Expression, Localization, and Processing of Chicken Sperm ADAM32L2 during the Acrosome Reaction: A Possible Function in the Sperm-Egg Interaction.

Sperm-egg interactions involve a complex series of molecular events. Among these, the acrosome reaction (AR) is a prerequisite for sperm penetration, facilitating the exposure of multiple acrosomal proteins that enhance sperm binding or penetration of the outer layer of the egg; however, the specific molecules involved in this process vary across species. A disintegrin and metalloproteinase (ADAM) proteins are transmembrane glycoproteins that play a role in sperm-egg interactions, with notable differences among ADAM isoforms. In a previous characterization of the chicken sperm membrane proteom

Inhibition of Potassium Channels Affects the Ability of Pig Spermatozoa to Elicit Capacitation and Trigger the Acrosome Exocytosis Induced by Progesterone.

During capacitation, sperm undergo a myriad of changes, including remodeling of plasma membrane, modification of sperm motility and kinematic parameters, membrane hyperpolarization, increase in intracellular calcium levels, and tyrosine phosphorylation of certain sperm proteins. While potassium channels have been reported to be crucial for capacitation of mouse and human sperm, their role in pigs has not been investigated. With this purpose, sperm samples from 15 boars were incubated in capacitation medium for 300 min with quinine, a general blocker of potassium channels (including voltage-gat

Sperm acrosome status before and during fertilization in the Chinese hamster (Cricetulus griseus), and observation of oviductal vesicles and globules.

The present study was conducted to determine exact location where the acrosome reaction of fertilizing spermatozoa begins in the oviduct of the Chinese hamster. Unlike spermatozoa of other rodent species, Chinese hamster spermatozoa did not spontaneously undergo the acrosome reaction in fertilization-supporting media. In naturally mated females, spermatozoa in the uterus had intact acrosomes, whereas those in the lower oviductal isthmus had visibly thin acrosomal caps. The acrosomal cap was lost when spermatozoa passed through the cumulus oophorus. Thus, Chinese hamster spermatozoa begin the a

Oviductal high concentration of K+ suppresses hyperpolarization but does not prevent hyperactivation, acrosome reaction and in vitro fertilization in hamsters.

Mammalian sperm have to undergo capacitation to be fertilization competent. Capacitated sperm in vitro show hyperpolarization of the membrane potential. It has been reported that in mouse membrane hyperpolarization is necessary for the acrosome reaction. We recently found that the fluid of the hamster oviduct, where fertilization occurs, contained a high potassium (K+) concentration (~20 mEq/l). This high K+ concentration could depolarize the membrane potential and prevent the acrosome reaction/fertilization. Conversely, some beneficial effects on capacitation of high K+ concentration or a hig

Processing, localization and binding activity of zonadhesin suggest a function in sperm adhesion to the zona pellucida during exocytosis of the acrosome.

Zonadhesin is a sperm protein that binds in a species-specific manner to the extracellular matrix ZP (zona pellucida) of the mammalian oocyte. The pig zonadhesin precursor is a 267000-Da mosaic protein with a Type I membrane topology and a large extracellular region comprising meprin/A5 antigen/mu receptor tyrosine phosphatase, mucin and five tandem von Willebrand D (VWD) domains. Multiple mature forms of zonadhesin in the sperm head differ in their avidities for the ZP. To determine the potential functions of zonadhesin forms in gamete adhesion, we characterized the processing, activation and

The Multi-PDZ domain protein MUPP1 as a lipid raft-associated scaffolding protein controlling the acrosome reaction in mammalian spermatozoa.

The success of acrosomal exocytosis, a complex process with a variety of interrelated steps, relies on the coordinated interaction of participating signaling molecules. Since scaffolding proteins are known to spatially organize sequential signaling pathways, we examined whether the Multi-PDZ domain protein MUPP1, recently identified in mammalian spermatozoa, is functionally active in controlling acrosomal secretion in mammalian sperm cells. To address this question, permeabilized mouse sperm were loaded with inhibitory antibodies against MUPP1 as well as with a photosensitive Ca(2+) chelator w

Tandem mass spectrometry identifies proteins phosphorylated by cyclic AMP-dependent protein kinase when sea urchin sperm undergo the acrosome reaction.

The exocytotic acrosome reaction (AR), which is required for fertilization, occurs when sea urchin sperm contact the egg jelly (EJ) layer. Among other physiological changes, increases in adenylyl cyclase activity, cAMP and cAMP-dependent protein kinase (PKA) activity occur coincident with the AR. By using inhibitors of PKA, a permeable analog of cAMP and the phosphodiesterase inhibitor IBMX, we show that PKA activity is required for AR induction by EJ. A minimum of six sperm proteins are phosphorylated by PKA upon exposure to EJ, as detected by a PKA substrate-specific antibody. The phosphoryl

Artemisia vulgaris extract causes precocious acrosome reaction and viability loss but low rate of membrane damage in mouse spermatozoa.

Several herbs including Artemisia are known to possess conceptive property. In the present study, mouse spermatozoa were incubated with ethanol extract of Artemisia vulgaris leaves. The effect of extract on acrosome exocytosis was studied by labeling spermatozoa with fluorescein isothiocyanate (FITC) peanut agglutinin and by staining with Coomassie blue. Viability and membrane integrity were studied by Trypan-blue staining and hypo-osmotic swelling test. Artemisia extract at very low concentration caused precocious acrosome reaction and loss of sperm viability. Acrosome reaction increased rema

Comprehensive functional analysis reveals that acrosome integrity and viability are key variables distinguishing artificial insemination bulls of varying fertility.

In vitro methods of assessing bull semen quality in artificial insemination (AI) centers are unable to consistently detect individuals of lower fertility, and attempts to reliably predict bull fertility are still ongoing. This highlights the need to identify robust biomarkers that can be readily measured in a practical setting and used to improve current predictions of bull fertility. In this study, we comprehensively analyzed a range of functional, morphological, and intracellular attributes in cryopreserved spermatozoa from a selected cohort of Holstein Friesian AI bulls classified as having
