Bigger is faster in the adaptive immune response.

Zoonotic pathogens represent a growing global risk, yet the speed of adaptive immune activation across mammalian species remains poorly understood. Despite orders-of-magnitude differences in size and metabolic rate, we show that the time to initiate adaptive immunity is remarkably consistent across species. To understand this invariance, we analyse empirical data showing how the numbers and sizes of lymph nodes scale with body mass, finding that larger animals have both more and larger lymph nodes. Using scaling theory and our mathematical model, we show that larger lymph nodes enable faster s

Adaptive immune function in critical illness.

PURPOSE OF REVIEW: This review is being published to update the literature on the function of the adaptive immune system in critical illness, specifically sepsis and acute lung injury. We have focused on the role of T cells in these syndromes. RECENT FINDINGS: The adaptive immune response becomes dysfunctional during sepsis and acute lung injury in very similar ways. Many of the abnormalities contribute to morbidity and mortality. Immunoparalysis captures the breadth of the dysfunction in that T-cell functions are broadly suppressed after the early proinflammatory stages of illness. Lymphocyt

Role of the adaptive immune system in atherosclerosis.

Atherosclerosis, the pathology underlying heart attacks, strokes and peripheral artery disease, is a chronic inflammatory disease of the artery wall initiated by elevated low-density lipoprotein (LDL) cholesterol levels. LDL accumulates in the artery wall, where it can become oxidized to oxLDL. T cell responses to ApoB, a core protein found in LDL and other lipoproteins, are detectable in healthy mice and people. Most of the ApoB-specific CD4T cells are FoxP3+ regulatory T cells (Treg). In the course of atherosclerosis development, the number of ApoB-reactive T cells expands. At the same time,

Role of the adaptive immune response in sepsis.

Sepsis is a syndrome of shock and dysfunction of multiple vital organs that is caused by an uncontrolled immune response to infection and has a high mortality rate. There are no therapies for sepsis, and it has become a global cause for concern. Advances in patient care and management now mean that most patients survive the initial hyper-inflammatory phase of sepsis but progress to a later immunosuppressed phase, where 30% of patients die due to secondary infection. Deficits in the adaptive immune response may play a major role in sepsis patient mortality. The adaptive immune response involves

Adaptive immune regulation in autoimmune diabetes.

Type 1 diabetes (T1D) is a chronic autoimmune disease characterized by the loss of insulin-producing pancreatic β-cells. The pathogenesis of T1D is complex and multifactorial and involves a genetic susceptibility that predisposes to abnormal immune responses in the presence of ill-defined environmental insults to the pancreatic islets. This review discusses how adaptive immunoregulatory T cells contribute to the modulation of the development and evolution of T1D, together with potential approaches that target these cells for new therapies in the disease.

The adaptive immune response in celiac disease.

Compared to other human leukocyte antigen (HLA)-associated diseases such as type 1 diabetes, multiple sclerosis, and rheumatoid arthritis, fundamental aspects of the pathogenesis in celiac disease are relatively well understood. This is mostly because the causative antigen in celiac disease-cereal gluten proteins-is known and the culprit HLA molecules are well defined. This has facilitated the dissection of the disease-relevant CD4+ T cells interacting with the disease-associated HLA molecules. In addition, celiac disease has distinct antibody responses to gluten and the autoantigen transgluta

Stress, adaptation, and immunity: studies in humans.

The notion that some combination of excessive "stress" and inadequate coping may increase susceptibility to illness has long been part of our conventional wisdom. Yet, until recently, there have been few data to support this contention. During the past decade, the relationship between stress, adaptation, and human immunity has come under closer scrutiny. There is now considerable evidence that certain types of experimental and naturally-occurring stress are associated with alterations of human cellular immune function. Furthermore, these observed changes generally are immunosuppressive. Howeve

Revised Adaptive Immune Receptor Data in the Immune Epitope Database.

The Immune Epitope Database (IEDB, iedb.org) is a freely available resource that catalogs experimentally defined immune epitopes and - if available - the immune receptors that recognize them. Currently, the IEDB records ~185,000 T cell receptors and ~5,000 B cell receptors/antibodies with experimentally verified epitope specificity. Because these receptor data were manually curated from ~3,300 references spanning decades, nomenclature inconsistencies present challenges for computational analyses and user queries. To support integrated analysis of the entire dataset, we revised the IEDB recepto

Trained Immunity: Long-Term Adaptation in Innate Immune Responses.

Adaptive immune responses are characterized by antigen specificity and induction of lifelong immunologic memory. Recently, it has been reported that innate immune cells can also build immune memory characteristics-a process termed trained immunity. Trained immunity describes the persistent hyperresponsive phenotype that innate immune cells can develop after brief stimulation. Pathogenic stimuli such as microorganisms, and also endogenous molecules including uric acid, oxidized LDL (low-density lipoprotein), and catecholamines, are capable of inducing memory in monocytes and macrophages. While

The T Cell Receptor: Molecular Sensor, Therapeutic Mediator and Probabilistic Driver of Adaptive Immunity.

Advances in high-throughput sequencing, single-cell profiling, and genome engineering have transformed the study of T cell receptors (TCRs), enabling the identification and functional interrogation of antigen-specific repertoires at an unprecedented scale. This review discusses how recent methodological developments-including high-dimensional TCR discovery strategies, physiological receptor engineering, and longitudinal in vivo analyses-have reshaped our understanding of TCR-driven immune responses. Recruitment into immune responses originates from diverse naïve precursor pools and results in

Guarding epithelial integrity: origins of adaptive immunity and cancer immunosurveillance over time and space.

While conventionally viewed as a defense against external pathogens, we posit that the adaptive immune system likely evolved primarily to preserve internal epithelial order. We propose that adaptive immunity, with combinatorial diversity emerging ~0.5 billion years ago during the Cambrian explosion, was based on extant and established complex cellular interactions, derived over the previous 3.5 billion years of life's history. These coordinate cellular processes originated to police epithelia against aberrant or rogue cells-inherent to complex multicellularity. Primordial surveillance relied o

Bispecific and multispecific immune engagers for redirecting innate and adaptive immunity against hematologic cancers.

Bispecific antibodies (BsAbs) and multispecific antibodies (MsAbs) represent a transformative class of immunotherapeutics in oncology. These engineered molecules possess the unique ability to simultaneously target two or more distinct antigens, thereby facilitating precise immune cell engagement and disrupting multiple signaling pathways. This comprehensive review focuses on their pivotal role in redirecting both innate (natural killer cells, phagocytes) and adaptive (T cells) immunity, with a particular emphasis on their application against hematologic cancers. Significant clinical successes,

Mesenchymal stromal/stem cells in inflammatory bowel disease: Crosstalk with innate and adaptive immunity.

Inflammatory bowel disease (IBD) is characterized by chronic, relapsing, inflammatory, and immune-mediated responses in the intestinal tract. IBD includes the two well-defined subsets Crohn's disease (CD) and ulcerative colitis (UC). Mesenchymal stromal/stem cells (MSCs) are non-hematopoietic progenitor cells known for their regenerative and immunomodulatory capabilities. They have gained prominence as therapeutic agents for immune-mediated disorders, such as IBD. This review summarizes and categorizes the therapeutic significance of MSCs for IBD in the context of their interactions with vario

LRRK2 G2019S reprograms innate and adaptive immunity to drive context-dependent host defense outcomes.

INTRODUCTION: Parkinson's disease (PD) is increasingly recognized as a neurological disorder characterized not only by neurodegeneration but also by chronic immune dysregulation across the lifespan. Although the initiating events underlying PD remain unclear, accumulating evidence suggests that inflammatory processes may contribute to disease susceptibility and progression. Mutations in leucine-rich repeat kinase 2 (LRRK2), particularly the gain-of-function G2019S variant, represent the common genetic cause of familial PD and have been implicated in immune regulation and infection susceptibili

Inhibition of SUMOylation by anacardic acid inhibits adaptive immunity and the development of EAE.

OBJECTIVE: Small ubiquitin-like modifiers (SUMO) are reversible post-translational modifiers of intracellular proteins. SUMOylation plays an important regulatory role in both innate and adaptive immunity. The natural compound anacardic acid (AA) has been shown to bind and inhibit the SUMO-activating enzyme E1, the first enzyme in the SUMOylation pathway. Here, we examined the consequences of AA treatment on the development of innate and adaptive immune responses in vitro and in vivo. METHODS: We examined the inhibitory effects of anacardic acid on SUMOylation and de-SUMOylation both in vitro

Modulation of lipid nanoparticle-formulated plasmid DNA drives innate immune activation promoting adaptive immunity.

Nucleic acid vaccines have grown in importance over the past several years, with the development of new approaches remaining a focus. We describe a lipid nanoparticle-formulated DNA (DNA-LNP) formulation which induces robust innate and adaptive immunity with similar serological potency to mRNA-LNPs and adjuvanted protein. Using an influenza hemagglutinin (HA)-encoding construct, we show that priming with our HA DNA-LNP demonstrated stimulator of interferon genes (STING)-dependent upregulation and activation of migratory dendritic cell (DC) subpopulations. HA DNA-LNP induced superior antigen-sp

Comparative analysis of adaptive immunity to SARS-CoV-2 in infected children and adults.

BACKGROUND: SARS-CoV-2 infection in children is most often mild and resembles that of seasonal coronaviruses. Profiling the adaptive immune response following infection may help to inform on the protective mechanisms mediating immunity in children and adults. METHODS: Humoral and cell-mediated immune responses from unvaccinated pediatric and adult participants were analyzed following non-Omicron SARS-CoV-2 infection. Specific T cell memory responses were investigated by quantifying interferon-gamma (IFN-γ) secreting cells after stimulation with ancestral and variant strains of SARS-CoV-2 and

Tumor Cell-Expressed Herpesvirus Entry Mediator Regulates Proliferation and Adaptive Immunity in Ovarian Cancer.

BACKGROUND: Ovarian cancer (OvCa) is a prevalent gynecological malignancy with an increasing incidence and high mortality rate. Although the role of the herpesvirus entry mediator (HVEM), encoded by the TNFRSF14 gene, is currently considered pivotal in various types of cancer, the regulation of tumor cell-expressed HVEM in OvCa remains inadequately understood. METHODS: Specimens were used to detect HVEM expression via quantitative RT-PCR and flow cytometry. The proliferation of the murine OvCa cell line ID8 was determined using the Cell Counting Kit-8, colony formation, and EdU staining assay

Lipolysis-microlipophagy cascade regulated by adipose triglyceride lipase drives pathogenic adaptive type 2 immunity.

A unique subpopulation of memory T helper 2 (TH2) cells expressing the interleukin-33 (IL-33) receptor ST2 drives allergic disease pathogenesis. However, the immunometabolic mechanisms that induce ST2hi memory TH2 cells remain unclear. We show using a mouse model of chronic allergic airway inflammation that long-chain unsaturated fatty acids (LC-UFAs) accumulate in the inflammatory milieu during chronic airway inflammation. Activated TH2 cells take up LC-UFAs, transiently store them in lipid droplets (LDs), and catabolize LDs through lipolysis and microlipophagy. LD catabolism regulated by adi

Innate and Adaptive Immunity during SARS-CoV-2 Infection: Biomolecular Cellular Markers and Mechanisms.

The coronavirus 2019 (COVID-19) pandemic was caused by a positive sense single-stranded RNA (ssRNA) severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). However, other human coronaviruses (hCoVs) exist. Historical pandemics include smallpox and influenza, with efficacious therapeutics utilized to reduce overall disease burden through effectively targeting a competent host immune system response. The immune system is composed of primary/secondary lymphoid structures with initially eight types of immune cell types, and many other subtypes, traversing cell membranes utilizing cell signal
