Sulfur uptake in the ectomycorrhizal fungus Laccaria bicolor S238N.

The importance of the ectomycorrhiza symbiosis for plant acquisition of phosphorus and nitrogen is well established whereas its contribution to sulfur nutrition is only marginally understood. In a first step to investigate the role of ectomycorrhiza in plant sulfur nutrition, we characterized sulfate and glutathione uptake in Laccaria bicolor. By studying the regulation of sulfate uptake in this ectomycorrhizal fungus, we found that in contrast to bacteria, yeast, and plants, sulfate uptake in L. bicolor was not feedback-inhibited by glutathione. On the other hand, sulfate uptake was increased

Antimicrobial activity of Streptomyces species against mushroom blotch disease pathogen.

Bacterial blotch caused by Pseudomonas tolaasii is still one of the main diseases in mushroom farms. Effective treatment of the infections caused by P. tolaasii is yet to be established. The isolation and identification of three Streptomyces species that showed antimicrobial activity against P. tolaasii NCPPB 2192(T) were described. Six strains were highly active with an inhibition zone more than 20 mm in diameter. They were assigned to the known Streptomyces cluster groups as S. rochei, S. lydicus and S. antibioticus. The fermentation, preliminary extraction and isolation of bioactive compone

Altered Bacterial-Fungal Interkingdom Networks in the Guts of Ankylosing Spondylitis Patients.

Intestinal bacterial dysbiosis has been increasingly linked to ankylosing spondylitis (AS), which is a prototypic and best studied subtype of spondyloarthritis (SpA). Fungi and bacteria coexist in the human gut and interact with each other. Although they have been shown to contribute actively to health or disease, no studies have investigated whether the fungal microbiota in AS patients is perturbed. In this study, fecal samples from 22 AS patients, with clinical and radiographic assessments, and 16 healthy controls (HCs) were collected to systematically characterize the gut microbiota and myc

Integrated Metabolomic and Bioreporter Screening of Five Thai Mushroom Extracts: Exploring Antibiofilm and Biocompatibility Profiles.

This study evaluated the biomedical potential of ethanolic extracts from five native Thai mushrooms, revealing distinct biological activities associated with their metabolomic profiles. Termitomyces clypeatus R. Heim demonstrated exceptional biocompatibility, effectively promoting human keratinocyte (HaCaT) proliferation without causing cytotoxicity to the cells. Its primary metabolome, rich in essential amino acids and nucleosides, supports its potential as a candidate for topical wound healing, pending further functional validation. Conversely, Lentinus squarrosulus exhibited strong antibiof

Distribution of incompatibility factors in natural populations of Hypsizygus marmoreus in Japan.

Hypsizygus marmoreus (bunashimeji in Japanese) is a major cultivated edible mushroom in Japan. Characterizing its mating incompatibility factors in natural populations is essential for improving breeding efficiency. In this study, 71 wild isolates of H. marmoreus collected from different regions in Japan were analyzed to determine the composition and distribution of incompatibility factors. Mating tests using tester monokaryotic strains identified 82 cases of monokaryons that were carrying different alleles at both the A and B mating locus, whereas another 60 factors were shared among isolates

Enhanced volatile fatty acid production from co-fermentation of spent mushroom waste and food waste with rumen microbes: Performance and mechanism.

OBJECTIVE: Co-fermentation of lignocellulosic biomass and food waste (FW) can enhance the production of volatile fatty acids (VFAs) during anaerobic fermentation. However, the effect and mechanism of co-fermentation of Auricularia auricula waste (AW) with FW remains unclear. This study aimed to investigate the effect and mechanism of co-fermentation of Auricularia auricula waste (AW) with food waste (FW) on volatile fatty acid (VFA) production. METHODS: Anaerobic fermentation experiments were conducted at different AW/FW ratios (1:0, 1:1, 2:1, 3:1, 4:1, 0:1) based on volatile solids (VS). VS

Biosynthesis of Crinipellin Diterpenes in Mushroom Marasmius fiardii PR-910.

Crinipellins are a distinctive family of 5/5/5/5 tetracyclic diterpenoids previously reported exclusively from mushrooms of the genus Crinipellis. Despite extensive synthetic studies, the biosynthetic machinery responsible for crinipellin formation has remained elusive. Here, we identify the crinipellin biosynthetic gene cluster (mfd) from the mushroom Marasmius fiardii PR-910, a member of the family Marasmiaceae to which Crinipellis also belongs, although M. fiardii PR-910 itself has not been previously reported to produce crinipellins. Using a combination of site-directed mutagenesis guided

Exploring of multi-functional umami peptides from Stropharia rugosoannulata: Saltiness-enhancing effect and mechanism, antioxidant activity and potential target sites.

Umami peptides enhance flavor and offer potential health benefits. We analyzed the taste-value profiles of five novel umami peptides from Stropharia rugosoannulata using E-tongue, exhibiting significant saltiness characteristics. While the peptides PHEMQ and SEPSHF exhibited higher saltiness, their mixture with salt did not enhance saltiness compared to individual peptides. Surprisingly, SGCVNEL, which was initially weak in saltiness, showed remarkably enhanced saltiness when mixed with salt, possibly due to have strong binding with receptors. Molecular docking elucidated the salt-forming mech

Mushroom-Derived Compounds as Metabolic Modulators in Cancer.

Cancer is responsible for lifelong disability and decreased quality of life. Cancer-associated changes in metabolism, in particular carbohydrate, lipid, and protein, offer a new paradigm of metabolic hits. Hence, targeting the latter, as well as related cross-linked signalling pathways, can reverse the malignant phenotype of transformed cells. The systemic toxicity and pharmacokinetic limitations of existing drugs prompt the discovery of multi-targeted and safe compounds from natural products. Mushrooms possess biological activities relevant to disease-fighting and to the prevention of cancer.

Endoscopy Deliverable and Mushroom-Cap-Inspired Hyperboloid-Shaped Drug-Laden Bioadhesive Hydrogel for Stomach Perforation Repair.

Gastrointestinal tract perforation is a full-thickness injury that causes bleeding and fatal infection of the peritoneum. This condition worsens in an acidic gastric environment which interferes with the normal coagulation cascade. Current endoscopic clips to repair gastric perforations are ineffective, and metal or plastic occluders need secondary surgery to remove them. Herein, we report a self-expandable, endoscopy deliverable, adhesive hydrogel to block gastric perforation. We found the nanosilica coating significantly enhanced the adhesive strength even under a simulated strong acidic sto

Regulation of Cholesterol Metabolism by Phytochemicals Derived from Algae and Edible Mushrooms in Non-Alcoholic Fatty Liver Disease.

Cholesterol synthesis occurs in almost all cells, but mainly in hepatocytes in the liver. Cholesterol is garnering increasing attention for its central role in various metabolic diseases. In addition, cholesterol is one of the most essential elements for cells as both a structural source and a player participating in various metabolic pathways. Accurate regulation of cholesterol is necessary for the proper metabolism of fats in the body. Disturbances in cholesterol homeostasis have been linked to various metabolic diseases, such as hyperlipidemia and non-alcoholic fatty liver disease (NAFLD).

Cadmium hyperaccumulating mushroom Cystoderma carcharias has two metallothionein isoforms usable for cadmium and copper storage.

Cystoderma carcharias is one of the few macrofungal species that can hyperaccumulate Cd. As we have previously documented in C. carcharias collected from a smelter-polluted area, it stores 40% of Cd and nearly 90% of Cu in sporocarps in complex(es) of identical size. In this paper we examined whether metallothionein (MT) peptides that bind Cd and Cu through cysteinyl-thiolate bonds were associated with the metals in these complexes. Screening of a sporocarp cDNA expression library in yeasts allowed the identification of two transcripts, CcMT1 and CcMT2, encoding functional 34-amino acid (AA) M

DARK Classics in Chemical Neuroscience: Psilocybin.

Psilocybin is found in a family of mushrooms commonly known as "magic mushrooms" that have been used throughout history to induce hallucinations. In the late 1950s Albert Hofmann, of Sandoz Laboratories, identified and synthesized the psychoactive compounds psilocybin and psilocin which are found in psilocybe mushrooms. Psilocybin was marketed by Sandoz as Indocybin for basic psychopharmacological and therapeutic clinical research. Psilocybin saw a rapid rise in popularity during the 1960s and was classed as a Schedule I drug in 1970. This led to a significant decrease in psilocybin research.
