In vitro metabolism of amphetamine: an apparent enantiomeric interaction.

The stereoselective formation of the major metabolites N-hydroxyamphetamine and 1-phenyl-2-propanol in incubations of amphetamine with 9,000 X g supernatant fractions of rabbit liver has been studied using deuterium labelling in conjunction with selected-ion-monitoring gas chromatography/mass spectrometry. Comparison of separate incubations of (R)-amphetamine with those of (S)-amphetamine-d3 and of (S)-amphetamine with those of (R)-amphetamine-d3 showed that larger amounts of the two metabolites were formed from the (R) enantiomer. When "pseudoracemic" mixture (R)-amphetamine/(S)-amphetamine-d

Amphetamine as an artifact of methamphetamine during periodate degradation of interfering ephedrine, pseudoephedrine, and phenylpropanolamine: an improved procedure for accurate quantitation of amphetamines in urine.

During periodate degradation of interfering ephedrine, pseudoephedrine, and phenylpropanolamine in the extraction of methamphetamine from urine, it was observed that a small amount of methamphetamine was demethylated to amphetamine. although all three interfering phenylpropanolamines could be degraded by periodate at pH 5.2 and above, this periodate-mediated transformation of methamphetamine to amphetamine was observed only at pH 9.1 and above. Therefore, to avoid this transformation, a pH of 6.2 was used for the oxidative degradation of phenylpropanolamines. The excess periodate was then redu

2D:4D-ratios among individuals with amphetamine use disorder, antisocial personality disorder and with both amphetamine use disorder and antisocial personality disorder.

BACKGROUND: There is sufficient evidence that the index-finger-to-ring-finger-ratio (2D:4D-ratio) is associated with testosterone and estrogen exposure during the fetal stage. More specifically, a lower 2D:4D-ratio (that is; a shorter index finger, compared to a longer ring finger) was associated with a prenatally higher testosterone and lower estrogen exposure during the first trimester of the fetal stage. At a behavioral level, among adults, a lower 2D:4D-ratio was associated with a higher competitive performance among both female and male professional athletes, and with personality traits s

Pneumomediastinum and subcutaneous emphysema post cocaine and amphetamine insufflation.

A 20-year-old gentleman presented with blood-streaked vomitus after insufflation of an unknown amount of powder cocaine and amphetamine. This was taken with an unspecified amount of alcohol. Other notable symptoms were dysphagia, chest pain, palpitations and the patient reported a 'crunchy' sensation in his chest. A chest x-ray revealed pneumomediastinum and a computerised tomography (CT) trauma confirmed these findings with associated subcutaneous emphysema without an identifiable cause. Follow up investigations included a barium swallow and gastroscopy which showed no obvious perforation but

Amphetamine-induced striatal dopamine release in schizotypal personality disorder.

RATIONALE: Previous research has suggested that schizotypal personality disorder (SPD), a condition that shares clinical and cognitive features with schizophrenia, may be associated with elevated striatal dopamine functioning; however, there are no published studies of dopamine release within subregions of the striatum in SPD. OBJECTIVES: To characterize dopamine release capacity in striatal subregions and its relation to clinical and cognitive features in SPD. METHODS: We used positron emission tomography with [11C]raclopride and an amphetamine challenge to measure dopamine D2-receptor avai

Narcolepsy Treated with Racemic Amphetamine during Pregnancy and Breastfeeding.

This case report describes a woman with narcolepsy treated with racemic amphetamine (rac-amphetamine) during pregnancy and breastfeeding with follow-up on the infant's development up to 10 months of age. The pregnancy outcome and the pharmacokinetics of rac-amphetamine were studied during breastfeeding. The pregnancy and the delivery were uneventful. Concentrations of rac-amphetamine were determined in the plasma of the mother and infant, and in the breast milk with a liquid chromatography-mass spectrometry method. Samples were obtained at 2, 5, and 9 weeks postpartum. The transfer of rac-amph

Fatal coronary artery intimal hyperplasia due to amphetamine use.

BACKGROUND: Amphetamines have been reported to cause myocardial infarct, cerebral hemorrhage, aortic dissection, hypertension, vasculitis, aneurysms, and, occasionally, death from direct toxicity. To date, there have been no reports of coronary intimal hyperplasia in an amphetamine user. METHODS: A 29-year-old male was found collapsed after jogging. He had been taking amphetamines for 11 years for attention deficit/hyperactivity disorder. Past medical history was positive for mild hypertension and migraine headaches. He underwent a complete autopsy and drug screen. RESULTS: At gross autopsy

Amphetamine-induced sensitization and spontaneous stereotypy in deer mice.

Stereotyped behavior is commonly observed in neurodevelopmental disorders (e.g., autism, intellectual and developmental disability) and in a wide variety of animal species maintained in restricted environments. Stereotyped behavior can also be induced by psychostimulants, an effect potentiated by repeated intermittent exposure to these drugs (behavioral sensitization). The present study evaluated whether similar neuroadaptations in cortical-basal ganglia circuitry underlie the expression and development of spontaneous stereotypy and psychostimulant-induced sensitization. Sensitization was indu

Mechanisms underlying aggressive and hostile behavior in amphetamine users.

PURPOSE OF REVIEW: To review current evidence linking amphetamine use with aggression and to consider possible factors that might underlie this association. RECENT FINDINGS: Although evidence that amphetamine use is related to increased levels of aggression continues to grow, the underlying processes or mechanisms remain somewhat elusive. In this review, three possible underlying factors are considered. Neurotoxic, pharmacological effects of amphetamine on the dopaminergic and serotonergic systems are related to aggressive, hostile behavior in both animal and human studies. Of particular inte

Long-lasting sensitization of reward-directed behavior by amphetamine.

Exposure to psychostimulant drugs of abuse such as amphetamine can result in long-lasting "sensitization" of reward-directed behavior, such that subjects display enhancements in behavior directed by and toward rewards and reward-predictive cues (i.e. "incentive sensitization"). The purpose of these experiments was to determine the degree to which such sensitization resulting from chronic amphetamine exposure influences both appetitive and consummatory food-motivated behavior. Adult male Long-Evans rats received daily i.p. injections of D-amphetamine (2.0 mg/kg) or saline vehicle for five conse

Individual differences in amphetamine sensitization, behavior and central monoamines.

Repeated amphetamine treatment results in behavioral sensitization in a high percentage of rats. Alterations to plasma corticosterone, neural monoamines and stress behavior can accompany amphetamine sensitization. Whether these changes occur following repeated amphetamine treatment in the absence of behavioral sensitization is not known. Male Sprague-Dawley rats were treated with amphetamine (2.5 mg/kg, i.p.) or saline once daily for 6 days. Amphetamine-induced locomotion and stereotypy, open-field anxiety behavior, plasma corticosterone and limbic monoamines were measured during withdrawal. S

Cocaine and amphetamine regulated transcript (CART) in suicide attempters.

Cocaine and amphetamine regulated transcript (CART) is a neuropeptide expressed in brain regions thought to regulate anxiety levels, depression, addiction and energy homeostasis. Individuals with a CART mutation display increased anxiety and depression. Severe anxiety is a core phenomenon of suicidality. We therefore studied levels of CART in the cerebrospinal fluid (CSF) of 98 patients with different psychiatric diagnoses, shortly after a suicide attempt. We also investigated the relationship between CSF-CART and relevant psychiatric symptoms. CART levels were determined using a radioimmunoas

Cocaine and amphetamine-like psychostimulants: neurocircuitry and glutamate neuroplasticity.

Although the pharmacology of amphetamine-like psychostimulants at dopamine transporters is well understood, addiction to this class of drugs has proven difficult to deal with. The reason for this disconnection is that while the molecular mechanism of amphetamine action is critical to reinforce drug use, it is only the first step in a sequence of widespread neuroplastic events in brain circuitry. This review outlines the affect of psychostimulants on mesocorticolimbic dopamine projections that mediate their reinforcing effect, and how this action ultimately leads to enduring pathological neurop
