BACKGROUND: Roberts syndrome (RBS) is a rare genetic disorder characterized by craniofacial abnormalities, limb malformation, and often severe mental retardation. RBS arises from mutations in ESCO2 that encodes an acetyltransferase and modifies the cohesin subunit SMC3. Mutations in SCC2/NIPBL (encodes a cohesin loader), SMC3 or other cohesin genes (SMC1, RAD21/MCD1) give rise to a related developmental malady termed Cornelia de Lange syndrome (CdLS). RBS and CdLS exhibit overlapping phenotypes, but RBS is thought to arise through mitotic failure and limited progenitor cell proliferation while

Reduced bone quality or mineral density predict susceptibility to fracture and also attenuate subsequent repair. Bone regrowth is also compromised by bacterial infection, which exacerbates fracture site inflammation. Because of the cellular complexity of fracture repair, as well as genetic and environmental influences, there is a need for models that permit visualisation of the fracture repair process under clinically relevant conditions. To characterise the process of fracture repair in zebrafish, we employed a crush fracture of fin rays, coupled with histological and transgenic labelling of

BACKGROUND: Sunitinib is a widely used multi-target tyrosine kinase inhibitor associated with side effects that may impair tissue repair and regeneration, potentially contributing to the onset of medication-related osteonecrosis of the jaw. Macrophages play a central role in tissue homeostasis and regeneration, and their dysfunction may be a key factor in this pathological process.
METHODS: In this study, a zebrafish tail fin injury regeneration model was used to investigate the effects of sunitinib on macrophage function and tissue regeneration. Transcriptomic analysis, immunofluorescence st

Salamanders and lungfishes are the only sarcopterygians (lobe-finned vertebrates) capable of paired appendage regeneration, regardless of the amputation level. Among actinopterygians (ray-finned fishes), regeneration after amputation at the fin endoskeleton has only been demonstrated in polypterid fishes (Cladistia). Whether this ability evolved independently in sarcopterygians and actinopterygians or has a common origin remains unknown. Here we combine fin regeneration assays and comparative RNA-sequencing (RNA-seq) analysis of Polypterus and axolotl blastemas to provide support for a common

Within Djibouti (Gulf of Aden), the scalloped hammerhead shark (Sphyrna lewini) and milk shark (Rhizoprionodon acutus) are important components of the artisanal fishery and they are caught to be exported or sold for local consumption. However, little scientific information exists on the contamination load of these species in this area of the world. With global populations of elasmobranchs in decline, understanding the extent of contaminant exposure is critical to future conservation as well as to assess the health risks for consumers of these species. The contaminants analyzed in this study co

BACKGROUND: Vibrio vulnificus infection is a rare, rapidly progressing condition often acquired through wounds exposed to aquatic environments or marine animals. It can cause septic shock, organ failure, and high mortality. Reports on limb salvage in necrotizing fasciitis caused by V. vulnificus are limited.
CASE PRESENTATION: A 62-year-old Vietnamese male with alcohol use and hepatitis B developed swelling, bullae, and necrosis in his left hand after a tilapia fin injury. He experienced septic shock requiring resuscitation, antibiotics, and ventilation. Cultures confirmed V. vulnificus. Surg

Lancefield serological group C Streptococcus dysgalactiae (GCSD) is an emerging cause of fish disease worldwide, largely associated with mariculture in Asia. In the United States (US), GCSD had been a pathogen of minimal concern for fish, but recent cases indicate a putative emergence in wild and cultured freshwater fish populations in the Americas. The current study discusses three novel cases of GCSD-associated streptococcosis in US farm-raised channel catfish (Ictalurus punctatus) occurring in Mississippi, US, in 2022 and 2023. Infected fish presented with pendulous abdomens, red swollen ve

A 25-day-old male common bottlenose dolphin (Tursiops truncatus) died suddenly while swimming at a dolphinarium. The gross examination revealed ulceration on the dorsal and pectoral fins and rostrum. Severe congestion, hemorrhage, and edema were observed in the gastrointestinal tract, liver, mesenteric lymph nodes, lungs, and kidneys. Fibrinosuppurative arthritis of the atlantooccipital joint and extension of fibrin into the spinal canal caused compression of the spinal cord. Histopathological examination revealed tracheitis, fibrinosuppurative bronchopneumonia and enteritis. In the central ne

INTRODUCTION/AIMS: The compound motor action potential (CMAP) is a very well-established output from standard motor conduction studies in patients. CMAP methods have also been developed for various animal models, including mice, rats, and dogs. Here, we describe a CMAP methodology for adult zebrafish.
METHODS: Using needle stimulating electrodes placed in proximity to the caudal spinal column and a fixed two-electrode surface array placed near the dorsal fin for recording, we obtained CMAPs in wildtype (WT) and symptomatic amyotrophic lateral sclerosis (ALS) SOD1G93A zebrafish, assessing repe

Respiratory infections are major health concerns in wild and managed cetaceans, yet reliable methods for assessing lower respiratory tract inflammation are limited. We aimed to evaluate the clinical utility of bronchoalveolar lavage fluid (BALF) neutrophil differentials in three managed bottlenose dolphins (Tursiops truncatus) and three managed beluga whales (Delphinapterus leucas) undergoing treatment for respiratory infections. Infection status was monitored by assessing isolated bacteria and fungi from BALF, BALF neutrophil differentials, and inflammatory markers in the blood, such as total

Rich fossil evidence suggests that many traits and functions related to terrestrial evolution were present long before the ancestor of lobe- and ray-finned fishes. Here, we present genome sequences of the bichir, paddlefish, bowfin, and alligator gar, covering all major early divergent lineages of ray-finned fishes. Our analyses show that these species exhibit many mosaic genomic features of lobe- and ray-finned fishes. In particular, many regulatory elements for limb development are present in these fishes, supporting the hypothesis that the relevant ancestral regulation networks emerged befo

Polyunsaturated fatty acids (PUFAs) play important roles in organisms, including the structure and liquidity of cell membranes, anti-oxidation and anti-inflammation. Very little has been done in terms of the effect of PUFAs on cell death, especially on DNA virus. In this study, we demonstrated that the infectious spleen and kidney necrosis virus (ISKNV) can induce host cell death via the apoptotic cell death pathway, which correlated to modulation by PUFAs in grouper fin cell line (GF-1) cells. We screened the PUFAs, including docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), for the

This study reports the results of seroepidemiological investigations carried out against Morbillivirus, Toxoplasma gondii and Brucella spp. on blood serum samples collected from 70 cetacean specimens found stranded along the Italian coastline between 1998 and 2014. A total number of 23 serum samples (32.8%) obtained from Stenella coeruleoalba, Tursiops truncatus, Balaenoptera physalus and Globicephala melas harboured anti-Morbillivirus neutralizing antibodies. Ten sera (16%) collected from S. coeruleoalba and T. truncatus were found positive against T. gondii, while no antibodies against Bruce

Narrow site specificity in parasites is assumed to be associated with fitness benefits, such as higher reproductive success, although this is never quantified. We linked the body mass and combined mass of egg sacs of female copepods, Neobrachiella spinicephala, parasitic on the sandperch, Pinguipes brasilianus, to attachment sites on the host. Adult females attach permanently either on the lips, the margins of the operculum, or the base of pectoral or pelvic fins. In addition to influences of sampling site, season and host body length, our analyses revealed important fitness effects. First, at

This study is the first investigation for using sex-related gene expression in tail fin tissues of seabass as early sex determination without killing the fish. The European seabass (Dicentrarchus labrax) is gonochoristic and lacks distinguishable sex chromosomes, so, sex determination is referred to molecular actions for some sex-related genes on autosomal chromosomes which are well known such as cyp19a1a, dmrt1a, and dmrt1b genes which play crucial role in gonads development and sex differentiation. cyp19a1a is expressed highly in females for ovarian development and dmrt1a and dmrt1b are for

INTRODUCTION: Type 2 diabetes mellitus (T2D) is a metabolic disorder characterized by chronic hyperglycemia, insulin resistance, and meta-inflammation, which significantly compromise tissue regeneration. Although macrophage dysfunction is implicated in impaired wound healing in T2D, the immunometabolic mechanisms linking hyperglycemia to defective tissue repair remain incompletely understood.
METHODS: Zebrafish larvae were exposed to hyperglycemic conditions (4% dextrose) to establish a T2D-like model. Survival, glycemic and biochemical parameters were assessed, followed by caudal fin amputat

We tested whether tetrodotoxin (TTX) functions as a stress relieving substance in puffer fish. We orally administered TTX to the juveniles of hatchery-reared non-toxic tiger puffer Takifugu rubripes and measured the effects of TTX on brain corticotropin-releasing hormone (CRH) mRNA expression and plasma cortisol levels in comparison with effects in non-toxic juveniles. Firstly, the reciprocal connections of CRH and adrenocorticotropic hormone (ACTH) were confirmed by dual-label immunohistochemistry. CRH-immunoreactive (ir) cell bodies were detected in the hypothalamus and CRH-ir fibers were ob

Aggressive behavior is important for animals to obtain limited resources. Understanding fish behavior and physiological response is of great significance to evaluate aquaculture production and fish welfare. Food is an important trigger of aggressive behavior in juvenile fish under high-density aquaculture conditions. The aim of this study was to investigate the aggressive behavior and monoamine levels of juvenile pufferfish (mean body mass of 6.29 ± 0.33 g) under normal feeding and restricted feeding. Our main results included the following: (1) The mortality and fin damage were higher and agg

Oxygen is essential for metabolism of animals and is a vital component of their natural habitats. Hypoxic conditions in tissue, when oxygen levels are lower than normal, change a variety of cellular processes, while environmental hypoxia can have physiological and behavioral effects on the whole animal. Larval zebrafish respond to oxygen deprivation with a characteristic set of physiological changes and motor behaviors, making them a convenient vertebrate model to study hypoxia responses. However, to date, hypoxia studies in zebrafish are limited by the existing experimental setups, which only
