Identification of Novel Toxin Genes from the Stinging Nettle Caterpillar Parasa lepida (Cramer, 1799): Insights into the Evolution of Lepidoptera Toxins.

Many animal species can produce venom for defense, predation, and competition. The venom usually contains diverse peptide and protein toxins, including neurotoxins, proteolytic enzymes, protease inhibitors, and allergens. Some drugs for cancer, neurological disorders, and analgesics were developed based on animal toxin structures and functions. Several caterpillar species possess venoms that cause varying effects on humans both locally and systemically. However, toxins from only a few species have been investigated, limiting the full understanding of the Lepidoptera toxin diversity and evoluti

Endoscopic skull base surgery II: a new animal model for surgery of the posterior fossa.

The field of skull base surgery has been influenced by a general philosophy that currently exists in modern surgical practice favoring less invasive means of managing surgical disease. Adapting techniques developed by general surgeons and other surgical subspecialists, skull base surgeons are now experimenting with endoscopy to resect tumors, manage vascular lesions, and manipulate critical intracranial structures. Lesions formerly requiring significant soft tissue dissection and craniotomy for exposure are now potentially amenable to treatment via a keyhole approach. As in other surgical spec

Effects of morphine on central catecholamine turnover, blood pressure and heart rate in the rat.

In the unanaesthetized rat morphine caused increased dopamine (DA) turnover, unchanged or possibly increased central noradrenaline (NA) turnover (utilization), hypertension and tachycardia. In the anaesthetized rat, brain DA turnover was not affected, whereas the NA-turnover was decelerated, particularly in some brain regions, e.g. cerebral cortex and medulla oblongata, and hypotension and bradycardia was obtained. Both biochemical and cardiovascular effects of morphine were antagonized by naloxone. A very small dose of morphine (1 mg/kg) caused tachycardia also in the anaesthetized rat. Decer

Variation in structural motifs within SARS-related coronavirus spike proteins.

SARS-CoV-2 is the third known coronavirus (CoV) that has crossed the animal-human barrier in the last two decades. However, little structural information exists related to the close genetic species within the SARS-related coronaviruses. Here, we present three novel SARS-related CoV spike protein structures solved by single particle cryo-electron microscopy analysis derived from bat (bat SL-CoV WIV1) and civet (cCoV-SZ3, cCoV-007) hosts. We report complex glycan trees that decorate the glycoproteins and density for water molecules which facilitated modeling of the water molecule coordination ne

A Review of Recent Progress on Collagen-Based Biomaterials.

Since the 2010s, the demand for healthcare models has exceeded the prevailing resources available due to the rapid increase in the aging population in China. However, a significant gap in development of biomedical materials remains, especially between China and the western developed countries. Collagen is the major protein of the extracellular matrix (ECM) and has been extensively applied in medical fields. Collagen-based biomaterials (CBBs) are used to prepare dressings and dermal substitutes, surgical sutures, plasma substitutes, tissue-engineered scaffolds, and drug delivery systems; this i

Structures of two core subunits of the bacterial type IV secretion system, VirB8 from Brucella suis and ComB10 from Helicobacter pylori.

Type IV secretion systems (T4SSs) are commonly used secretion machineries in Gram-negative bacteria. They are used in the infection of human, animal, or plant cells and the propagation of antibiotic resistance. The T4SS apparatus spans both membranes of the bacterium and generally is composed of 12 proteins, named VirB1-11 and VirD4 after proteins of the canonical Agrobacterium tumefaciens T4SS. The periplasmic core complex of VirB8/VirB10 structurally and functionally links the cytoplasmic NTPases of the system with its outer membrane and pilus components. Here we present crystal structures o

Animal toxins and NETosis: A new frontier in innate immune modulation.

Neutrophil extracellular traps are molecular structures that play a dual role in host defense and pathology. While NETs effectively capture and neutralize pathogens, excessive or dysregulated NET formation contributes to autoimmune and inflammatory disorders. Animal venoms contain bioactive molecules that can modulate neutrophil functions, including NETosis. They are capable of inducing, inhibiting, or degrading NETs through different mechanisms, such as signaling pathway modulation, or direct DNA hydrolysis. Such effects not only shape the course of envenomation but also reveal novel pathways

Proprioception in Muscle Hydrostats.

Proprioception can be seen as a somatic sense stimulated by the action of the body itself. It is perceived through proprioceptors and is tightly linked to the animal body, as it is influenced by the biomechanical properties of the structures in which it is embedded. A specific class of these receptors, the muscle proprioceptors, project at several levels of the nervous system and provide information about limb position, whether in the presence or absence of movement, as well as muscle length, the sense of effort, and the sense of balance. In skeletal systems, proprioception is involved in post

The curly tail mouse model of human neural tube defects demonstrates normal spinal cord differentiation at the level of the meningomyelocele: implications for fetal surgery.

The paralysis associated with lumbosacral meningomyelocele has been attributed both to myelodysplasia and to degeneration of the exposed neural tissue. Surgically created dysraphism shows that exposure of an intact spinal cord in a genetically normal animal results in degeneration of the normal nervous tissue and subsequent paralysis. Our objective was to study neuronal differentiation in the curly tail mouse mutant model, which develops lumbosacral meningomyelocele naturally and is a phenocopy of nonsyndromic human neural tube defects. Prenatal repair of meningomyelocele assumes that the norm

Structural and resting state functional connectivity beyond the cortex.

Mapping the structural and functional connectivity of the central nervous system has become a key area within neuroimaging research. While detailed network structures across the entire brain have been probed using animal models, non-invasive neuroimaging in humans has thus far been dominated by cortical investigations. Beyond the cortex, subcortical nuclei have traditionally been less accessible due to their smaller size and greater distance from radio frequency coils. However, major neuroimaging developments now provide improved signal and the resolution required to study these structures. He

The Riveting Cellular Structures of Apicomplexan Parasites.

Parasitic protozoa of the phylum Apicomplexa cause a range of human and animal diseases. Their complex life cycles - often heteroxenous with sexual and asexual phases in different hosts - rely on elaborate cytoskeletal structures to enable morphogenesis and motility, organize cell division, and withstand diverse environmental forces. This review primarily focuses on studies using Toxoplasma gondii and Plasmodium spp. as the best studied apicomplexans; however, many cytoskeletal adaptations are broadly conserved and predate the emergence of the parasitic phylum. After decades cataloguing the co

Feasibility and therapeutic strategies of vaccines against Porphyromonas gingivalis.

Periodontitis is a chronic infectious disease that is highly prevalent worldwide and is characterized by inflammation of the gums, and loss of connective tissue and bone support. The Gram-negative anerobic bacterium Porphyromonas gingivalis is generally accepted as the main etiological agent for chronic periodontitis. The objective of this paper is to elucidate the feasibility of achieving protection against periodontitis though immunization against P. gingivalis. Until now, animal studies have showed no complete protection against P. gingivalis. However, current knowledge about P. gingivalis

A miniaturized cryoprobe for functional neuronal blockade in freely moving animals.

We have developed a miniature cryoprobe for blockade of neuronal structures. The cryoprobe uses Freon as a cooling medium and consists of concentric double stainless steel cannulae, a shaft heater coil and a tip thermocouple. The cryoprobe tip temperature is controlled by adjusting Freon flow supplying the cryoprobe by an automobile fuel injector and electronic control circuit, and is displayed on a temperature monitor. Tissue temperatures, determined by thermocouples, ranged from 8.5 degrees C near the cryoprobe tip to 20 degrees C 2.5 mm distal to the tip. The cryoprobe shaft is maintained a

Design with nonhumans takes wildlife agency into account to reduce conflicts.

The concept of nonhuman animal agency has recently been introduced in fields from more-than-human geography to wildlife conservation. Nonhuman animal agency refers to the capacity of animals to create, set in motion, and cause or influence actions through their relations with humans and other nonhumans. Ecologists have long recognized that animal species are active components of ecosystems and have important ecological functions, yet most wildlife management treats species as objects that are (or should be) passive and controllable. We argue that explicit recognition of nonhuman agency can red

Generating Free-floating Normal Human Epithelial-Fibroblast Spheroid Co-Cultures.

Three-dimensional (3D) culture systems have become increasingly popular due to their ability to reproduce natural cell properties and architectures, thus mimicking tissue-like structures in vitro. Among these models, the culture of spherical cell aggregates (so-called spheroids) embedded in a semi-solid extracellular matrix (ECM) represents an advanced near-in vivo cell culture model, as it allows for different functional cell states as a result of cell-cell and cell-ECM interactions, as well as oxygen and nutrient gradients. Because spheroids are technically less demanding and relatively inex

The role of prefrontal systems in sexual behavior.

Research from a variety of methodologies is demonstrating a role for neuroanatomical structures in different aspects of sexual behavior. This article reviews the particular contribution of prefrontal-subcortical systems to sexual behavior, including regions of prefrontal cortex (i.e., dorsolateral, medial, and orbitofrontal cortex), and associated subcortical structures (i.e., basal ganglia and thalamus). Findings are integrated from functional neuroimaging and clinical studies in humans, as well as animal studies, which convergently illustrate the role of different prefrontal systems. In addi

Physiological and Behavioral Mechanisms of Thermoregulation in Mammals.

This review analyzes the main anatomical structures and neural pathways that allow the generation of autonomous and behavioral mechanisms that regulate body heat in mammals. The study of the hypothalamic neuromodulation of thermoregulation offers broad areas of opportunity with practical applications that are currently being strengthened by the availability of efficacious tools like infrared thermography (IRT). These areas could include the following: understanding the effect of climate change on behavior and productivity; analyzing the effects of exercise on animals involved in sporting activ

Cross-Sectoral Zoonotic Disease Surveillance in Western Kenya: Identifying Drivers and Barriers Within a Resource Constrained Setting.

Background: Collaboration between the human and animal health sectors, including the sharing of disease surveillance data, has the potential to improve public health outcomes through the rapid detection of zoonotic disease events prior to widespread transmission in humans. Kenya has been at the forefront of embracing a collaborative approach in Africa with the inception of the Zoonotic Disease Unit in 2011. Joint outbreak responses have been coordinated at the national level, yet little is currently documented on cross-sectoral collaboration at the sub-national level. Methods: Key informant in
