Evaluation of Novel Isatin-Schiff Base Derivatives: Monoamine Oxidase Inhibition and Antimicrobial Assessment.

Five novel series of isatin-Schiff base derivatives (1a-5e) were synthesized, and their neuroprotective and anti-infective potential was evaluated. Their half-maximal inhibitory concentration (IC50) was determined against both monoamine oxidase A (MAO-A) and MAO-B targets for the treatment of depression and Parkinson's disease, respectively. The anti-infective potential of the synthesized derivatives was evaluated by measuring their IC50 values against urease, an enzyme utilized by specific pathogenic bacteria as a virulence factor. The minimum inhibitory concentration for each compound was as

The antifungal, antibacterial, and antiviral activities of a Echinosterol isolated from the red sea Echinoclathria sponge.

The emergence of drug-resistant pathogenic microbes is a global medical concern. Our aim was to evaluate the anti-infective activities of a novel marine sterol isolated from the Red Sea Echinoclathria sponge. An agar-plate diffusion assay was used to evaluate antifungal and antibacterial activity, while a rapid antiviral assay was used to assess antiviral activity. In silico analysis via the PASS-Online platform was used to predict and explain the potential anti-infective mechanisms of action. Echinosterol was effective against Candida albicans, Cryptococcus neoformans, Aspergillus fumigatus,

Inhaled fixed-dose combination powders for the treatment of respiratory infections.

INTRODUCTION: Respiratory infections are a major cause of morbidity and mortality. As an alternative to systemic drug administration, inhaled drug delivery can produce high drug concentrations in the lung tissue to overcome resistant bacteria. The development of inhaled fixed-dose combination powders (I-FDCs) is promising next step in this field, as it would enable simultaneous drug-drug or drug-adjuvant delivery at the site of infection, thereby promoting synergistic activity and improving patient compliance. AREAS COVERED: This review covers the clinical and pharmaceutical rationales for th

[11 cases of erythema multiforme. The etiological correlations and therapy].

Erythema multiforme (EM) is characterised by a polymorphous eruptive complex which many involve the cutis and oral, genital and conjunctival mucous. Its etiopathogenesis is unclear: it is thought to be associated with various viral or bacterial infective agents (herpes virus, Coxsackie virus, mycoplasmas, etc.), numerous drugs, physical therapy, systemic pathologies of various types. The authors report their experience in relation to 11 patients suffering from EM; the clinical characteristics of each case are reported, together with the course of disease, and the diagnostic and therapeutic pro

Crovirin, a snake venom cysteine-rich secretory protein (CRISP) with promising activity against Trypanosomes and Leishmania.

BACKGROUND: The neglected human diseases caused by trypanosomatids are currently treated with toxic therapy with limited efficacy. In search for novel anti-trypanosomatid agents, we showed previously that the Crotalus viridis viridis (Cvv) snake venom was active against infective forms of Trypanosoma cruzi. Here, we describe the purification of crovirin, a cysteine-rich secretory protein (CRISP) from Cvv venom with promising activity against trypanosomes and Leishmania. METHODOLOGY/PRINCIPAL FINDINGS: Crude venom extract was loaded onto a reverse phase analytical (C8) column using a high perf

[Pharmacologic therapy of bronchial asthma].

The main feature in asthma is bronchial eosinophilic inflammation, induced, in genetically predisposed subjects, by known (aeroallergens, viral infection, occupational agents) and unknown agents. Inflammation critically increases bronchial responsiveness so that even rather mild stimuli (exercise, cold air, mist, allergens) are able to induce obstructive reactions. When preventive measures (specific immunotherapy, avoidance of inflammatory and precipitating factors) are inadequate, a pharmacological treatment is necessary. This should, in addition relieve bronchospasm, neutralize precipitating

Limitations of current antibiotics for the treatment of bacterial conjunctivitis.

Bacterial conjunctivitis is a common ocular infection that is generally treated empirically with a broad-spectrum antibiotic. The more common pathogens causing bacterial conjunctivitis include Staphylococcus aureus, Haemophilus influenzae, Streptococcus pneumoniae, Staphylococcus epidermidis, and Moraxella species. Several antibiotics traditionally used to treat bacterial conjunctivitis are no longer widely prescribed because of increased bacterial resistance and/or safety concerns. The introduction of the fluoroquinolone class of anti-infectives offered effective and better tolerated treatmen

2-(Nitroaryl)-5-Substituted-1,3,4-Thiadiazole Derivatives with Antiprotozoal Activities: In Vitro and In Vivo Study.

Nitro-containing compounds are a well-known class of anti-infective agents, especially in the field of anti-parasitic drug discovery. HAT or sleeping sickness is a neglected tropical disease caused by a protozoan parasite, Trypanosoma brucei. Following the approval of fexinidazole as the first oral treatment for both stages of T. b. gambiense HAT, there is an increased interest in developing new nitro-containing compounds against parasitic diseases. In our previous projects, we synthesized several megazole derivatives that presented high activity against Leishmania major promastigotes. Here, w

GC-MS profiling and antivirulence potential of limonene-rich Citrus medica cv. 'Rugosa' essential oil: in vitro and in silico evaluations.

Antimicrobial therapy is a primary approach for treating microbial infections, yet increasing antibiotic resistance continues to hinder effective disease management, highlighting natural products as promising alternative sources of novel anti-infective agents. In this context, Citrus medica cv. ‘rugosa’ EO was chemically profiled by GC-MS, and its antimicrobial, antibiofilm, and anti-quorum sensing inhibitory action was firstly tested using both in vitro and computational studies. C. medica cv ‘‘rugosa ‘’ EO revealed a predominance of monoterpene hydrocarbons (79.12%) with limonene (76.15%) as

Neutropenia possibly caused by cefoperazone/sulbactam.

Neutropenia is a rare complication of drug therapy and is usually underdiagnosed. Cefoperazone/sulbactam is a combination of broad-spectrum antibacterial agents. Data on cefoperazone/sulbactam-induced neutropenia are limited. Herein, we report the case of a 35 year-old female patient who was admitted to the hospital due to an appendiceal abscess. After anti-infective treatment with cefoperazone/sulbactam, the patient developed neutropenia on day 4. After discontinuing treatment with cefoperazone/sulbactam, the patient's white blood cells and neutrophils gradually returned to normal. Hence, cli

Enantioselective Total Syntheses of Sannamycins A and B.

Aminoglycoside antibiotics are one of the oldest and most clinically relevant classes of anti-infective agents, yet their complex molecular architectures have restricted access through bottom-up syntheses for decades. Herein we report enantioselective syntheses of 2-deoxyfortamine-type aminoglycosides sannamycins A and B. The described strategy involves an enantioselective dearomative hydroamination, rapid stereo- and chemoselective introduction of heteroatom functionalities, and a unique skeletal rearrangement to forge the aminocyclitol core. The carbohydrate fragment was elaborated from Cyre

Bacterial Infections in the Stem Cell Transplant Recipient and Hematologic Malignancy Patient.

Bacteremia (bloodstream infection) is frequent (20%-30% incidence) in the stem cell transplant and hematologic malignancy population and often occurs in the early post-transplant engraftment period. In most studies, Gram-positive bacteria occur at greater frequency than gram-negative bacteria, although some centers report that rates of gram-negative bloodstream infections have recently increased. In many centers, resistance rates among Enterococci and gram-negative bacteria, especially the Enterobacteriaceae, are extensive and associated with increased mortality. Better prediction tools, enhan

[Therapeutic approaches to cryptosporidiosis. A review of the literature].

Cryptosporidiosis is a coccidian infection that usually occurs in children an immunocompromised patients. With the AIDS (Acquired Immunodeficiency Syndrome) epidemic there have been an increased number of clinical cases and still we don't have an optimal therapeutic regimen to eradicate the infection. Since 1907 when the organism was first described, a large amount of anti-infective agents have been used without success. We present herein a review of the new therapeutic approaches, although none of them is satisfactory and new studies are required for the development of an optimal treatment. S

Effect of pretreatment with Mycobacterium bovis (strain BCG) and immune syphilitic serum on rabbit resistance to Treponema pallidum.

Stimulation of the rabbit reticuloendothelial system with viable Mycobacterium bovis (strain BCG), and other agents, had no effect on the development of syphilitic lesions after intradermal or intravenous inoculation with graded doses of Treponema pallidum (virulent Nichol's strain; mean infective doses less than 10). The simultaneous administration of immune syphilitic rabbit serum retarded the development of lesions, but this appeared to be due solely to the immune serum, suggesting no synergism between the activated reticuloendothelial system and the anti-T. pallidum antibodies. Evaluation of Short-Chain Antimicrobial Peptides With Combined Antimicrobial and Anti-inflammatory Bioactivities for the Treatment of Zoonotic Skin Pathogens From Canines.

The incidence of zoonotic Staphylococcus pseudintermedius and Microsporum canis infections is rapidly growing worldwide in the context of an increasing frequency of close contact between animals and humans, presenting challenges in both human and veterinary medicine. Moreover, the development of microbial resistance and emergence of recalcitrant biofilms, accompanied by the insufficiency of new antimicrobial agents, have become major obstacles in treating superficial skin infections caused by various microbes including S. pseudintermedius and M. canis. Over recent years, the prospects of antim

Next-generation-sequencing technology used for the detection of Mycoplasma hominis in renal cyst fluid: a case report.

Background: Mycoplasma is an opportunistic pathogen causing both urogenital and extragenital infections. The lack of cell wall renders Mycoplasma difficult to culture and identify with ordinary methods. Next-generation sequencing (NGS) is a new technology helping a lot in the diagnosis of infective diseases. In this case, NGS played a key role in the diagnosis of Mycoplasma infection. Case presentation: A mid-aged man suffering from renal cyst underwent cyst incision followed by invasive treatments to eliminate hematoma caused by renal artery hemorrhage. After the cyst incision operation, the

[4.3.1]Bicyclic FKBP Ligands Inhibit Legionella Pneumophila Infection by LpMip-Dependent and LpMip-Independent Mechanisms.

Legionella pneumophila is the causative agent of Legionnaires' disease, a serious form of pneumonia. Its macrophage infectivity potentiator (Mip), a member of a highly conserved family of FK506-binding proteins (FKBPs), plays a major role in the proliferation of the gram-negative bacterium in host organisms. In this work, we test our library of >1000 FKBP-focused ligands for inhibition of LpMip. The [4.3.1]-bicyclic sulfonamide turned out as a highly preferred scaffold and provided the most potent LpMip inhibitors known so far. Selected compounds were non-toxic to human cells, displayed antiba
