Effects of harmane and other β-carbolines on apomorphine-induced licking behavior in rat.

Harmane, harmine and norharmane are β-carboline compounds which have been referred to as inverse agonists of benzodiazepine receptors. The effect of these compounds on apomorphine-induced licking behavior was studied in rats. Subcutaneous (s.c.) injection of apomorphine (0.5 mg/kg) induced licking. The licking behavior was counted with a hand counter and recorded for a period of 75 min by direct observation. Intraperitoneal (i.p.) injections of harmane (1.25-5 mg/kg), harmine (2.5-10 mg/kg) and norharmane (1.25-5 mg/kg) significantly reduced the licking behavior. In rats pretreated with reserp

Microdialysis study of striatal dopamine in MPTP-hemilesioned rats challenged with apomorphine and amphetamine.

Motor impairments of Parkinson's disease (PD) appear only after the loss of more than 70% of the DAergic neurons of the substantia nigra pars compacta (SNc). An earlier phase of this disease can be modeled in rats that received a unilateral infusion of the neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyrindine (MPTP) into the SNc. Though these animals do not present gross motor impairments, they rotate towards the lesioned side when challenged with DAergic drugs, like amphetamine and apomorphine. The present study aimed to test whether these effects occur because the drugs disrupt compensato

The effect of apomorphine on blink kinematics in subhuman primates with and without facial nerve palsy.

PURPOSE: The purpose of this study was to document the effect of acutely delivered apomorphine, a dopamine receptor agonist with both D1 and D2 properties, on blink rate and the amplitude-velocity characteristics of eyelid kinematics in a group of nonhuman primates. METHODS: Three cynomolgus and two rhesus macaques underwent baseline recordings for eyelid kinematics, using the Robinson search coil technique. Next, each animal received a 0.15-mg/kg subcutaneous injection of apomorphine. Recordings were taken at 45 and 90 minutes, respectively, after injection. Blink rates per minute were obtai

Potent neuroprotective and antioxidant activity of apomorphine in MPTP and 6-hydroxydopamine induced neurotoxicity.

Apomorphine is a potent radical scavenger and iron chelator. In vitro apomorphine acts as a potent iron chelator and radical scavenger with IC50 of 0.3 microM for iron (2.5 microM) induced lipid peroxidation in rat brain mitochondrial preparation, and it inhibits mice striatal MAO-A and MAO-B activities with IC50 values of 93 microM and 241 microM. Apomorphine (1-10 microM) protects rat pheochromocytoma (PC12) cells from 6-hydroxydopamine (150 microM) and H2O2 (0.6 mM) induced cytotoxicity and cell death. The neuroprotective property of (R)-apomorphine, a dopamine D1-D2 receptor agonist, has b

Autosomal and Y chromosomal effects on the stereotyped response to apomorphine in wild house mice.

The behavioral response to apomorphine, a dopamine agonist, was shown to be different between a selection line characterized by Short Attack Latencies (SAL) and a selection line having Long Attack Latencies (LAL) (4). Aggressive SAL mice were more sensitive to apomorphine than nonaggressive LAL males. The aim of this research was to determine whether the stereotyped response to apomorphine is affected by the Y chromosome in the same way as it influences attack latency. For this purpose, F1 reciprocal hybrids as well as congenic lines (SAL.LY and LAL.SY) were used. The major difference between

Psychotic and depressive symptoms in Parkinson's disease. A study of the growth hormone response to apomorphine.

BACKGROUND: The growth hormone (GH) response to apomorphine, thought to reflect central dopaminergic receptor sensitivity, has been reported as enhanced in acute schizophrenia. We investigated this response in relation to the psychotic episodes associated with Parkinson's disease (PD). METHOD: The GH response to apomorphine was measured in three groups of patients with Parkinson's disease: those currently psychotic (n = 9), those with a past history of psychosis (n = 7) and those who had never been psychotic (n = 8). RESULTS: Apomorphine-induced GH response was not related to psychosis but w

The effect of a putative antidepressant, an O2-substituted derivative of barbitone on apomorphine induced hypomotility.

The effects of the chronic administration of barbitone and its O2-substituted isopropyl derivative (O2IB) was studied on the hypomotility induced in rats by the acute administration of a low dose of apomorphine. Neither drug was found to selectively antagonize the hypomotility caused by acute apomorphine administration. No correlation could be found between the behavioural effects of the barbiturates and changes in the concentrations of biogenic amines and GABA in four discrete brain regions. The results of this study do not support the conclusions of previous acute studies in which O2IB has b

Midification of tardive dyskinesia and spasmodic torticollis by apomorphine. Possible role of dopamine autoreceptors.

A group of 16 patients afflicted with involuntary movement disorders received subcutaneous injections of the direct dopamine agonist, apomorphine hydrochloride. Paradoxically, these injections were generally followed by a reduction of dyskinesia; this was most noticeable in patients with tardive dyskinesia and was only mild in some patients with spasmodic torticollis. Preferential stimulation by apomorphine of inhibitory dopamine presynaptic receptors (so-called dopamine autoreceptors) is proposed as the most likely explanation for the observed antidyskinetic effect of this drug. The results o

Sublingual apomorphine for Parkinsonian tremor: a post-hoc analysis of pivotal trial data.

Tremor is a common and disabling feature of Parkinson's disease (PD) and often responds suboptimally to oral dopaminergic therapy. This post‑hoc analysis evaluated tremor outcomes associated with apomorphine sublingual film (SL‑APO) in patients with a tremor‑dominant PD phenotype, using data from a 12‑week randomised, double‑blind, placebo‑controlled trial (NCT02469090) and a 12‑month open‑label study (NCT02542696) conducted in levodopa‑responsive patients with OFF episodes. Tremor‑dominant phenotype was defined according to Movement Disorder Society-Unified Parkinson's Disease Rating Scale (M

Two distinctive apomorphine-induced phenotypes in the Roman high- and low-avoidance rats.

Strain differences in spontaneous locomotor activity and the behavioral patterns induced by dopaminergic agonists in rodents can disclose differential genetic susceptibilities to dopaminergic dysfunction (i.e. vulnerability to psychosis). Psychogenetic selection of hypoemotional Roman High-Avoidance (RHA) and hyperemotional Low-Avoidance (RLA) rats leads to divergence in dopaminergic function as well. The present study was designed to characterize their spontaneous activity and their responses to apomorphine (0.067-3 mg/kg, s.c.) as compared to those of the standard Sprague-Dawley (SD) strain.

Mixed D2/5-HT2 antagonism differentially affects apomorphine- and amphetamine-induced stereotyped behavior.

Evidence supports the hypothesis that psychostimulant stereotypy is mediated through postsynaptic dopamine receptors. Given the recent findings of behavioral, neurochemical and electrophysiological studies showing 5-HT2 modulation of dopamine systems, a series of experiments were undertaken to assess the ability of D2 and 5-HT2 antagonists to reverse apomorphine and amphetamine stereotypy in the rat. Haloperidol reduced stereotyped behavior induced by d-amphetamine (50% reduction with 0.162 mg/kg) and apomorphine (50% reduction with 0.112 mg/kg) MDL 28,133A, a mixed D2/5-HT2 antagonist, also r

Early dumping reaction after partial gastrectomy and its relation to preoperative apomorphine testing.

Risk factors for postoperative early dumping symptoms were studied prospectively in 289 consecutive peptic ulcer patients followed up for 5 years after antrectomy and gastroduodenostomy with or without vagotomy. Grade 2 or 3 dumping was present in 14% of the patients and was about four times as frequent in women as in men after antrectomy alone. The incidence of such dumping grades was twice as high after combined antrectomy and vagotomy as after antrectomy alone. Both differences were statistically significant. Milk intolerance, which often appeared as a symptom of dumping, was more common af

"On-off" phenomena, dyskinesias and dystonias. Comparison of lisuride versus apomorphine acute treatment.

In this study the effects of an acute injection of lisuride and apomorphine in 12 subjects affected by dystonic-dyskinetic syndromes of different aetiology are evaluated: 3 patients with spasmodic torticollis, 4 with tardive dyskinesia and 5 Parkinson patients suffering from "on-off" attacks with prominent dyskinesias during the mobile phase. In the last group drugs were administered during the "on" phase. In 11 out of 12 patients both lisuride and apomorphine induced a marked improvement of the abnormal involuntary movements. How to manage the initiation of apomorphine therapy without antiemetic pretreatment: A review of the literature.

INTRODUCTION: Pretreatment with the antiemetic trimethobenzamide has been recommended practice in the United States (US) to address the risk of nausea and vomiting during initiation of apomorphine treatment. However, trimethobenzamide is no longer being manufactured in the US, and despite the recent update to the US prescribing information, there may be uncertainty regarding how to initiate apomorphine. METHODS: To better understand why antiemetic pretreatment was recommended and if it is necessary when initiating apomorphine therapy, we performed a literature review of subcutaneous apomorphi

Involvement of NMDA receptors in the ventrolateral striatum of rats in apomorphine-induced jaw movements.

The role of NMDA receptors in the ventrolateral striatum to modulate dopamine receptor-mediated jaw movements was investigated in freely moving rats, using a magnetic sensor system combined with intracerebral microinjection of drugs. Apomorphine (1mg/kg i.v.) induced repetitive jaw movements that were reduced, in a dose-dependent manner, by bilateral microinjections of the NMDA receptor agonist NMDA (0.1 and 1mug/0.2mul bilaterally) into the ventrolateral striatum. Apomorphine-induced repetitive jaw movements were also reduced, in a dose-dependent manner, by bilateral microinjections of the NM

Protection by apomorphine in two independent models of acute inhibition of oxidative metabolism in rodents.

Apomorphine was administered by continuous infusion in the mouse following acute inhibition of oxidative metabolism induced by systemic administration of MPTP, and in the gerbil following transient occlusion of the carotid arteries. The dosage employed was comparable to the one used in the treatment of severe on-off fluctuations in Parkinson's disease. The results show that apomorphine significantly diminishes the striatal lesion caused by MPTP and the size of the infarct associated with the transient global ischemia. These data suggest that apomorphine is neuroprotective, probably by means of

Excellent response to apomorphine in Parkinsonism with optic atrophy unresponsive to oral antiparkinsonian medication.

Two patients presented with the cardinal symptoms of Parkinson's disease (PD), which had developed slowly and progressively. One patient underwent brain imaging of the dopaminergic system, the results of which were compatible with PD. However, both patients showed no response to L-dopa or oral dopaminergic medication. They also had optic nerve atrophy of unknown etiology. The severe motor symptoms of both patients were conspicuously alleviated by apomorphine delivered subcutaneously. One patient underwent bilateral subthalamic nucleus deep brain stimulation, which also proved effective.

Differential effects of 7-OH-DPAT on the development of behavioral sensitization to apomorphine and cocaine.

The primary objective of this study was to determine whether concurrent treatments with a low dose of the dopamine D(3)-preferring receptor agonist 7-OH-DPAT would attenuate the development of behavioral sensitization to the indirect dopamine receptor agonist, cocaine, or the direct dopamine receptor agonist, apomorphine. In two experiments, male Wistar rats (250-350 g) were given seven daily injections of 7-OH-DPAT (0.05 mg/kg sc) or vehicle in combination with either cocaine (15 mg/kg ip), apomorphine (1.0 mg/kg sc), or vehicle. Systemic administration of apomorphine improves the hemodynamic mechanism of clitoral and vaginal engorgement in the rabbit.

Clitoral and vaginal engorgement during sexual stimulation depend in part on the increase of arterial inflow. It has been shown that apomorphine (APO), a non-selective dopamine receptor agonist, produces penile erection by activating dopaminergic receptors in the central nervous system. Our aim was to study whether systemic administration of APO improves the hemodynamic mechanism of clitoral and vaginal engorgement in the rabbit. Female New Zealand white rabbits (3.5-4 kg, n=6) were anesthetized. To examine sexual arousal function, the vaginal/clitoral branch of the pelvic nerve was stimulated
