Cambarus (C.) hatfieldi, a new species of crayfish (Decapoda:Cambaridae) from the Tug Fork River Basin of Kentucky, Virginia and West Virginia, USA.

Cambarus (Cambarus) hatfieldi is a stream-dwelling crayfish that appears to be endemic to the Tug Fork River system of West Virginia, Virginia, and Kentucky. Within this region, it is prevalent in all major tributaries in the basin as well as the Tug Fork River's mainstem. The new species is morphologically most similar to Cambarus sciotensis and Cambarus angularis. It can be differentiated from C. sciotensis by its squamous, subtrinagular chelae compared to the elongate triangular chelae of C. sciotensis; its shorter palm length/palm depth ratio (1.9) compared to C. sciotensis (2.3); and a sm

Detection of Aphanomyces astaci in North American crayfish by polymerase chain reaction.

We present a PCR based method to detect Aphanomyces astaci in North American crayfish. Primers were designed to specifically amplify parts of the internal transcribed spacer (ITS) regions and the 5.8 rRNA gene of A. astaci. A single round and a semi-nested assay were tested for their sensitivity and specificity. Specificity of the PCR assays was tested against several closely related Aphanomyces species, other Oomycetes and some non-A. astaci DNA that might be found in or on crayfish. The single round assay was fully specific against all DNA tested. In the semi-nested assay, cross-reaction was

Factors affecting food chain transfer of mercury in the vicinity of the Nyanza Site, Sudbury River, Massachusetts.

The influence of the Nyanza Chemical Waste Dump Superfund Site on the Sudbury River, Massachusetts, was assessed by analysis of sediment, fish prey organisms, and predator fish from four locations in the river system. Whitehall Reservoir is an impoundment upstream of the site, and Reservoir #2 is an impoundment downstream of the site. Cedar Street is a flowing reach upstream of the site, and Sherman Bridge is a flowing reach downstream of the site. Collections of material for analysis were made three times, in May, July, and October. Sediment was analyzed for acid-volatile sulfide (AVS), simul

[The biology of Paragonimus westermani ichunensis in the Maritime Territory. Experimental research].

It has been experimentally found that larval and pulmonary paragonimiasis types are caused by the trematode Paragonimus westermani ichunensis. The development of either paragonimiasis in nontypical definitive hosts depends on the stage of larval development when they penetrate into the mammal. Only the P. westermani ichunensis trematode is likely to develop with the participation of Juga mollusks on the territory of the Primorye.

Dependence of intracellular free calcium and tension on membrane potential and intracellular pH in single crayfish muscle fibres.

The dependence of intracellular free calcium ([Ca2+]i) and tension on membrane potential and intracellular pH (pHi) was studied in single isolated fibres of the crayfish claw-opener muscle using ion-selective microelectrodes. Tension (T) was quantified as a percentage of the maximum force, or as force per cross-sectional area (N/cm2). In resting fibres, pHi had a mean value of 7.06. Contractions evoked by an increase extracellular potassium [( K+]0) produced a fall in pHi of 0.01-0.05 units. The lowest measured levels of resting [Ca2+]i corresponded to a pCai (= -log [Ca2+]i) of 6.8. Intracell

Evaluation of the new role of the invasive species (Procambarus clarkii) as an environmental indicator in polluted watersheds.

Procambarus clarkii represents an invasive species of crayfish common in the rivers of the Iberian Peninsula, which replaced a previous invasive species of crayfish (Austropotamobius pallipes) and is now being replaced by a new invasive species (Pacifastacus leniusculus) in northern rivers. Leaving aside the ecological problems that these invasive species can cause, these organisms can be excellent candidates to act as environmental indicators in pollution processes that involves the interface water-sediment. In this work, the potential role of Procambarus clarkii as a bioindicator of pollutio

Population Pharmacokinetics of Florfenicol in Crayfish (Procambarus clarkii) Based on the Sparse Sampling Method and a Nonlinear Mixed-Effect Model.

The present study was conducted to establish population pharmacokinetics (PPK) of florfenicol (FLO) in crayfish (Procambarus clarkii) after a single oral administration at a dose of 15 mg/kg at 25°C based on a nonlinear mixed effect model. The sparse sampling method was used to collect the blood samples. Thirty-two crayfish were divided into four groups, and one group included four male crayfish and four female crayfish. One animal undertook three sampling time points. All samples were quantified using high-performance liquid chromatography with an ultraviolet detector. The initial pharmacokin

Elements in Invasive Redclaw Crayfish Cherax quadricarinatus Pose Human Health Risks in the Largest Floodplain System of South Africa.

The Australian redclaw crayfish, Cherax quadricarinatus, has been introduced globally for aquacultural purposes, and invasive wild populations have established in several countries. An increase in population growth, has caused several low-income and rural communities to utilise it as an inexpensive protein source. Despite this introduction, limited research has been done on element accumulation, and the risks for human consumption. This study focused on elemental accumulation in C. quadricarinatus from two rivers in the Phongolo River Floodplain (PRF), South Africa. The non-carcinogenic and ca

Enrofloxacin-induced transfer of multiple-antibiotic resistance genes and emergence of novel resistant bacteria in red swamp crayfish guts and pond sediments.

Antibiotic resistance genes (ARGs) can be transferred from environmental microbes to human pathogens, thus leading to bacterial infection treatment failures. The aquaculture polluted by over-used antibiotics is considered as a notorious reservoir of ARGs. However, the origin, diachronic changes, and mobility of ARGs under antibiotic exposure in aquaculture systems remain elusive. Our findings showed that enrofloxacin application also increased the relative abundance of various ARGs in addition to quinolone-resistance genes and induced ARG dissemination in crayfish gut and sediment bacteria. The response of phenoloxidase to cadmium-disturbed hepatopancreatic immune-related molecules in freshwater crayfish Procambarus clarkii.

There has been extensive research on local and systemic oxidative stress and immunosuppression in cadmium exposed crustaceans, but the underlying mechanisms remain to be elucidated. Because of multiple functions of epithelial cells, such as storing and detoxifying heavy metals, producing and secreting immune-related molecules (i.e. hemocyanin, NF-κB and CBS/H2S et al.), hepatopancreas may play an important role in immune system. In the present study, as an indication of systemic and local immune status in crayfish Procambarus clarkii, the relationship between PO activities in haemolymph and le

A tentative new species Aphanomyces fennicus sp. nov. interferes with molecular diagnostic methods for crayfish plague.

Several isolates of an unknown oomycete resembling the genus Aphanomyces were obtained into laboratory culture from samples of noble crayfish (Astacus astacus) in 2016-2017. The crayfish were kept in cages in connection with a study on an eventually persistent crayfish plague infection in a small Finnish lake, following an acute episode of the disease in 2010. Despite the close resemblance of the isolates to the causative agent of crayfish plague, Aphanomyces astaci, and the positive results obtained in OIE recommended A. astaci-specific ITS-based conventional PCR and qPCR molecular assays, th

Characterization of locomotor response to psychostimulants in the parthenogenetic marbled crayfish (Procambarus fallax forma virginalis): A promising model for studying the neural and molecular mechanisms of drug addiction.

Although scientific research using mammalian models has made great strides in uncovering the enigmatic neural and molecular mechanisms orchestrating the state of drug addiction, a complete understanding has thus far eluded researchers. The complexity of the task has led to the use of invertebrate model systems to complement the research of drug-induced reward in mammalian systems. Invertebrates, such as crayfish, offer excellent model systems to help reveal the underlying mechanisms of drug addiction as they retain the ancestral neural reward circuit that is evolutionarily conserved across tax

Optimization of the incubation time and temperature for spermatozoa extraction in freshwater crayfish Pontastacus leptodactylus (Eschscholtz, 1823).

Determination and control of spermatozoa quality in crustacean aquaculture is an important issue for successful and controlled reproduction. Investigation of spermatozoa number in spermatophores is a basic and common parameter for determining the reproductive quality in farmed decapods. In the present study, spermatozoa extraction from spermatophores located in the ductus deferens was conducted in Pontastacus leptodactylus using different incubation times and temperatures. The results indicate that the duration of incubation and temperature affected (P < 0.05) spermatozoa extraction. Greater t

Detection of Listeria in crawfish processing plants and in raw, whole crawfish and processed crawfish (Procambarus spp.).

The foodborne pathogen Listeria monocytogenes represents a major concern to the food industry and particularly to producers of ready-to-eat (RTE) foods because of the severity of human listeriosis infections and because of the ubiquitous nature of this organism. Although several studies on the prevalence and sources of L monocytogenes in various RTE seafoods have been conducted, limited information is available on the presence and potential sources of this organism in RTE crawfish products. We thus monitored the presence of L monocytogenes and other Listeria spp. in the processing environment,

Effects of black widow spider venom and latrocrustatoxin on crustacean nerve cells: electrophysiological and ultrastructural study.

1. Black widow spider venom (BWSV), or crustacean-specific toxin purified from it (named latrocrustatoxin, LCT) were lethal for the crayfish. 2. Distortions in the EMG and motor activity pattern after LCT injection indicated venom effects on the crayfish nervous system. 3. In the isolated ventral nerve cord, transient activation of the "silent" interneurons and motoneurons occurred after BWSV or LCT application. 4. In the isolated nerve-muscle preparation, LCT application elicited an intense transmitter release resulting in the complete blockade of neuromuscular transmission. 5. Electron micro

Local anaesthetics potentiate GABA-mediated Cl- currents by inhibiting GABA uptake.

A two-electrode voltage clamp was used to examine the effects of the local anaesthetics (LAs) lidocaine, tetracaine and bupivacaine, and the meta-isomer of lidocaine (LL33), on the gamma-aminobutyric acid (GABA)-gated Cl- channel in the crayfish stretch receptor neurone. The voltage-induced current changes were recorded while exposing the neurone to GABA in saline containing different LAs. All LAs enhanced the voltage-induced Cl- current equivalent to a shift of the conductance vs GABA curve towards lower concentrations. The mechanism of the facilitation of GABA-induced conductance was found t

Fictive locomotion in the fourth thoracic ganglion of the crayfish, Procambarus clarkii.

Bath application of muscarinic agonists induced rhythmic motor activity in an in vitro preparation of the thoracic nervous system of the crayfish, Procambarus clarkii. In 70% of the cases, the rhythm was organized into 1 of the 2 normal patterns: "backward" walking or "forward" walking. In the rest (30%), the ganglion produced either a series of bursts of impulses or no rhythm at all, just an increase in the tonic activity. When it was isolated from all ascending and descending afferents, the fourth thoracic ganglion was still able to generate rhythmic motor output during bath application of m

Feedforward afferent excitation of peripheral inhibitors in the crayfish escape system.

1. Each abdominal ganglion of the crayfish contains peripheral inhibitors of the fast flexor muscles. These flexor inhibitors (FIs), which can effectively inhibit tension development in the tailflip powerstroke muscles, are excited by a delayed central pathway from the same giant axons which trigger escape (33). The FIs also received sensory input, which increases in efficacy in the more posterior segments (4), but until now neither the origin of the input nor its central pathways had been well described. We have used intracellular recording and staining techniques to investigate the afferent
