Age-related changes in mitochondrial antioxidant enzyme Trx2 and TXNIP-Trx2-ASK1 signal pathways in the auditory cortex of a mimetic aging rat model: changes to Trx2 in the auditory cortex.

Age-associated degeneration in the central auditory system, which is defined as central presbycusis, can impair sound localization and speech perception. Research has shown that oxidative stress plays a central role in the pathological process of central presbycusis. Thioredoxin 2 (Trx2), one member of thioredoxin family, plays a key role in regulating the homeostasis of cellular reactive oxygen species and anti-apoptosis. The purpose of this study was to explore the association between Trx2 and the phenotype of central presbycusis using a mimetic aging animal model induced by long-term exposu

Increased functional connectivity between the auditory cortex and the frontoparietal network compensates for impaired visuomotor transformation after early auditory deprivation.

Early auditory deprivation leads to a reorganization of large-scale brain networks involving and extending beyond the auditory system. It has been documented that visuomotor transformation is impaired after early deafness, associated with a hyper-crosstalk between the task-critical frontoparietal network and the default-mode network. However, it remains unknown whether and how the reorganized large-scale brain networks involving the auditory cortex contribute to impaired visuomotor transformation after early deafness. Here, we asked deaf and early hard of hearing participants and normal hearin

Cross-Modal Plasticity in Higher-Order Auditory Cortex of Congenitally Deaf Cats Does Not Limit Auditory Responsiveness to Cochlear Implants.

UNLABELLED: Congenital sensory deprivation can lead to reorganization of the deprived cortical regions by another sensory system. Such cross-modal reorganization may either compete with or complement the "original" inputs to the deprived area after sensory restoration and can thus be either adverse or beneficial for sensory restoration. In congenital deafness, a previous inactivation study documented that supranormal visual behavior was mediated by higher-order auditory fields in congenitally deaf cats (CDCs). However, both the auditory responsiveness of "deaf" higher-order fields and interact

Transcranial direct current stimulation of posterior temporal cortex modulates electrophysiological correlates of auditory selective spatial attention in posterior parietal cortex.

Speech perception in "cocktail-party" situations, in which a sound source of interest has to be extracted out of multiple irrelevant sounds, poses a remarkable challenge to the human auditory system. Studies on structural and electrophysiological correlates of auditory selective spatial attention revealed critical roles of the posterior temporal cortex and the N2 event-related potential (ERP) component in the underlying processes. Here, we explored effects of transcranial direct current stimulation (tDCS) to posterior temporal cortex on neurophysiological correlates of auditory selective spati

Neural mechanisms of fear memory precision and generalization: from auditory cortex to amygdala.

Identifying neural networks that infer safety versus danger is of great interest across multiple disciplines. Recent research has started to elucidate the brain networks mediating fear discrimination and generalization. Here, we focus on the contribution of the auditory cortices to auditory fear memory and discrimination processes. We first review studies showing that the primary auditory cortex (Te1) is involved in fear memory for tones with complex perceptual features only. Conversely, Te1 is essential for fear discrimination across all types of acoustic stimuli. These studies support the id

Chronic nicotine reverses age-related decline of interneuron markers in mouse auditory cortex.

Inhibition in central auditory pathways declines in old age, which contributes to age-related auditory processing deficits. In auditory cortex, most inhibitory interneurons can be identified by one of three marker proteins: parvalbumin (PV), somatostatin (SOM), or vasoactive intestinal peptide (VIP). About 10% of interneurons contain the α2 nicotinic acetylcholine receptor (α2 cells), and most of these are layer 5 Martinotti cells (α2-MCs). We examined the distribution of these four interneuron subtypes in auditory cortex of FVB mice, from young adult through old age, and the extent to which α

Hippocampal and amygdalar structures preferentially target somatostatin-expressing interneurons in the auditory cortex.

The auditory cortex integrates sensory and contextual inputs in emotion and fear driven situations that can be consolidated over learning. Somatostatin-expressing (SST+) interneurons play a key role in inhibitory modulation of sensory processing, yet their long-range inputs remain poorly characterised. We performed rabies-based retrograde monosynaptic tracing to compare the specific inputs to SST+ interneurons with the general input connectome of neurons in the mouse auditory cortex. Overall, inputs were similar, with the majority of presynaptic neurons originating from the ipsilateral auditor

Disturbing the sound of silence: Bilateral temporal cortex stimulation and auditory mental imagery.

Auditory imagery depends on temporal-cortical mechanisms that generate and sustain internal sound representations. If these mechanisms are causally involved, externally perturbing temporal cortex should alter the quality of imagery. We tested whether bilateral high-frequency transcranial random noise stimulation (hf-tRNS) over temporal cortex alters the vividness and control of auditory imagery. Forty-nine healthy adults completed two sessions on separate days, receiving Active hf-tRNS in one session and Sham in the other (order counterbalanced). The Bucknell Auditory Imagery Scale (BAIS; Vivi

Music can be reconstructed from human auditory cortex activity using nonlinear decoding models.

Music is core to human experience, yet the precise neural dynamics underlying music perception remain unknown. We analyzed a unique intracranial electroencephalography (iEEG) dataset of 29 patients who listened to a Pink Floyd song and applied a stimulus reconstruction approach previously used in the speech domain. We successfully reconstructed a recognizable song from direct neural recordings and quantified the impact of different factors on decoding accuracy. Combining encoding and decoding analyses, we found a right-hemisphere dominance for music perception with a primary role of the superi

Focal tDCS of auditory cortex in chronic tinnitus: A randomized controlled mechanistic trial.

OBJECTIVE: The goal of this pilot study was to understand how focal transcranial direct current stimulation (tDCS) targeting auditory cortex changes brain function in chronic tinnitus using magnetic resonance imaging (MRI). METHODS: People with chronic tinnitus were randomized to active or sham tDCS on five consecutive days in this mechanistic trial (n = 10/group). Focal 4x1 tDCS (central anode, surround cathodes) targeted left auditory cortex, with single-blind 2 mA current during twenty-minute sessions. Arterial spin-labeled and blood oxygenation level dependent MRI occurred immediately bef

Electrocorticography reveals continuous auditory and visual speech tracking in temporal and occipital cortex.

During natural speech perception, humans must parse temporally continuous auditory and visual speech signals into sequences of words. However, most studies of speech perception present only single words or syllables. We used electrocorticography (subdural electrodes implanted on the brains of epileptic patients) to investigate the neural mechanisms for processing continuous audiovisual speech signals consisting of individual sentences. Using partial correlation analysis, we found that posterior superior temporal gyrus (pSTG) and medial occipital cortex tracked both the auditory and the visual

Selective Neuronal Activation by Cochlear Implant Stimulation in Auditory Cortex of Awake Primate.

UNLABELLED: Despite the success of cochlear implants (CIs) in human populations, most users perform poorly in noisy environments and music and tonal language perception. How CI devices engage the brain at the single neuron level has remained largely unknown, in particular in the primate brain. By comparing neuronal responses with acoustic and CI stimulation in marmoset monkeys unilaterally implanted with a CI electrode array, we discovered that CI stimulation was surprisingly ineffective at activating many neurons in auditory cortex, particularly in the hemisphere ipsilateral to the CI. Furthe

Spatial pattern of BOLD fMRI activation reveals cross-modal information in auditory cortex.

Recent findings suggest that neural representations in early auditory cortex reflect not only the physical properties of a stimulus, but also high-level, top-down, and even cross-modal information. However, the nature of cross-modal information in auditory cortex remains poorly understood. Here, we used pattern analyses of fMRI data to ask whether early auditory cortex contains information about the visual environment. Our data show that 1) early auditory cortex contained information about a visual stimulus when there was no bottom-up auditory signal, and that 2) no influence of visual stimula

Auditory cortex asymmetry, altered minicolumn spacing and absence of ageing effects in schizophrenia.

The superior temporal gyrus, which contains the auditory cortex, including the planum temporale, is the most consistently altered neocortical structure in schizophrenia (Shenton ME, Dickey CC, Frumin M, McCarley RW. A review of MRI findings in schizophrenia. Schizophr Res 2001; 49: 1-52). Auditory hallucinations are associated with abnormalities in this region and activation in Heschl's gyrus. Our review of 34 MRI and 5 post-mortem studies of planum temporale reveals that half of those measuring region size reported a change in schizophrenia, usually consistent with a reduction in the left hem

Inhibition of voltage-gated channel currents in rat auditory cortex neurons by salicylate.

Salicylate is a medicine for anti-inflammation with a side effect of tinnitus. To understand the mechanisms of tinnitus induced by salicylate, we studied the effects of salicylate on voltage-gated ion channels and action potential firing rates in freshly dissociated rat pyramidal neurons in auditory cortex (AC) using the whole-cell patch technique. We found that salicylate reduced the voltage-gated sodium current (I(Na)), the delayed rectifier potassium current (I(K(DR))) and the L-type voltage-gated calcium current (I(Ca,L)) in concentration-dependent manner. An amount of 1mM salicylate shift

Image-to-sound conversion: experience-induced plasticity in auditory cortex of blindfolded adults.

The ability to adapt to environmental changes is based on the impressive capacity of the central nervous system for plasticity changes. A better understanding of the requirements of neuroplasticity will help to apprehend and predict the success of sensory prostheses. To investigate neuroplastic changes associated with (1) blindfolding and (2) the use of a mobile visual-auditory substitution system, five normally sighted adults underwent weekly measurements of neuromagnetic activity using a 122-channel whole head neuromagnetometer. The substitution device converted visual images into sound patt

When vision listens: auditory predictive processing in primary visual cortex of anesthetized rats.

Primary visual cortex (V1), traditionally considered a purely visual area, is increasingly recognized for its role in multisensory processing. According to predictive coding theory, cortical circuits generate expectations about sensory inputs (predictions) and signal deviations from them (prediction errors). To test whether V1 encodes auditory prediction errors, we recorded neuronal activity and LFP from layers IV-VI in urethane-anesthetized rats during auditory oddball and no-repetition control sequences. Recorded neurons from layers V-VI in our sample responded to tones, with many showing st

Left inferior parietal lobe and auditory cortex jointly contribute to sound knowledge retrieval.

BACKGROUND: Conceptual knowledge is central to human cognition. Neuroimaging studies suggest that conceptual processing relies on the joint contribution of modality-specific perceptual-motor and multimodal brain regions. In particular, the multimodal left inferior parietal lobe (IPL) couples with auditory association cortex during sound knowledge retrieval and with somatomotor cortex during action feature retrieval. However, it remains unknown whether the interaction between modality-specific and multimodal cortices is causally relevant for conceptually-guided behavior. METHODS: To tackle thi
