Advanced aptamer-based sensors for monitoring theophylline.

Theophylline is a widely used bronchodilator for treating asthma-related symptoms like shortness of breath and chest tightness. However, its narrow therapeutic plasma range (20-200 μM) necessitates careful monitoring of blood levels to prevent toxicity. Various clinical laboratory techniques have been developed for detecting theophylline, including spectroscopy, high-performance liquid chromatography (HPLC), fluorescence polarization immunoassay, and radioimmunoassay. Despite their utility, these methods are limited by complex sample preparation, long processing times, large sample volumes, an

Pathophysiological Associations and Measurement Techniques of Red Blood Cell Deformability.

Red blood cell (RBC), accounting for approximately 45% of total blood volume, are essential for oxygen delivery and carbon dioxide removal. Their unique biconcave morphology, high surface area-to-volume ratio, and remarkable deformability enable them to navigate microvessels narrower than their resting diameter, ensuring efficient microcirculation. RBC deformability is primarily determined by membrane viscoelasticity, cytoplasmic viscosity, and cell geometry, all of which can be altered under various physiological and pathological conditions. Reduced deformability is a hallmark of numerous dis

Fabrication of a Solution-gated Indium-Tin-Oxide-based One-piece Transistor Enabling Sensitive Biosensing.

A simple and rapid method to fabricate a solution-gated one-piece indium-tin-oxide (ITO)-based ion-sensitive field-effect transistor (ISFET) (ITO-ISFET) for biosensing is presented in this report. Although ITO is a conductive oxide, it exhibits semiconducting properties at a depletion layer thickness of approximately 20 nm or less, making it suitable as the channel of a pH-sensitive solution-gated ISFET. Specifically, by etching the middle part of a conductive ITO single thin film with an acidic solution to make it ultrathin, a one-piece ITO-ISFET with a semiconductive channel can be fabricate

Rapid identity testing of antibody-based hot targeted radionuclide therapies by bio-layer interferometry.

Targeted Radionuclide Therapies (TRT) involve the tailored combination of a therapeutic radionuclide and a targeting molecule, as for instance antibodies or fragments thereof. Despite their short shelf-life, these drug products must meet stringent regulatory standards before use. We introduce a novel, efficient method utilizing Bio-Layer Interferometry (BLI) for rapid identity testing of TRT drug products in less than five minutes. This approach not only reduces radioactive waste but also minimizes operator exposure to radiation. This label-free method has been successfully developed and valid

Multi-Degree-of-Freedom Stretchable Metasurface Terahertz Sensor for Trace Cinnamoylglycine Detection.

Terahertz (THz) spectroscopy, an advanced label-free sensing method, offers significant potential for biomolecular detection and quantitative analysis in biological samples. Although broadband fingerprint enhancement compensates for limitations in detection capability and sensitivity, the complex optical path design in operation restricts its broader adoption. This paper proposes a multi-degree-of-freedom stretchable metasurface that supports magnetic dipole resonance to enhance the broadband THz fingerprint detection of trace analytes. The metasurface substrate and unit cell structures are co

Protonation-mediated DNA tile self-assembly with nuclease resistance characteristic for signal-amplified detection of microRNAs.

DNA nanotechnology, developing rapidly in recent years, has unprecedented superiorities in biological application-oriented research including high programmability, convenient functionalization, reconfigurable structure, and intrinsic biocompatibility. However, the susceptibility to nucleases in the physiological environment has been an obstacle to applying DNA nanostructures in biological science research. In this study, a new DNA self-assembly strategy, mediated by double-protonated small molecules instead of classical metal ions, is developed to enhance the nuclease resistance of DNA nanostr

Scalable Functionalization of Polyaniline-Grafted rGO Field-Effect Transistors for a Highly Sensitive Enzymatic Acetylcholine Biosensor.

For decades, acetylcholine (Ach) has been considered a critical biomarker for several degenerative brain diseases, including Alzheimer's, Parkinson's disease, Huntington's disease, and schizophrenia. Here, we propose a wafer-scale fabrication of polyaniline (PAni)-grafted graphene-based field-effect transistors (PGFET) and their biosensing applications for highly sensitive and reliable real-time monitoring of Ach in flow configuration. The grafted PAni provides suitable electrostatic binding sites for enzyme immobilization and enhances the pH sensitivity (2.68%/pH), compared to that of bare gr

Rapid bacteria-detection platform based on magnetophoretic concentration, dielectrophoretic separation, and impedimetric detection.

Most biosensors employ small sample quantities (less than 100 μL) for bacteria detection, thereby resulting in inaccurate low-concentration measurements. Detection performed using small sample volumes with low bacteria concentration may produce false-negative results. Therefore, sample pretreatment plays a critical role in accurate bacteria detection. This paper presents an impedimetric bacteria-detection sensor integrated with bacteria concentration and separation devices for rapid bacteria detection. Post conjugation using magnetic particles (MPs), the MP-conjugated bacteria (MP/Bac) are con

Sensitive and selective methanol biosensor using two-enzyme cascade reaction and fluorometry for non-invasive assessment of intestinal bacteria activity.

For understanding the status of intestinal flora non-invasively, methanol (MeOH) has been attracting the attention. In this study, we have developed and compared two different liquid-phase methanol biosensors. One, referred to as the AOD electrosensor, utilized alcohol oxidase (AOD) and an oxygen electrode. It electrochemically measured the decrease in oxygen through AOD-catalyzed oxidation of MeOH. The other, referred to as the AOD-FALDH fluorosensor, exploited a cascade reaction of AOD and formaldehyde dehydrogenase (FALDH) in conjunction with a fiber-optic sensor. It measured increase in th

Semi-circular sweep voltammetry. Bio-analytical applications.

A novel voltammetric method applying a semi-circular potential sweep is applied to the simultaneous electroanalytical determination of solutions containing two components with similar oxidation potentials which precludes their resolution using conventional voltammetric methods including linear sweep, square wave and pulse voltammetries. Three such biologically important mixtures, ascorbic acid/acetaminophen, glucose/ethanol and hydroquinone/catechol were studied, analytical methods developed and the method of semi-circular sweep voltammetry shown to give notable advantages over the other conve

Biosensors on the road to early diagnostic and surveillance of Alzheimer's disease.

Alzheimer's disease is a debilitating and largely untreatable condition with subtle onset and slow progression over an extensive period of time, which culminate in increasing levels of disability. As Alzheimer's disease prevalence is expected to grow exponentially in the upcoming decades, there is an urgency to develop analytical technologies for the sensitive, reliable and cost-effective detection of Alzheimer's disease biomarkers. Biosensors are powerful analytical devices that translate events of biological recognition on physical or chemical transducers into electrical, thermal or optical

Biosensors for Non-Invasive Detection of Celiac Disease Biomarkers in Body Fluids.

Celiac disease is a chronic gluten-initiated autoimmune disorder that predominantly damages the mucosa of the small intestine in genetically-susceptible individuals. It affects a large and increasing number of the world’s population. The diagnosis of this disease and monitoring the response of patients to the therapy, which is currently a life-long gluten-free diet, require the application of reliable, rapid, sensitive, selective, simple, and cost-effective analytical tools. Celiac disease biomarker detection in full blood, serum, or plasma offers a non-invasive way to do this and is wel

Electrochemical impedance spectroscopy in label-free biosensor applications: multivariate data analysis for an objective interpretation.

Electrochemical impedance spectroscopy plays an important role in biosensor science thanks to the possibility of finding specific information from processes with different kinetics at a chosen electrode potential in one experiment. In this paper we briefly discuss label-free impedimetric biosensors described in the literature. A novel method for neutral interpretation of impedance data is presented that includes complex number chemometrics. Three examples are given based on impedance measurements on synthetic biomembranes, in this case a lipid monolayer deposited on a mercury electrode. The in
