Migration of Bisphenol A and Its Derivatives From Epoxy Coatings and Demand for BPA-NI Products: Scientific Insights and Perspectives Leading to Regulation (EU) 2024/3190.

Bisphenol A (BPA) is a widely used industrial chemical used in the production of polycarbonate and epoxy resins; however, it is also recognized as an endocrine-disrupting chemical. This review examines scientific evidence on the migration of bisphenol A and its derivatives from epoxy can coatings into food products, highlighting the factors that have prompted recent regulatory shifts. The newly enacted Commission Regulation (EU) 2024/3190 addresses health concerns by promoting the use of BPA-Non-Intent (BPA-NI) materials, which are formulated to avoid intentional BPA use and minimize contamina

Increased gut serotonin production in response to bisphenol A structural analogs may contribute to their obesogenic effects.

Obesogens are synthetic, environmental chemicals that can disrupt endocrine control of metabolism and contribute to the risk of obesity and metabolic disease. Bisphenol A (BPA) is one of the most studied obesogens. There is considerable evidence that BPA exposure is associated with weight gain, increased adiposity, poor blood glucose control, and nonalcoholic fatty liver disease in animal models and human populations. Increased usage of structural analogs of BPA has occurred in response to legislation banning their use in some commercial products. However, BPA analogs may also cause some of th

Lipidomic biomarkers: Potential mediators of associations between urinary bisphenol A exposure and colorectal cancer.

Previous research reported associations between bisphenol A (BPA) exposure and some malignant tumor incidences, yet the underlying mechanism remains largely uncertain. This investigation was aimed to explore the association of BPA exposure burden with colorectal cancer (CRC) and the role of tumor tissue lipid metabolism in the associations between BPA and CRC using lipidomic approach. Urinary BPA levels in CRC cases were significantly higher than those in controls (P value < 0.05). BPA was positively correlated with all three serum CRC biomarkers, with an estimated odds ratio (OR) of 4.45 (95%

Chronic exposure to low doses of bisphenol A alters hydromineral responses in rats.

Bisphenol A (BPA) is a chemical commonly used in the industrial sectors, hence humans are exposed to the compound repetitively. BPA is an endocrine disruptor and has been anticipated to interfere on chemical estrogen receptor functions and other nuclear hormone receptors. Estrogens are steroid hormones that, in addition to their neuroendocrine roles, affect water and salt intakes in numerous species, including humans and rodents. Changes in the hydrosaline balance produce compensatory behavioral and physiological responses, which serve to preserve or restore osmolarity and blood volume to opti

Cardiovascular Effects and Molecular Mechanisms of Bisphenol A and Its Metabolite MBP in Zebrafish.

The plastic monomer bisphenol A (BPA) is one of the highest production volume chemicals in the world and is frequently detected in wildlife and humans, particularly children. BPA has been associated with numerous adverse health outcomes relating to its estrogenic and other hormonal properties, but direct causal links are unclear in humans and animal models. Here we simulated measured (1×) and predicted worst-case (10× ) maximum fetal exposures for BPA, or equivalent concentrations of its metabolite MBP, using fluorescent reporter embryo-larval zebrafish, capable of quantifying Estrogen Respons

Bisphenol A degradation by Lactiplantibacillus plantarum AM employing Fenton chemistry.

Lactiplantibacillus plantarum, Gram-positive facultative anaerobic strain with antioxidant potential, can degrade and detoxify xenobiotic compounds. In this study, we evaluated Lpb. plantarum AM for its antioxidant properties, bisphenol A (BPA) degradation, and Lpb. plantarum AM mediated-Fenton reaction for BPA degradation. ≤ 43.8 µmol BPA did not affect the growth of Lpb. plantarum AM even in glucose-deficient MRS medium. BPA at ≥ 52.56 µmol significantly inhibited growth (p < 0.001) of Lpb. plantarum AM and delayed transition from the logarithmic (log) phase to the stationary phase. When ≥ 5

Nature-Derived Epoxy Resin Monomers with Reduced Sensitizing Capacity─Isosorbide-Based Bis-Epoxides.

Epoxy resin systems (ERSs) are a class of thermosetting resins that become thermostable and insoluble polymers upon curing. They are widely used as components of protective surfaces, adhesives, and paints and in the manufacturing of composites in the plastics industry. The diglycidyl ether of bisphenol A (DGEBA) is used in 75-90% of ERSs and is thus by far the most used epoxy resin monomer (ERM). Unfortunately, DGEBA is a strong skin sensitizer and it is one of the most common causes of occupational contact dermatitis. Furthermore, DGEBA is synthesized from bisphenol A (BPA), which is a petrol

Exploring the Effects of Metabolism-Disrupting Chemicals on Pancreatic α-Cell Viability, Gene Expression and Function: A Screening Testing Approach.

Humans are constantly exposed to many environmental pollutants, some of which have been largely acknowledged as key factors in the development of metabolic disorders such as diabetes and obesity. These chemicals have been classified as endocrine-disrupting chemicals (EDCs) and, more recently, since they can interfere with metabolic functions, they have been renamed as metabolism-disrupting chemicals (MDCs). MDCs are present in many consumer products, including food packaging, personal care products, plastic bottles and containers, and detergents. The scientific literature has ever-increasingly

BPA and its alternatives BPF and BPAF exaggerate hepatic lipid metabolism disorders in male mice fed a high fat diet.

Alternatives to Bisphenol A (BPA), such as BPF and BPAF, have found increasing industrial applications. However, toxicological research on these BPA analogues remains limited. This study aimed to investigate the effects of BPA, BPF, and BPAF exposure on hepatotoxicity in mice fed with high-fat diets (HFD). Male mice were exposed to the bisphenols at a dose of 0.05 mg per kg body weight per day (mg/kg bw/day) for eight consecutive weeks, or 5 mg/kg bw/day for the first week followed by 0.05 mg/kg bw/day for seven weeks under HFD. The low dose (0.05 mg/kg bw/day) was corresponding to the tolerab

Platelet mitochondrial DNA methylation mediates the association of bisphenol, phthalate, and paraben exposures with type 2 diabetes mellitus: An exploratory nested case-control study.

Exposure to environmental pollutants has been found to be associated with epigenetic modifications of platelet mitochondria, which may influence the risk of type 2 diabetes mellitus (T2DM). However, research on the relationship between exposure to environmental endocrine disrupting chemicals (EDCs) and T2DM remains very limited at the molecular level of mitochondrial epigenetic alterations. This study aims to investigate the impact of mixed exposure to bisphenols (BPs), phthalates (PAEs), and parabens (PBs) on T2DM and platelet mitochondrial DNA (mtDNA) methylation, using a nested case-control

Parabens, bisphenols, and other environmental phenols exposure from cosmetics use in Korean adolescent girls: Findings from a 2-day intervention study.

Endocrine-disrupting chemicals (EDCs), such as bisphenols, parabens, and benzophenones, are commonly found in personal care products (PCPs) and pose a significant public health concern, particularly in adolescent girls. This study aimed to investigate the association between PCP use and urinary concentrations of environmental phenols in Korean adolescent girls, focusing on the potential role of PCPs as a major source of EDC exposure. A total of 112 female adolescents (aged 13-17 years) were recruited to examine the associations between urinary concentrations of parabens, bisphenols, benzopheno

Evaluating the potential of foodborne chemicals to modulate intestinal barrier function: a combined in vitro/in silico toolbox to study the effects on tight junction protein claudin-4.

Intestinal barrier integrity is of paramount importance for our health. Several food constituents and contaminants have been described for positive or detrimental effects, however, a correlation between gut associated disorders and xenobiotics exposures remains challenging and modeling options limited. This study presents a NAMs-based hybrid in vitro/in silico workflow to investigate, and eventually prioritize, foodborne xenobiotics for their effect on gut barrier homeostasis. Model compounds comprised endocrine-active chemicals, such as plastic components (bisphenol A, bisphenol F), phytoestr

Prepubertal exposure to compounds that increase prolactin secretion in the male rat: effects on the adult prostate.

To test the hypothesis that a transient increase in prolactin (PRL) secretion prior to puberty can result in an alteration of the adult prostate, male rats were exposed from postnatal Days (PND) 22 to 32 to compounds that increase PRL secretion. These compounds included pimozide (a dopamine antagonist), estradiol-17beta, and bisphenol A (a monomer of polycarbonate plastics reported to have weak estrogenic activity). During dosing, pimozide (PIM), bisphenol A (BPA), and estradiol-17beta (E(2)) stimulated an increased secretion of PRL. At 120 days of age, the lateral prostate weight was increase

Characterisation, release and migration of phenolic compounds from resoles used in polyester-phenol coatings intended for food contact materials.

Resoles are multifarious pre-polymeric resins produced by the condensation of basic chemicals phenols, formaldehyde and optionally aliphatic alcohols like butanol. They are widely used as cross-linkers to form resistant internal coatings on metal surfaces of cans, containers or closures. Although the application of resoles is common in food contact, usually little is known about their exact composition, the toxicological hazards of their individual constituents and the migration of phenolic compounds, e.g., of the potentially endocrine-disrupting chemical bisphenol F. Our study fills major gap

Neuroactive compounds in tomatoes: metabolic fate during in vitro digestion and colonic fermentation.

Neurodegenerative diseases are hard to treat, and their progression is linked to diet and the gut microbiota. Tomatoes contain potentially neuroactive compounds, but their fate during digestion and colonic fermentation remains unclear. This study tracked neuroactive compounds and fecal fatty acids (<C12) during in vitro gastrointestinal digestion and fecal fermentation. Conditions included a fecal blank, digested inulin, digested tomato, tomato with neuro-disrupting compounds, and individual neuroprotective compounds. Targeted analyses and linear mixed-effects models showed samples were primar

PD-1/PD-L1 mediated glycolysis-driven M1 polarization of Hofbauer cells impairs placental and fetal development following maternal mixed bisphenols exposure.

Real-world gestational bisphenols (BPs) exposure typically occurs as complex mixtures and is associated with placental dysfunction as well as impaired fetal growth. However, the underlying mechanisms remain poorly defined. This study investigated how environmentally relevant mixed BPs disrupt placental immune homeostasis and fetal development. Maternal mixed BPs exposure induced placental pro-inflammatory milieu and impaired placental development, leading to fetal growth restriction (FGR) by promoting M1 polarization of Hofbauer cells (HBCs), particularly at mid-gestation. Functionally, M1 mac

Bisphenol Z at environmentally relevant dose dysregulates mitochondrial metabolism and proliferative capacity in human ovarian granulosa cells by inducing changes in metabolic substrate utilization.

The new generation of bisphenols, including bisphenol AF (BPAF), B (BPB), P (BPP) and Z (BPZ), has been detected within the environment of human ovarian follicles. It is therefore hypothesized that these bisphenols may interact directly with ovarian granulosa cells (GCs), which have high metabolic and proliferative activity that is crucial for oocyte development. Therefore, this study aimed to evaluate the effects of environmentally relevant concentrations (0.1–10 nM) of BPB, BPAF, BPP and BPZ on the mitochondrial function, metabolic fuel utilization and proliferation capacity of human GCs.
