Helicobacter hepaticus CdtB promoted brain impairment in BALB/c mice via gut-brain axis.

BACKGROUND: Helicobacter hepaticus infection has been linked to neurodegenerative diseases, but the underlying molecular mechanism is still unclear. In this study, we established an animal model of neurodegeneration via infecting BALB/c mice with wild-type H. hepaticus (WT) and CdtB-mutant (ΔCdtB) strains to investigate the influence of CdtB on the progression of cerebral injury. METHODS: BALB/c mice were infected with either WT or ΔCdtB, and then were euthanized at 6- and 12- months post of infection (MPI). By means of histopathology and molecular biology techniques, we evaluated the coloniza

Radiation-Induced Brain Injury: Mechanistic Insights and the Promise of Gut-Brain Axis Therapies.

Radiation therapy is widely recognized as an efficacious modality for treating neoplasms located within the craniofacial region. Nevertheless, this approach is not devoid of risks, predominantly concerning potential harm to the neural structures. Adverse effects may encompass focal cerebral necrosis, cognitive function compromise, cerebrovascular pathology, spinal cord injury, and detriment to the neural fibers constituting the brachial plexus. With increasing survival rates among oncology patients, evaluating post-treatment quality of life has become crucial in assessing the benefits of radia

Gut-brain axis in Alzheimer's disease: neuroinflammatory mechanisms and immunopharmacological interventions.

Despite being defined pathologically by amyloid-β plaques, chronic neuroinflammation and neurofibrillary tangles, the drugs that target these processes directly (lecanemab and donanemab) have only modest effects on the clinical course of Alzheimer's disease (AD) and significant safety concerns have been identified, so there is renewed interest in upstream, systemic contributors to disease. This review aims to provide a synthesis of the latest mechanistic and clinical evidence that gut microbiota dysbiosis is associated with Alzheimer's disease with a specifically immunopharmacological focus. Myokines, Microbiota, and Neuroinflammation: Physical Activity Modulates the Gut-Brain Axis.

BACKGROUND: Neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis are increasingly recognized as disorders influenced not only by intrinsic neural pathology but also by systemic physiological networks, including the gut-brain axis. Emerging evidence highlights physical activity as a potent modulator of this bidirectional communication system, with muscle-derived signals particularly myokines, metabolites, and extracellular vesicles playing a central role. METHODS: This narrative review synthesizes current knowledge on how exercise-induc

Exploring the gut-brain axis: dietary influences on Alzheimer's disease pathogenesis.

Alzheimer's disease (AD) is one of the most diagnosed neurodegenerative disorders worldwide and presents a significant challenge for both affected individuals and their caregivers. Alzheimer's disease is characterized by the accumulation of amyloid plaques and dysfunctional tau protein in the brain, along with the final development of dementia. Recently, in addition to the strongly developing ischemic etiology of AD, it is suggested that the gut and oral microbiota may also participate in the development of this disease. This involvement may stem from an unbalanced diet and the consumption of

The Adolescent Microbiome-Gut-Brain Axis: Development, Mechanisms, and Translational Perspectives.

BACKGROUND: Adolescence represents a pivotal, yet understudied, developmental window in which maturation of the gut microbiome, neural circuits, endocrine systems, and behavioral processes converge. Although early-life microbiota-gut-brain interactions are well established, emerging evidence demonstrates that adolescence constitutes a "second sensitive period" during which microbial composition, metabolic output, and immune function undergo substantial restructuring. These microbial transitions parallel key neurodevelopmental events, including synaptic pruning, dopaminergic refinement, and hei

The brain-bone-gut axis: a microbial bridge underlying multisystem comorbidities.

Multi-axis interactions among the skeletal system, immune system, and gut microbiota (GM) have become a prominent focus of interdisciplinary research. The brain-bone-gut axis, proposed in recent years, provides an integrative physiological framework describing a bidirectional regulatory network linking the central nervous system, bone metabolism, and the GM via neural, endocrine, and immune pathways, thereby offering a unified perspective on multi-organ comorbidities. This article systematically examines the interconnections and synergistic effects across three core pathways within this framew

Revisiting Rosacea Through the Skin-Gut-Brain Axis: A Neuroimmune Perspective.

Rosacea is increasingly recognized as a complex inflammatory disorder extending beyond isolated cutaneous pathology, involving dysregulated interactions between the skin, gastrointestinal system, and central nervous system. The skin-gut-brain axis has emerged as a relevant conceptual framework for understanding this multifactorial disease, integrating gut microbiota dysbiosis, neuroimmune signaling, autonomic nervous system dysfunction, and stress-related mechanisms. The aim of this narrative hypothesis-driven overview is to reframe rosacea as a neuroimmune disorder in which central nervous sy

Targeting the microbiota-gut-brain axis in neurodegeneration: Zebrafish-based investigations.

The microbiota-gut-brain axis (MGBA) is a bidirectional signaling pathway regulated by the gut microbiome and the central nervous system (CNS). Moreover, MGBA is crucial for normal growth, development, and physiology of the brain and gut of the host; dysfunction in MGBA has been closely linked with the development of neurological disorders. Gut dysbiosis and its metabolites modulate barrier permeability and cause gastrointestinal tract (GIT) inflammation, which is followed by an increase in pro-inflammatory cytokines, immune cell infiltration into the brain, and vagus nerve dysfunction, result

Microbiota-gut-brain axis pathogenesis and targeted therapeutics in sleep disorders.

Sleep constitutes an essential physiological process that is vital for maintaining physical and mental wellbeing. However, the science of sleep focusing on basic questions such as "how" we sleep and "why" we sleep is still not clear. Over the past decade, substantial progress has also been made in elucidating the interactions between sleep and other biological processes, providing insights into the basic questions of sleep. Among these, emerging evidence highlights the microbiota-gut-brain axis (MGBA) as a pivotal bidirectional network that connects gut microorganisms with the central nervous

The gut-brain vagal axis scales hippocampal memory processes and plasticity.

The vagus nerve serves as an interoceptive relay between the body and the brain. Despite its well-established role in feeding behaviors, energy metabolism, and cognitive functions, the intricate functional processes linking the vagus nerve to the hippocampus and its contribution to learning and memory dynamics remain still elusive. Here, we investigated whether and how the gut-brain vagal axis contributes to hippocampal learning and memory processes at behavioral, functional, cellular, and molecular levels. Our results indicate that the integrity of the vagal axis is essential for long-term re

Does gut brain axis has an impact on Parkinson's disease (PD)?

Parkinson's Disease (PD) is becoming a growing global concern by being the second most prevalent disease next to Alzheimer's Disease (AD). Henceforth new exploration is needed in search of new aspects towards the disease mechanism and origin. Evidence from recent studies has clearly stated the role of Gut Microbiota (GM) in the maintenance of the brain and as a root cause of various diseases and disorders including other neurological conditions. In the case of PD, with an unknown etiology, the GM is said to have a larger impact on the disease pathophysiology. Although GM and its metabolites ar

Targeting gut-brain axis through scalp-abdominal electroacupuncture in Parkinson's disease.

BACKGROUND: Evidence suggests that the pathogenesis of Parkinson's disease (PD) is initiated in the gut rather than in the brain. Thus, targeting the gut in early stages may have the potential to halt disease progression and alleviate symptoms. Various acupuncture techniques have been used to treat patients with PD and have shown promising results. However, previous acupuncture techniques focused on the brain and motor symptoms. We aimed to determine if targeting PD patients' gut-brain axis through electroacupuncture could be an effective, safe, and low-cost therapeutic option for management o

Neonatal Enteropathogenic Escherichia coli Infection Disrupts Microbiota-Gut-Brain Axis Signaling.

Diarrheal diseases are a leading cause of death in children under the age of 5 years worldwide. Repeated early-life exposures to diarrheal pathogens can result in comorbidities including stunted growth and cognitive deficits, suggesting an impairment in the microbiota-gut-brain (MGB) axis. Neonatal C57BL/6 mice were infected with enteropathogenic Escherichia coli (EPEC) (strain e2348/69; ΔescV [type III secretion system {T3SS} mutant]) or the vehicle (Luria-Bertani [LB] broth) via orogastric gavage at postnatal day 7 (P7). Curcumin attenuates collagen-induced inflammatory response through the "gut-brain axis".

BACKGROUND: Previous studies have demonstrated that oral administration of curcumin exhibited an anti-arthritic effect despite its poor bioavailability. The present study aimed to explore whether the gut-brain axis is involved in the therapeutic effect of curcumin. METHODS: The collagen-induced arthritis (CIA) rat model was induced by immunization with an emulsion of collagen II and complete Freund's adjuvant. Sympathetic and parasympathetic tones were measured by electrocardiographic recordings. Unilateral cervical vagotomy (VGX) was performed before the induction of CIA. The ChAT, AChE acti

Brain-Gut-Bone Marrow Axis: Implications for Hypertension and Related Therapeutics.

Hypertension is the most prevalent modifiable risk factor for cardiovascular disease and disorders directly influencing cardiovascular disease morbidity and mortality, such as diabetes mellitus, chronic kidney disease, obstructive sleep apnea, etc. Despite aggressive attempts to influence lifestyle modifications and advances in pharmacotherapeutics, a large percentage of patients still do not achieve recommended blood pressure control worldwide. Thus, we think that mechanism-based novel strategies should be considered to significantly improve control and management of hypertension. The overall

Maternal Weaning Modulates Emotional Behavior and Regulates the Gut-Brain Axis.

Evidence shows that nutritional and environmental stress stimuli during postnatal period influence brain development and interactions between gut and brain. In this study we show that in rats, prevention of weaning from maternal milk results in depressive-like behavior, which is accompanied by changes in the gut bacteria and host metabolism. Depressive-like behavior was studied using the forced-swim test on postnatal day (PND) 25 in rats either weaned on PND 21, or left with their mother until PND 25 (non-weaned). Non-weaned rats showed an increased immobility time consistent with a depressive

DPP-4 inhibitor alleviates gut-brain axis pathology in Parkinson's disease.

BACKGROUND: Dipeptidyl peptidase-4 inhibitors (DPP-4is) have been reported to exhibit therapeutic effects in Parkinson's disease (PD), increasing their potential for drug repurposing. One aspect of PD pathogenesis is thought to be associated with the gut-brain axis, where α-synuclein from the gut is transmitted to the brain via the vagus nerve (VN). OBJECTIVE: We explored whether sitagliptin, a DPP-4i, exhibits a protective effect in a low-dose rotenone-treated gut-brain axis-associated PD model. DESIGN: To explore the effect of sitagliptin, we used the oral rotenone-treated mouse model, whi

Microbiota-gut-brain axis in cerebral palsy: from mechanisms to interventions.

Cerebral palsy (CP) is the most common cause of chronic motor disability in childhood and arises from non-progressive injury to the developing brain. Despite the static nature of the primary lesion, children with CP frequently experience evolving gastrointestinal, nutritional, inflammatory, sleep, cognitive, and seizure-related comorbidities that substantially influence functional recovery and caregiver burden. The microbiota-gut-brain axis (MGBA) provides a biologically plausible framework linking these multisystem manifestations. Emerging evidence suggests that children with CP may exhibit r
