Preparation of metal ion buffers for biological experimentation: a methods approach with emphasis on iron and zinc.

Transition metal ions are a challenge to study in physiology because of problems associated with solubility, oxidation, binding, and attaining appropriate free activities in solution. This review discusses these problems and potential ways of accommodating them. Special attention is given to iron and zinc ions, but many of the concepts can be applied for studying other transition metals. Selection of reagents appropriate for metal work (including water, salts, noncomplexing pH buffers) is briefly discussed. Calculation of the solubility product (K(sp)) for common iron and zinc precipitates is

The roles of intracellular buffers and bone mineral in the regulation of acid-base balance in mammals.

1. Regulation of intracellular and extracellular pH may conflict in their requirements for movement of acid or base. 2. Cells make a positive or a negative contribution to 'tissue buffering' of extracellular fluid (ECF), depending on their internal buffer value, on the tightness of their internal pH control by membrane mechanisms, and on the nature of the acid-base disturbance. 3. A role is suggested for electrogenic Na-HCO3 co-transport in some of the ion shifts that occur in acid-base disturbances. 4. The time course of 'tissue buffering' in nephrectomized mammals in hypercapnia is variable,

Psychological capital and health behaviours: Can it buffer against fatigue?

This study explores how psychological capital (PsyCap) and self-regulatory fatigue (SRF) predict discretionary food intake and physical activity over time. Discretionary food includes cakes, confectionery, and processed meat. They are regarded as delectable but are also high in saturated fat and added sugars. Physical activity is similarly crucial for a healthy lifestyle. About 387 Australian adults participated in a three-wave online survey over 4 weeks that measured four aspects of PsyCap (hope, optimism, self-efficacy, and resilience) and SRF. The outcome variables were daily discretionary

Psychological consequences of AI-assisted training and the buffering role of mindfulness.

The integration of artificial intelligence (AI) into athletic training is accelerating, yet its psychological implications for athletes remain insufficiently understood. Drawing on the transactional model of stress and the stress-buffering framework of mindfulness, this study examined whether mindfulness training can mitigate adverse psychological responses associated with AI-assisted training. Using a randomized controlled factorial design, 160 collegiate athletes were assigned to AI-assisted training or standard training, with or without concurrent mindfulness intervention, and assessed at b

Ketamine Metabolites Promote Anxiolysis and Hydrocortisone Stress Buffering in Zebrafish.

Anxiety and stress-related disorders affect over 280 million individuals worldwide and remain inadequately treated by current pharmacotherapies, which are often limited in efficacy and tolerability. Ketamine produces rapid therapeutic effects in mood and anxiety disorders, but its clinical use is constrained by dissociative side effects and abuse liability. Ketamine metabolites, particularly (2 R,6 R)-hydroxynorketamine (RANK) and (2S,6S)-hydroxynorketamine (SHANK), may retain therapeutic benefits while minimizing adverse effects, but their behavioral and stress-dependent actions remain incomp

Degradation kinetics of apomorphine hydrochloride in buffered formulations and the effect of antioxidants.

INTRODUCTION: Apomorphine (APO) is inherently unstable in aqueous solutions, as it forms auto-oxidation products. The study investigates the degradation kinetics of APO as the function of pH and evaluates the stabilizing effects of antioxidants (i.e. ascorbic acid (AA) and sodium metabisulfite (SMB)) on APO solution formulations. SIGNIFICANCE: To understand combined impact of pH-adjustment, buffer concentrations and antioxidant addition on the degradation kinetics of APO. METHODS: The degradation kinetics of APO in solutions and the antioxidant effect were investigated with a reversed-phase

Limited capacity of habitat features to buffer grassland bird declines.

Grassland birds are among the most rapidly declining groups of species, largely due to historical habitat losses that have led to extinction debts. Beyond direct habitat manipulations (e.g., prescribed fire), conservation strategies for grassland birds often adopt a landscape-scale perspective, particularly when guiding decisions on reserve acquisition, prioritizing management across existing reserves, and restoring habitat. Additionally, climate is increasingly recognized as a critical consideration for grassland bird conservation, since temperature influences survival and nest success, while

Macroinvertebrates, a forgotten player in the nutrient dynamics of riparian buffer zones.

One of the main causes of poor ecological surface water quality in Europe is nutrient enrichment due to agricultural activities. Riparian buffer zones, the vegetative zones between agricultural land and surface waters, have proven to retain and remove nutrients, yet with highly variable effectivity. So far, the role of invertebrates as an explanation for this variability remains severely overlooked. Therefore, this study aimed to assess the contribution of invertebrates to nutrient retention and removal processes of riparian buffer zones. Only four articles on invertebrate activity in riparian

Food conscientiousness as a buffer against college students' weight gain.

INTRODUCTION: A variety of psychological factors may influence weight gain among undergraduates. As one of the psychological factors that might influence such weight gain, this research introduces food conscientiousness, a behavioral tendency toward making healthier food choices. METHODS: In Phase 1 of the study, we developed a food conscientiousness scale. In Phase 2, we examined whether undergraduates demonstrated weight gain and whether it was smaller among those high in food conscientiousness. RESULTS: The results indicated that college students demonstrated weight gain (2 lbs, on averag

Chronic stress, social support, and symptoms at midlife. Is there a buffering effect?

OBJECTIVES: This study was designed to examine associations among measures of stress, social support, and symptoms at midlife. Specifically, the study examined whether support buffered against the negative effects of stress on severity of symptoms grouped via factor analyses into emotional instability, vaso-somatic symptoms, mood disturbances, and aches and pains. METHODS: We used cross-sectional data from n = 119 women aged 40-55 in Nagaland, India. Midlife symptoms were measured with the help of questionnaires, and factor analysis was used to identify latent factors. Stress and social suppo

Nuclear proteasomes buffer cytoplasmic proteins during autophagy compromise.

Autophagy is a conserved pathway where cytoplasmic contents are engulfed by autophagosomes, which then fuse with lysosomes enabling their degradation. Mutations in core autophagy genes cause neurological conditions, and autophagy defects are seen in neurodegenerative diseases such as Parkinson's disease and Huntington's disease. Thus, we have sought to understand the cellular pathway perturbations that autophagy-perturbed cells are vulnerable to by seeking negative genetic interactions such as synthetic lethality in autophagy-null human cells using available data from yeast screens. These reve

A new hypothesis on vascular calcification: the exhausting buffer syndrome (EBS).

Here we propose that the Western world lifestyle disrupts phosphate metabolism and homeostasis due to caloric or acidic hyperphagia. Psychic factors such as social defeat due to stressed social coexistence characterized by reduced activity and chronic hypoventilation (hypercapnia) also play a role. At least two mechanisms mediate the harmful vascular effects of phosphates with intracellular acidosis being a feature in both of them. First, insufficient lifestyle and adjacent diet together with the psychosomatic mechanism of social defeat (mainly through chronic hypercapnic acidosis) lead to ins

A roadmap for potassium buffering/dispersion via the glial network of the CNS.

Glia use multiple mechanisms to mediate potassium fluxes that support neuronal function. In addition to changes in potassium levels within synapses, these ions are dynamically dispersed through the interstitial parenchyma, perivascular spaces, leptomeninges, cerebrospinal fluid, choroid plexus, blood, vitreous, and endolymph. Neural circuits drive diversity in the glia that buffer potassium and this is reciprocal. Glia mediate buffering of potassium locally at glial-neuronal interfaces and via widespread networked connections. Control of potassium levels in the central nervous system is mediat

Light-Induced Covalent Buffer Adducts to Histidine in a Model Protein.

PURPOSE: Light is known to induce histidine (His) oxidation and His-His crosslinking in proteins. The crosslinking is resulted from the nucleophilic attack of a His to a photooxidized His from another protein. The goal of this work is to understand if covalent buffer adducts on His residues can be generated by light through similar mechanisms in nucleophilic buffers such as Tris and His. METHODS: A model protein (DNase) was buffer exchanged into nucleophilic buffers before light exposure. Photogenerated products were characterized by tryptic peptide mapping with mass spectrometry (MS) analysi

Proposal for Alzheimer's diagnosis using molecular buffer and bus network.

A novel design of an optical trapping tool for tangle protein (tau tangles, β-amyloid plaques) and molecular motor storage and delivery using a PANDA ring resonator is proposed. The optical vortices can be generated and controlled to form the trapping tools in the same way as the optical tweezers. In theory, the trapping force is formed by the combination between the gradient field and scattering photons, and is reviewed. By using the intense optical vortices generated within the PANDA ring resonator, the required molecular volumes can be trapped and moved dynamically within the molecular buff

Electrotransfer of proteins in an environmentally friendly methanol-free transfer buffer.

Western blot transfer buffer was modified to substitute the acute poison methanol, with the common rubbing alcohol, isopropanol in concentrations of as low as 5 % for protein electrotransfer. Commercially available molecular weight markers and rabbit serum were run on polyacrylamide gels and shown to be transferred adequately to both nitrocellulose and polyvinylidene difluoride membranes under either wet or semi-dry conditions with similar results in all cases. This procedure was successfully used for immunodetection of the rabbit IgG heavy chain from serum. Therefore, this represents a good a
