[Prevention of adrenaline-induced arrhythmia by a calmodulin blocker, trifluoroperazine].

Adrenergic arrhythmias were induced in isolated rat hearts with epinephrine in a concentration of 5.10(-5) M in the perfusing solution. The rhythm disturbance was accompanied by a pronounced depression of contractility. It was found that the preliminary administration of the calmodulin blocker trifluoperazine (10(-6) M) decreased the duration of arrhythmias and the latency of their development, the contractility remaining at a higher level than in control. Verapamil administered into the perfusing solution did not significantly affect the latency of arrhythmias and reduced the duration of arrh

Calmodulin is labeled at lysine 148 by a chemically reactive phenothiazine.

10-(3-Propionyloxysuccinimide)-2-(trifluoromethyl)phenothiazine (POS-TP) is a chemically reactive calmodulin antagonist: 2 mol are incorporated per mol of calmodulin when excess reagent is used, and only lysyl side chains are modified. Tryptic peptide mapping demonstrated that a single unique site on calmodulin reacts at low molar ratios of POS-TP. Labeled peptides were isolated and analyzed by amino acid composition and sequence analysis. The unique site was identified as Lys148 of calmodulin, the carboxyl-terminal residue. At higher molar ratios of the reagent Lys21, Lys75, and Lys77 are lab

Effects of calmodulin antagonists on secretion of bile and bile acid.

In the isolated perfused rat liver, the effects of calmodulin (CaM) antagonists, chlorpromazine (CPZ), trifluoperazine (TFP), W-7 and W-5, on secretion of bile and bile acid were compared. Without addition of taurocholic acid to the perfusate, TFP (200 microM or higher), CPZ (200 microM) and W-7 (400 microM) decreased the bile flow transiently. In contrast, W-5 did not decrease the bile flow. Taking into consideration the binding of TFP and CPZ to bovine serum albumin in the perfusate, they diminished the bile flow by inhibiting CaM function. This was also supported by the difference between t

Paradoxical effect of trifluoperazine, a calmodulin antagonist, on pepsinogen secretion.

Pepsinogen secretion (PS) is modulated at the intracellular level by both cAMP and calcium ion. Cholecystokinin octapeptide (CCK-8), a potent stimulus for PS, is believed to act through calcium. The most extensively studied pathway for calcium-mediated modulation involves the formation of calcium/calmodulin complexes, leading to activation of calmodulin. We have therefore examined the hypothesis that an inhibitor of calmodulin might inhibit PS stimulated by CCK-8. The phenothiazine derivative trifluoperazine (TFP) was chosen as a calmodulin antagonist. We measured in vitro secretion of pepsino

A role for calmodulin in renal production of 1,25-dihydroxyvitamin D3.

The conversion of circulating 25-hydroxyvitamin D3 (25OHD3) to its active metabolite 1,25-dihydroxyvitamin D3 [1,25-(OH)2D3] in the renal tubule mitochondrion by the enzyme 25OHD3-1 alpha-hydroxylase is closely regulated in vivo according to the physiological need for calcium and phosphorus. The mechanism by which this regulation is achieved at the cellular level has not been clarified, although a number of lines of evidence suggest that calcium ions (Ca2+) are involved. This study was designed to determine whether calmodulin, the ubiquitous cell protein that binds and mediates many of the reg

Inhibition of growth of C6 astrocytoma cells by inhibitors of calmodulin.

We evaluated the effect of several classes of calmodulin inhibitors on the activity of calmodulin prepared from C6 astrocytoma cells and studied the activity of these drugs as inhibitors of the growth of C6 cells in tissue culture. There was a good correlation between the activity of the drugs as inhibitors of calmodulin and their activity as inhibitors of cell growth. The most potent compounds were calmidazolium and melittin as compared to the phenothiazines, trifluoperazine, chlorpromazine, chlorpromazine-sulfoxide or the diphenylbutylpiperidine, pimozide. The mechanism by which the inhibiti

Calmodulin in mast cells and its role in histamine secretion.

Calmodulin content and distribution in rat peritoneal mast cells was determined by radioimmunoassay. Isolated pure mast cells were disrupted by sonication and the total calmodulin content and its distribution were determined. Calmodulin bound to the membranes was released by heating with 0.1% Lubrol PX to 95 degrees C for 5 min. The total calmodulin content of mast cells was found to be 160 +/- 14 ng/10(6) cells (9.4 +/- 0.82 pmoles/10(6) cells). The highest amount (68%) was present in the supernatant representing the cytosol. The next highest amount (26%) was found in the composite fraction c

Studies on calmodulin-binding proteins (CaMBPs) in the cilia of Tetrahymena.

As a first step to elucidate the involvement of calmodulin in Ca2+-dependent regulation of ciliary motility, molecular species and properties of calmodulin-binding proteins (CaMBPs) in Tetrahymena cilia were investigated by a modified [125I]calmodulin overlay method. At least 36 kinds of CaMBPs were detected. All the CaMBPs bound to calmodulin in Ca2+-dependent and calmodulin-specific manners, but they showed different Ca2+-dependencies. Several of CaMBPs bound to calmodulin in the presence of 100 microM trifluoperazine, several did in the presence of 8 M urea, and a few of them were highly se

Effects of trifluoperazine on calcium binding by calmodulin. A microcalorimetric study.

Microcalorimetric titrations of calmodulin with Ca2+ and trifluoperazine (TFP) at various molar ratios have been carried out at 25 degrees C and at pH 7.0. Ca2+ binding to calmodulin produces heat (-delta H) in the presence of TFP, while heat is absorbed in the absence of TFP. The total heat produced by Ca2+ binding to all four sites is increased at increasing TFP-to-calmodulin ratios, attaining a plateau at about 7. These results indicate that at the higher ratios, the enthalpy changes (delta H) associated with Ca2+ binding are affected by TFP molecules bound at both high- and low-affinity si

Lead and other metals can substitute for Ca2+ in calmodulin.

We have studied the interaction between some heavy metal ions, as compared with earth alkali ions, and calmodulin, a tissue protein which binds Ca2+ and mediates some of its effects. 1. Calmodulin dependent phosphodiesterase was activated with Pb2+, Ca2+, Sr2+, Ba2+, and Cd2+ (EC50 about 0.8 microM). The maximal activation achieved decreases in the order given. Hg2+, Sn2+, Fe2+, Cu2+, Ni2+, Bi3+, and Sb3+ up to 20 microM did not activate. 2. Pb2+ can replace Ca2+ with respect to the calmodulin-dependent phosphorylation of brain membranes. With high Pb2+ concentrations, phosphorylation was inhi

Possible involvement of calmodulin in maturation and activation of Chaetopterus eggs.

We report the isolation of calmodulin from oocytes of Chaetopterus pergamentaceus. The identification of this protein is based on (1) activation of beef heart cAMP phosphodiesterase, (2) heat stability, (3) sensitivity to chlorpromazine, and (4) electrophoretic mobility identical to that of porcine brain calmodulin after sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the presence of either Ca2+ or EGTA. We treated oocytes with chlorpromazine and W-7 to investigate the involvement of calmodulin in meiosis initiation and egg activation. Very low concentrations of chlorpromazine inh

H-NMR studies of calmodulin: the effect of W-7 (N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide) and Ca2+ on conformational changes of calmodulin.

The effect of W-7 (N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide), a calmodulin antagonist, on the structure of calmodulin was studied with 400 MHz H-NMR. W-7 affected the calcium-induced conformational change of calmodulin in several resonances. One resonant peak was assigned to the His-107 H2 proton. The other peaks were seen in the area of the methionine methyl group (around 2 ppm) and the high-field methyl group (0-1 ppm), these peaks cannot be assigned. The modifying effect of W-7 on the methyl-group resonances of calmodulin fully bound with Ca2+ was similar to that of trifluoperazin

G protein-coupled receptor activation rapidly stimulates focal adhesion kinase phosphorylation at Ser-843. Mediation by Ca2+, calmodulin, and Ca2+/calmodulin-dependent kinase II.

A rapid increase in the tyrosine phosphorylation of focal adhesion kinase (FAK) has been extensively documented in cells stimulated by multiple signaling molecules, but little is known about the regulation of FAK phosphorylation at serine residues. Stimulation of Swiss 3T3 cells with the G protein-coupled receptor agonists bombesin, vasopressin, or bradykinin induced an extremely rapid (within 5 s) increase in FAK phosphorylation at Ser-843. The phosphorylation of this residue preceded FAK phosphorylation at Tyr-397, the major autophosphorylation site, and FAK phosphorylation at Ser-910. Treat

Calmodulin Binding Domains in Critical Risk Proteins Involved in Neurodegeneration.

Neurodegeneration leads to multiple early changes in cognitive, emotional, and social behaviours and ultimately progresses to dementia. The dysregulation of calcium is one of the earliest potentially initiating events in the development of neurodegenerative diseases. A primary neuronal target of calcium is the small sensor and effector protein calmodulin that, in response to calcium levels, binds to and regulates hundreds of calmodulin binding proteins. The intimate and entangled relationship between calmodulin binding proteins and all phases of Alzheimer's disease has been established, but th

Inhibition of bacterial ubiquitin ligases by SidJ-calmodulin catalysed glutamylation.

The family of bacterial SidE enzymes catalyses phosphoribosyl-linked serine ubiquitination and promotes infectivity of Legionella pneumophila, a pathogenic bacteria that causes Legionnaires' disease1-3. SidE enzymes share the genetic locus with the Legionella effector SidJ that spatiotemporally opposes the toxicity of these enzymes in yeast and mammalian cells, through a mechanism that is currently unknown4-6. Deletion of SidJ leads to a substantial defect in the growth of Legionella in both its natural hosts (amoebae) and in mouse macrophages4,5. Here we demonstrate that SidJ is a glutamylase

Interaction of antagonists with calmodulin: insights from molecular dynamics simulations.

We report results of 12 ns, all-atom molecular dynamics simulation (MDS) and Poisson-Boltzmann free energy calculations (PBFE) on calmodulin (CaM) bound to two molecules of trifluoperazine (TFP) and of N-(3,3, diphenylpropyl)- N'-[1- R-(3,4-bis-butoxyphenyl)-ethyl]-propylenediamine (DPD). X-ray data show very similar structures for the two complexes, yet the antagonists significantly differ with respect to their CaM binding affinities, the neutral DPD is much more potent. The goal of the study was to unravel the reason why TFP is less potent although its positive charge should facilitate bindi

Overexpression of calmodulin in pancreatic beta cells induces diabetic nephropathy.

Recently, endothelial dysfunction induced by an uncoupling of vascular endothelial growth factor (VEGF) and nitric oxide has been implicated in the pathogenesis of diabetic nephropathy (DN). Investigating the pathogenesis of DN has been limited, however, because of the lack of animal models that mimic the human disease. In this report, pancreatic beta cell-specific calmodulin-overexpressing transgenic (CaMTg) mice, a potential new model of DN, are characterized with particular emphasis on VEGF and related molecules. CaMTg mice developed hyperglycemia at 3 wk and persistent proteinuria by 3 mo.

Targeting calmodulin in reversing multi drug resistance in cancer cells.

Calmodulin is a Ca2+ binding protein found in many eukaryotic cells. It is one of the most important intracellular mediators of Ca2+-dependant signaling in eukaryotic cells. It regulates diverse processes including mitosis, muscle contraction and nucleotide metabolism by modulating the activity of at least 30 different target enzymes in a calcium-dependant manner. Calmodulin plays an important role in the regulation of processes, such as the assembly and disassembly of microtubules by controlling protein kinase activities, by exerting an indirect influence upon a wide variety of cellular proce
