Perivascular epithelioid cell tumor of the descending colon mimicking a gastrointestinal stromal tumor: a case report.

BACKGROUND: We present a case of perivascular epithelioid cell tumor (PEComa), which clinically and histologically mimics a gastrointestinal stromal tumor (GIST). CASE PRESENTATION: A 42-year-old woman was found to have a mass in the left flank during her annual medical checkup. Computed tomography examination revealed a submucosal tumor of the descending colon. Surgeons and radiologists suspected that the lesion was a GIST, and left hemicolectomy was performed without biopsy. Microscopic examination showed that the lesion was composed of spindle and epithelioid cells, which were immunohistoc

Tubular-trabecular type Basal cell adenoma of the parotid gland: a patient report.

Basal cell adenoma (BCA) is an uncommon benign salivary gland neoplasm that includes isomorphic basaloid cells. We report on a female patient with BCA that developed in the right parotid gland in her 50s. The present patient demonstrated a few tumor nests in the fibrous capsule, and her tumor was larger than usual. These facts made us suspect of malignancy. Histopathologically, the tumor was characterized by multiple duct-like structures and tubular-trabecular masses composed of small isomorphic cells with hyperchromatic, round nuclei and an eosinophilic cytoplasm. It was difficult to determin

Transcriptome dysregulation by anthrax lethal toxin plays a key role in induction of human endothelial cell cytotoxicity.

We have investigated how Bacillus anthracis lethal toxin (LT) triggers caspase-3 activation and the formation of thick actin cables in human endothelial cells. By DNA array analysis we show that LT has a major impact on the cell transcriptome and we identify key host genes involved in LT cytotoxic effects. Indeed, upregulation of TRAIL and downregulation of XIAP both participate in LT-induced caspase-3 activation. LT induces a downregulation of the immediate early gene and master regulator of transcription egr1. Importantly, its re-expression in LT-intoxicated cells blocks caspase-3 activation

Perivascular epithelioid cell tumour of the vagina in an 8 year old girl.

Perivascular epithelioid cell tumor (PEComa) is a rare mesenchymal tumor with an unpredictable natural history. We present the first reported case of PEComa of the vagina diagnosed in an 8-year-old girl. This was initially diagnosed on biopsy as an embryonal rhabdomyosarcoma, and the patient underwent 3 cycles of chemotherapy according to the Mesenchymal Malignant Tumour 1989 protocol. A repeat scan, however, revealed an increase in tumor size. Total resection was achieved via a transvaginal (vertical sleeve) partial vaginectomy. The resected tumor had the typical appearance of a PEComa. Immun

Structure-Based Design of First Ndc80 Loop-Targeting Peptide Disrupting Ndc80-Nuf2 Interaction for Hepatocellular Carcinoma Therapy.

Ndc80 is essential for kinetochore-microtubule attachment, but its broad protein-protein interaction interface remains difficult to target. Here, we used structure-guided peptide optimization to develop Peptide-4, a loop-directed peptide designed to modulate the Ndc80-Nuf2 interaction. In contrast to reported Ndc80-directed compounds acting through the Hec1-Nek2 axis or the calponin homology domain, Peptide-4 addresses an underexplored loop region. Peptide-4 bound Ndc80 with a Kd of 0.45 ± 0.01 nM and inhibited the Ndc80-Nuf2 interaction with an IC50 of 0.76 ± 0.03 nM. In Huh7 cells, Peptide-4

Gastric plexiform fibromyxoma: a case report and literature review.

Plexiform fibromyxoma (PF) is a rare mesenchymal tumour that primarily occurs in the stomach, with the antrum and pyloric region being the most common sites. A few cases have also been reported in the duodenum, jejunum, mediastinum, gallbladder, and other locations. Over 100 cases have been reported in the literature, with more than 30 cases reported in our country. A rare case of PF occurring in the cardia and fundus of the stomach is reported in this study. The tumour tissue showed a multinodular, plexiform growth pattern between the muscle bundles of the gastric wall, in which a myxoid matr

Novel Use for DOG1 in Discriminating Breast Invasive Carcinoma from Noninvasive Breast Lesions.

AIMS: DOG1 has proven to be a useful marker of gastrointestinal stromal tumors (GISTs). Recently, DOG1 expression has also been reported in some non-GIST malignant tumors, but the details related to DOG1 expression in breast tissue remain unclear. The aim of this study was to detect the expression of DOG1 in the human breast and to evaluate the feasibility of using DOG1 to discriminate between invasive breast carcinoma and noninvasive breast lesions. METHODS AND RESULTS: A total of 210 cases, including both invasive and noninvasive breast lesions, were collected to assess DOG1 expression immu

Salivary duct carcinoma of the parotid gland: A case report and review of the literature.

Salivary duct carcinoma (SDC) is a rare and aggressive parotid malignancy that most commonly affects males in the fifth and sixth decades of life. Histopathology specimens obtained from SDC patients demonstrate a resemblance to ductal carcinoma of the breast. Therefore, to distinguish SDC from breast ductal carcinoma, several immunohistochemical markers exist that may enable surgeons to make an accurate diagnosis. In this study, the case of a 54-year-old male with salivary duct carcinoma of the right parotid gland is presented. The results of the present case study revealed that the SDC sample

Ca2+ and calmodulin regulate the binding of filamin A to actin filaments.

Filamin A (FLNa) cross-links actin filaments (F-actin) into three-dimensional gels in cells, attaches F-actin to membrane proteins, and is a scaffold that collects numerous and diverse proteins. We report that Ca(2+)-calmodulin binds the actin-binding domain (ABD) of FLNa and dissociates FLNa from F-actin, thereby dissolving FLNa.F-actin gels. The FLNa ABD has two calponin homology domains (CH1 and CH2) separated by a linker. Recombinant CH1 but neither FLNa nor its ABD binds Ca(2+)-calmodulin in the absence of F-actin. Extending recombinant CH1 to include the negatively charged region linker

Congenital diaphragmatic hernia as a prominent feature of a SPECC1L-related syndrome.

Congenital diaphragmatic hernias (CDH) confer substantial morbidity and mortality. Genetic defects, including chromosomal anomalies, copy number variants, and sequence variants are identified in ~30% of patients with CDH. A genetic etiology is not yet found in 70% of patients, however there is a growing number of genetic syndromes and single gene disorders associated with CDH. While there have been two reported individuals with X-linked Opitz G/BBB syndrome with MID1 mutations who have CDH as an associated feature, CDH appears to be a much more prominent feature of a SPECC1L-related autosomal

LIMCH1-enriched extracellular vesicles promote vascular permeability in early-onset preeclampsia.

Preeclampsia (PE) is a major pregnancy complication characterized by hypertension and multiple end-organ dysfunctions; however, its detailed pathogenesis remains unclear. Extracellular vesicles (EVs) play diverse and critical roles in intercellular communication, and we have demonstrated interaction between EVs and vascular endothelial cells. Through serum proteomic analysis, we identified LIM and calponin homology domain-containing protein 1 (LIMCH1) as a PE-associated EV protein that is highly expressed in PE placentas, particularly in syncytiotrophoblasts, which release EVs into the materna

An ulcerated ileal leiomyosarcoma identified by single-balloon enteroscopy despite nondiagnostic mucosal biopsies.

Gastrointestinal leiomyosarcoma is a rare malignant smooth-muscle neoplasm that may be difficult to diagnose using superficial mucosal biopsy. A 44-year-old woman presented with chronic intermittent abdominal pain and microcytic hypochromic anemia. Retrograde single-balloon enteroscopy identified an ulcerated, stenosing lesion in the distal ileum, whereas six forceps biopsy specimens showed only inflammation and granulation tissue. Segmental ileal resection was therefore performed because malignancy remained strongly suspected. Histological examination demonstrated a spindle-cell neoplasm with

Hypercalcemia of malignancy in a dog with cutaneous apocrine gland carcinoma and malignant myoepithelioma.

Humoral Hypercalcemia of Malignancy (HHM) is a commonly reported condition in dogs affected by several cancers, mostly T-cell lymphoma and anal sac gland carcinomas, but also other type of tumors. We report the case of a 6-years-old, female neutered Border Collie with an ulcered nodule on the right retromandibular region, that had mild serum total calcium elevation (14,1 mg/dl; reference range (RR): 8,4 to 11 mg/dl) in the absence of clinicopathological abnormalities or imaging finding suggesting any known cause of hypercalcemia. Histologically, the tumor had a tubular epithelial component, ad
