Fungicidal effects of eight disinfectants against resistant Candidozyma auris (formerly Candida auris), Aspergillus fumigatus, and Trichophyton indotineae.

UNLABELLED: Disinfectants are widely used in hospital environments to control bacterial, viral, and fungal hospital-acquired infections. In recent years, nosocomial infections caused by drug-resistant fungi, such as Candidozyma auris, Aspergillus fumigatus, and Trichophyton indotineae, have posed an increasing transmission risk among patients and healthcare workers, making treatment more challenging. Although many disinfectants are routinely applied in hospital settings, data on their efficacy against drug-resistant fungi remain limited. In this study, we evaluated the antifungal efficacy of e

CRISPR-Cas9 RNP-Mediated Deletion of ERG25 in Non-albicans Candida Species, Including Candida auris.

The incidence of infections caused by non-albicans Candida (NAC) species, including Candida glabrata and Candida tropicalis, has recently increased. Furthermore, Candida auris, a multidrug-resistant yeast, poses a serious threat to global health. The development of novel antifungal agents with alternative mechanisms of action is necessary to combat these fungi. However, genetic analyses of the virulence factors in these NAC species are insufficient for this purpose. Recent advancements in the clustered regularly interspaced short palindromic repeat (CRISPR)-Cas9 system have facilitated enhance

Updated Genomic Epidemiologic Description of Candida (Candidozyma) auris, United States.

The multidrug-resistant yeast Candida (Candidozyma) auris has caused several healthcare-associated outbreaks in the United States. We provide a genomic epidemiologic description of 1,535 C. auris isolates collected in the United States during 2013-2022. We identified clades I, II, III, and IV but not clades V or VI. Median pairwise single-nucleotide polymorphism distances indicated lower intraclade relatedness for clades I (91), III (43), and IV (43), compared with clade II (1,455). Phylogenetic analysis showed regional clusters with varying predominant clades. Of 809 isolates that underwent a

Ethical Priorities for Candida auris Screening in Solid Organ Transplantation.

Candida auris is an emerging, multidrug-resistant fungal pathogen of growing concern in solid organ transplantation, where colonization may have implications for pretransplant candidacy and post-transplant outcomes. Although C. auris screening is increasingly common in presurgical and critical care settings, no national or professional guidelines in the United States specify who, how, or when to screen, nor is there consensus on how screening results should impact patient care. Here, we examine the ethical significance of C. auris colonization in transplant candidacy decisions and outline the

Growth phase influences virulence in Candida auris systemic infection models.

Candida auris is a growing public health concern, capable of causing long-term contamination of healthcare settings, skin colonization, and life-threatening bloodstream infections. However, C. auris pathogenesis is not well understood, which is exacerbated by limitations and discrepancies in existing animal infection models. Further, the effects of C. auris growth phase on virulence have not been examined, despite growth phase being linked to virulence in many bacterial species. To address this question, and to develop an immunocompetent murine model of infection, we directly compared log and

Genomic analysis of Candida auris transmission within an urban region.

Candida auris emerged in Chicago, IL, USA, in 2016 and has since become endemic. We used whole-genome sequencing (WGS) of 494 isolates, epidemiologic metadata and patient transfer data to describe the transmission of C. auris among Chicago healthcare facilities between 2016 and 2021. In total, 99% of isolates formed a single clade IV phylogenetic lineage, suggesting a single introduction. The remaining isolates were clade III. Isolates were grouped into genomic clusters based on phylogenetic clustering and relatedness. Isolates from 14 facilities were included in 77 genomic clusters with evide

Environmental reservoirs of the drug-resistant pathogenic yeast Candida auris.

Candia auris is an emerging human pathogenic yeast; yet, despite phenotypic attributes and genomic evidence suggesting that it probably emerged from a natural reservoir, we know nothing about the environmental phase of its life cycle and the transmission pathways associated with it. The thermotolerant characteristics of C. auris have been hypothesised to be an environmental adaptation to increasing temperatures due to global warming (which may have facilitated its ability to tolerate the mammalian thermal barrier that is considered a protective strategy for humans against colonisation by envir

Detection and Identification of Candida auris from Clinical Skin Swabs.

Candida auris is a recently emerged pathogenic fungus posing an urgent global health threat. Given its propensity to cause nosocomial outbreaks, rapid detection and accurate identification of C. auris have become essential for an effective implementation of infection prevention and control measures in healthcare facilities. Unfortunately, the requirement of culturing a yeast colony from a patient sample, central for most of the available diagnostic tools, results in substantial delays (several days or longer) in diagnosis. However, nucleic acid-based techniques can deliver accurate diagnostic

Optimization and Validation of Candida auris Short Tandem Repeat Analysis.

Candida auris is an easily transmissible yeast with resistance to different antifungal compounds. Outbreaks of C. auris are mostly observed in intensive care units. To take adequate measures during an outbreak, it is essential to understand the transmission route, which requires isolate genotyping. In 2019, a short tandem repeat (STR) genotyping analysis was developed for C. auris. To determine the discriminatory power of this method, we performed STR analysis of 171 isolates with known whole-genome sequencing (WGS) data using Illumina reads, and we compared their resolutions. We found that ST

Candida auris Invasive Infections during a COVID-19 Case Surge.

Clinical cases of C. auris noted during a COVID-19 surge led to an epidemiological, clinical, and genomic investigation. Evaluation identified a close genetic relationship but inconclusive epidemiologic link between all cases. Prolonged hospitalization due to critical illness from COVID-19 and use of antimicrobials may have contributed to clinical infections.

Candida auris-an impending threat: A case report from home.

Candida auris has been identified as an invasive fungal organism of outbreaks globally. Due to its striking ability to colonize the skin and readily transmit from person-to-person, it is being categorized as a public health threat. Infected patients are not only characteristically critically ill, but delayed identification along with limited options for effective antifungals makes this nosocomial infection worrisome for clinicians. The risk of persistence of fungus as a contaminant in hospital rooms and on medical equipment is also a concern for health care facilities. In this case report, we

The Postbiotic Activity of Lactobacillus paracasei 28.4 Against Candida auris.

Candida auris has emerged as a medically important pathogen with considerable resistance to antifungal agents. The ability to produce biofilms is an important pathogenicity feature of this species that aids escape of host immune responses and antimicrobial agents. The objective of this study was to verify antifungal action using in vitro and in vivo models of the Lactobacillus paracasei 28.4 probiotic cells and postbiotic activity of crude extract (LPCE) and fraction 1 (LPF1), derived from L. paracasei 28.4 supernatant. Both live cells and cells free supernatant of L. paracasei 28.4 inhibited
