Multi-omic analysis reveals that the CARNS1-mediated carnosine synthesis pathway modulates muscle metabolic responses to nutrient restriction in goats.

Seasonal feed shortages pose critical challenges to ruminant productivity, yet the mechanisms underlying muscle metabolic responses to varying degrees of restriction remain unclear. This study simulated feed shortage in goats using three dietary treatments: normal feeding (RF0), 25% restricted feeding (RF25), and 50% restricted feeding (RF50). A total of 30 male Xiangdong black goats with similar initial body weight (19.14 ± 2.75 kg) and age (4.0 ± 0.3 months) were randomly assigned into 3 groups (n = 10 per group) for an 87-d trial (7-d preliminary period followed by an 80-d experimental peri

Carnosine-modified gelatin-hyaluronic acid hydrogel comprising fenofibrate-loaded nanoparticles targeting chondrocyte ferroptosis and macrophage polarization for synergistic osteoarthritis therapy.

Osteoarthritis (OA), a debilitating and progressive joint disease, presents major therapeutic challenges due to chondrocyte ferroptosis, chronic synovial inflammation, and the pharmacokinetic limitations of conventional intra-articular therapies, such as rapid clearance and poor retention. To overcome these barriers, we developed a dual-functional intra-articular delivery platform composed of a gelatin-hyaluronic acid hydrogel encapsulating fenofibrate-loaded, cartilage-targeting nanoparticles (FNPs-GelHA hydrogel). This system synergistically inhibits OA progression by simultaneously suppress

Prevention of radiation esophagitis by polaprezinc (zinc L-carnosine) in patients with non-small cell lung cancer who received chemoradiotherapy.

BACKGROUND: Concurrent chemoradiotherapy (CCRT) plays an important role in multimodality therapy for non-small cell lung cancer. However, esophagitis often develops as a complication of CCRT, causing treatment delays and reducing the patient's quality of life. We examined the efficacy of polaprezinc (PZ), zinc L-carnosine used for the therapy of gastric ulcer, against the onset of esophagitis caused by CCRT for lung cancer. PATIENTS AND METHODS: Patients who concurrently underwent chemotherapy with carboplatin and paclitaxel and thoracic radiotherapy at Gifu University Hospital during a perio

1 H NMRS of carnosine combined with 31 P NMRS to better characterize skeletal muscle pH dysregulation in Duchenne muscular dystrophy.

In recent years, quantitative nuclear magnetic resonance imaging and spectroscopy (NMRI and NMRS) have been used more systematically as outcome measures in natural history and clinical trial studies for Duchenne muscular dystrophy (DMD). Whereas most of these studies have emphasized the evaluation of the fat fraction as an assessment for disease severity, less focus has been placed on metabolic indices measured by NMRS. 31 P NMRS in DMD reveals an alkaline inorganic phosphate (Pi ) pool, originating from either leaky dystrophic myocytes or an increased interstitial space. 1 H NMRS, exploiting

Biomolecular Interaction of Carnosine and Anti-TB Drug: Preparation of Functional Biopeptide-Based Nanocomposites and Characterization through In Vitro and In Silico Investigations.

Host-directed therapies (HDTs) resolve excessive inflammation during tuberculosis (TB) disease, which leads to irreversible lung tissue damage. The peptide-based nanostructures possess intrinsic anti-inflammatory and antioxidant properties among HDTs. Native carnosine, a natural dipeptide with superior self-organization and functionalities, was chosen for nanoformulation. In the present work, multiscale self-assembly approaches of carnosine were developed using a solvent-mediated process (hexafluoro-2-propanol) and further linked with first-line anti-TB drugs. The organofluorine compound in a

The Role and Mechanism of Carnosine in Alleviating Type 2 Diabetic Sarcopenia in Mice Through PI3K/AMPK/PGC-1α Signaling Pathway.

Type 2 diabetes mellitus (T2DM) accounts for over 90% of diabetes cases, and T2DM-related sarcopenia is a growing concern. Carnosine, abundant in human skeletal muscle, helps maintain muscle quality and function. This study investigated whether carnosine deficiency contributes to T2DM-related sarcopenia and whether exogenous carnosine supplementation alleviates muscle atrophy. A mouse model of T2DM sarcopenia was established using streptozotocin combined with a high-fat diet. LC-MS metabolomics revealed a significant reduction in carnosine content in the gastrocnemius muscle of model mice. A C

Dietary supplementation with carnosine reduces the prevalence of breast muscle myopathies without altering performance, meat yield or quality in broiler chickens.

Breast muscle myopathies such as White striping and Wooden breast represent a major challenge for the poultry industry due to the deterioration of breast meat quality induced by these myopathies. Previous research has demonstrated that oxidative stress and subsequent oxidative damage in the Pectoralis major muscle were predisposing factors for the development of breast muscle myopathies. More specifically, research has shown that myopathic muscle content of histidine-containing dipeptides such as carnosine that is well-known for its antioxidant properties was almost entirely depleted, exposing

A Catecholaldehyde Metabolite of Norepinephrine Induces Myofibroblast Activation and Toxicity via the Receptor for Advanced Glycation Endproducts: Mitigating Role of l-Carnosine.

Monoamine oxidase (MAO) is rapidly gaining appreciation for its pathophysiologic role in cardiac injury and failure. Oxidative deamination of norepinephrine by MAO generates H2O2 and the catecholaldehyde 3,4-dihydroxyphenylglycolaldehyde (DOPEGAL), the latter of which is a highly potent and reactive electrophile that has been linked to cardiotoxicity. However, many questions remain as to whether catecholaldehydes regulate basic physiological processes in the myocardium and the pathways involved. Here, we examined the role of MAO-derived oxidative metabolites in mediating the activation of card

L-carnosine alleviates wooden breast myopathy in broilers fed oxidized soybean oil: Focus on antioxidant defense, calcium homeostasis, and satellite cell function.

Feeding oxidized soybean oil (OSO) can disrupt the redox homeostasis in broilers and increase the incidence and severity of wooden breast (WB) myopathy. The antioxidant activity of L-carnosine (LC) has been demonstrated to help alleviate oxidative stress generated during metabolic processes. Therefore, the effects of dietary LC supplementation on WB myopathy were investigated in this study. One hundred and twenty-six 21-d-old white-feathered broilers (1.017 ± 0.039 kg) were randomly allocated into three groups (7 replicates per group with 6 broilers per replicate): fresh soybean oil group (CON

Carnosine alleviates oxidative stress to prevent cellular senescence by regulating Nrf2/HO-1 pathway: a promising anti-aging strategy for oral mucosa.

Introduction: Aging is associated with significant metabolic alterations that contribute to cellular senescence and age-related functional decline. As individuals age, an increased prevalence of oral diseases and a gradual decline in oral functions are observed. However, the metabolic shifts underlying oral mucosal aging remain unexplored. Methods: We initially conducted histological analyses on the tongues from young (4-week-old), adult (4-month-old) and old (20-month-old) C57BL/6 mice to identify age-related alterations in the tongue mucosa. Subsequently, metabolomics analysis was performed

Carnosine improves colonic hyperpermeability through the brain histamine H1 receptor, basal forebrain cholinergic neurons, adenosine A2B receptors and vagus nerve in rats.

Leaky gut is implicated in disorders such as irritable bowel syndrome (IBS) and Alzheimer's disease (AD). Our previous study demonstrated that brain histamine H1 receptor signaling-mediated via basal forebrain cholinergic neurons (BFCNs), adenosine A2B receptors, and the vagus nerve-regulates intestinal barrier function. In this study, we investigated the role of carnosine, a dipeptide composed of beta-alanine and L-histidine, in modulating intestinal barrier integrity. In an LPS-induced leaky gut rat model, intracisternal administration of carnosine improved colonic permeability as determined
