Pharmacologic Comparison of Clinical Neutral Endopeptidase Inhibitors in a Rat Model of Acute Secretory Diarrhea.

Racecadotril (acetorphan) is a neutral endopeptidase (NEP) inhibitor with known antidiarrheal activity in animals and humans; however, in humans, it suffers from shortcomings that might be improved with newer drugs in this class that have progressed to the clinic for nonenteric disease indications. To identify potentially superior NEP inhibitors with immediate clinical utility for diarrhea treatment, we compared their efficacy and pharmacologic properties in a rat intestinal hypersecretion model. Racecadotril and seven other clinical-stage inhibitors of NEP were obtained or synthesized. Enzyme

Scalable Production of Self-Toughening Plant Oil-Based Polyurethane Elastomers with Multistimuli-Responsive Functionalities.

Plant oils are becoming of high industrial importance due to the persisting challenges befalling with the utilization of fossil fuels. Thus, developing methodologies to produce multifunctional materials by taking advantage of the unique structure of plant oil is highly desired. In this study, castor oil served as a cross-linker and soft segments, by incorporating scalable rhodamine 6G derivatives, to systematically synthesize a series of smart polymers that possess self-toughening and multistimuli-responsive capabilities. The polyurethane elastomers showed 10 times and 60 times increases in te

Oleogel-mediated Topical Administration of Roflumilast and Paclitaxel as a Synergistic Strategy to Combat Imiquimod-induced Psoriasis.

Psoriasis is a systemic immune disease with severe inflammation and skin thickening. Roflumilast (ROF) blocks cAMP hydrolysis, and paclitaxel (PTX) inhibits cell proliferation; both are effective in topical psoriasis treatment. However, the combination of ROF and PTX has not been reported. This study explored their synergistic mechanism and formulated a ROF-PTX oleogel with strong skin adhesion, low viscosity, enhanced skin penetration, and increased retention. The oleogel, prepared via direct gelation with jojoba oil as oil phase, PPG-15 as solvent, Transcutol as solubilizer, and hydrogenated

Anti-diarrheal effect of water extract of Evodiae fructus in mice.

Our previous study showed that Evodiae fructus (the dried, unripe fruit of Evodia rutaecarpa) has an inhibitory effect on the intestinal transit (anti-transit effect) in mice. In the present study, a water extract of Evodiae fructus was used to examine its effect on castor oil-induced diarrhea and to compare with its anti-transit effect in mice. The results indicated that Evodiae fructus had both anti-transit and anti-diarrheal effects with comparable ID(50) (the dose for 50% inhibition) values of 54+/-7 and 76+/-17 mg/kg. The time-courses of Evodiae fructus pretreatment for both anti-transit

CYX-5, a G-protein biassed MOP receptor agonist, DOP receptor antagonist and KOP receptor agonist, evokes constipation but not respiratory depression relative to morphine in rats.

BACKGROUND: Strong opioid analgesics such as morphine alleviate moderate to severe acute nociceptive pain (e.g. post-surgical or post-trauma pain) as well as chronic cancer pain. However, they evoke many adverse effects and so there is an unmet need for opioid analgesics with improved tolerability. Recently, a prominent hypothesis has been that opioid-related adverse effects are mediated by β-arrestin2 recruitment at the µ-opioid (MOP) receptor and this stimulated research on discovery of G-protein biassed opioid analgesics. In other efforts, opioids with MOP agonist and δ-opioid (DOP) recepto

A possible mode of solubilization of coenzyme Q10 with HCO-60.

The redox level of [14C]coenzyme Q10 solubilized with HCO-60 in the livers of guinea pigs at 24 h after intravenous injection was approximately 1/4 (18.2% vs. 63.4%) of that of [14C]coenzyme Q10 solubilized with ethanol-water (1:5 by vol). Further, the redox level of coenzyme Q10 (Q10) solubilized with HCO-60 or ethanol-water was 20.2 or 82.3%, respectively, after a 30-min incubation in the liver cytosol. Q10 solubilized with HCO-60 was thus reduced only slightly in vivo or in vitro. The critical micelle concentrations of HCO-60 in the absence and presence of Q10 were 0.02 and 0.002% (w/v), re

Synthesis and pharmacological activity of 2-(substituted)-3-{2-[(4-phenyl-4-cyano)piperidino]ethyl}-1,3-thiazolidin-4-ones.

Loperamide is a well-known peripherally acting opiate used for the treatment of diarrhoea. To gain more knowledge on the structure-activity relationships of antidiarrhoeal drugs and to develop new active molecules, a series of aryl-cyano-piperidinoalkyl-thiazolidinones related to Loperamide was synthesized and screened for antidiarrhoeal activity in mice by castor oil test. To characterize the potency and toxicity of the synthesized compounds ED50 and LD50 values were also determined. The thiazolidinones 2-6 displayed antidiarrhoeal activity at doses ranging between 15 and 82 mg/kg. Although t

Comparison of Efficacy of Two Different Topical 0.05% Cyclosporine A Formulations in the Treatment of Adenoviral Keratoconjunctivitis-Related Subepithelial Infiltrates.

Subepithelial infiltrates secondary to adenoviral keratoconjunctivitis may persist for years and cause blurred vision, halos, glare, and photophobia. These infiltrates arise from immune reaction against the virus, and few studies have reported topical cyclosporine A to be effective in the treatment of subepithelial infiltrates. Herein, we describe a patient with adenoviral keratoconjunctivitis-related subepithelial infiltrates who did not respond to treatment with a new topical cyclosporine A emulsion prepared with castor oil (Depores 0.05%; Deva İlaç, Kocaeli, Turkey), while the FDA-approved

Cutaneous leishmaniasis in Kenya: Sergentomyia garnhami (Diptera Psychodidae), a possible vector of Leishmania major in Kitui District: a new focus of the disease.

Investigations on phlebotomine sandflies in Tseikuru, Kitui District, Kenya were carried out to determine the vectors of Leishmania major. Sandflies were trapped from animal burrows over a period of one year using castor oil-smeared sticky traps. Nineteen phlebotomine sandfly species were identified and their prevalence in this habitat determined. Dissections on the parous sandflies resulted in the isolation of Leishmania parasites from Phlebotomus martini and Sergentomyia garnhami. The isolated parasites were cultured in NNN medium and were consequently mass cultured in RPMI medium for bioche
