Association between the Catechol O-methyltransferase (COMT) Val158met polymorphism and different dimensions of impulsivity.

BACKGROUND: Impulsivity is a multidimensional construct which has been associated with dopaminergic neurotransmission. Nonetheless, until this moment, few studies addressed the relationship between different types of impulsivity and the single nucleotide polymorphism caused by a substitution of valine (val) with methionine (met) in the 158 codon of the Catechol-o-Methyltransferase gene (COMT-val158met). The present study aimed to investigate the association between val158met COMT polymorphism and impulsive behavior measured by two neuropsychological tests. METHODOLOGY/PRINCIPAL FINDINGS: We a

Catechol O-methyltransferase variants and cognitive performance in schizophrenia and bipolar disorder versus controls.

BACKGROUND: The gene encoding Catechol O-methyltransferase (COMT), a dopamine catabolic enzyme, is an important candidate gene in several psychiatric disorders. Several studies have shown an association between the functional Val(158)Met polymorphism and cognitive performance. However, the results have been inconsistent and there are few studies addressing other COMT single nucleotide polymorphisms (SNPs). METHODS: We investigated SNPs across the whole COMT gene, including the Val(158)Met polymorphism, for a putative effect on working memory, executive function and IQ in 315 patients with sch

Catechol and humic acid sorption onto a range of laboratory-produced black carbons (biochars).

Although the major influence of black carbon (BC) on soil and sediment organic contaminant sorption is widely accepted, an understanding of the mechanisms and natural variation in pyrogenic carbon interaction with natural organic matter (NOM) is lacking. The sorption of a phenolic NOM monomer (catechol) and humic acids (HA) onto BC was examined using biochars made from oak, pine, and grass at 250, 400, and 650 degrees C. Catechol sorption equilibrium occurred after 14 d and was described by a diffusion kinetic model, while HA required only 1 d and followed pseudo-second-order kinetics. Catecho

Nonsynonimous mutation of catechol-O-methyl-transferase (COMT) gene in a patient with temporomandibular disorder.

We report a case of temporomandibular disorder patient with disc displacement without reduction, myofascial pain, limited opening and a novel, never described, nonsynonimous mutation of catechol-O-methyl-transferase (COMT) gene. COMT is one of the enzymes that metabolizes catecholamines, thereby acting as a key modulator of dopaminergic and adrenergic/noradrenergic neurotransmissions, which play a key role in pain modulation. This novel mutation, p.R58S, changed a codon (58 from arginine to serine) in the COMT protein. The introduction of a serine residue in a highly organised secondary struct

Expression of catechol-O-methyltransferase in the brain and periphery of normal and MPTP-treated common marmosets.

Catechol-O-methyltransferase (COMT) inhibition is widely used to potentiate the effects of levodopa in Parkinson's disease but the effects of nigral dopaminergic cell loss and levodopa treatment on COMT activity are not known. The present study investigated the expression of COMT in the brain and liver of normal common marmosets, and animals treated with MPTP and those treated with levodopa to induce dyskinesia. Reverse transcript PCR demonstrated the expression of COMT mRNA in the liver, cortex and striatum of normal marmosets. Using Western blotting, the presence of two subunits of COMT prot

Anodic electrochemiluminescence of CdTe quantum dots and its energy transfer for detection of catechol derivatives.

This work reported for the first time the anodic electrochemiluminescence (ECL) of CdTe quantum dots (QDs) in aqueous system and its analytical application based on the ECL energy transfer to analytes. The CdTe QDs were modified with mercaptopropionic acid to obtain water-soluble QDs and stable and intensive anodic ECL emission with a peak value at +1.17 V (vs Ag/AgCl) in pH 9.3 PBS at an indium tin oxide (ITO) electrode. The ECL emission was demonstrated to involve the participation of superoxide ion produced at the ITO surface, which could inject an electron into the 1Se quantum-confined orb

Association of the functional V158M catechol-O-methyl-transferase polymorphism with panic disorder in women.

Panic disorder is an anxiety disorder with an estimated heritability of up to 48%. Pharmacological and genetic studies suggest that genes coding for proteins involved in the catecholaminergic system might be relevant for the pathogenesis of the disease. In the present study, we genotyped a single nucleotide polymorphism (472G/A=V158M) in the coding region of the catechol-O-methyl-transferase (COMT) gene in 115 patients with panic disorder and age- and sex-matched controls. Association analysis revealed a significant excess of the more active COMT allele (472G=V158) in patients with panic disor

Oxidation of N-methyl(R)salsolinol: involvement to neurotoxicity and neuroprotection by endogenous catechol isoquinolines.

1(R), 2(N)-Dimethyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, N-methyl(R)salsolinol, is a potent dopaminergic neurotoxin to induce parkinsonism in rats. The cytotoxicity of N-methyl(R)salsolinol proved to be ascribed to its oxidation into cytotoxic 1,2-dimethyl-6,7-dihydroxyisoquinolinium ion with generation of hydroxyl radical. The isoquinolinium ion caused massive necrosis in the striatum, whereas N-methyl(R)salsolinol depleted selectively dopaminergic neurons in the substantia nigra without necrotic tissue reaction. N-Methyl(R)salsolinol induced DNA damage to human neuroblastoma SH-SY5Y

Effects of catechol compound administration on nerve growth factor synthesis in the peripheral nervous system.

The effects of the intraperitoneal administration of catechol compounds on nerve growth factor (NGF) synthesis in the peripheral nervous system were examined in Wistar male rats. Five injections of 4-methylcatechol (4-MC) during a three-day period caused a threefold to fourfold increase in the NGF content of organs with sympathetic innervation (heart, submandibular gland) and the sciatic nerve. Next, we investigated time-dependent changes in the NGF content after a single injection of 2 micrograms of 4-MC. A transient increase in the NGF content was detected in the heart and submandibular glan

Inhibition of catechol estrogen formation in rat liver microsomes by hormonal steroids and related compounds.

The inhibitory action of a number of different hormonal steroids and related compounds on the 2-hydroxylation of estradiol by male rat liver microsomes was examined by a radiometric assay. Progesterone, Diethylstilbestrol, testosterone and 4-androstenedione were found to be the most potent of the compounds tested but inhibition was also observed with other steroids and a group of androgen analogs which are aromatization inhibitors. The kinetic constant Ki for those steroids which gave linear double reciprocal plots when added to [2-3H]estradiol was determined and the products from [14C]estradi

A pharmacological investigation of the electrically evoked convulsive activity induced by administration of catechol in the anaesthetized rat.

1 The response evoked by electrical stimulation at the wrist has been recorded from muscles of the forelimb of anaesthetized rats induced to convulse by administration of catechol.2 This response can be divided into three temporally distinct components, the characteristics of which have been described.3 The probability of occurrence of the two early components of the response has been measured before and after administration of various drugs. The results show that the first component is not affected by cholinoceptor or adrenoceptor blocking drugs or anticholinesterase agents. The probability o

Structure-guided discovery of non-catechol dopamine D1 receptor ligands with biased agonism and antagonism.

The catechol L-DOPA, a cornerstone of Parkinson's disease (PD) treatment, has two major drawbacks: poor pharmacokinetics and, more significantly, debilitating dyskinesias from chronic dopamine D1 receptor (D1R) activation. Preclinical rodent studies suggest that D1R antagonism or β-arrestin-biased agonism can alleviate these motor complications, highlighting the need for next-generation non-catechol ligands. Through virtual screening, we identified eight novel chemotypes as D1R ligands, including two G protein-biased agonists, two β-arrestin-biased agonists and four antagonists. Structure-acti

Mussel-Inspired Copolyether Brushes: Synergistic Catechol-Amine Interactions for Enhanced Adhesion and Antifouling Performance.

Biofouling poses major challenges in marine and biomedical sectors. To overcome the substrate-specific limitations of conventional poly(ethylene glycol) (PEG) coatings, we report herein a versatile, mussel-inspired anchoring strategy. Using catechol-amine functionalized copolyethers, we created substrate-independent antifouling PEG brushes. To elucidate the synergistic role of catechol and amine groups, three control copolymers (catechol-only, phenol-amine, and phenyl-amine) were synthesized via anionic ring-opening polymerization using a PEG macroinitiator. Comprehensive characterization reve

Multifunctional Catechol-Functionalized Cellulose Hydrogels for the Minimally Invasive Treatment of Acute Optic Nerve Injuries.

Oxidative stress-induced retinal ganglion cell degeneration is a major pathological feature of acute optic nerve injury, yet current posterior-segment therapies are limited by poor local retention and repeated invasive administration. Here, we developed an injectable catechol-functionalized carboxymethyl cellulose hydrogel, CMCDA, as a bioadhesive and antioxidative intravitreal platform. Through dopamine grafting and oxidative crosslinking, CMCDA exhibited shear-thinning injectability, self-healing behavior, wet-tissue adhesion, controlled biodegradability, and good biocompatibility. Empagliflozin reduces liver fibrosis by restoring catechol-O-methyltransferase activity associated with magnesium levels.

Catechol-O-methyltransferase (COMT), a magnesium (Mg)-dependent enzyme, metabolises catecholamines. Diabetic patients exhibit hypomagnesemia and sympathetic overactivity compared with non-diabetics. Sodium-glucose cotransporter 2 (SGLT2) inhibitors increase serum Mg levels in diabetic patients. Sympathetic overactivity is associated with diabetic complications; however, the entire mechanism has not been elucidated. Type 2 diabetes model BKSdb/db male mice were fed either a control or an empagliflozin-supplemented diet. Mg2+ concentrations, catecholamines, and COMT activity and protein levels w
