Stage-Dependent Inhibitory Connectivity in Striatal-Motor Circuit in Huntington's Disease.

BACKGROUND: Elucidating dysfunctional connectivity patterns among key brain regions in Huntington's disease (HD) underlying progression may have implications for developing treatment and therapeutic evaluation. OBJECTIVE: Explore the relationship between abnormal spontaneous resting-state activity and atrophy in HD-specific brain regions and clarify effective connectivity changes among them across different stages of HD. METHODS: Amplitude of low-frequency fluctuation (ALFF) analysis was used to detect abnormal spontaneous neural activity; voxel-based morphometry analysis was applied to asse

Multimodality neuroimaging in Tourette syndrome: alpha-[11C] methyl-L-tryptophan positron emission tomography and diffusion tensor imaging studies.

Previous studies in Tourette syndrome have reported lateralized abnormalities of neurotransmitters and microstructure of the cortico-striato-thalamo-cortical circuit. The authors analyzed the relationship between serotonin synthesis and microstructural changes in the subcortical structures (caudate nucleus, lentiform nucleus, and thalamus) related to this circuit, using alpha-[(11)C]methyl-L-tryptophan positron emission tomography (PET) and diffusion tensor imaging, respectively, in 16 children with Tourette syndrome. Correlations between diffusion tensor imaging and alpha-[(11)C]methyl-L-tryp

Positron emission tomography reveals elevated D2 dopamine receptors in drug-naive schizophrenics.

In postmortem studies of patients with schizophrenia, D2 dopamine receptors in the basal ganglia have been observed to be more numerous than in patients with no history of neurological or psychiatric disease. Because most patients with schizophrenia are treated with neuroleptic drugs that block D2 dopamine receptors in the caudate nucleus, it has been suggested that this increase in the number of receptors is a result of adaptation to these drugs rather than a biochemical abnormality intrinsic to schizophrenia. With positron emission tomography (PET), the D2 dopamine receptor density in the ca

Neural compensation preserves white matter integrity in early Parkinson's disease but not atypical Parkinsonism: a diffusion tensor imaging study.

Parkinsonism is a common clinical manifestation of Parkinson's disease (PD) and other atypical parkinsonism (AP). Diffusion tensor imaging (DTI) may help characterize pathological alterations in cerebral white matter (WM). This study aimed to analyze the differences in early white matter microstructural alterations between PD and AP using DTI and to explore the potential value of DTI in the early differential diagnosis of PD and AP. DTI data were acquired from 12 patients with early PD, 11 with late PD, 12 with AP, and 11 healthy controls (HCs). Fractional anisotropy (FA) and apparent diffusio

Systems-Level Transcriptomic and Cellular Architecture of the Human Striatum in Schizophrenia and Bipolar Psychosis.

Psychosis involves striatal circuit dysfunction, but the regional molecular and cellular changes remain unclear. We re-analyzed 215 postmortem RNA-seq samples (72 donors: 36 controls, 28 schizophrenia, 8 bipolar disorder with psychosis) from the nucleus accumbens, caudate and putamen, asking whether diagnosis-associated transcriptional changes are region-specific and whether they reflect cellular composition or cell-intrinsic state. Region-specific limma-voom models identified 0, 141 and 2152 schizophrenia-associated genes (FDR < 0.10) in the nucleus accumbens, caudate and putamen; the small b

[Effect of electroacupuncture on electroconvulsive shock--an autoradiographic study for opioid receptors].

Radioreceptor-binding assay combined with autoradiography technique in the brain slices of rats was used to investigate cerebral areas related to electroconvulsive shock (ECS) and to analyze changes of opioid receptors of these areas in electroacupuncture (EA) anticonvulsion. The results from image processing and detection of optic density (O.D.) showed that repeated ECS resulted in an increase of opioid receptor densities in the caudate nucleus, hippocampus, habenulae nucleus and amygdala. In the group of EA plus ECS, however, the opioid receptor densities in the hippocampus, habenula nucleus

[Electroencephalographic analysis of the central action of benzonal].

Chronic experiments on conscious rabbits demonstrated that benzonal suppresses the spontaneous bioelectrical activity of the motor division of the cortex of large hemispheres while failing to affect the spontaneous biorhythms of the nonspecific nuclei of the thalamus, caudate nucleus, dorsal hippocamp and the basolateral nuclei of the tonsil. The drug decreased the electroexcitability of the motor cortex and caudate nucleus, exerted a mild depressant action on the reticular formation of the midbrain, tonsil, and hippocamp and had no effect on the excitability of nonspecific nuclei of the thala

Acute changes in regional brain water content following experimental closed head injury.

A Remington humane stunner was used to deliver blows to the skulls of anesthetized cats. The animals were sacrificed at 30 minutes or 1, 2, or 6 hours after trauma and selected for data collection on the basis of the following two categories of gross intracranial pathology: 1) unilateral contusion, with subarachnoid hemorrhage (SAH); or 2) SAH only. For selected cats, specific gravity was measured in 5- to 10-mg samples of uncontused tissue taken from coronal slices at the level of the frontoparietal suture. The regions tested included dorsal cerebral cortex, subcortical white matter, deep whi

Age-related dopamine deficiency in the mesostriatal dopamine system of zitter mutant rats: regional fiber vulnerability in the striatum and the olfactory tubercle.

Oxidant stress has been implicated in the pathogenesis of Parkinson's disease. To test the oxidant stress hypothesis of dopaminergic degeneration, age-related changes in the mesostriatal dopamine neuron system were compared between zitter mutant rats which have abnormal metabolism of oxygen species in the brain and Sprague-Dawley rat as a control using the neurochemistry and immunohistochemistry. Dopamine content in the caudate-putamen, nucleus accumbens and olfactory tubercle of zitter rats decreased significantly with age, and was lower than that found in corresponding age-matched controls.

Impact of Visual Corticostriatal Loop Disruption on Neural Processing within the Parahippocampal Place Area.

UNLABELLED: The caudate nucleus is a part of the visual corticostriatal loop (VCSL), receiving input from different visual areas and projecting back to the same cortical areas via globus pallidus, substantia nigra, and thalamus. Despite perceptual and navigation impairments in patients with VCSL disruption due to caudate atrophy (e.g., Huntington's disease, HD), the relevance of the caudate nucleus and VCSL on cortical visual processing is not fully understood. In a series of fMRI experiments, we found that the caudate showed a stronger functional connection to parahippocampal place area (PPA)

Transient tic disorder following carbon monoxide poisoning.

We report a 12-year-old male patient who developed transient motor and vocal tics twelve days after carbon monoxide (CO) poisoning. Cranial magnetic resonance image (MRI) of the patient showed bilateral symmetric hyperintensity in the caudate nucleus and putamen. Tic disorder was successfully treated with haloperidol. Thirty-three months after CO poisoning, the patient was asymptomatic and MRI revealed atrophy in caudate nucleus and putamen. The mechanism of tic disorder in CO intoxication is discussed.

Electrophysiological study of corticocaudate projections in cats.

The potential waves evoked in the caudate nucleus (CN) of cats by stimulation of the cerebral neocortex were sterotactically recorded. The head and the body of the caudate nucleus were systematically explored. Stimulation of the ipsilateral sigmoid gyrus and the orbitofrontal cortex evoked waves with the largest amplitude in the CN. Smaller potentials were evoked from the ipsilateral ectosylvian and suprasylvian gyri and from the sigmoid gyrus on the contralateral side. Antidromic conduction from the caudate nucleus to the cortex demonstrated the directness of the corticocaudate pathway. By st

Aneurysm of Distal Medial Lenticulostriate Artery: Report of 2 Cases.

BACKGROUND: Aneurysms originating from the distal portion of the lenticulostriate artery (LSA) are uncommon. Distal medial LSA (MLSA) aneurysms are particularly uncommon when compared with distal lateral LSA aneurysms, and their clinical features are unclear. Here, we present 2 patients with aneurysms of the distal MLSA who exhibited hemorrhages of the caudate nucleus and intraventricular region (intraventricular hemorrhage [IVH]). CASE DESCRIPTION: Patient 1 is a 50-year-old woman who was admitted to our hospital because of a hemorrhage in the left caudate nucleus and ventricles. She underwe

Longitudinal metabolite changes in Huntington's disease during disease onset.

BACKGROUND: Previous cross-sectional magnetic resonance spectroscopy (MRS) studies in Huntington's disease (HD) have demonstrated differences in metabolite concentrations in several regions of interest, especially the putamen and caudate nucleus. OBJECTIVE: To assess metabolite changes in both premanifest and early HD over a two year follow up period using MRS at 7 Tesla in several regions of interest. METHODS: In 13 HD gene carriers (10 premanifest and 3 manifest HD) proton MRS was performed at baseline and after 24 months. At follow up, four of the premanifest HD gene carriers had progress

Striatal FP-CIT uptake differs in the subtypes of early Parkinson's disease.

In idiopathic Parkinson's disease (PD), a tremor-dominant type (TDT), an akinetic-rigid type (ART), and a mixed type (MT) are distinguished. We compared cerebral [I-123]FP-CIT SPECT in the PD subtypes (67 patients Hoehn and Yahr stage 1:26 with ART, 19 with MT, 22 with TDT). We measured the ratios putamen/occipital lobe binding and caudate nucleus/occipital lobe binding. Parkinsonian motor symptoms were quantified by UPDRS motor scale. In both putamen and caudate nucleus contralateral to the clinically affected body side TDT patients showed a significantly higher FP-CIT uptake than ART or MT p

Positron emission tomography study of human narcolepsy: no increase in striatal dopamine D2 receptors.

We investigated dopamine D2 receptors in the caudate nucleus and putamen with positron emission tomography in seven patients with narcolepsy and seven healthy controls by using [11C]raclopride as a ligand. We applied an equilibrium method and Scatchard principle to give a quantitative estimation of the number (Bmax) and dissociation constant (Kd) of the receptors. Both the Bmax and Kd were within the normal range in the caudate nucleus and putamen in narcoleptic patients. We found no evidence for increased D2 receptor binding in narcolepsy.

MPTP-induced parkinsonism in primates: pattern of striatal dopamine loss following acute and chronic administration.

We analyzed the effect of two different schedules of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) treatment on dopaminergic systems in the striatum of cynomolgus monkeys. Acute MPTP treatment produced a marked dopamine (DA) depletion, more severe in the caudate nucleus than in the putamen. Chronic MPTP induced a more pronounced reduction in DA levels, the putamen being slightly more affected than the caudate nucleus, in accord with immunohistochemical findings that showed a higher loss of tyrosine-hydroxylase positive neurons in ventral subpopulations of the substantia nigra pars compac

Trust versus paranoia: abnormal response to social reward in psychotic illness.

Psychosis is characterized by an elementary lack of trust in others. Trust is an inherently rewarding aspect of successful social interactions and can be examined using neuroeconomic paradigms. This study was aimed at investigating the underlying neural basis of diminished trust in psychosis. Functional magnetic resonance imaging data were acquired from 20 patients with psychosis and 20 healthy control subjects during two multiple-round trust games; one with a cooperative and the other with a deceptive counterpart. [Bilateral hyperintensity of the pulvinar and dorsomedial nucleus of the thalamus in sporadic Creutzfeldt-Jakob disease].

INTRODUCTION: Creutzfedt-Jakob Disease (CJD) is a rapidly progressive neurodegenerative disease caused by prions. Early diagnosis and the determination of its form are epidemiologically important, with strong impact on public health. Bilateral pulvinar hyperintensity, either alone (pulvinar sign) or in association with the dorsomedial nucleus of the thalamus (double hockey stick sign) on T2, FLAIR and diffusion weighted imaging (DWI), is a criterion for the probable diagnosis of the variant CJD (vCJD). Bilateral hyperintensity of the caudate, putamina and cortex is the usual pattern found in t
