CD4+ T cells cross-compete for MHC class II-restricted peptide antigen complexes on the surface of antigen presenting cells.

CD4(+) T cells are activated upon recognition of peptide antigen in the context of MHC class II molecules, expressed by specialized APC. In this study, we show that CD4(+) T cells cross-compete for antigenic complexes on the surface of APC, inhibiting activation of other potentially reactive T cells of the same and differing specificities. T cells with either a higher affinity receptor for antigen or which have undergone prior activation compete more efficiently than low affinity or resting T cells. This implies that T-cell avidity for the APC is primarily responsible for the competitive advan

Antagonistic Effects of Actin-Specific Toxins on Salmonella Typhimurium Invasion into Mammalian Cells.

Competition between bacterial species is a major factor shaping microbial communities. It is possible but remains largely unexplored that competition between bacterial pathogens can be mediated through antagonistic effects of bacterial effector proteins on host systems, particularly the actin cytoskeleton. Using Salmonella Typhimurium invasion into cells as a model, we demonstrate that invasion is inhibited if the host actin cytoskeleton is disturbed by actin-specific toxins, namely, Vibrio cholerae MARTX actin crosslinking (ACD) and Rho GTPase inactivation (RID) domains, Photorhabdus luminesc

Antagonistic effects of actin-specific toxins on Salmonella Typhimurium invasion into mammalian cells.

Competition between bacterial species is a major factor shaping microbial communities. In this work, we explored the hypothesis that competition between bacterial pathogens can be mediated through antagonistic effects of bacterial effector proteins on host systems, particularly the actin cytoskeleton. Using Salmonella Typhimurium invasion into cells as a model, we demonstrate that invasion is inhibited if the host actin cytoskeleton is disturbed by any of the four tested actin-specific toxins: Vibrio cholerae MARTX actin crosslinking and Rho GTPase inactivation domains (ACD and RID, respective

Cancer susceptibility and reproductive trade-offs: a model of the evolution of cancer defences.

The factors influencing cancer susceptibility and why it varies across species are major open questions in the field of cancer biology. One underexplored source of variation in cancer susceptibility may arise from trade-offs between reproductive competitiveness (e.g. sexually selected traits, earlier reproduction and higher fertility) and cancer defence. We build a model that contrasts the probabilistic onset of cancer with other, extrinsic causes of mortality and use it to predict that intense reproductive competition will lower cancer defences and increase cancer incidence. We explore the tr

Exploring regulatory mechanisms of CD8+ T cell contraction.

A small fraction of the CD8 T cell effector population that responds to an infection progresses through the contraction phase to the memory stage. The factors regulating the extent of contraction are poorly understood. Competition for limited resources has been widely postulated to be the cause of cell death during the contraction phase, but our data show that competition does not affect contraction kinetics. We go on to demonstrate that all effector cells present at the peak of the response have the potential to become bona fide memory cells, thus excluding selection on the basis of functiona

Can Match-Mimicking Intermittent Practice Be Used as a Simulatory Training Mode of Competition Using Olympic Time Frame in Elite Taekwondo Athletes?

Aim: The physiological realities between Taekwondo (TKD) simulation kicking training and TKD competition according to the Olympic time frame remain unclear. The purpose of this study is to establish an Olympic match-simulated kicking model and compare its effects with real TKD competition on physiological challenges and hormonal responses during serial matches in elite athletes. Method: Sixteen elite TKD athletes randomly were assigned into either a TKD match-simulated kicking group (TMSK; N = 8, age: 21.3 ± 0.2 years) or a simulated TKD competition group (STC; N = 8, age: 21.6 ± 0.5 years). Murine cytomegalovirus strains co-replicate at multiple tissue sites and establish co-persistence in salivary glands in the absence of Ly49H-mediated competition.

Infection with multiple genetically distinct strains of pathogen is common and can lead to positive (complementation) or negative (competitive) within-host interactions. These interactions can alter aspects of the disease process and help shape pathogen evolution. Infection of the host with multiple strains of cytomegalovirus (CMV) occurs frequently in humans and mice. Profound, NK-cell-mediated (apparent) competition has been identified in C57BL/6 mice, and prevented the replication and shedding of certain co-infecting CMV strains. However, the frequency of such strong competition has not bee

Gene cloning using cDNA libraries in a differential competition hybridization strategy: application to cloning XP-A related genes.

A competition hybridization strategy using size-cut cDNA libraries as both probe and competitor was designed for the cloning of genes whose mRNAs are either regulated transcriptionally or vary in abundance as a function of cell line or cell cycle. We used this strategy to construct cDNA libraries from a particular size fraction of mRNA from three members of a xeroderma pigmentosum (XP), complementation group A family. Size-cut cDNA libraries derived from the father's, mother's, and child's fibroblast cell line were used in a competition scheme to screen two lambda gt11 human cDNA libraries. Of

Inhibition of immunological components by antigenic competition and other immuno-depressing procedures.

Four experimental procedures leading to lower antibody production, namely administration of a minute amount of antigen or excess of antigen, competition with an unrelated antigen and treatment with methotrexate, were studied for their effect on each of the forms of immunological responses in guinea-pigs to a DNP-BSA conjugate. It was found that delayed hypersensitivity and 2-ME sensitive antibody production were more reduced by administration of excess of antigen or by competition than by giving a minute amount of antigen or methotrexate. On the other hand, IgG1 production was more suppressed

Carbamylated-oxidized LDL: proatherosclerotic effects on endothelial cells and macrophages.

AIM: Both oxidized LDL and carbamylated LDL are considered important for initiating atherosclerosis in patients with end-stage kidney disease through vascular endothelial cell dysfunction or injury. However their effects on each other and their relationship related to pro-atherosclerotic effects on endothelial cells and macrophages have not been investigated. In this study, we analyzed the competition between LDL carbamylation and oxidation, tested biological effects of carbamylated-oxidized LDL (coxLDL) toward the endothelial cells, assessed its ability to cause foam cell development, and det

Host-Like Conditions Are Required for T6SS-Mediated Competition among Vibrio fischeri Light Organ Symbionts.

Bacteria employ diverse competitive strategies to enhance fitness and promote their own propagation. However, little is known about how symbiotic bacteria modulate competitive mechanisms as they compete for a host niche. The bacterium Vibrio fischeri forms a symbiotic relationship with marine animals and encodes a type VI secretion system (T6SS), which is a contact-dependent killing mechanism used to eliminate competitors during colonization of the Euprymna scolopes squid light organ. Like other horizontally acquired symbionts, V. fischeri experiences changes in its physical and chemical envir

Competition/antagonism associations of biofilm formation among Staphylococcus epidermidis Agr groups I, II, and III.

Staphylococci have quorum-sensing (QS) systems that enable cell-to-cell communication, as well as the regulation of numerous colonization and virulence factors. The accessory gene regulator (Agr) operon is one of the Staphylococcus genus QS systems. Three groups (I, II, and III) are present in Staphylococcus epidermidis Agr operon. To date, it is unknown whether Agr groups can interact symbiotically during biofilm development. This study analyzed a symbiotic association among Agr groups during biofilm formation in clinical and commensal isolates. Different combinations among Agr group isolates

Receptor for pre-S1(21-47) component of hepatitis B virus on the liver cell: role in virus cell interaction.

Attachment of hepatitis B virus to a hepatoblastoma cell line (HepG2) was examined using a synthetic peptide corresponding to the pre-S1 (21-47) region of the envelope protein. Scatchard analysis revealed a single class binding site of Kd 104 +/- 27 nM/l and 5.4 +/- 1.2 x 10(5) sites per cell. Competition of HBV with pre-S1 peptides was dose dependent, and demonstrated it as the dominant binding site. In view of the suggested sequence homology between the peptide and IgA, cross-competition studies were carried out. The results indicate no direct role of IgA receptor in HBV binding. The recepto

Highly Invasive Listeria monocytogenes Strains Have Growth and Invasion Advantages in Strain Competition.

Multiple Listeria monocytogenes strains can be present in the same food sample; moreover, infection with more than one L. monocytogenes strain can also occur. In this study we investigated the impact of strain competition on the growth and in vitro virulence potential of L. monocytogenes. We identified two strong competitor strains, whose growth was not (or only slightly) influenced by the presence of other strains and two weak competitor strains, which were outcompeted by other strains. Cell contact was essential for growth inhibition. In vitro virulence assays using human intestinal epitheli

High Iron-Sequestrating Bifidobacteria Inhibit Enteropathogen Growth and Adhesion to Intestinal Epithelial Cells In vitro.

The gut microbiota plays an important role in host health, in particular by its barrier effect and competition with exogenous pathogenic bacteria. In the present study, the competition of Bifidobacterium pseudolongum PV8-2 (Bp PV8-2) and Bifidobacterium kashiwanohense PV20-2 (Bk PV20-2), isolated from anemic infant gut microbiota and selected for their high iron sequestration properties, was investigated against Salmonella Typhimurium (S. RNA-Seq reveals that Pseudomonas aeruginosa mounts growth medium-dependent competitive responses when sensing diffusible cues from Burkholderia cenocepacia.

Most habitats host diverse bacterial communities, offering opportunities for inter-species interactions. While competition might often dominate such interactions, little is known about whether bacteria can sense competitors and mount adequate responses. The competition sensing hypothesis proposes that bacteria can use cues such as nutrient stress and cell damage to prepare for battle. Here, we tested this hypothesis by measuring transcriptome changes in Pseudomonas aeruginosa exposed to the supernatant of its competitor Burkholderia cenocepacia. We found that P. aeruginosa exhibited significan

A hybrid multiscale model for predicting CAR-T therapy outcomes in solid tumors.

T cell distribution within tumors ("tumor hotness") critically determines the success of immunotherapy. However, despite numerous strategies to enhance intratumoral T cell accumulation - such as multi-target CAR-Ts and combinatorial approaches - limited mechanistic understanding of T cell-microenvironment interactions has constrained progress. To address this, we developed a mechanistic physiological model of the 3D tumor microenvironment (TME) to evaluate CAR-T performance under environmental fluctuations and across different infusion strategies. The model integrates key vascular (rolling, fi

Metabolic Competition-Driven Antitumor Immunity of glpX-Deficient Engineered Salmonella via Glucose Deprivation and STING Activation in Bladder Cancer.

Background: Bacteria-mediated cancer therapy is emerging as a promising approach due to its low cost, scalability, and sustained efficacy. While bacterial immunomodulation has been extensively studied, the role of bacterial metabolic competition, particularly for glucose, in tumor suppression remains unclear. We aimed to investigate how engineered Salmonella strains exploit metabolic competition to trigger antitumor immunity and improve therapeutic outcomes in bladder cancer models. Methods: Using genetic screening, we generated a gluconeogenesis-deficient Salmonella strain, the AISIH strain w

Enhanced fitness of adult spermatogonial stem cells bearing a paternal age-associated FGFR2 mutation.

Pathogenic de novo mutations increase with fathers' age and could be amplified through competition between genetically distinct subpopulations of spermatogonial stem cells (SSCs). Here, we tested the fitness of SSCs bearing wild-type human FGFR2 or an Apert syndrome mutant, FGFR2 (S252W), to provide experimental evidence for SSC competition. The S252W allele conferred enhanced FGFR2-mediated signaling, particularly at very low concentrations of ligand, and also subtle changes in gene expression. Mutant SSCs exhibited improved competitiveness in vitro and increased stem cell activity in vivo up

A Putative Lipoprotein Mediates Cell-Cell Contact for Type VI Secretion System-Dependent Killing of Specific Competitors.

Interbacterial competition is prevalent in host-associated microbiota, where it can shape community structure and function, impacting host health in both positive and negative ways. However, the factors that permit bacteria to discriminate among their various neighbors for targeted elimination of competitors remain elusive. We identified a putative lipoprotein (TasL) in Vibrio species that mediates cell-cell attachment with a subset of target strains, allowing inhibitors to target specific competitors for elimination. Here, we describe this putative lipoprotein, which is associated with the br
