Correlation of acetabular chondrocyte apoptosis with caspase-3 and Bcl-2 expression in developmental dislocations of the hip.

This study aimed to determine whether abnormal apoptosis is present in acetabular cartilage in early developmental dislocations of the hip (DDH), and if so, whether it is correlated with the expression of caspase-3 and Bcl-2. DDH was induced in 24 4-week-old New Zealand white rabbits. Acetabular cartilage specimens from the experimental and control groups were stained with hematoxylin and eosin (H&E). Animals from the experimental group developed acetabular dysplasia. Apoptotic chondrocytes were observed by ultrastructural electron microscopy and H&E. TUNEL assays revealed significantly greate

Cytoskeletal proteins connecting intermediate filaments to cytoplasmic and nuclear periphery.

Intermediate filaments (IFs), together with microtubules and microfilaments build up the cytoskeleton of most eukaryotic cells. Cytoplasmic IFs form a dense filament network radiating from the nucleus and extending to the plasma membrane. The association between the cytoplasmic and nuclear surfaces appears to provide a continuous link important for the organisation of the cytoplasm, for cellular communication, and possibly for the transport into and out of the nucleus. Cytoplasmic IFs approach the nuclear surface, thin fibrils seem to connect the IFs with the nuclear pore complexes and a direc

Single-cell morphodynamics predict cell fate decisions during mucociliary epithelial differentiation.

Cell state transitions underlie the emergence of diverse cell types and are traditionally defined by changes in gene expression. Yet these transitions also involve coordinated shifts in cell morphology and behavior, which remain poorly characterized in densely packed epithelia. We developed a quantitative live-imaging and computational framework to track thousands of individual cells over time in the rapidly differentiating Xenopus mucociliary epithelium (MCE). From segmentations and trajectories, we extracted dynamic features-cell and nuclear shape, movement, and position-to create a time-res

Intraflagellar transport-associated CCDC92 is required for spermiogenesis and male fertility in mice.

The differentiation of a round spermatid into a streamlined sperm cell involves a series of remarkable morphological changes, such as sperm head shaping and flagellum formation. However, the underlying mechanism of spermatid shaping remains unclear. In this study, we find that CCDC92 deficiency in mice leads to severe abnormalities of the sperm head and flagellum and causes male infertility. Ultrastructural analyses of testicular elongating Ccdc92 knockout spermatids reveal severely deformed manchette structures. The manchette defects impair the subsequent sperm nucleus elongation and acrosome

Morphometric comparison of the motor trigeminal neurons.

Comparative morphological and morphometric studies of the motor trigeminal nucleus (MTN) were carried out on the brains of an alligator, a human being and a rat for the first time. Differences in neurons regarding sizes, areas, numbers, column volumes, neuropil indices and circulatory ratios were found. The results showed that the positions and shapes of the MTN were different, and the shapes of motor neurons were different, too. The average area of motor trigeminal neurons was the largest in the alligator, compared with the area in the human being, and the rat; the number of neurons and the c

Two distinct populations of neurons expressing nitric oxide synthase mRNA in the female rat preoptic area: site specific changes induced by sex steroids.

Non-isotopic in situ hybridization histochemistry in the basal forebrain of gonadectomized juvenile female rats visualized neuronal nitric oxide synthase (nNOS) mRNA in two distinct cellular populations, one in the organum vasculosum of the lamina terminals (OVLT) and the other in the rostral preoptic area at the level of the anteroventral periventricular nucleus (rPOA). In the rPOA, digoxigenin-labeled nNOS mRNA positive cells were in close proximity to the cell body of gonadotropin-releasing hormone (GnRH) -immunoreactive neurons. In the OVLT, the labeled cells were in an area rich in GnRH f

RNA Concentration and Content in the Nucleoli and Cytoplasmic Rim in Differentiating Lymphocytes of Patients Suffering from B Chronic Lymphocytic Leukaemia - a Cytochemical Note.

Nucleolar RNA optical density (concentration) measurements at the single cell level indicated that differentiation of lymphocytes is accompanied by a slightly decreased nucleolar RNA concentration in contrast to the cytoplasmic rim around the nucleus. On the other hand, the nucleolar size was markedly reduced and the cytoplasmic rim surrounding the nucleus was reduced only weakly. Concerning the calculated rough estimate of the RNA content, the differentiation induced its larger decrease in the nucleoli than in the cytoplasmic rim. These observations indicated that the nucleolar RNA concentrat

Localization of brainstem motoneurons involved in dewlap extension in the lizard, Anolis equestris.

Dewlap extension is a characteristic component of inter- and intraspecific displays of Anolis lizards. Dewlap extension is accomplished by the contraction of ceratohyoid muscles associated with the hyoid apparatus. Retrograde transport of horseradish peroxidase (HRP) was used to investigate the brainstem origins of efferent fibers to the ceratohyoid muscles in Anolis equestris. Following application of HRP to the ceratohyoid muscle or to its nerve supply on one side of the throat, large polygonal motoneurons were found ipsilaterally in the vagal (Amb X) and glossopharyngeal (Amb IX) parts of n

Acute Toxicity of Profenofos on Hemato-Biochemical Parameters and Peripheral Erythrocytes in Banded Gourami (Trichogaster fasciata).

The indiscriminate and excessive use of pesticides ultimately reaches the aquatic ecosystems and results in detrimental impacts on fish and other aquatic species. The current study focused on elucidating the acute toxic effects of the organophosphate pesticide, profenofos, on various blood biomarkers in banded gourami (Trichogaster fasciata). Fish were subjected to different sub-lethal concentrations (0%, 5%, 10%, 20%, and 40% of the 96-h LC50 = 2972.7 μg/L) of profenofos for 1, 24, and 96 h. Fish exposed to profenofos induced a significant rise (p < 0.05) in glucose, white blood cell levels,

Connections of the dorsal nucleus of the lateral lemniscus: an inhibitory parallel pathway in the ascending auditory system?

This study examines the dorsal nucleus of the lateral lemniscus (DNLL) and its afferent and efferent connections. In Nissl-stained material, DNLL has three parts: dorsal, ventral, and lateral. Although each part contains neurons with similar Nissl patterns, the subdivisions may be distinguished by the size, shape, and orientation of the cells. The lateral DNLL contains a mixture of DNLL neurons and cells from the sagulum. Afferent connections to DNLL were investigated with anterograde axonal transport techniques. Bilateral inputs to DNLL arise from the anteroventral cochlear nucleus and latera

Immunohistochemical Localization of Somatostatin in the Brain of Chinese Alligator Alligator sinensis.

In this study, the regional distribution and histological localization of somatostatin (SS) immunoreactive (IR) perikarya and fibers was investigated for the first time in the brain of adult Chinese alligator by immunohistochemical method. The results showed SS-IR perikarya and fibers were widely distributed in various parts of the brain except for olfactory bulbs. In the telencephalon, SS-IR perikarya were predominantly located in the cellular layer and deep plexiform layer of dorsomedial and medial cortex, less in the dorsal and lateral cortex, while SS-IR fibers were found in all layers of

MSH6- or PMS2-deficiency causes re-replication in DT40 B cells, but it has little effect on immunoglobulin gene conversion or on repair of AID-generated uracils.

The mammalian antibody repertoire is shaped by somatic hypermutation (SHM) and class switch recombination (CSR) of the immunoglobulin (Ig) loci of B lymphocytes. SHM and CSR are triggered by non-canonical, error-prone processing of G/U mismatches generated by activation-induced deaminase (AID). In birds, AID does not trigger SHM, but it triggers Ig gene conversion (GC), a 'homeologous' recombination process involving the Ig variable region and proximal pseudogenes. Because recombination fidelity is controlled by the mismatch repair (MMR) system, we investigated whether MMR affects GC in the ch

Cyto-, dendro- and fibro-architectonic studies on the chicken hypothalamus.

Based on the chicken hypothalamic analysis using the Nissl, Golgi-Cox and silver staining methods, a first attempt to make the cyto-, dendro- and fibro-architectonic atlases of the frontal planes at the same level was performed. In the preparations stained with Nissl method, the hypothalamus was subdivisible basically into the preoptic, rostral and caudal hypothalamic regions, each of which was composed of medial and lateral parts respectively. A well-defined boundaries between the preoptic and rostral hypothalamic regions was decided at the level of the rostral margin of the commissura rostra

A novel subdivision of the bed nucleus of stria terminalis in monkey, rat, and mouse brains.

The bed nucleus of stria terminalis (BST) is a critical structure that mediates sustained vigilant responses to contextual, diffuse, and unpredictable threats. Dysfunction of the BST could lead to excessive anxiety and hypervigilance, which are often observed in posttraumatic stress disorder and anxiety disorders. Vigilance of potential future threats from the external environment is a basic brain function and probably requires rapid and/or short neural circuits, which enable both quick detection of the potential threats and fast adaptive responses. However, the BST in literature does not appe

Single-nucleus multimodal spatial transcriptomics reveals spatial colocalization of neoantigen-expressing tumor cells and cognate T cells.

Improved methods to identify therapeutically relevant tumor neoantigens and their cognate T cells would aid the development of precision medicines for cancer. Here, we developed Slide-GoTags, a droplet-based single-nucleus spatial transcriptomics approach that characterizes neoantigen-specific immunity by integrating targeted transcript genotyping and T cell receptor (TCR) sequencing with single-nucleus RNA sequencing from the same slice of frozen tissue. Application of Slide-GoTags to mouse and human tumors revealed colocalization of clonally expanded, neoantigen-specific T cells with tumor c

The Untouchable Ventral Nucleus of the Trapezoid Body: Preservation of a Nucleus in an Animal Model of Autism Spectrum Disorder.

Autism spectrum disorder (ASD) is a neurodevelopmental condition characterized by repetitive behaviors, poor social skills, and difficulties with communication and hearing. The hearing deficits in ASD range from deafness to extreme sensitivity to routine environmental sounds. Previous research from our lab has shown drastic hypoplasia in the superior olivary complex (SOC) in both human cases of ASD and in an animal model of autism. However, in our study of the human SOC, we failed to find any changes in the total number of neurons in the ventral nucleus of the trapezoid body (VNTB) or any chan

Deciphering the longitudinal trajectories of glioblastoma ecosystems by integrative single-cell genomics.

The evolution of isocitrate dehydrogenase (IDH)-wildtype glioblastoma (GBM) after standard-of-care therapy remains poorly understood. Here we analyzed matched primary and recurrent GBMs from 59 patients using single-nucleus RNA sequencing and bulk DNA sequencing, assessing the longitudinal evolution of the GBM ecosystem across layers of cellular and molecular heterogeneity. The most consistent change was a lower malignant cell fraction at recurrence and a reciprocal increase in glial and neuronal cell types in the tumor microenvironment (TME). The predominant malignant cell state differed betw

Location and morphology of parasympathetic preganglionic neurons in the sacral spinal cord of the cat revealed by retrograde axonal transport of horseradish peroxidase.

The distribution and morphology of preganglionic neurons in the sacral parasympathetic nucleus (SPN) of the cat have been studied with the horseradish peroxidase (HRP) tracing technique. HRP applied to the cut pelvic nerve was identified in cells located ipsilaterally, primarily in the intermediate gray matter. They formed a column approximately 10 mm long, usually contained within two, but occasionally three, sacral segments, S2 contained a majority of cells. In transverse sections the SPN had the appearance of an inverted "L". Cells were medium-sized, oval or spindle-shaped, and transversely

Postsynaptic targets of type II auditory nerve fibers in the cochlear nucleus.

Type II auditory nerve fibers, which provide the primary afferent innervation of outer hair cells of the cochlea, project thin fibers centrally and form synapses in the cochlear nucleus. We investigated the postsynaptic targets of these synapses, which are unknown. Using serial-section electron microscopy of fibers labeled with horseradish peroxidase, we examined the border of the granule-cell lamina in mice, an area of type II termination that receives branches having swellings with complex shapes. About 70% of the swellings examined with the electron microscope formed morphological synapses,
