The immune cells in modulating osteoclast formation and bone metabolism.

Osteoclasts are pivotal in regulating bone metabolism, with immune cells significantly influencing both physiological and pathological processes by modulating osteoclast functions. This is particularly evident in conditions of inflammatory bone resorption, such as rheumatoid arthritis and periodontitis. This review summarizes and comprehensively analyzes the research progress on the regulation of osteoclast formation by immune cells, aiming to unveil the underlying mechanisms and pathways through which diseases, such as rheumatoid arthritis and periodontitis, impact bone metabolism.

Corynebacterium jeikeium Dormant Cell Formation and Photodynamic Inactivation.

Pathogenic non-spore forming bacteria enter a dormant state under stressful conditions, which likely allows them to acquire resistance to various antibiotics. This work revealed the efficient formation of dormant "non-culturable" (NC) Corynebacterium jeikeium cells in stationary phase upon gradual acidification of the growth medium. Such cells were unable to form colonies and existed in a prolonged stationary phase. At an early stage of dormancy (approximately 14 days post-inoculation), dormant cells are able for resuscitation in liquid medium. However, those stored for long time in dormant st

Titan cell formation is unique to Cryptococcus species complex.

Members of the Cryptococcus species complex stand out by unique virulence factors that allowed evolutionary transition to pathogenesis. Among the factors contributing to cryptococcosis is a morphological transformation into giant (Titan) cells. It remains unclear whether species outside of the C. neoformans/C. gattii species complex are capable of titanization. We utilized two recently developed protocols that allow obtaining Titan cells in vitro to test if titanization occurs in non-C. neoformans/C. gattii species. We find that none of the tested strains, representing 10 species of basidiomyc

TRRAP is a central regulator of human multiciliated cell formation.

The multiciliated cell (MCC) is an evolutionarily conserved cell type, which in vertebrates functions to promote directional fluid flow across epithelial tissues. In the conducting airway, MCCs are generated by basal stem/progenitor cells and act in concert with secretory cells to perform mucociliary clearance to expel pathogens from the lung. Studies in multiple systems, including Xenopus laevis epidermis, murine trachea, and zebrafish kidney, have uncovered a transcriptional network that regulates multiple steps of multiciliogenesis, ultimately leading to an MCC with hundreds of motile cilia

Formation, release, and internalization of stable tau oligomers in cells.

Tau is a neuronal microtubule-binding protein that, in Alzheimer's disease and other neurodegenerative diseases, can form oligomeric and large fibrillar aggregates, which deposit in neurofibrillary tangles. Tau's physiological state of multimerization appears to vary across conditions, and a stable dimeric form of soluble tau has been suggested from experiments using recombinant tau in vitro. We tested if tau dimerization or oligomerization, also occurs in cells, and if soluble tau oligomers are relevant for the release and internalization of tau. We developed a sensitive tau split-luciferase

Bartonella-host-cell interactions and vascular tumour formation.

Bartonellae are arthropod-borne bacterial pathogens that typically cause persistent infection of erythrocytes and endothelial cells in their mammalian hosts. In human infection, these host-cell interactions result in a broad range of clinical manifestations. Most remarkably, bartonellae can trigger massive proliferation of endothelial cells, leading to vascular tumour formation. The recent availability of infection models and bacterial molecular genetic techniques has fostered research on the pathogenesis of the bartonellae and has advanced our understanding of the virulence mechanisms that un

Cell-mediated gene therapy for bone formation and regeneration.

Cell-mediated gene therapy is one of the new modalities branching out from the wide-ranging field of gene transfer and therapy. When applied to bone formation and regeneration, it has particular advantages depending on the type of cell used as a platform for gene delivery. When utilizing adult mesenchymal stem cells or osteoprogenitor cells for the expression of bone-promoting osteogenic factors, the cells not only express the factors promoting bone growth, but can respond, differentiate and participate in the bone formation process. The ability of engineered cells to respond to the transgene,

The role of Müller cells in the formation of the blood-retinal barrier.

We have compared the ability of Müller cells and astrocytes to induce the formation of barrier properties in blood vessels. Müller cells cultured from the rabbit retina, and astrocytes and meningeal cells cultured from the rat cerebral cortex, were injected into the anterior chamber of the rat eye, where they formed aggregates on the iris. We have examined the barrier properties of the vessels in those aggregates and, for comparison, the barrier properties of vessels in the retina, ciliary processes and iris. Two tracers were perfused intravascularly to test barrier properties. The movement of

Formation and release of dinitrosyl iron complexes by endothelial cells.

The release of dinitrosyl non-heme iron complexes from cytotoxic macrophages accounts for NO-mediated iron loss. We have now investigated whether or not a similar mechanism operates in endothelial cells. Following stimulation with bradykinin or calcium ionophore A23187 NO and intracellular dinitrosyl iron complexes were detected by ESR spectroscopic analysis of frozen cells. In addition, endothelial cells released dinitrosyl iron complexes which bound to extracellular albumin. In transferrin and iron-free medium stimulation of endothelial cells by bradykinin or thimerosal resulted in a loss of

Membrane formations in the pineal cells of the teleost Gambusia affinis.

Highly organized membrane formations in the cytoplasm of photoreceptor and supporting cells of the pineal organ of Gambusia affinis are described. These membranous structures show different characteristics depending on the cell type where they are located. Some of the morphological characteristics of these membranous formations are similar to those of myeloid bodies, but their presence in the photoreceptor cells raises some doubt about their being myeloid bodies. The three-dimensional interpretation of these structures is discussed.

T-cell independent transfer of organized granuloma formation.

A direct role for delayed hypersensitivity in organized granuloma formation has been emphasized in the literature, but some clinical and experimental studies have suggested a more complicated relationship. In order to further evaluate T-cell function in cutaneous granulomatous inflammation, hepatic granulomas isolated from athymic mice with schistosomiasis were grafted into the skin of athymic and euthymic mice. Granulomas developed in athymic mice, as seen previously after transplantation of euthymic hepatic granulomas, indicating that granulomas develop in the skin without T-cell function. H

Candida albicans Goliath cells pioneer biofilm formation.

Candida albicans is a fungal pathogen that can cause life-threatening bloodstream infections. Indwelling medical devices, such as catheters, play a critical role in their progression, as C. albicans can colonize these devices and form biofilms, which can then seed the bloodstream, resulting in recurrent candidemia. Under conditions of zinc restriction, C. albicans forms enlarged Goliath cells. Here, we show that Goliath cells exhibit enhanced adhesion to host tissue under static conditions, particularly to vaginal and oral epithelial cell monolayers. Moreover, Goliath cells are fully capable o

Basal Cell Carcinoma of the Eyelid With Metaplastic Bone Formation.

Basal cell carcinoma accounts for 90% of malignant tumors of the eyelid. Basal cell carcinoma has been reported to rarely occur in conjunction with osteoma cutis or bone formation in the skin. The mechanism of this secondary osteoma cutis has yet to be explained. Herein, the authors present the case of a 68-year-old woman with a rapidly enlarging basal cell carcinoma with secondary osteoma cutis of the left lower eyelid.

[The role of epithelial cells in the formation and development of nasal polyps].

Nasal polyps, as a subtype of chronic sinusitis, is prone to clinical recurrence and seriously affects the quality of life of patients, but its pathogenesis is unclear. It is considered to be a chronic inflammation of the nasal-sinus mucosa under the combined action of multiple factors. Among them, nasal mucosal epithelial injury and shedding are the main pathological features. Nasal-sinus epithelial separated nasal sinuses with the outside world, is the complex natural biological barrier to resist external stimuli and the key to maintaining homeostasis in the nasal sinuses. Trichinella: what is going on during nurse cell formation?

Recent molecular studies have revealed that many genes are mobilized during nurse cell formation, including those involved in activation and proliferation of satellite cells, dedifferentiation, cell cycle re-entry and arrest, apoptosis and transformation. This article reviews the kinetics of gene expression from a cellular biology point-of-view in an effort to dissect the complex events that lead to unusual pathological changes after a Trichinella infection.

Formation and toxicity of soluble polyglutamine oligomers in living cells.

BACKGROUND: Aggregation and cytotoxicity of mutant proteins containing an expanded number of polyglutamine (polyQ) repeats is a hallmark of several diseases, including Huntington's disease (HD). Within cells, mutant Huntingtin (mHtt) and other polyglutamine expansion mutant proteins exist as monomers, soluble oligomers, and insoluble inclusion bodies (IBs). Determining which of these forms constitute a toxic species has proven difficult. Recent studies support a role for IBs as a cellular coping mechanism to sequester levels of potentially toxic soluble monomeric and oligomeric species of mHtt
