Molecular insights into the hydrolysis and transglycosylation of a deep-sea Planctomycetota-derived GH16 family laminarinase.

The biochemical and structural characteristics of PtLam, a laminarinase from deep-sea Planctomycetota, have been extensively elucidated, unveiling the fundamental molecular mechanisms governing substrate recognition and enzymatic catalysis. PtLam functions as an exo-laminarinase with the ability to sequentially hydrolyze laminarin, cleaving glucose units individually. Notably, PtLam exhibits proficient transglycosylation capabilities, utilizing various sugar alcohols as acceptors, with lyxose, in particular, yielding exclusively transglycosylated products. Structural analysis of both apo-PtLam

Evaluation of organosolv pretreatment on the structural characteristics of lignin polymers and follow-up enzymatic hydrolysis of the substrates from Eucalyptus wood.

In the present study, Eucalyptus was subjected to organosolv pretreatment processes with aqueous 2-propanol at 200-220°C to obtain lignin with benign characters for its valorization and digestible substrates for bioethanol production. Results showed that different delignification ratios (64.00%-81.26%) and molecular weights (Mw=610-2680g/mol) of lignin fractions were dissociated from various pretreatment conditions, and the glucose yields of all the pretreated substrates significantly increased to 54.65-88.59% as compared to that of raw material (9.20%). Additionally, the amounts of β-O-4, β-β

Substrate specific synergism between CTec 2 and galactoglucomannan-hydrolysing enzymes from Penicillium rotoruae improves softwood saccharification.

The substrate-specific synergistic action of hemicellulolytic enzymes supplemented to the commercial cellulase cocktail CTec 2 was demonstrated using softwood galactoglucomannan (GGM) containing substrates. Submerged fermentation of the novel fungus Penicillium rotoruae produced three key GGM-hydrolysing enzymes, namely β-mannanase, β-mannosidase, and α-galactosidase, which were partially purified using sequential ultrafiltration and fast protein liquid chromatography (FPLC). A systematic enzyme combination screen was employed to evaluate dual and triple enzyme combinations supplemented to CTe

Mitigation effects of solar-driven metalloporphyrins on CO2 and N2O emissions from an agricultural soil.

Developing green technologies to mitigate soil carbon dioxide (CO2) and nitrous oxide (N2O) emissions is critical for sustainable agriculture. Here, we investigated the effects of two water-soluble metalloporphyrins (FeTPPS and MnTPPS), with and without glucose addition, on CO2 and N2O emissions and the associated mechanisms in an agricultural soil under natural solar irradiation. Metalloporphyrin effects depended on metal identity and labile carbon (C) availability. FeTPPS alone significantly reduced cumulative CO2 emissions by 9.9%, and FeTPPS with glucose reduced emissions by 7.2% relative

A thermostable and inhibitor resistant β-glucosidase from Rasamsonia emersonii for efficient hydrolysis of lignocellulosics biomass.

The present study reports a highly thermostable β-glucosidase (GH3) from Rasamsonia emersonii that was heterologously expressed in Pichia pastoris. Extracellular β-glucosidase was purified to homogeneity using single step affinity chromatography with molecular weight of ~ 110 kDa. Intriguingly, the purified enzyme displayed high tolerance to inhibitors mainly acetic acid, formic acid, ferulic acid, vanillin and 5-hydroxymethyl furfural at concentrations exceeding those present in acid steam pretreated rice straw slurry used for hydrolysis and subsequent fermentation in 2G ethanol plants. Decomposition mechanisms of Pinus koraiensis logs in a mixed broad leaved-Korean pine forest.

We analyzed the dynamic changes of the decomposition of fallen logs of Pinus koraiensis with different decay classes (ⅰ-ⅴ) and diameter classes (Ⅰ-Ⅲ) following the space-for-time substitution method. We further elucidated the decomposition mechanism by examining the interrelationships of chemical components, microbial communities, and related degradation enzyme activities during the decomposition process. The results showed that lignocellulose contents and the activities of related degrading enzymes were significantly varied across decay class and diameter class. The contents of lignin, cellul

Synergistic microwave enzymatic immersion pretreatment of apple slices: Effects on mass transfer, enzyme activity, and digestive bioaccessibility.

Herein, we identified an optimal parameter combination for microwave-assisted cellulase immersion (MA-CI) based on the retention of water-soluble bioactive compounds and the extent of cell wall enzymatic hydrolysis. MA-CI showed cell wall remodeling and improved mass transfer, while MA9-2CI30 produced a more uniform pore network, facilitating moisture migration and limiting nutrient loss. Based on these results, the effects of penetrative microwave blanching (PMB) on the physicochemical properties and water migration of apple slices pretreated with MA-CI were examined. Results revealed that PM

Structural Regulation of Lignin Using an Al3+/Zn2+ Bimetallic Lewis Acid System to Enhance Enzymatic Saccharification.

This study employed AlCl3/ZnCl2 bimetallic salt catalysis with ethanol organic solvent for sugar cane bagasse pretreatment. The process retained 80.12% cellulose while removing ∼70% xylan and 80.56% lignin, achieving 90.18% glucose conversion during enzymatic hydrolysis. Comprehensive characterization and DFT calculations revealed a synergistic dual-action mechanism: strong Lewis acid Al3+ extensively cleaved β-O-4 bonds (reducing them to 40.68%), whereas weak Lewis acid Zn2+ interacted broadly with side chains and aromatic rings. This synergy facilitated substantial lignin removal and minimiz

Construction of lignin humic-like biochar composites for intensifying thermophilic biohydrogen production from lignocellulosic residues.

Dark fermentative hydrogen production from lignocellulosic residues is often limited by inefficient electron distribution and competing metabolic pathways. In this study, a lignin-derived humic substance-biochar (LHS-BC) was developed to enhance thermophilic hydrogen production from lignocellulosic residues. Among the tested materials, the lignin-derived humic-like substance obtained via alkaline oxidative humification and subsequently combined with biochar (HSH@BC) exhibited the best performance, increasing hydrogen production (mL/L) by 29.76% compared with the control. Kinetic analysis showe

[Use of the presence of cellulose in cellular wall of Acanthamoeba cysts for diagnostic purposes].

Species identification within the genus Acanthamoeba is based predominantly on morphological and biochemical features. It is labor-intensive and requires cloning and axenization. We described a novel immunocytochemical method for the identification of Acanthamoeba spp. based on selective binding of Clostridium cellulovorans cellulase to protozoan cyst wall cellulose. Free-living amoebae isolated from different water sources by filtration and subsequent cultivation on non-nutrient agar were assigned to genera Acanthamoeba, Naegleria or Hartmannella using morphological taxonomic criteria. [Experimental induction of rotund bodies in Thermus equaticus].

With several isolates of T. aquaticus rotund bodies (rbs) have been artificially induced. The most active agents were lysozyme, pronase (not trypsin), cellulase (not alpha-amylase or beta-glucosidase), penicillin and some mineral salts. Two modifications of rbs were observed: the vesticular type arising from single cells or filaments and the aggregate-rbs resulting from secondary associations of several cells or filaments. In both cases the primary event is a partial detachment of the outer layer of cell envelopes followed by its swelling to form a hyaline bleb. Within these osmotically stable

Sitosterol-beta-glucoside as primer for cellulose synthesis in plants.

Cellulose synthesis in plants requires beta-1,4-glucan chain initiation, elongation, and termination. The process of chain elongation is likely to be distinct from the process of chain initiation. We demonstrate that a CesA glucosyltransferase initiates glucan polymerization by using sitosterol-beta-glucoside (SG) as primer. Cotton fiber membranes synthesize sitosterol-cellodextrins (SCDs) from SG and uridine 5'-diphosphate-glucose (UDP-Glc) under conditions that also favor cellulose synthesis. The cellulase encoded by the Korrigan (Kor) gene, required for cellulose synthesis in plants, may fu

Enhanced high-solids hydrolysis of pretreated wheat straw at low enzyme dosage via dual mechanisms of lignin surface passivation and aqueous environment modulation.

The enzymatic hydrolysis of lignocellulosic biomass under high-solid loading is hindered by inefficient mass transfer and high enzyme dosage. In this study, two polymeric additives, polyvinyl alcohol (PVA) and polyethylene glycol 4000 (PEG4000), were employed to enhance the saccharification of dilute-acid pretreated wheat straw (PWS) under high-solid loading (15% w/w) and low enzyme dosage (2.7 FPU/g dry substrate). Using a multi-scale characterization approach, these PVA and PEG4000 were found to overcome different hydrolysis barriers through distinct mechanisms. PVA primarily acts through co

In vivo application of potent probiotics for enhancing potato growth and controlling Ralstonia solanacearum and Fusarium oxysporum infections.

Plant probiotics are live microbial cells or cultures that support plant growth and control plant pathogens through different mechanisms. They have various effects on plants, including plant growth promotion through the production of indole acetic acid (IAA), biological control activity (BCA), and production of cellulase enzymes, thus inducing systemic resistance and increasing the availability of mineral elements. The present work aimed to study the potential of Achromobacter marplatensis and Bacillus velezensis as plant probiotics for the field cultivation of potatoes. In vitro studies have

Enhanced enzymatic hydrolysis of eucalyptus by synergy of zinc chloride hydrate pretreatment and bovine serum albumin.

Enhancement of eucalyptus enzymatic saccharification by synergy of ZnCl2 hydrate pretreatment and bovine serum albumin (BSA) was investigated in this study. The result showed that the ZnCl2 hydrate pretreatment could not only selectively extract up to ∼100% of the hemicellulose from eucalyptus, but also convert portion of high crystalline cellulose I into low crystalline cellulose II, which both beneficial for enhancing subsequent pretreated solids enzymatic saccharification. The addition of BSA into enzymatic hydrolysis step could significantly promote the glucose release from pretreated soli
