Intestinal interleukin-10 mobilization as a contributor to the anti-arthritis effect of orally administered madecassoside: a unique action mode of saponin compounds with poor bioavailability.

Madecassoside, a triterpenoid saponin present in Centella asiatica herbs with extremely low bioavailability, possesses excellent anti-rheumatoid arthritis property after oral administration. Such a disconnection between poor pharmacokinetic property and undoubted bioactivity also exists in many other herbal medicines. However, there is no reasonable explanation for this phenomenon to date. Here we showed that orally administered madecassoside displayed marked therapeutic effect on collagen-induced arthritis (CIA) in rats, which was accompanied by a systemic downregulation of inflammatory cytok

Extracellular vesicles derived from the lactic acid bacterium Enterococcus rotai suppress melanogenesis by modulating the cAMP-MITF pathway.

Enterococcus rotai CMTB-CA6-derived extracellular vesicles (CA6-ErEVs), isolated from a Centella asiatica-associated lactic acid bacterium, were investigated for their potential to modulate oxidative stress, inflammation, and melanogenesis in skin cells. CA6-ErEVs were purified and characterized using standardized nanoparticle analyses. Functional assays demonstrated that CA6-ErEVs displayed robust antioxidant activity by reducing free radical accumulation and intracellular reactive oxygen species levels. In tumor necrosis factor-α-stimulated dermal fibroblasts, CA6-ErEVs attenuated the secret

Asiaticoside attenuates oxygen-glucose deprivation/reoxygenation-caused injury of cardiomyocytes by inhibiting autophagy.

Asiaticoside is a natural triterpene compound derived from Centella asiatica, possessing confirmed cardioprotective property. However, the roles of asiaticoside in regulating oxygen-glucose deprivation/reoxygenation (OGD/R)-caused cardiomyocyte dysfunction remain largely obscure. Human cardiomyocyte AC16 cells were stimulated with OGD/R to mimic myocardial ischemia/reperfusion injury and treated with asiaticoside. Cytotoxicity was investigated by CCK-8 assay and lactate dehydrogenase (LDH) release analysis. Autophagy- and Wnt/β-catenin signaling-related protein levels were measured via western

Review of neuroprotective potential of natural products against hypoxia-induced neuronal injury.

Neurological disorders such as neurodegenerative diseases (NDDs) and stroke have become a major global health burden. Evidences from several studies suggest that their pathogenesis is related to hypoxia. However, there are certain limitations and adverse effects associated with the current treatments for neurological disorders. Studies have shown that some natural products and their extracts-such as (-)-epigallocatechin-3-gallate, Centella asiatica, ginkgolides, quercetin, berberine, and curcumin, which are the focus of this paper, along with briefly mentioned resveratrol and compounded prepar

Asiaticoside nitric oxide gel accelerates diabetic cutaneous ulcers healing by activating Wnt/β-catenin signaling pathway.

Diabetic ulcers, gangrene, local infections and other traumatic symptoms of wound healing are all directly related. Promoting the early healing of diabetic cutaneous ulcers (DCU) and reducing the disability and treatment costs is an important research project integrating traditional Chinese and Western medicine. Nitric oxide (NO) is a key component of wound healing, and endogenous NO secretion is insufficient during the development of DCU. It has been reported that exogenous NO can promote wound healing, but exogenous NO has a short half-life and is difficult to adhere to the skin. Asiaticosid

Development and evaluation of madecassoside-loaded microemulsion for enhanced topical drug delivery.

Madecassoside, a major triterpenoid glycoside from Centella asiatica, has potential value for dermatological applications; however, its poor stratum corneum permeability is due to high hydrophilicity and large molecular size. This work focused on designing a microemulsion carrier to enhance the topical delivery of madecassoside. Formulation regions were first screened using pseudo-ternary phase diagrams, followed by preparation of madecassoside-loaded systems with different oil phases, surfactant HLB values, cosurfactant compositions, and drug loadings. The resulting formulations were characte
