Endothelin-1-like immunoreactivity in cerebral cortex of Alzheimer-type dementia.

1. Endothelin-1-like immunoreactivity (ET-1-LI) in cerebral cortex in postmortem brains obtained from patients with Alzheimer-type dementia (ATD) was measured by enzyme-immunoassay. 2. The ET-1-LI in the ATD brains was significantly increased in frontal and occipital cortex than those in the control brains and a significant correlation was found between frontal and temporal lobe of ATD brains. 3. These findings may explain the clinico-radiological results that the cerebral blood flow is decreased in ATD patients, the mechanism of which is still unknown.

[Glutamate metabolism in cerebral cortex obtained from chronic hepatic failure rats].

The present investigation was carried out in order to elucidate the amino acid metabolism in hepatic failure with particular emphasis placed on glutamate. For this purpose, chronic hepatic failure models were produced in adult male Wistar rats by successive carbontetrachloride injection (0.20 ml/100 g. B. W., twice/week) for 13 weeks. They were confirmed to develop chemical changes compartible with hepatic failure, showing markedly elevated serum levels of NH3, GOT and ALP. Animals were killed by decapitation during fasting and the brains were removed immediately. After the parietal cortical s

Human cerebral cortex organization characterized by functional PET-FDG "Metabolic Connectivity".

PURPOSE: In this study, we characterize the spatiotemporal organization of resting-state metabolic connectivity (RSMC) in the human brain, as measured by [18F]-fluorodeoxyglucose (FDG) functional PET (fPET-FDG). We examine the relationship between RSMC organization and resting-state functional connectivity (RSFC) derived from functional magnetic resonance imaging and other known cortical organizational principles. METHODS: Resting-state fPET-FDG data from 24 individuals were obtained from a publicly available repository (Monash rsPET-MR Dataset). We characterized local metabolic organization

Connectivity-based parcellation of normal and anatomically distorted human cerebral cortex.

For over a century, neuroscientists have been working toward parcellating the human cortex into distinct neurobiological regions. Modern technologies offer many parcellation methods for healthy cortices acquired through magnetic resonance imaging. However, these methods are suboptimal for personalized neurosurgical application given that pathology and resection distort the cerebrum. We sought to overcome this problem by developing a novel connectivity-based parcellation approach that can be applied at the single-subject level. Utilizing normative diffusion data, we first developed a machine-le

Cholinergic Switch between Two Types of Slow Waves in Cerebral Cortex.

Sleep slow waves are known to participate in memory consolidation, yet slow waves occurring under anesthesia present no positive effects on memory. Here, we shed light onto this paradox, based on a combination of extracellular recordings in vivo, in vitro, and computational models. We find two types of slow waves, based on analyzing the temporal patterns of successive slow-wave events. The first type is consistently observed in natural slow-wave sleep, while the second is shown to be ubiquitous under anesthesia. Network models of spiking neurons predict that the two slow wave types emerge due

Individual Variability of the Human Cerebral Cortex Identified Using Intraoperative Mapping.

BACKGROUND: Intraoperative functional cortical mapping using direct electrical stimulation may show a wider individual variability than suggested by noninvasive imaging data of healthy subjects. METHODS: We assessed intraoperative variability of the frontal eye fields and the speech arrest sites in adult patients who underwent awake craniotomy with direct electrostimulation for treatment of diffuse gliomas located within eloquent regions, and we compared findings with human cortical parcellation of the Human Connectome Project. RESULTS: The frontal eye fields were defined by intraoperative d

Teaching neuroimages: Pseudohypertrophic cerebral cortex in end-stage Creutzfeldt-Jakob disease.

A 43-year-old woman presented with 1 month of progressive lower limb burning sensation, blurred vision, and gait disturbance. Her mother died of Creutzfeldt-Jakob disease (CJD). Neurologic examination revealed only cerebellar ataxia. EEG revealed periodic generalized discharges. MRI (figure 1) showed FLAIR hyperintensity in caudate and lenticular nuclei. Molecular analysis confirmed genetic CJD (PRNP E200K mutation). One month later, she became comatose. After 10 months, EEG showed diffuse background flattening without periodic abnormalities, and MRI (figure 2) disclosed diffuse pseudohypertro

Energy metabolism of rat cerebral cortex, hypothalamus and hypophysis during ageing.

Ageing is one of the main risk factors for brain disorders. According to the neuroendocrine theory, ageing modifies the sensitivity of hypothalamus-pituitary-adrenal axis to homoeostatic signals coming from the cerebral cortex. The relationships between the energy metabolism of these areas have not been considered yet, in particular with respect to ageing. For these reasons, this study was undertaken to systematically investigate in female Sprague-Dawley rats aged 4, 6, 12, 18, 24, 28 months and in 4-month-old male ones, the catalytic properties of energy-linked enzymes of the Krebs' cycle, el

N-acetylaspartic acid promotes oxidative stress in cerebral cortex of rats.

N-acetylaspartic acid accumulates in Canavan Disease, a severe leukodystrophy characterized by swelling and spongy degeneration of the white matter of the brain. This inherited metabolic disease, caused by deficiency of the enzyme aspartoacylase, is clinically characterized by severe mental retardation, hypotonia and macrocephaly, and also generalized tonic and clonic type seizures in about half of the patients. Considering that the mechanisms of brain damage in this disease remain not fully understood, in the present study we investigated whether oxidative stress is elicited by N-acetylaspart

Distribution of N-cadherin in human cerebral cortex during prenatal development.

An important subgroup of adhesion molecules is the superfamily of cadherins, which takes part in cell recognition and differentiation during development. To our knowledge only one study describing N-cadherin expression in developing human brain has been performed so far. Our aim is to identify N-cadherin expression to establish a relationship between its expression and function in human cerebral cortex during prenatal development. In the present study, localization and intensity of N-cadherin was investigated in developing cerebral cortex. Fetuses from spontaneous abortions (n=13) were obtaine

GABAA receptors in damaged cerebral cortex areas in human chronic alcoholics.

Chronic alcoholics without complicating disease showed greater densities of GABA agonist sites (labelled with the selective ligand [3H]muscimol) on the GABAA receptor in the superior frontal gyrus, in comparison with both precentral gyrus in the same cases, and with superior frontal gyrus in matched controls. Whereas cases with concomitant Wernicke encephalopathy may also have had greater numbers of superior frontal [3H]muscimol binding sites than controls, alcoholics with cirrhosis of the liver showed more muted differences. Since the GABAA receptor is a multimeric complex which also possesse

How does the cerebral cortex work? Development, learning, attention, and 3-D vision by laminar circuits of visual cortex.

A key goal of behavioral and cognitive neuroscience is to link brain mechanisms to behavioral functions. The present article describes recent progress toward explaining how the visual cortex sees. Visual cortex, like many parts of perceptual and cognitive neocortex, is organized into six main layers of cells, as well as characteristic sublamina. Here it is proposed how these layered circuits help to realize processes of development, learning, perceptual grouping, attention, and 3-D vision through a combination of bottom-up, horizontal, and top-down interactions. A main theme is that the mechan

Chronic stepwise cerebral hypoperfusion differentially induces synaptic proteome changes in the frontal cortex, occipital cortex, and hippocampus in rats.

During chronic cerebral hypoperfusion (CCH), the cerebral blood flow gradually decreases, leading to cognitive impairments and neurodegenerative disorders, such as vascular dementia. The reduced oxygenation, energy supply induced metabolic changes, and insufficient neuroplasticity could be reflected in the synaptic proteome. We performed stepwise bilateral common carotid occlusions on rats and studied the synaptic proteome changes of the hippocampus, occipital and frontal cortices. Samples were prepared and separated by 2-D DIGE and significantly altered protein spots were identified by HPLC-M

[Ultrastructural changes in the cerebral cortex, cerebellum and ventro-oral nuclei of the optic thalamus in infantile cerebral palsy].

The authors describe 3-, 5-, 7- and 9-layer linear and concentric structures discovered in brain bioptates of patients with cerebral infantile paralysis. They were found to be specific to this disease. The assumptions on the origin and role of these ultrastructures in the pathogenesis of cerebral infantile paralysis are formulated.

Wdr5 Modulates Synaptic Development and Memory Consolidation in the Hippocampus and Cerebral Cortex.

WD-repeat domain 5 (Wdr5), a core component of the SET1/MLL family of histone methyltransferases, is widely involved in the regulation of development, epigenetic control, transcription, and cell division. Wdr5 has been linked to neurodevelopmental disorders characterized by intellectual disability and developmental delay-yet its regulatory roles in neurons remain poorly understood. Here, we report that postnatal deletion of Wdr5 in excitatory forebrain neurons impairs learning and memory in male Wdr5fl/fl; CaMKIIα-iCre conditional knockout (Wdr5 cKO) mice. We showed that the conditional Wdr5 k

Spatial transcriptomics reveal developmental dynamics of the human cerebral cortex and striatum.

The human fetal brain undergoes morphological changes that contribute to the development of regional functionalities. However, the features of structural development, the underlying molecular and cellular signatures in the fetal brain, remain unclear. With spatial transcriptomics and single-nucleus RNA sequencing (snRNA-seq), we identified 25 forebrain regions and characterized the dynamic changes in the cortex and striatum during the late first and early second trimesters. In particular, we discovered that the temporal lobe enriched NPY-expressing L2/3 excitatory (EX) neurons potentially inte

Effect of Cu2+ on the Nucleoli of Cultured Astrocytes from the Rat Cerebral Cortex.

Cultured astrocytes were incubated with CuCl2, which caused dose-dependent cell death (25-200 μM, 24 h). Immunocytochemical detection of the nucleolar protein nucleophosmin/B23 (NPM/B23) demonstrated that exposure to Cu2+ (100 μM, 24 h) caused a significant increase in the surface area of NPM/B23 clusters, which was accompanied by the changes in the nucleolar ultrastructure characteristic of nucleolar stress. Longer incubation of astrocytes with Cu2+ (100 μM, 48 h) led to accumulation of the endoplasmic reticulum (ER) stress marker GRP78, which was accompanied by the increase in nucleolar size
