Ca2+ signaling of pancreatic acinar cells in malignant hyperthermia susceptibility.

BACKGROUND: Malignant hyperthermia susceptibility (MHS) and acute pancreatitis (AP) share a common cellular pathomechanism that is Ca2+-overload of the muscle fiber and the pancreatic acinar cell (PAC). In the muscle, gain-of-function mutations of the ryanodine receptor (RyR1) make the Ca2+-release mechanism hypersensitive to certain ligands, including Ca2+, volatile anaesthetics and succinylcholine, creating a medical emergency when the patient is exposed to these drugs. As RyR1 was shown to contribute to Ca2+-overload in PAC, we presumed that pancreata of MHS individuals are more prone to AP

Dynamic imaging of pancreatic nuclear factor κB (NF-κB) activation in live mice using adeno-associated virus (AAV) infusion and bioluminescence.

Nuclear factor κB (NF-κB) is an important signaling molecule that plays a critical role in the development of acute pancreatitis. Current methods for examining NF-κB activation involve infection of an adenoviral NF-κB-luciferase reporter into cell lines or electrophoretic mobility shift assay of lysate. The use of adeno-associated viruses (AAVs) has proven to be an effective method of transfecting whole organs in live animals. We examined whether intrapancreatic duct infusion of AAV containing an NF-κB-luciferase reporter (AAV-NF-κB-luciferase) can reliably measure pancreatic NF-κB activation.

Alpha-1-antichymotrypsin is an effective inhibitor of pancreatitis-induced lung injury.

BACKGROUND: Pancreatitis-induced adult respiratory distress syndrome (ARDS) may result from an imbalance between leucocyte proteases, produced by infiltrating neutrophils, and endogenous protease inhibitors. OBJECTIVE: The aim of this study was to evaluate the role of recombinant alpha-1-antichymotrypsin (rACT P3-P3'), an endogenous serine protease inhibitor, in ameliorating lung injury associated with pancreatitis. DESIGN: Sprague-Dawley rats were randomly divided into control (saline infusion) and pancreatitis groups, which were treated immediately with saline or rACT P3-P3' (50 mg/kg body

Conduction of activity between muscles in the terminal region of the common bile duct and in the neighboring duodenum.

The relationship between muscle activity at the terminal region of the common bile duct and the duodenal muscle was examined in rabbits. The rhythmic muscle activity in the terminal region was synchronous with duodenal muscle activity. The activity of the latter muscle preceded the former. The activity at the terminal region synchronous with the rhythmic activity of the duodenal muscle sometimes disappeared spontaneously. The muscle activity of the ampulla and the spincter at the terminal region was sometimes independently lost. The conduction of excitation from the duodenal muscle to the term

Nicotinamide N-Methyltransferase Inhibition Mitigates Cerulein-Induced Pancreatic Fibrosis via Galectin-3-Mediated Regulation of Stellate Cell Activation and Macrophage M2 Polarization in Mice.

Chronic pancreatitis (CP) is a chronic disease characterized by pancreatic fibrosis driven by pancreatic stellate cell activation and M2 polarization of macrophages. Nicotinamide n-methyltransferase (NNMT) is a methylase critically involved in fibrosis. Galectin-3 (LGALS3), a member of the galectin family, drives M2 macrophage polarization. This study explores the roles of NNMT and LGALS3 in CP. CP was induced in C57BL/6 mice via repeated intraperitoneal injections of 50 µg/kg cerulein. NNMT was upregulated in pancreatic tissues of CP mice, especially in mouse pancreatic stellate cells (mPSCs)

Development of a Novel Mouse Model of Severe Acute Pancreatitis with Pathological Features Recapitulating Human Disease.

BACKGROUND: Current severe acute pancreatitis (SAP) murine models face significant limitations. The sodium taurocholate method consistently induces pancreatic necrosis but demands intricate surgical procedures, leading to high variability and infection risks. In contrast, the arginine approach causes acinar cell damage without fully mimicking clinical injury pathways. Furthermore, the caerulein-lipopolysaccharide (CAE-LPS) model, while operationally straightforward, fails to replicate persistent organ failure due to LPS's rapid clearance, thereby inadequately capturing the lethal trajectory ob

Circulating cyclophilin A levels elevate in animal models and can predict mortality in patients with acute pancreatitis.

BACKGROUND/OBJECTIVES: Cyclophilin A (CypA) is released into the blood following cellular injury and correlates with acute pancreatitis (AP) severity, however, investigations into circulating CypA in experimental AP and hypertriglyceridaemia-associated AP (HTG-AP) prevalent cohorts are lacking. METHODS: C57Bl/6 mice received 4, 8, and 12 caerulein injections to induce escalating severity of AP models (CER-AP). Mice were sacrificed 1 h after the last injection of caerulein to assess severity of CER-AP through biochemical and histopathological analyses, and serum CypA levels were also measured.

Emodin: an alveolar macrophage protector in acute pancreatitis induced lung injury.

Background: Emodin (EMO), an anthraquinone derivative from roots and leaves of various plants, has been widely used in many inflammatory diseases. Alveolar macrophages (AMs) play a critical role in maintaining alveolar homeostasis in the lung. However, the comprehensive mechanisms of EMO therapy on AMs during acute pancreatitis-associated lung injury (AP-ALI) have not been reported. Methods: Both in vivo [caerulein/ lipopolysaccharides (LPS)-induced AP-ALI in mice] and in vitro MH-S models were generated to assess the protective features of EMO on mitochondrial damage and mitophagy dysfunction

Deubiquitinase Josephin Domain-containing Protein 2 Promotes Acute Pancreatitis by Removing K63-linked Poly-ubiqutin Chain on Proliferating Cell Nuclear Antigen in Pancreatic Acinar Cells.

BACKGROUND & AIMS: Acute pancreatitis (AP) results in localized pancreatic injury or systemic inflammatory responses, contributing to high morbidity and mortality worldwide. Acinar cell death and inflammation are critical key drivers of AP progression. Some deubiquitinases (DUBs), which regulate the stability and/or activity of substrate proteins, may play a role in the development of AP. Here, we aimed to investigate how Josephin domain-containing protein 2 (JOSD2) involved in AP progression. METHODS: JOSD2 expression was assessed in mouse pancreatic tissues with cerulein-induced AP. Lonicerin protects pancreatic acinar cells from caerulein-induced apoptosis, inflammation, and ferroptosis by activating the SIRT1/GPX4 signaling pathway.

Acute pancreatitis (AP) is a familiar emergency of digestive system characterized by pancreatic inflammation. Lonicerin (LCR) has been reported to exert anti-inflammatory and immunomodulatory characteristics in several inflammatory diseases. Nevertheless, its role and mechanism involved in AP are still unknown. This study was designed to explore the protective effect and potential mechanism of LCR in AP. In this study, LCR and ferrostatin-1 alleviated, but erastin aggravated caerulein (CAE) exposure-induced cytotoxicity and reduction of cell viability in AR42J cells. LCR exhibited a protective

Protective effects of Coenzyme Q10 against acute pancreatitis.

Acute pancreatitis (AP) refers to inflammation in the pancreas, which may lead to death in severe cases. Coenzyme Q10 (Q10), generally known to generate energy, plays an important role as an anti-oxidant and anti-inflammatory effector. Here, we showed the effect of Q10 on inflammatory response in murine AP model. For this study, we induced AP by injection of cerulein intraperitoneally or pancreatic duct ligation (PDL) in mice. The level of cytokines and digestive enzymes were measured in pancreas, and blood. All pancreatic tissues were excised for investigation such as histological changes, in

Abnormalities of caerulein- and carbamylcholine-stimulated pancreatic enzyme secretion in the obese Zucker rat.

The secretory function of the exocrine pancreas has been studied in dispersed pancreatic acini from obese and homozygous lean Zucker rats at 6 and 22 wk. No abnormality was found in acini from young rats. Acini from 22 wk obese and lean rats were equally responsive to secretagogues which stimulate cAMP, i.e. vasoactive intestinal peptide (VIP) and secretin. By contrast, there was a reduction in the maximum responsiveness to caerulein and carbamylcholine in acini from obese rats. These latter secretagogues act through mobilization of intracellular Ca2+. Since obese animals are insulin resistant

Berberine Impedes Acute Pancreatitis Development by Suppressing VNN1 Expression and the NF-κB Signaling Pathway.

The global incidence of acute pancreatitis (AP) is progressively increasing with rising risk factors such as hyperlipidemia and alcoholism. Berberine (BBR), a quaternary ammonium alkaloid and primary antibacterial agent isolated from the traditional Chinese herb, Coptis chinensis, was investigated for its therapeutic mechanisms in AP. Cell viability was first assessed using the MTT assay at various BBR concentrations, with lactate dehydrogenase (LDH) levels measured in parallel. Then, ELISA quantified inflammatory mediators (caspase-1, IL-1β, IL-18, TNF-α, and IL-6), while western blot analyze

Protease Inhibitor VR23 Alleviates Severe Acute Pancreatitis with High Trypsin Synthesis in Mice.

BACKGROUND: Acute pancreatitis (AP), characterized by pancreatic autodigestion and systemic inflammation, remains a life-threatening condition with a high mortality rate in severe cases. Cholinergic signaling through muscarinic receptors (mAChRs) regulates pancreatic exocrine secretion, yet the role of chronic cholinergic hyperactivation in AP progression is poorly understood. Protease inhibitors like VR23, a potent trypsin-selective compound, hold therapeutic potential but lack evaluation in AP models. OBJECTIVE: This study aimed to investigate the effects of prolonged cholinergic stimulatio
