Inner ear development in cetaceans.

Cetaceans face the challenge of maintaining equilibrium underwater and obtaining sensory input within a dense, low-visibility medium. The cetacean ear represents a key innovation that marked their evolution from terrestrial artiodactyls to among the most fully aquatic mammals in existence. Using micro-CT and histological data, we document shape and size changes in the cetacean inner ear during ontogeny, and demonstrate that, as a proportion of gestation time, the cetacean inner ear is precocial in its growth compared with that of suid artiodactyls. Cetacean inner ears begin ossifying and reach

A multiplex real-time polymerase chain reaction assay with two internal controls for the detection of Brucella species in tissues, blood, and feces from marine mammals.

Brucellosis has emerged as a disease of concern in marine mammals in the last 2 decades. Molecular detection techniques have the potential to address limitations of other methods for detecting infection with Brucella in these species. Presented herein is a real-time polymerase chain reaction (PCR) method targeting the Brucella genus-specific bcsp31 gene. The method also includes a target to a conserved region of the eukaryotic mitochondrial 16S ribosomal RNA gene to assess suitability of extracted DNA and a plasmid-based internal control to detect failure of PCR due to inhibition. This method

Emerging infectious diseases in cetaceans worldwide and the possible role of environmental stressors.

We reviewed prominent emerging infectious diseases of cetaceans, examined their potential to impact populations, re-assessed zoonotic risk and evaluated the role of environmental stressors. Cetacean morbilliviruses and papillomaviruses as well as Brucella spp. and Toxoplasma gondii are thought to interfere with population abundance by inducing high mortalities, lowering reproductive success or by synergistically increasing the virulence of other diseases. Severe cases of lobomycosis and lobomycosis-like disease (LLD) may contribute to the death of some dolphins. The zoonotic hazard of marine m

Antibodies to Brucella in marine mammals around the coast of England and Wales.

Following the isolation of previously unrecognised species of Brucella from stranded seals and cetaceans in Scotland and northern England, a serological survey was carried out to investigate the range of marine mammal species which may have been exposed to Brucella species around the coasts of England and Wales, the prevalence of infection and the temporal and geographical distribution of seropositive animals. Serum collected from 153 stranded marine mammals from the coasts of England and Wales between 1989 and 1995 were tested by competitive and indirect ELISA. Positive titres were recorded f

Comparative analysis of the myoglobin gene in whales and humans reveals evolutionary changes in regulatory elements and expression levels.

Cetacea and other diving mammals have undergone numerous adaptations to their aquatic environment, among them high levels of the oxygen-carrying intracellular hemoprotein myoglobin in skeletal muscles. Hypotheses regarding the mechanisms leading to these high myoglobin levels often invoke the induction of gene expression by exercise, hypoxia, and other physiological gene regulatory pathways. Here we explore an alternative hypothesis: that cetacean myoglobin genes have evolved high levels of transcription driven by the intrinsic developmental mechanisms that drive muscle cell differentiation. Environmental contaminants and the HPA axis: Advances in adrenocortical hormone research in cetaceans.

Environmental pollution is one of the main threats to marine life, and chemical contaminants are known endocrine disruptors capable of altering the physiological balance of exposed organisms. Among marine mammals, cetaceans stand out as environmental sentinels due to their high trophic level, longevity, and ability to bioaccumulate lipophilic substances. This review explores the impact of environmental contamination on the hypothalamic-pituitary-adrenal (HPA) axis, a key endocrine system involved in stress responses, whose chronic dysregulation may lead to adverse health outcomes. Existing evi

Anthropogenic litter in terrestrial flora and fauna: Is the situation as bad as in the ocean? A field study in Southern Germany on five meadows and 150 ruminants in comparison with marine debris.

In contrast to the abundance of research on marine debris, terrestrial anthropogenic litter and its impacts are largely lacking scientific attention. Therefore, the main objective of the present study is to find out whether ingested litter produces pathological consequences to the health of domestic ruminants, as it does in their relatives in the ocean, the cetaceans. For this purpose, five meadows (49°18'N, 10°24'E) with a total survey area of 139,050 m2 as well as the gastric content of 100 slaughtered cattle and 50 slaughtered sheep have been examined for persistent man-made debris in North

Brucella ceti sp. nov. and Brucella pinnipedialis sp. nov. for Brucella strains with cetaceans and seals as their preferred hosts.

Small Gram-negative cocco-bacilli resembling Brucella strains have been reported from marine mammals since the mid-1990s. Their placement in the genus Brucella has been supported by the following characteristics: they are aerobic, non-motile and catalase-positive, do not produce acid from carbohydrates and have a DNA-DNA relatedness value of >77% with the six established members of the genus. Twenty-eight European isolates of the genus Brucella from marine mammals were distinguished from the six recognized species by their pattern of utilization of eleven substrates in oxidative metabolism tes

Urinary concentrations of ovarian steroid hormone metabolites and bioactive follicle-stimulating hormone in killer whales (Orcinus orchus) during ovarian cycles and pregnancy.

Reproductive hormone profiles of six captive killer whales (Orcinu orcus) from three Sea World aquaria were studied for intervals up to 2 yr. Daily urine samples and bimonthly blood samples were collected and analyzed for hormone concentration. Immunoreactive estrone conjugates, pregnanediol-3-glucoruonide, 20-alpha-hydroxyprogesterone as well as bioactive follicle-stimulating hormone (FSH) were measured in urine samples and indexed by creatinine concentrations of the same sample. In selected cases, serum progesterone concentrations were also measured. Three of the animals in the study became

Habitat-associated detection of Toxoplasma gondii and Sarcocystis spp. in Cetaceans from the Brazilian coast.

Cetaceans are sentinels of marine ecosystem health and are increasingly exposed to protozoal pathogens. This study investigated the occurrence and molecular identity of Sarcocystidae protozoa in 159 stranded cetaceans representing 21 species along the Brazilian coast. Molecular analysis based on PCR amplification and sequencing of the sarcocystid internal transcribed spacer 1 (ITS1) region revealed infections in 14 individuals, showing a clear habitat-associated distribution. Toxoplasma gondii and Sarcocystis neurona were detected in nine coastal cetaceans, consistent with transmission from te

Serologic survey of Brucella infection in cetaceans inhabiting along the coast of Japan.

A serologic investigation of Brucella infection was performed in 7 species of cetaceans inhabiting along the coast of Japan. A total of 32 serum samples were examined by enzyme-linked immunosorbent assay (ELISA) using Brucella abortus and B. canis antigens. One serum sample from five melon-headed whales (Peponocephala electra) was positive for B. abortus. No serum sample showed positive for B. canis. The ELISA-positive melon-headed whale serum demonstrated a strong band appearance only against B. abortus antigens in Western blot analysis. Many detected bands were discrete, while some of them h

Comparative pathology of nocardiosis in marine mammals.

Nocardia spp. infections in mammals cause pyogranulomatous lesions in a variety of organs, most typically the lung. Members of the Nocardia asteroides complex are the most frequently recognized pathogens. Nine cases of nocardiosis in free-ranging pinnipeds and 10 cases of nocardiosis in cetaceans were evaluated. Host species included the hooded seal (Cystophora cristata, n = 8), leopard seal (Hydrurga leptonyx, n = 1), Atlantic bottlenose dolphin (Tursiops truncatus, n = 4), beluga whale (Delphinapterus leucas, n = 4), and killer whale (Orcinus orca, n = 2). Herpes-like viral dermatitis in a beluga whale (Delphinapterus leucas).

Approximately 3.5 mo following its capture, a beluga whale (Delphinapterus leucas) developed focal pale gray skin lesions. These lesions persisted for at least 8 mo. A biopsy from one of these sites revealed epithelial intranuclear inclusions. Herpes-like viral particles were seen by transmission electron microscopy. The eventual regression of skin lesions and lack of other clinical signs suggests the virus was only mildly pathogenic in this animal.

Variations in lipid composition and sound velocity in melon from the North Atlantic Pilot whale, globicephala melaena melaena.

Nonpolar lipids and sound velocity at different distances from the skin surface within a sample of pilot whale melon were studied. Thin layer chromatography, sensitive radioisotopic methods, and an ultrasonic echo scanner were used. Wax esters had a maximum concentration at 11-12 cm from the skin surface. Non- and monoisovalero triglycerides both exhibited a minimum at 8-10 cm, whereas the diisovalero species increased steadily from the skin surface towards the center of the pilot whale head. A minimum of sound velocity, ca. 1340 m/s, was found at 9-11 cm, thus coinciding with a region rich in

Leptospira spp. and Toxoplasma gondii in stranded representatives of wild cetaceans in the Philippines.

BACKGROUND: The stranding events of cetaceans in the Philippines provide opportunities for gathering biological information and specimens, especially from the pelagic forms. As part of an effort to monitor the health of wild cetaceans, this study detected Leptospira spp. and Toxoplasma gondii, causative agents of the emerging zoonotic diseases leptospirosis and toxoplasmosis respectively, in their stranded representatives. From October 2016-August 2018, 40 cetaceans (representing 14 species) that stranded nationwide were sampled for brain, cardiac muscle, skeletal muscle, kidney, and blood tis

Sexual selection targets cetacean pelvic bones.

Male genitalia evolve rapidly, probably as a result of sexual selection. Whether this pattern extends to the internal infrastructure that influences genital movements remains unknown. Cetaceans (whales and dolphins) offer a unique opportunity to test this hypothesis: since evolving from land-dwelling ancestors, they lost external hind limbs and evolved a highly reduced pelvis that seems to serve no other function except to anchor muscles that maneuver the penis. Here, we create a novel morphometric pipeline to analyze the size and shape evolution of pelvic bones from 130 individuals (29 specie

Deep breath out: molecular survey of selected pathogens in blow and skin biopsies from North Atlantic cetaceans.

BACKGROUND: Cetacean morbillivirus, herpesvirus, avian influenza virus (AIV) and Brucella spp. have been linked to numerous cetacean strandings in the Northeast (NE) Atlantic. Yet, their prevalence in free-living cetaceans remains insufficiently investigated, particularly in northern regions.
