Development of increased serum immunoblot reactivity against a 45,000-dalton polypeptide of Treponema pallidum (Nichols) correlates with establishment of chancre immunity in syphilitic rabbits.

Rabbits developed chancre immunity 5.0 to 7.5 weeks after intradermal infection with 10(3) Treponema pallidum (Nichols). The serological response against T. pallidum antigen during this 2.5-week period was examined by Western immunoblotting. Sera from rabbits infected for 5.0 weeks contained antibodies against 7 of 13 major T. pallidum immunogens, with strongest binding detected against a polypeptide of Mr 47,000. By 7.5 weeks of infection, syphilitic rabbit sera recognized 10 of 13 antigens; the most evident increase in serological reactivity was directed against a polypeptide of Mr 45,000, s

A mixed infection of syphilis and chancroid.

The clinical differentiation between a hard chancre and a soft chancre is well known to dermatologists. We present a patient with both a hard and a soft chancre developing on the penis. Thorough investigational studies should be made of all patients with venereal disease to determine whether more than one venereal disease is present.

Early events following challenge of cattle with tsetse infected with Trypanosoma congolense: development of the local skin reaction.

A local skin reaction (chancre) was elicited in susceptible cattle after the successful feeding of Glossina morsitans morsitans infected with one of two different cloned isolates of Trypanosoma congolense. The chancre first appeared as a small 2 to 3 mm nodule at the site of the challenge as early as day 5 and reached maximum activity by days 10 to 13 when it had developed into a raised, indurated, hot, painful swelling measuring up to 100 mm in diameter. Thereafter it declined in size and activity and by days 20 to 30 was undetectable. Histologically the lesion was characterised by an intense

Parasite development and host responses during the establishment of Trypanosoma brucei infection transmitted by tsetse fly.

Following inoculation of Trypanosoma brucei into large mammals by the tsetse fly a local skin reaction, the 'chancre', develops due to trypanosome proliferation. We have cannulated the afferent and efferent lymphatics of the draining lymph node in goats and examined the onset of a cellular reaction, the emigration of the parasite from the chancre and the development of both antigenic variation and the specific immune response. The chancre first became detectable by day 3 post-infection, peaked by day 6 and then subsided. Lymphocyte output increased 6- to 8-fold by day 10 and the number of lymp

Trypanosoma congolense infection in sheep: cellular phenotypes in lymph and lymph nodes associated with skin reactions.

Intradermal inoculation of sheep with culture-derived metacyclic forms of Trypanosoma congolense resulted in the development of localized skin reactions (chancres) and enlargement of the draining lymph nodes 7 days after infection. Changes in the expression of surface antigens of lymphocytes in lymph leaving the affected skin reactions and in the associated lymph nodes were monitored by cannulating the afferent and efferent lymphatic ducts. Trypanosomes appeared in afferent and efferent lymph 3 to 5 days after infection and persisted even as the chancres regressed. The cellular output in both

Chancroid: a review for the family practitioner.

Chancroid, as the name implies, is like a chancre. Unlike the painless chancre of syphilis, it is painful, darkfield negative, and does not respond to penicillin therapy. The number of cases have continued to rise in recent years. Because it can cause irreversible anatomical destruction, making the correct diagnosis is important and can prevent chronic morbidity.

Primary syphilis of the fingers.

Six patients were seen with primary syphilitic chancres on their fingers between 1965 and 1980. Of these, two had bipolar chancres on their fingers and genitals resulting from sexual foreplay. Because syphilis is rarely suspected in such cases diagnostic errors are common.

Immunity to syphilis. I. Passive transfer in rabbits with hyperimmune serum.

Passive transfer of humoral immunity to experimental syphilis in rabbits was accomplished with large doses of hyperimmune serum. When the serum was administered before inoculation of Treponema pallidum, the onset of chancres was delayed; when it was given early in the course of infection, a transient period of arrest and healing of the chancres was imposed.

Trypanosoma congolense: host responses following tsetse transmitted infection of Kilifi isolates in goats.

East African x Galla goats, when infected with Trypanosoma congolense isolates from the Kilifi area of Kenya by Glossina morsitans centralis, did not develop the characteristic chancre reaction at the bite sites, whereas bites of tsetse infected with the cloned T. congolense IL.1180 from Serengeti, Tanzania, resulted in chancres in the same goats. Histological changes could not be observed in skin biopsies collected 8 or 9 days after infection with Kilifi isolates. However, all goats became parasitemic about 10 days after challenge. It is concluded that the absence of chancre development is a

Primary syphilis of the finger: report of four cases.

Although primary syphilis generally involves the genitalia, literature reports that 12% of chancres are extragenital, with the anus and oral cavity as the most frequent locations. We present hereby a case series of four chancres of the finger observed at the sexually transmitted infection centre of Milan between 2010 and 2021.

Clinical and laboratory features of 244 men with primary syphilis: a 5-year single-centre retrospective study.

BACKGROUND: Syphilis incidence has exponentially increased in recent decades, particularly among men who have sex with men (MSM). Primary syphilis is characterised by a chancre appearing at the site of Treponema pallidum (TP) inoculation. Atypical morphological variants of syphilitic chancre are frequent. Clinical suspicion must be confirmed either by the demonstration of TP within the lesion through direct tests, such as dark field microscopy (DFM) or T. pallidum nucleic acid amplification technique (TP-NAAT), or by serological tests. OBJECTIVES: To analyse the clinical features, the sexual

An extraordinary case of syphilis presenting with a labial ulcer.

In the differential diagnosis of patients with ulcers on the lips characteristics like the duration of the ulcer, number, size, depth, shape, base, margins, and distribution are considered. Such ulcers arise from many diseases particularly, viral and bacterial infections, malignancies can also be responsible. Classic syphilitic chancres are painless erosions settled on hard papule; these are evident in the genital area in more than 90% of patients. This study describes a case of a 38-year-old female patient presenting with a painful ulcer covering 3 quarters of the upper lip showing settlement

Stability of metacyclic variable antigen types (M-VATs) during the early stages of infection with Trypanosoma congolense.

Expression of nine metacyclic variable antigen types (M-VATs) of Trypanosoma congolense in chancres from infected rabbits was determined using monoclonal antibodies raised against metacyclic forms of trypanosomes. Trypanosomes present in chancres 7-9 days post infection expressed M-VATs present in metacyclic populations of the parasites. The majority of M-VATs expressed showed little proportional change from those observed on metacyclic trypanosomes during this period although expression of one M-VAT increased, and another decreased. Although trypanosomes in chancres continued to express M-VAT

Early stages of infection with Trypanosoma congolense: parasite kinetics and expression of metacyclic variable antigen types.

Trypanosoma congolense develops in the skin of sheep at the site of inoculation of metacyclic trypanosomes, forming a chancre containing large numbers of parasites. By cannulating the afferent and efferent lymphatic ducts draining the skin and regional lymph node, the progressive development and migration of trypanosomes from the chancre was monitored and the expression of metacyclic antigen types (M-VATs) was determined. The kinetics of development of parasitosis in the afferent and efferent lymph was similar. Trypanosomes were detected in lymph 5 to 6 days after the inoculation of cultured m

Cell and tissue interactions of Treponema pallidum in primary and secondary syphilitic skin lesions: an ultrastructural study of serial sections.

There are limited reports on the ultrastructure of syphilis skin lesions. The aim of this study has been to perform an electron microscopic investigation of the morphology and the tissue distribution of treponemes in primary and secondary cutaneous lesions. Three cases of primary syphilitic chancre and one case of secondary syphilis were included. Prominent epidermal abnormalities in the primary chancre and a perivascular inflammatory infiltrate in the secondary lesion were found by light microscopy. Ultrastructurally, spirochetes were located mainly in the blood vessel walls and dermal tissue

Development of Trypanosoma congolense, T vivax and T brucei in the skin reaction induced in goats by infected Glossina morsitans centralis: a light and electron microscopical study.

The development and distribution of Trypanosoma congolense, T vivax and T brucei in the skin of goats was examined after the animals were bitten by infected Glossina morsitans centralis. Following the tsetse bite, the trypanosomes in the skin multiplied, reaching maximum numbers when the skin reaction (chancre) of the host attained its maximum size. In goats infected with T vivax and T brucei, trypanosomes were observed circulating in the blood before the peak of the chancre, while in T congolense-infected goats microscopically detectable parasites were found in blood only during the decline o

Epidemic of venereal treponematosis in wild monkeys: a paradigm for syphilis origin.

Treponema pallidum infections have been primarily known as slightly contagious mucocutaneous infections called yaws (tropical Africa and America) and bejel (subtropical North Africa). T. pallidum emerged as a highly infectious venereal syphilis agent in South America, probably about 500 years ago, and because of its venereal transmission, it quickly caused a worldwide pandemic. The disease manifests as lesions, including a chancre; then antibodies become detectable when or slightly after the chancre appears, and before the development of a rash and other systemic manifestations. Venereal disea

Trypanosoma congolense infection in sheep: ultrastructural changes in the skin prior to development of local skin reactions.

Events occurring in the skin of sheep prior to development of Trypanosoma congolense-induced local skin reactions (chancres) were studied using electron microscopy. Three days after infection, few trypanosomes were present in the dermal collagen. However, these parasites were more abundant 5 days after infection, and were also found in dermal lymphatics and in the connective tissue matrix between collagen bundles. Mast cells in the skin obtained 5 days after infection showed evidence of degranulation. These events may play a role during the induction phase of trypanosomal chancres.
