Depression of neutrophil chemotaxis in atopic individuals. An H2 histamine receptor response.

Neutrophil chemotaxis was compared in normal and atopic individuals using a modified Boyden chamber with as chemotactant, autologous serum either unactivated or activated by zymosan or an endotoxin-containing house dust preparation. A high incidence of defective leukotaxis was found in atopics when cells were opposed to activated serum. The cause of this abnormality is not intrinsic to the leukocytes since random migration and chemotaxis towards unactivated serum were comparable in normal and atopic subjects. The defect persisted when atopic leukocytes were opposed to activated normal serum an

Clinical syndrome of chemotaxis defect, infections, and hyperimmunoglobulinemia E. A case report.

A 17-year-old girl had a clinical syndrome that consisted of neutrophil chemotaxis defect, recurrent bacterial infections, and hyperimmunoglobulinemia E. Neutrophil chemotaxis was quantitated by a radioassay technique that used 51Cr-labeled neutrophils and a double micropore filter chemotaxis chamber. A table of the features of other cases of this syndrome is presented. Since ten of the 13 patients who are described had a chronic eczematoid dermatitis, we suggest that this may be a fourth feature of this syndrome. Red hair and fair skin were not consistent findings in these patients. In the ev

Enhancement of neutrophil chemotaxis and alteration of levels of cellular cyclic nucleotides by levamisole.

Levamisole, an antihelminthic agent reported to enhance nonspecifically various parameters of the immune response, was examined for its effect on chemotaxis of human neutrophils and on levels of cellular cyclic nucleotides. This agent was found, in most instances, to enhance chemotactic responses of neutrophils to a bacterial chemotactic factor derived from Escherichia coli. At similar concentrations, levamisole produced increases in levels of guanosine 3':5'-cyclic phosphate in neutrophils. In contrast, a decrease in concentrations of adenosine 3':5'-cyclic phosphate was observed when neutrop

A hyperimmunoglobulin E syndrome with normal chemotaxis in vitro and defective leukotaxis in vivo.

A 26-yr-old male with a lifelong history of atopic dermatitis and recurrent severe staphylococcal abscesses was found to have hyperimmunoglobulinemia E. Evaluation of both the humoral and cellular aspects of chemotaxis in vitro showed both neutrophils and monocytes to be normal. However, quantitative neutrophil migration in vivo was significantly suppressed using the patient's own serum as the attractant. This defective migration in vivo was partially corrected by serum from normal donors as the attractant and also partially corrected following plasma infusion in this patient. Evaluation of qu

TAXISCAN, Optimizing throughput and behavioral depth in standard C. elegans chemotaxis assays.

Chemosensory signaling is crucial for organisms to respond to their environment and dysfunctions in these complex pathways have been implicated in diseases such as neurological disorders. The nematode C. elegans is a useful tool for studying chemosensation as it offers a fully-mapped nervous system with relatively simple chemosensory circuits. One of the most common methods to quantify chemosensory function in worms is through chemotaxis assays, which require extensive time and manual effort. In this work, we sought to capture novel chemosensory metrics and increase the throughput of these ass

Baicalin Nasal Irrigation Mitigates Allergic Rhinitis in Mice by Inhibiting Eosinophil Chemotaxis.

INTRODUCTION: Allergic rhinitis (AR) significantly impairs quality of life. Although baicalin is known for its anti-inflammatory properties, its therapeutic potential via localized nasal delivery remains unexplored. This study evaluated the efficacy and potential mechanisms of baicalin nasal irrigation in an AR mouse model. METHODS: Ovalbumin (OVA)-induced BALB/c mice (n = 17 per group) were randomized into six groups: control, AR model, baicalin, loratadine, combination of baicalin and loratadine, and excipient. Nasal symptom scores were recorded pre- and post-treatment. Serum and nasal lava

Mammalian sperm hyperactivation regulates navigation via physical boundaries and promotes pseudo-chemotaxis.

Mammalian sperm migration within the complex and dynamic environment of the female reproductive tract toward the fertilization site requires navigational mechanisms, through which sperm respond to the tract environment and maintain the appropriate swimming behavior. In the oviduct (fallopian tube), sperm undergo a process called "hyperactivation," which involves switching from a nearly symmetrical, low-amplitude, and flagellar beating pattern to an asymmetrical, high-amplitude beating pattern that is required for fertilization in vivo. Here, exploring bovine sperm motion in high-aspect ratio m

Tonsillitis exacerbates renal injury in IgA nephropathy through promoting Th22 cells chemotaxis.

BACKGROUND: Tonsillitis can promote the progression of IgA nephropathy (IgAN) by aggravating immunopathologic response. Th22 cell disorder is involved in the pathogenesis of IgAN with tonsillitis. This study was determined to explore the possible mechanism of IgAN with tonsillitis underlying Th22 cell chemotaxis response to the effect of CCL20, CCL22, and CCL27. METHODS: This research was conducted on 65 subjects including 16 healthy controls (HC group), 5 patients with  renal carcinoma (HTC group) and 44 patients with IgAN between 2015 and 2016. According to clinical symptoms and results of

-NOD Mice Having a Lyn Tyrosine Kinase Mutation Exhibit Abnormal Neutrophil Chemotaxis.

Neutrophils from NOD (Non-Obese Diabetic) mice exhibited reduced migration speed, decreased frequency of directional changes, and loss of directionality during chemotaxis (compared to wild-type [WT] C57BL/6 mice). Additionally, F-actin of chemotaxing NOD neutrophils failed to orient toward the chemoattractant gradient and NOD neutrophil adhesion was impaired. A point mutation near the autophosphorylation site of Lyn in NOD mice was identified. Point mutations of G to A (G1412 in LynA and G1199 in LynB) cause a change of amino acid E393 (glutamic acid) to K (lysine) in LynA (E393 →K) (E372 of L

Metal ions activate vascular endothelial cells and increase lymphocyte chemotaxis and binding.

Metal on metal articulations in hip arthroplasty offer advantages, including lower volumetric wear compared to conventional metalonpolyethylene bearings, and increased resistance to dislocation. Reports described early failures, with histologic features similar to a Type IV immune response. Mechanisms by which metal wear products cause this reaction are not completely understood. We hypothesized a mechanism through direct activation of endothelial cells (ECs) by metal ions, resulting in both vasculitis and accumulation of lymphocytes without prior immune sensitization. Effects of metal ions we

Rhamnolipid (RL) from Pseudomonas aeruginosa OBP1: a novel chemotaxis and antibacterial agent.

In the present study, the interaction of rhamnolipid produced by Pseudomonas aeruginosa OBP1 with the cell surfaces of Staphylococcus aureus (MTCC 3160) and Klebsiella pneumoniae (MTCC 618) were studied. Rhamnolipid concentration below critical micelle concentration (CMC) did not exhibit significant antibacterial activity. However, on increasing rhamnolipid concentration beyond CMC a prominent antibacterial activity was observed. The results demonstrated different degree of rhamnolipid interaction with both the bacteria. This might be due to the changes in their cell wall composition. The anti

Naturally occurring IgM anti-leukocyte autoantibodies inhibit T-cell activation and chemotaxis.

INTRODUCTION: Naturally occurring IgM antileukocyte autoantibodies (IgM-ALA) are present from birth and increase during inflammatory processes of diverse etiologies. The clinical observation demonstrating a significant correlation (P < 01) between lack of acute rejections and presence of high levels of IgM-ALA in recipients of kidney allografts prompted us to study if IgM-ALA alters T-cell function and leukocyte chemotaxis. METHODS: In-vitro functional assays were performed using leucocytes isolated from human peripheral blood. In-vivo studies were performed in C57BL6 mice. RESULT: Human stu

Uncoupling protein-2 negatively regulates polymorphonuclear leukocytes chemotaxis via modulating [Ca2+] influx.

OBJECTIVE: Previous studies demonstrated that uncoupling protein 2 (UCP2) plays a negative role in modulating leukocyte inflammatory responses. The mechanism underneath the role of UCP2 in modulating leukocyte inflammatory responses, however, is incompletely understood. Here, we investigated the effect of UCP2 in polymorphonuclear leukocyte (PMN) chemotaxis. METHODS AND RESULTS: First, we assessed PMN chemotaxis in zymosan-induced murine peritonitis and found that UCP2(-/-) mice had significantly more migrated PMN in peritoneal lavage compared to their wild-type littermates. Dobutamine inhibits monocyte chemoattractant protein-1 production and chemotaxis in human monocytes.

UNLABELLED: It has been reported that, in patients with acute myocardial infarction or congestive heart failure, monocyte chemoattractant protein-1 (MCP-1) plays an important role in the development of inflammatory responses and that the level of MCP-1 is correlated with the severity of the disease. We conducted this study to investigate the effects of dobutamine and dopamine on lipopolysaccharide (LPS)-induced MCP-1 production in human monocytic THP-1 cells. Monocytes were incubated in vitro with LPS for 16 h at 37 degrees C in the presence or absence of dobutamine or dopamine. Enzyme-linked

Eotaxin and RANTES enhance 5-oxo-6,8,11,14-eicosatetraenoic acid-induced eosinophil chemotaxis.

BACKGROUND: The 5-lipoxygenase product 5-oxo-6,8,11,14-eicosatetraenoic acid (5-oxo-ETE) is a potent activator of human eosinophils and, among lipid mediators, is the most active chemoattractant for these cells. Studies have demonstrated the importance of 5-lipoxygenase products in allergen-induced pulmonary eosinophilia. Because CC chemokines such as eotaxin and RANTES also play critical roles in this phenomenon, it would seem likely that members of both classes of mediators contribute to this response. OBJECTIVE: The study was designed to directly compare the effects of 5-oxo-ETE on eosinop
