Estimating bioaugmentation efficacy of TCE dechlorination using long-term field well-to-well tests in a highly recharged and TCE-contaminated aquifer.

This study demonstrates a combined field method accurately assessing the extent of trichloroethylene (TCE) reductive dechlorination activity and the mass fraction of its by-products. A combined method of injecting a known concentration of 1,1,2-trichloro-2-fluoroethene (TCFE) as a TCE bio-surrogate and a data processing technique of forced mass balance (FMB), considering the sorption effect on the mass fraction of chloroethene was evaluated by performing soil column and field bioaugmentation tests. In the soil column test, the FMB resulted in the mass fraction of 6% TCE, 48.3% cis-1,2-dichloro

Quantitative and functional dynamics of Dehalococcoides spp. and its tceA and vcrA genes under TCE exposure.

This study aimed at monitoring the dynamics of phylogenetic and catabolic genes of a dechlorinating enrichment culture before, during, and after complete dechlorination of chlorinated compounds. More specifically, the effect of 40 μM trichloroethene (TCE) and 5.6 mM lactate on the gene abundance and activity of an enrichment culture was investigated for 40 days. Although tceA and vcrA gene copy numbers were relatively stable in DNA extracts over time, tceA and vcrA mRNA abundances were upregulated from undetectable levels to 2.96 × and 6.33 × 10⁴ transcripts/mL, respectively, only after exposu

Effects of elevated temperature on Dehalococcoides dechlorination performance and DNA and RNA biomarker abundance.

Coupling thermal treatment with microbial reductive dechlorination is a promising remedy for tetrachloroethene (PCE) and trichloroethene (TCE) contaminated source zones. Laboratory experiments evaluated Dehalococcoides (Dhc) dechlorination performance, viability, and biomarker gene (DNA) and transcript (mRNA) abundances during exposure to elevated temperatures. The PCE-dechlorinating consortia BDI and OW produced ethene when incubated at temperatures of 30 °C, but vinyl chloride (VC) accumulated when cultures were incubated at 35 or 40 °C. Cultures incubated at 40 °C for less than 49 days resu

Bioreduction of trichloroethene using a hydrogen-based membrane biofilm reactor.

A H2-based, denitrifying membrane-biofilm reactor (MBfR) was effective for removing trichloroethene (TCE) by reductive dechlorination. When TCE was first added to the MBfR, reductive dechlorination took place immediately and then increased over 18 weeks, and TCE was completely dechlorinated to ethene by about 120 days. These results indicate that TCE-dechlorinating bacteria were present naturally in the H2-based biofilm, and that enrichment for TCE-dechlorinating bacteria occurred. Dehalococcoides were documented in the MBfR biofilm before and after TCE feeding. Their proportion, quantified us

Simultaneous removal of manganese and nitrate by manganese redox driven moving bed biofilm reactor: Characteristic, biological community and mechanism.

This study investigated the effects of different manganese forms (MnCl2 and MnO2) on the nitrogen and phosphorus removal performance of moving bed biofilm reactor (MBBR). Compared to the control without manganese, the addition of MnCl2 and MnO2 increased NO3--N removal efficiency by 11.47 % and 9.54 %, and total nitrogen (TN) removal efficiency by 17.91 % and 15.45 %, respectively. The average accumulation of NO2--N decreased from 3.02 to 0.04 mg/L and 0.18 mg/L, respectively. The manganese redox system induced by MnCl2 enhanced total phosephorus (TP) removal efficiency by approximately 3.5 ti

The pesticides carbofuran and picloram alter the diversity and abundance of soil microbial communities.

Many countries widely use pesticides to increase crop productivity in agriculture. However, their excessive and indiscriminate use contaminates soil and other environments and affects edaphic microbial communities. We aimed to examine how the pesticides carbofuran and picloram affect the structure and functionality of soil microbiota using cultivation-independent methods. Total DNA was extracted from microcosms (treated or not with pesticides) for amplification and metabarcoding sequencing for bacteria (16S gene) and fungi (28S gene) using Illumina-MiSeq platform. Data analysis resulted in 6,7

Microbial community acclimation during anaerobic digestion of high-oil food waste.

Anaerobic digestion is one of the most promising options for the disposal of biodegradable food waste. However, the relatively high content of oil in food waste inhibits the conversion efficiency of anaerobic digestion because of the accumulation of long-chain fatty acids (LCFAs). In this study, activated anaerobic sludge was acclimated to accommodate high-oil conditions. The methane yield of high-oil food waste digested by the acclimated sludge increased by 24.9% compared to that digested by the raw sludge. To determine the internal changes in the acclimated sludge, the shifts in the microbia

Magnetite Alters the Metabolic Interaction between Methanogens and Sulfate-Reducing Bacteria.

It is known that the presence of sulfate decreases the methane yield in the anaerobic digestion systems. Sulfate-reducing bacteria can convert sulfate to hydrogen sulfide competing with methanogens for substrates such as H2 and acetate. The present work aims to elucidate the microbial interactions in biogas production and assess the effectiveness of electron-conductive materials in restoring methane production after exposure to high sulfate concentrations. The addition of magnetite led to a higher methane content in the biogas and a sharp decrease in the level of hydrogen sulfide, indicating i

Depth-dependent microbial communities potentially mediating mercury methylation and various geochemical processes in anthropogenically affected sediments.

Metal contamination and other geochemical alterations affect microbial composition and functional activities, disturbing natural biogeochemical cycles. Therefore, it is essential to understand the influences of multi-metal and geochemical interactions on microbial communities. This work investigated the distributions of total mercury (THg), methylmercury (MeHg), and trace metals in the anthropogenically affected sediment. The microbial communities and functional genes profiles were further determined to explore their association with Hg-methylation and geochemical features. The levels of THg a

Antibiotic contamination amplifies the impact of foreign antibiotic-resistant bacteria on soil bacterial community.

Human activities are stimulating the presence of foreign antibiotic-resistance bacteria (ARB) in soils and antibiotic-contaminated soils are increasing continuously in the world. However, little is known about the impacts of foreign ARB on the indigenous bacterial community in antibiotic-contaminated soil. Herein, using a microcosm experiment we studied the soil bacterial community composition and function (presented with niche structure and niche breadth) in the response to a model ARB (multidrug-resistant Escherichia coli) amendment in the absence and presence of tetracycline contamination.

Development and Characterization of PCE-to-Ethene Dechlorinating Microcosms with Contaminated River Sediment.

An industrial complex in Wonju, contaminated with trichloroethene (TCE), was one of the most problematic sites in Korea. Despite repeated remedial trials for decades, chlorinated ethenes remained as sources of down-gradient groundwater contamination. Recent efforts were being made to remove the contaminants of the area, but knowledge of the indigenous microbial communities and their dechlorination abilities were unknown. Thus, the objectives of the present study were (i) to evaluate the dechlorination abilities of indigenous microbes at the contaminated site, (ii) to characterize which microbe

Sediment prokaryote communities in different sites of eutrophic Lake Taihu and their interactions with environmental factors.

To investigate the temporal variation of the sediment prokaryote communities and their relation with the rapid increase of algae population in Taihu, a shallow eutrophic freshwater Lake, water and surface sediments were sampled from seven sites in different stages of algal bloom. The physicochemical characterization revealed positive correlations among the nutrient (N, P) parameters in the water and sediments, as well as TN/TP ratio 30.79 in average in water and 0.13 in sediments, demonstrating that P content was the limit factor for bloom in Taihu and sediment was an important nutrient resour

Using electron balances and molecular techniques to assess trichoroethene-induced shifts to a dechlorinating microbial community.

This study demonstrated the utility in correlating performance and community structure of a trichloroethene (TCE)-dechlorinating microbial consortium; specifically dechlorinators, fermenters, homoacetogens, and methanogens. Two complementary approaches were applied: predicting trends in the microbial community structure based on an electron balance analysis and experimentally assessing the community structure via pyrosequencing and quantitative polymerase chain reaction (qPCR). Fill-and-draw reactors inoculated with the DehaloR^2 consortium were operated at five TCE-pulsing rates between 14 an

Soil flooding suppresses Fusarium wilt in watermelon by modulating soil chemical properties and microbial community richness.

Soil flooding is a well-documented agricultural practice for disease control, but the underlying mechanisms are still poorly understood. The objective of this study was to investigate the effect of soil flooding on watermelon growth, soil chemical properties, enzyme activities, and the diversity and richness of microbial community structure, and to examine the relationships between these factors and wilt incidence. The results showed that soil flooding reduced the population of FON and Fusarium wilt incidence, promoted plant growth, increased soil AP and AK contents, increased UA and ALPA acti
