Quantification of chemotherapy-induced changes in body composition in pediatric, adolescent, and young adult lymphoma using standard of care CT imaging.

OBJECTIVES: The objective of this study was to use computed tomography (CT) imaging to quantify chemotherapy-induced changes in body composition (BC) in pediatric, adolescent, and young adult (AYA) patients with lymphoma and to compare image-derived changes in BC measures to changes in traditional body mass index (BMI) measures. METHODS: Skeletal muscle (SkM), subcutaneous adipose tissue (SAT), and visceral adipose tissue (VAT) volumes were manually segmented using low-dose CT images acquired from a 10-year retrospective, single-site cohort of 110 patients with lymphoma. CT images and BMI per

Prognostic value of baseline body composition parameters from 18F-FDG PET/CT in angioimmunoblastic T-cell lymphoma patients.

This retrospective study investigated preliminarily the prognostic value of body composition parameters derived from baseline 18F-FDG PET/CT in angioimmunoblastic T-cell lymphoma (AITL) patients. We included 94 treatment-naïve AITL patients diagnosed by histopathology. Based on the axial CT images of the third lumbar vertebra, the areas of skeletal muscle (SM), visceral adipose tissue (VAT), and subcutaneous adipose tissue (SAT) were semi-automatically delineated and standardized to finally obtain SM index (SMI), VAT index (VATI), and SAT index (SATI). Body composition density characterized by

Colon composited B cell lymphoma involving intestinal mesenteric lymph nodes combined with adenocarcinoma and schistosome infection: a case report.

Here we reported one rare case of colon composited B cell lymphoma associated with adenocarcinoma. The composited B cell lymphoma composed of extranodal marginal zone B-cell lymphoma of mucosa-associated lymphoid tissue (MALT lymphoma) and large B-cell lymphoma. Additionally, adjacent intestinal mesenteric lymph modes were involved. And interestingly, schistosome infection was also found in lymphoma and adjacent tissues. The patient was given a usual local intestinal resection for colon adenocarcinoma. But in the site of one surgical margin, 10 cm away from intestinal carcinoma, we found anoth

Composite angioimmunoblastic T cell/diffuse large B-cell lymphoma treated with reduced-intensity conditioning HLA-haploidentical allo-HSCT: a case report and review of the literature.

Cases of diffuse large B-cell lymphoma (DLBCL) occurring together with angioimmunoblastic T cell lymphoma (AITL) are rare. Treatments for AITL and DLBCL composite lymphoma include chemotherapy, targeted therapy, immunomodulatory therapy and hematopoietic stem cell transplantation, but no standard treatment for this aggressive disease has yet been defined. There are no case reports on AITL/DLBCL composite lymphoma treated with allogeneic hematopoietic stem cell transplantation (allo-HSCT). Herein, we report a case of AITL/DLBCL composite lymphoma treated with reduced-intensity conditioning HLA-

HIV and prior exposure to antiretroviral therapy alter tumour composition and tumour: T-cell associations in diffuse large B-cell lymphoma.

Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of lymphoma worldwide, accounting for up to 40% of new non-Hodgkin Lymphoma (NHL) globally. People living with HIV are up to 17 times more likely to develop NHL, and as such, DLBCL is the leading cause of cancer death in this high-risk population. While histologically indistinguishable, HIV-associated (HIV+) and HIV-negative (HIV-) DLBCL are molecularly distinct, and biological differences may have implications for the development of future therapeutic interventions. Further, the impact of immunologic differences in people with H

Increased expression and altered subunit composition of proteasomes induced by continuous proteasome inhibition establish apoptosis resistance and hyperproliferation of Burkitt lymphoma cells.

The proteasome is the main protease for extralysosomal protein degradation in eukaryotic cells, and constitutes a sophisticated high molecular mass proteinase complex underlying a tightly coordinated expression and assembly of multiple subunits and subcomplexes. Here we show that continuous inhibition of proteasomal chymotrypsin-like peptidase activity by the proteasome inhibitor bortezomib induces in human Namalwa Burkitt lymphoma cells increased de novo biogenesis of proteasomes accompanied by increased expression of the proteasome maturation protein POMP, increased expression of 19S-20S-19S

Role of body composition and metabolic parameters extracted from baseline 18F-FDG PET/CT in patients with diffuse large B-cell lymphoma.

This study aimed to clarify the clinical and prognostic role of body composition and metabolic parameters extracted from baseline 18F-FDG PET/CT in patients with diffuse large B-cell lymphoma (DLBCL). We retrospectively collected the clinicopathological and 18F-FDG PET/CT parameters of 181 DLBCL patients. The indexes of skeletal muscle, subcutaneous adipose tissue, and visceral adipose tissue were calculated using the area measured at the 3rd lumbar level normalized for height. Additionally, the metabolic activity of corresponding muscle and adipose tissue, and maximum standardized uptake valu

Tumor Microenvironment Composition and Severe Cytokine Release Syndrome (CRS) Influence Toxicity in Patients with Large B-Cell Lymphoma Treated with Axicabtagene Ciloleucel.

PURPOSE: One of the challenges of adoptive T-cell therapy is the development of immune-mediated toxicities including cytokine release syndrome (CRS) and neurotoxicity (NT). We aimed to identify factors that place patients at high risk of severe toxicity or treatment-related death in a cohort of 75 patients with large B-cell lymphoma treated with a standard of care CD19 targeted CAR T-cell product (axicabtagene ciloleucel). EXPERIMENTAL DESIGN: Serum cytokine and catecholamine levels were measured prior to lymphodepleting chemotherapy, on the day of CAR T infusion and daily thereafter while pa

Composite Epstein-Barr Virus-Associated B-Cell Lymphoproliferative Disorder and Tubular Adenoma in a Rectal Polyp.

Composite tumors are formed when there is intermingling between two components of separate tumors seen histologically. Cases demonstrating composite tubular adenoma with other types of tumors in the colon are rare. Composite tubular adenomas with nonlymphoid tumors including carcinoids, microcarcinoids, and small cell undifferentiated carcinoma have been reported in the literature. The occurrence of composite lymphoma and tubular adenoma within the colorectal tract is extremely rare. Only three cases have been reported and include one case of mantle cell lymphoma and two cases of diffuse large

High-resolution genomic profiling reveals clonal evolution and competition in gastrointestinal marginal zone B-cell lymphoma and its large cell variant.

We studied marginal zone B-cell lymphomas of the gastrointestinal tract including seven small cell lymphomas, eight large cell areas of composite lymphomas and 13 large cell variants using SNP array profiling. We found an increase of genomic complexity with lymphoma progression from small to large cytology, and identified gains of prominent (proto) oncogenes such as REL, BCL11A, ETS1, PTPN1, PTEN and KRAS which were found exclusively in the large cell variants. Copy numbers of ADAM3A, SCAPER and SIRPB1 were varying between the three different modes of presentation, hence suggestive for aberrat

Plasticity of Mature B Cells Between Follicular and Classic Hodgkin Lymphomas: A Series of 22 Cases Expanding the Spectrum of Transdifferentiation.

Follicular lymphoma and classic Hodgkin lymphoma can be associated in composite and/or sequential lymphomas. Common IGH and BCL2 rearrangements have already been identified between both contingents of these entities, but mutation profiles have not yet been investigated. The main objective of this study was to analyze the transdifferentiation process that may occur between Hodgkin and follicular contingents in sequential and composite lymphomas to better characterize these entities. From 2004 to 2020, a retrospective multicentric study was performed, including 9 composite and 13 sequential lymp

A rare presentation of in situ mantle cell lymphoma and follicular lymphoma: a case report and review of the literature.

A 65-year-old gentleman presented with left groin swelling over the course of two months. Physical exam revealed nontender left inguinal adenopathy, and computed tomography scans detected multiple lymph nodes in the mesenteric, aortocaval, and right common iliac regions. An excisional lymph node biopsy was performed. Pathologic evaluation demonstrated follicular center site which stained positive for PAX5, CD20, CD10, Bcl-2, Bcl-6, and mantle zone cells. These findings demonstrated CCND1 and CD5 positivity, suggesting composite lymphoma comprising follicular lymphoma (FL) with in situ mantle c

Development of follicular lymphoma after treatment of diffuse large B-cell lymphoma: two case reports with review of literature.

Diffuse large B-cell lymphoma (DLBCL) is one of the most frequent subtypes of aggressive lymphomas, which can be transformed lymphoma that mostly develops from other types of lymphoma. Cases of DLBCL arising developing after the initial diagnosis of follicular lymphoma (FL) have been reported. However, until now, little few studies have been reported were conducted on the patients with DLBCL and with subsequent development of FL in patients with DLBCL. Here we presented this study with two rare cases of, namely, FL and Composite concurrently occurs with DLBCL and FL developing after the initia

[Combination lymphomas, grey zone lymphomas and future challenges].

Lymphomas are basically divided into Hodgkin and B‑ and T‑cell lymphomas, with diagnosis based on morphological, immunohistochemical and, increasingly, molecular methods. Classic Hodgkin lymphoma is characterised by the presence of Hodgkin-Reed-Sternberg cells, which have a typical marker profile (CD30+, CD15+, Pax5+) and are diagnostically decisive despite their low number. Molecularly, these tumours can be identified as B‑cell lymphomas, with the tumour cells often possessing non-functional immunoglobulin genes. In B‑ and T‑cell lymphomas, the tumour cells in most cases are predominant in ter

[Current aspects of the pathology and differentiation of extranodal marginal zone B-cell lymphoma, MALT-Type, and gastrointestinal diffuse large B-cell lymphoma].

The marginal zone B-cell lymphoma, MALT-type (MZBL, MT) is a low-grade B-cell lymphoma which is predominantly localised in the stomach with a typical morphology and cytogenetic pattern. The coexistence of a diffuse large B-cell lymphoma (DLBCL) with an MZBL, MT in the gastrointestinal tract is defined as a composite lymphoma (ComL) and represents a fascinating model of lymphoma progression. In this review we focus on current aspects regarding the molecular characterisation of MZBL, MT and gastrointestinal DLBCL and their mutual relationships.

Primary thyroid B-cell lymphoma: molecular insights into its clonal evolution and relapse.

Primary thyroid lymphomas comprise largely extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue (EMZL) and diffuse large B-cell lymphoma (DLBCL), followed by follicular lymphoma (FL). They commonly develop from a background of Hashimoto's thyroiditis (HT), where dysregulated immune responses trigger autoreactive infiltrates and drive clonal B-cell evolution. To understand how these lymphomas and their relapse evolve, we investigated 10 cases by mutation profiling, including five with metachronous lymphomas [primary lymphoma (EMZL = 4, DLBCL = 1) with local relapse (EMZL = 3,

Metachronous EBV-positive classical Hodgkin lymphoma following EBV-positive diffuse large B-cell lymphoma: a case report and literature review.

Epstein-Barr virus (EBV) is closely associated with the development of various malignancies, particularly in lymphomas. Diffuse large B-cell lymphoma (DLBCL), as the most common subtype of non-Hodgkin lymphoma, and classical Hodgkin lymphoma (CHL) both originate from B cells, yet they exhibit significant differences in pathological diagnosis and clinical management. While composite lymphoma (CL) with sequential or simultaneous DLBCL and CHL has been reported, cases where both components are EBV-positive remain rare. Here we report a rare case of composite lymphoma with EBV-positive DLBCL follo
