A report of the investigation and control measures instituted after the isolation of toxin-producing Corynebacterium diphtheriae mitis from a child in Leeds.

This report describes the control measures instituted and further investigations undertaken after the isolation of toxin-producing Corynebacterium diphtheriae from a throat swab taken routinely from a nine-year-old boy, recently returned from Pakistan, who had been admitted to hospital in Leeds with hepatitis A infection. Four of his siblings were subsequently shown to have identical Corynebacterium diphtheriae on throat swabbing, except that in one child the organism did not produce toxin. All the children were asymptomatic carriers.

First isolation of nontoxigenic tox gene-bearing Corynebacterium ulcerans from a diphtheria case in Minas Gerais, Brazil.

INTRODUCTION: Diphtheria is a severe, vaccine-preventable infection caused mainly by Corynebacterium diphtheriae, but other species within the C. diphtheriae complex, including C. ulcerans, can also carry the diphtheria toxin (DT) coding gene (tox). Strains classified as “nontoxigenic toxin-gene bearing” (NTTB) harbor the tox gene but do not produce functional DT, although reactivation remains possible. This study reports the first isolation of a NTTB C. ulcerans strain from a diphtheria case in Brazil. METHODS: A bacterial strain was isolated in Minas Gerais (2019) and subjected to phenotypic

Analysis of Corynebacterium silvaticum genomes from Portugal reveals a single cluster and a clade suggested to produce diphtheria toxin.

BACKGROUND: Corynebacterium silvaticum is a pathogenic, gram-positive bacterial species that causes caseous lymphadenitis in wild boars, domestic pigs and roe deer in Western Europe. It can affect animal production and cause zoonosis. Genome analysis has suggested that one strain from Portugal and one from Austria could probably produce the diphtheria toxin (DT), which inhibits protein synthesis and can cause death. METHODS: To further investigate the species genetic diversity and probable production of DT by Portuguese strains, eight isolates from this country were sequenced and compared to

Detection and virulence potential of a phospholipase D-negative Corynebacterium ulcerans from a concurrent diphtheria and infectious mononucleosis case.

Diphtheria by Corynebacterium ulcerans is increasingly occurring in children, adolescents and adults. In addition to diphtheria toxin (DT), phospholipase D (PLD) is considered a virulence factor of C. ulcerans. In the present study, a first case of concurrent diphtheria by a PLD-negative C. ulcerans and infectious mononucleosis (IM) was verified. Clinical and microbiological profiles and binding properties to human Fibrinogen (Fbg), Fibronectin (Fn) and type I collagen (col I) biotinylated proteins and virulence to Caenorhabditis elegans were investigated for C. ulcerans strain 2590 (clinical

Utilization of host iron sources by Corynebacterium diphtheriae: multiple hemoglobin-binding proteins are essential for the use of iron from the hemoglobin-haptoglobin complex.

The use of hemin iron by Corynebacterium diphtheriae requires the DtxR- and iron-regulated ABC hemin transporter HmuTUV and the secreted Hb-binding protein HtaA. We recently described two surface anchored proteins, ChtA and ChtC, which also bind hemin and Hb. ChtA and ChtC share structural similarities to HtaA; however, a function for ChtA and ChtC was not determined. In this study, we identified additional host iron sources that are utilized by C. diphtheriae. We show that several C. diphtheriae strains use the hemoglobin-haptoglobin (Hb-Hp) complex as an iron source. We report that an htaA d

Lack of Detectable Anti-Diphtheria Toxin Antibodies in European Hedgehogs (Erinaceus europaeus) from a Corynebacterium ulcerans Hotspot.

Corynebacterium ulcerans is a zoonotic pathogen causing cutaneous diphtheria. In Belgium, the bacterium is the cause of hedgehog diphtheric disease (HDD) that appears to have emerged from an endemic state within the European hedgehog (Erinaceus europaeus) population. In spring 2025, 11 hedgehogs admitted to a wildlife rehabilitation center were euthanized due to their severe condition. Blood was collected for anti-diphtheria toxin antibody detection, and lesion swabs (if present) and oral cavity swabs were taken, followed by necropsy. Of nine hedgehogs exhibiting HDD-like lesions, five were co

Diphtheria Toxin-Producing Corynebacterium ramonii in Inner-City Population, Vancouver, British Columbia, Canada, 2019-2023.

We conducted patient chart reviews and whole-genome sequencing of wound specimens containing presumptive Corynebacterium ulcerans from Vancouver, British Columbia, Canada, during July 2019-July 2023. Sequencing confirmed 8/14 isolates were C. ramonii and identified 2 distinct clusters. Molecular methods should be used to clinically differentiate potential toxin-producing Corynebacterium spp.

Identification of a two-component signal transduction system from Corynebacterium diphtheriae that activates gene expression in response to the presence of heme and hemoglobin.

Corynebacterium diphtheriae, the causative agent of diphtheria, utilizes various host compounds to acquire iron. The C. diphtheriae hmuO gene encodes a heme oxygenase that is involved in the utilization of heme and hemoglobin as iron sources. Transcription of the hmuO gene in C. diphtheriae is controlled under a dual regulatory mechanism in which the diphtheria toxin repressor protein (DtxR) and iron repress expression while either heme or hemoglobin is needed to activate transcription. In this study, two clones isolated from a C. diphtheriae chromosomal library were shown to activate transcri

Toxigenic Corynebacterium ulcerans in raw milk of a cow with acute mastitis: case report and historical review on milk-transmitted diphtheria.

PURPOSE: Historically, a considerable number of diphtheria outbreaks, mainly in the UK and the USA, have been epidemiologically or bacteriologically linked to the consumption of milk and dairy products. However, with the introduction of milk pasteurization and hygienic improvements in livestock farming and animal welfare, no cases of milk-linked human diphtheria outbreaks or bovine mastitis due to toxigenic corynebacterial have been reported in the recent decades. Here we report the first isolation of a toxigenic Corynebacterium ulcerans strain from the milk of a cow with acute mastitis within

Solution 1H NMR investigation of the active site molecular and electronic structures of substrate-bound, cyanide-inhibited HmuO, a bacterial heme oxygenase from Corynebacterium diphtheriae.

The molecular structure and dynamic properties of the active site environment of HmuO, a heme oxygenase (HO) from the pathogenic bacterium Corynebacterium diphtheriae, have been investigated by (1)H NMR spectroscopy using the human HO (hHO) complex as a homology model. It is demonstrated that not only the spatial contacts among residues and between residues and heme, but the magnetic axes that can be related to the direction and magnitude of the steric tilt of the FeCN unit are strongly conserved in the two HO complexes. The results indicate that very similar contributions of steric blockage o

NGS-based phylogeny of diphtheria-related pathogenicity factors in different Corynebacterium spp. implies species-specific virulence transmission.

BACKGROUND: Diphtheria toxin (DT) is produced by toxigenic strains of the human pathogen Corynebacterium diphtheriae as well as zoonotic C. ulcerans and C. pseudotuberculosis. Toxigenic strains may cause severe respiratory diphtheria, myocarditis, neurological damage or cutaneous diphtheria. The DT encoding tox gene is located in a mobile genomic region and tox variability between C. diphtheriae and C. ulcerans has been postulated based on sequences of a few isolates. In contrast, species-specific sequence analysis of the diphtheria toxin repressor gene (dtxR), occurring both in toxigenic and

An increase in non-toxigenic Corynebacterium diphtheriae infections in Poland--molecular epidemiology and antimicrobial susceptibility of strains isolated from past outbreaks and those currently circulating in Poland.

OBJECTIVES: An increase in non-toxigenic Corynebacterium diphtheriae infections--mainly invasive infections--has been observed in countries with high vaccination coverage. However, reasons for this situation are unknown. In this study we characterized and compared human clinical isolates of non-toxigenic C. diphtheriae strains isolated from infections that have occurred over recent years and C. diphtheriae strains isolated from diphtheria cases from past outbreaks in Poland. METHODS: We determined biotypes, genotypes, the occurrence of plasmids, and antimicrobial susceptibilities of 19 clinic
