A ferritin-based mosaic-like nanovaccine elicits effective cross-protection against H1N1 and H3N2 swine influenza viruses.

Swine influenza viruses (SIVs), particularly the H1N1 and H3N2 subtypes, pose ongoing threats to the swine industry and global public health. Current commercial vaccines generally provide limited cross-protection against heterologous viruses due to the frequent antigenic drift and shift in SIVs, highlighting the need for broad-spectrum vaccines. Self-assembling nanoparticles (NPs) can elicit strong and broad immune responses, representing a promising platform for the development of broad-spectrum nanovaccines. In this study, two newly-screened conserved epitopes (H1-3 and H1-5) and (H3-1 and H

A STING-adjuvanted outer membrane vesicle nanoparticle vaccine vs. Klebsiella pneumoniae elicits broad capsule-type cross-protection.

Klebsiella pneumoniae (Kp) is a leading cause of hospital- and community-acquired infections and is increasingly resistant to last-line antibiotics, positioning it as a World Health Organization-designated priority pathogen. Vaccine development has been hindered by the extensive diversity of Kp exopolysaccharide capsules. We developed a cellular nanoparticle (CNP) platform in which bacterial outer membrane vesicles (OMVs) are stably coated onto a STING (stimulator of interferon genes)-adjuvanted nanoparticle core (CNP-KpSTING). The resulting ∼100-nm nanoparticles are homogenous and optimized f

Cross-species blood transcriptional correlates of BCG-mediated protection against tuberculosis include innate and adaptive immune processes.

The immune mechanisms induced by the Bacillus Calmette-Guérin (BCG) vaccine, and the subset of which mediate protection against tuberculosis (TB), remain poorly understood. This is further complicated by difficulties to verify vaccine-induced protection in humans. Although research in animal models, namely mice and non-human primates (NHPs), has begun to close this knowledge gap, discrepancies in the relative importance of biological pathways across species limit the utility of animal model-derived biological insights in humans. To address these challenges, we applied a systems modeling framew

Development and evaluation of neutralizing antibodies for cross-protection against West Nile virus and Japanese encephalitis virus.

BACKGROUND: West Nile virus is a severe zoonotic pathogen that can cause severe central nervous system symptoms in humans and horses, and is fatal for birds, chickens and other poultry. With no specific drugs or vaccines available, antibody-based therapy is a promising treatment. This study aims to develop neutralizing antibodies against West Nile virus and assess their cross-protective potential against Japanese encephalitis virus. METHODS: Monoclonal antibodies against WNV and JEV were isolated by hybridoma technology. The therapeutic efficacy of these antibodies was evaluated using a mouse

Sun exposure and protection practices among Florida college marching band members and alumni: a cross-sectional study.

In the USA, skin cancer is the most common cancer, more than all other cancers combined. Skin cancer may be prevented by using sun protection. This is particularly important in 18-29-year-olds who, compared to older individuals, experience sunburns more frequently. Moreover, in locations close to the equator, such as Florida, people are at an increased risk of developing melanoma. College marching band members spend a significant amount of time in the sun, as much as 25 h weekly practicing and performing at games, yet little is known about this population. We examined and compared sun exposure

Do modified live virus vaccines against bovine viral diarrhea induce fetal cross-protection against HoBi-like Pestivirus?

Bovine Pestivirus heterogeneity is a major challenge for vaccines against bovine viral diarrhea (BVD). In breeding herds, fetal protection is a high priority issue. To some degree, fetal infections in vaccinated heifers have been attributed to the antigenic diversity of bovine Pestiviruses. The purpose of this study was to assess fetal protection against a divergent bovine Pestivirus (Hobi-like Pestivirus, HoBiPeV) with a commercially available modified live vaccine (MLV) claiming fetal protection against BVDV 1 and BVDV 2 up to one year after the first inoculation. Five vaccinated and four un

Live Attenuated African Swine Fever Viruses as Ideal Tools to Dissect the Mechanisms Involved in Cross-Protection.

African swine fever (ASF) has become the major threat for the global swine industry. Furthermore, the epidemiological situation of African swine fever virus (ASFV) in some endemic regions of Sub-Saharan Africa is worse than ever, with multiple virus strains and genotypes currently circulating in a given area. Despite the recent advances on ASF vaccine development, there are no commercial vaccines yet, and most of the promising vaccine prototypes available today have been specifically designed to fight the genotype II strains currently circulating in Europe, Asia, and Oceania. The serogroup B meningococcal outer membrane vesicle-based vaccine 4CMenB induces cross-species protection against Neisseria gonorrhoeae.

There is a pressing need for a gonorrhea vaccine due to the high disease burden associated with gonococcal infections globally and the rapid evolution of antibiotic resistance in Neisseria gonorrhoeae (Ng). Current gonorrhea vaccine research is in the stages of antigen discovery and the identification of protective immune responses, and no vaccine has been tested in clinical trials in over 30 years. Recently, however, it was reported in a retrospective case-control study that vaccination of humans with a serogroup B Neisseria meningitidis (Nm) outer membrane vesicle (OMV) vaccine (MeNZB) was a

A canine vaccine against Leptospira serovars Icterohaemorrhagiae, Canicola and Grippotyphosa provides cross protection against Leptospira serovar Copenhageni.

Efficacy of the Leptospira components of multivalent vaccine DAPPi-L was previously demonstrated against virulent challenge with three serovars of Leptospira interrogans (Canicola, Icterohaemorrhagiae and Grippotyphosa) carried out 14 days after primary vaccination. In this study we demonstrate that this vaccine provides, two weeks after vaccination, an additional protection (prevention of mortality, clinical signs, renal infection, bacterial excretion, renal carriage and renal lesions) against fatal leptospirosis due to Leptospira interrogans serovar Copenhageni (serovar of major medical impo

Recombinant influenza H7 hemagglutinin containing CFLLC minidomain in the transmembrane domain showed enhanced cross-protection in mice.

Since February 2013, H7N9 influenza virus, causing human infections with high mortality in China, has been a potential pandemic threat. The H7N9 viruses are found to diverge into distinct genotypes as other influenza viruses; thus a vaccine that can provide sufficient cross-protection against different genotypes of H7N9 viruses is urgently needed. Our previous studies demonstrated that the HA-based structural design approach by introducing a CFLLC minidomain into transmembrane domain (TM) of H1, H5 or H9 hemagglutinin (HA) proteins by replacing with H3 subtype HA TM could enhance their cross-p

Cross-protection elicited by primary and booster vaccinations against Japanese encephalitis: a two-year follow-up study.

BACKGROUND: The inactivated Vero cell-derived vaccine (JE-VC, IXIARO) has replaced the traditional mouse brain-derived preparations (JE-MB) in travelers' vaccinations against Japanese encephalitis. We showed recently that a single JE-VC dose efficiently boosts immunity in JE-MB-primed vaccinees, and that JE-VC elicits cross-protective immunity against non-vaccine genotypes, including the emerging genotype I. While these studies only provided short-term data, the present investigation evaluates the longevity of seroprotection in the same volunteers. METHODS: The study comprised 48 travelers wh

Cross-species protection mediated by a Bordetella bronchiseptica strain lacking antigenic homologs present in acellular pertussis vaccines.

The Bordetella species are Gram-negative bacterial pathogens that are characterized by long-term colonization of the mammalian respiratory tract and are causative agents of respiratory diseases in humans and animals. Despite widespread and efficient vaccination, there has been a world-wide resurgence of pertussis, which remains the leading cause of vaccine-preventable death in developed countries. It has been proposed that current acellular vaccines (Pa) composed of only a few bacterial proteins may be less efficacious because of vaccine-induced antigenic shifts and adaptations. [Lassa and Mozambique viruses: cross protection in experiments on mice and action of immunosuppressants on experimental infections].

The pathogenicity of Mozambique virus for random-bred mice and CBA mice was studied. In contrast to Lassa virus, intracerebral inoculation of newborn zandom-bred mice with Mozambique virus (1000 PFU/mouse) results in death of the animals. The pathogenic properties of both viruses for adult CBA mice were found to be similar: intracerebral inoculation of the viruses caused death of the animals within 6-8 days, but not intraperitoneal inoculation. With the latter, they produced a population of immunocompetent cells protecting syngeneic recipient mice against the lethal intracerebral inoculation o

A Multivalent Dengue Fusion Protein ΔcNS1-cEDIII-ΔnNS3 Confers Cross-Serotype Protection and Durable Immunity in Mice.

Current dengue vaccines remain limited by serotype-dependent efficacy and interference from preexisting anti-dengue immunity. We developed a novel multivalent fusion protein vaccine composed of three engineered dengue virus (DENV) components: a C-terminal truncated nonstructural protein 1 (ΔcNS1) to block NS1-mediated pathologic effects without harmful cross-reactivity, a consensus envelope protein domain III (cEDIII) to induce broad neutralizing antibodies, and an N-terminal truncated NS3 (ΔnNS3) to enhance cellular immune responses. In a murine dengue disease model, three-dose immunization w

An advanced graph algorithm-based protection strategy for detecting kilometric and cross-country faults in DC microgrid.

Low voltage DC microgrids (LVDC) are on rise, because of increase in usage of electronics-based utility loads. However, the protection and safety aspects of these grids remain unresolved due to the fault current magnitude, unnecessary tripping, and blinding of protection. In this paper, an adaptive differential protection system incorporated with an advanced graph algorithm is proposed for DC microgrid. This graph algorithm is a combination of Fenwick tree and Bidirectional Dijkstra algorithm. Fenwick tree algorithm is used to determine the network configuration and Bidirectional Dijkstra algo

Vaccination with ancestral SARS-CoV-2 spike adjuvanted with TLR agonists provides cross-protection against XBB.1.

Many different platforms have been used to develop highly protective vaccines against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in humans. However, protection has eroded over time due to the emergence of antigenically diverse viral variants, especially the Omicron subvariants. One successful platform for the generation of SARS-CoV-2 vaccines are recombinant spike protein vaccines, of which two are licensed in the United States and Europe. Typically, purified recombinant protein antigens are poorly immunogenic and adjuvants must be included in the formulation. Here, we adjuva

Vaccination with nanoparticles displaying gH/gL from Epstein-Barr virus elicits limited cross-protection against rhesus lymphocryptovirus.

Epstein-Barr virus (EBV) is associated with infectious mononucleosis, cancer, and multiple sclerosis. A vaccine that prevents infection and/or EBV-associated morbidity is an unmet need. The viral gH/gL glycoprotein complex is essential for infectivity, making it an attractive vaccine target. Here, we evaluate the immunogenicity of a gH/gL nanoparticle vaccine adjuvanted with the Sigma Adjuvant System (SAS) or a saponin/monophosphoryl lipid A nanoparticle (SMNP) in rhesus macaques. Formulation with SMNP elicits higher titers of neutralizing antibodies and more vaccine-specific CD4+ T cells. All

Commercial vaccine provides cross-protection by reducing colonization of Salmonella enterica serovars Infantis and Hadar in turkeys.

Human foodborne outbreaks with antibiotic-resistant Salmonella enterica associated with contaminated poultry products have recently involved serogroup C serovars Infantis and Hadar. The current study evaluated a commercially available Salmonella vaccine for cross-protection against Infantis and Hadar serovars in turkeys. The live, attenuated S. Typhimurium (serogroup B) vaccine significantly reduced colonization of intestinal tissues (cecum, cecal tonsils, and cloaca) by serovars Infantis (C1) and Hadar (C2) and significantly limited systemic dissemination to the spleen.
