Aggregates-on-chip: a microfluidic device for crowding plant cells in culture.

INTRODUCTION: Microfluidic devices enable the study of cellular properties in a highly controlled environment while allowing close observation. One important aspect of a plant cell's environment is the influence of neighboring cells on cell growth through chemical signals and mechanical forces. However, while the use of microfluidics in plant cell biology has expanded in recent years, very few systems provide cell-cell contact together with long-term growth and imaging. METHODS: Here, we develop a microfluidic device that uses arrays of cylindrical cups to gently capture and confine multiple

Influence of Molecular Crowding on Aptamers and Application in Lateral Flow Assays.

It is generally believed that the practical application of aptamers in food safety monitoring is often hindered by their attenuated performance in complex, crowded food matrices. As a fundamental characteristic of realistic sample matrices, how molecular crowding influences aptamer functionality was investigated. Using non-pretreated milk as a model crowded environment, it was found that molecular crowding significantly enhanced the performance of an Escherichia coli O157:H7-specific aptamer, which is contrary to common sense. Compared to a dilute buffer, the crowding condition increased bindi

Unraveling the Molecular Jam: How Crowding Shapes Protein Aggregation in Neurodegenerative Disorders.

Protein misfolding and aggregation are the hallmarks of neurodegenerative diseases including Huntington's disease, Parkinson's disease, Alzheimer's disease, and prion diseases. A crowded cellular environment plays a crucial role in modulating protein aggregation processes in vivo and the pathological aggregation of proteins linked to different neurodegenerative disorders. Here, we review recent studies examining the effects of various crowding agents, such as polysaccharides, polyethylene glycol, and proteins like BSA and lysozyme on the behaviors of aggregation of several amyloidogenic peptid

Macromolecular crowding potently stimulates DNA supercoiling activity of Mycobacterium tuberculosis DNA gyrase.

Macromolecular crowding, manifested by high concentrations of proteins and nucleic acids in living cells, significantly influences biological processes such as enzymatic reactions. Studying these reactions in vitro, using agents such as polyetthylene glycols (PEGs) and polyvinyl alcohols (PVAs) to mimic intracellular crowding conditions, is essential due to the notable differences from enzyme behaviors observed in diluted aqueous solutions. In this article, we studied Mycobacterium tuberculosis (Mtb) DNA gyrase under macromolecular crowding conditions by incorporating PEGs and PVAs into the DN

Trisomy 21 induces pericentrosomal crowding delaying primary ciliogenesis and mouse cerebellar development.

Trisomy 21, the genetic cause of Down syndrome, disrupts primary cilia formation and function, in part through elevated Pericentrin, a centrosome protein encoded on chromosome 21. Yet how trisomy 21 and elevated Pericentrin disrupt cilia-related molecules and pathways, and the in vivo phenotypic relevance remain unclear. Utilizing ciliogenesis time course experiments combined with light microscopy and electron tomography, we reveal that chromosome 21 polyploidy elevates Pericentrin and microtubules away from the centrosome that corral MyosinVA and EHD1, delaying ciliary membrane delivery and m

A clinical approach to managing open-bite malocclusion associated with severe crowding.

Open bite has been identified as one of the most challenging malocclusions in orthodontics. The treatment approach is complex, the resulting esthetics may not meet the patient's expectations, and the chance of recurrence is high. A 13-year-old girl presented a skeletal Class II malocclusion associated with anterior and posterior open bite, maxillary transverse deficiency, severe anterior crowding, and a hyperdivergent facial pattern. Orthodontic treatment was performed with a Haas expander and subsequent standard edgewise appliances, and the 4 first premolars were extracted. The vertical contr

Lytic transglycosylases mitigate periplasmic crowding by degrading soluble cell wall turnover products.

The peptidoglycan cell wall is a predominant structure of bacteria, determining cell shape and supporting survival in diverse conditions. Peptidoglycan is dynamic and requires regulated synthesis of new material, remodeling, and turnover - or autolysis - of old material. Despite exploitation of peptidoglycan synthesis as an antibiotic target, we lack a fundamental understanding of how peptidoglycan synthesis and autolysis intersect to maintain the cell wall. Here, we uncover a critical physiological role for a widely misunderstood class of autolytic enzymes, lytic transglycosylases (LTGs). We

Relationship between oral function and mandibular anterior crowding in early mixed dentition.

The position of the dentition is considered to be determined by a combination of forces exerted by the perioral muscles and tongue. We hypothesized that mandibular anterior crowding may be related to abnormalities in the development of oral function. To determine the relationship between oral function and mandibular anterior crowding in early mixed dentition. A total of 61 children (30 boys, 31 girls) with early mixed dentition were included in this study. Height, body weight, maximum occlusal force, lip-closing strength, and maximum tongue pressure were measured in all participants, and their

Crowding and Pausing Strongly Affect Dynamics of Kinesin-1 Motors along Microtubules.

Molecular motors of the kinesin-1 family move in a directed and processive fashion along microtubules. It is generally accepted that steric hindrance of motors leads to crowding effects; however, little is known about the specific interactions involved. We employ an agent-based lattice gas model to study the impact of interactions that enhance the detachment of motors from crowded filaments on their collective dynamics. The predictions of our model quantitatively agree with the experimentally observed concentration dependence of key motor characteristics including their run length, dwell time,

Three-dimensional evaluation of mandibular anterior dental crowding in digital dental casts.

INTRODUCTION:: Digital dental models provide a more accurate and comprehensive assessment of orthodontic cases. Although this technique is quite promising, there are few three-dimensional measurements methods described in the literature. OBJECTIVE:: The aim of this study was to propose a method for assessing the degree of mandibular anterior dental crowding in the three planes of space, using digital dental models. METHODS:: Thirty dental casts were selected and scanned by Maestro 3D Dental Scanner and imported by Geomagic Qualify 2013 software. The degree of crowding was calculated by two e

Crowding out of long-term care insurance: evidence from European expectations data.

Long-term care (LTC) is the largest insurable risk that old-age individuals face in most western societies. However, the demand for LTC insurance is still ostensibly small in comparison with the financial risk. One explanation that has received limited support is that expectations of either 'public sector funding' and 'family support' crowd out individual incentives to seek insurance. This paper aims to investigate further the aforementioned motivational crowding-out hypothesis by developing a theoretical model and by drawing on an innovative empirical analysis of representative European surve

Study: hospitals struggle to implement proven strategies for eliminating ED boarding, crowding.

A new study suggests that proven strategies for eliminating boarding and crowding in the ED are being left on the table in many hospitals because leadership has not stepped forward to eliminate pockets of resistance. Further, there is new evidence that changes in practice intensity in the ED are contributing to crowding even though some of these changes were designed to do the opposite. Strategies that can ease crowding include smoothing out the schedule for elective procedures, moving boarded patients up to the hallways on inpatient floors, and appointing a bed czar to oversee bed utilization

In vivo effects of putative crowding factors on development of Hymenolepis diminuta.

During in vitro incubation, Hymenolepsis diminuta secretes substances into the medium that inhibit DNA synthesis in the germinative region of freshly isolated, uncrowded worms. Of the many substances that are released by H. diminuta into the medium, earlier studies indicate that only succinate, acetate, glucosaminic acid, and cGMP are responsible for the inhibition. In the present report, effects of these putative crowding factors on worm development in vivo were examined. At 7 days postinfection the proximal end of the host's intestine was catheterized and perfused with test solution. The tes

Glandular Crowding of the Endometrium: Clinical Outcomes of a Diagnostic Gray Zone.

Endometrioid intraepithelial neoplasia (EIN) is a well-defined premalignant lesion of the endometrium, characterized by crowded glands exceeding the amount of stroma, cytologic atypia distinct from background endometrium, and a sufficiently sized lesion (often designated as 1 mm). However, some endometrial samples contain foci of crowded glands that fall short of these criteria. These diagnostically indeterminate areas, often described as "atypical glandular crowding," present challenges in both interpretation and clinical management. To assess their significance, we conducted a retrospective

Dental arch morphology and factors associated with dental crowding: a systematic review.

INTRODUCTION: Dental crowding is considered the most common type of malocclusion, ranking third among the most prevalent oral pathologies. Several genetic and environmental factors are associated with dental crowding. Dental arch dimensions are essential for determining the degree of crowding in the dentition. This review aims to assess the influence of dental arch dimensions on the prevalence of crowding. MATERIAL AND METHODS: A study was conducted in accordance with PRISMA guidelines and registered in PROSPERO (CRD420251082613). An electronic literature search from 2010 to 2025 was conducte
