Differential role of melatonin in restoration of age-induced alterations in daily rhythms of expression of various clock genes in suprachiasmatic nucleus of male Wistar rats.

Aging is associated with changes in several basic parameters of circadian rhythms in mammals leading to circadian dysfunction. The hypothalamic Suprachiasmatic nucleus (SCN) regulates neuronal, endocrine and behavioral rhythms through the expression of various clock genes and release of melatonin from pineal gland. In the present study, we investigated the effect of aging on daily rhythms of various clock genes such as rPer1, rPer2, rCry1, rCry2 and rBmal1 in the SCN of male Wistar rats. The m-RNA expression levels of these genes were studied by using quantitative Polymerase Chain Reaction (qP

Liquiritigenin ameliorates circadian rhythm-mediated osteoblast differentiation through Rorα and Nr1d1 signaling pathways.

Circadian rhythm disorders can destroy the balance of bone metabolism and hence closely related to bone metabolic diseases. To further explore the relationship between circadian rhythm and bone metabolism, the sleep deprivation model of zebrafish induced by continuous light and the MC3T3-EI cell model induced by Nr1d1-specific agonist STL1267 (STL1267) were used to study the intervention mechanism of liquiritigenin on bone metabolism imbalance mediated by circadian rhythm. Melatonin and curcumin reestablish disturbed circadian gene expressions and restore locomotion ability and eclosion behavior in Drosophila model of Huntington's disease.

Deficit in locomotion (motor) ability and disturbance of the circadian behavior and sleep-wake pattern characterize Huntington's disease (HD). Here, we examined the disturbance of circadian timing with the progression of HD pathogenesis, and tested the efficacy of melatonin and curcumin in preventing the motor deficit and disturbed eclosion behavior in the Drosophila model of HD. To examine circadian timing, we assayed mRNA expression of genes of the transcriptional feedback (TF) loop that generates the near 24-h rhythmicity. We performed qPCR of the Period, Timeless, Clock, Cycle, Clockwork,

Targeting the xylosyltransferase TMEM5 in glioblastoma to modulate CLOCK and CRY1 expression and restore temozolomide sensitivity via the circadian signaling axis.

BACKGROUND: Glioblastoma (GBM) is the most aggressive primary brain tumor and exhibits marked resistance to standard therapies, including temozolomide (TMZ). Circadian rhythm disruption is increasingly recognized in GBM; however, the molecular factors linking circadian dysregulation to metabolic adaptability and therapy resistance remain poorly defined. This study investigates the role of transmembrane protein 5 (TMEM5; ribitol xylosyltransferase 1, RXYLT1) as a circadian-associated regulator in GBM. METHODS: Integrative in silico analyses of TCGA-GBM and CircaDB datasets were performed to ev

Infrared-A (IR-A) modulates the p53 pathway and diurnal clock in human skin and subcutaneous adipose tissue in vivo.

Infrared-A (IR-A) radiation is thought to protect the skin from damage due to ultraviolet radiation and alter its carcinogenic potential, possibly by preventing keratinocyte apoptosis via p53 pathway modulation. Solar radiation, including IR-A, is a major factor in contributing to the entrainment of circadian rhythms. We examined the impacts of IR-A on the intrinsic clock and transcriptomics in individuals with different diurnal preference to understand the mechanisms by which IR-A acts in the human skin and subcutaneous adipose tissue. IR-A caused multiple gene expressional changes, mainly as

Expression of circadian core clock genes in fibroblasts of human gingiva and periodontal ligament is modulated by L-Mimosine and hypoxia in monolayer and spheroid cultures.

OBJECTIVE: The circadian clock is involved in a plethora of physiological processes including bone formation and tooth development. While expression of circadian core clock genes was observed in various tissues, their role in the periodontium is unclear. We hypothesized that periodontal cells express circadian core clock genes and that their levels are modulated by hypoxia mimetic agents and hypoxia. MATERIAL AND METHODS: Fibroblasts of human gingiva (GF) and periodontal ligament (PDLF) in monolayer and spheroid cultures were treated with the hypoxia mimetic agent L-Mimosine (L-MIM) or hypoxi

Aminooxyacetic acid up-regulates the Cry1 and Bmal1 clock gene in a sirtuin 1 dependent manner. In vitro study.

The regulation of the cyclic oscillation of the components of the circadian clock is complex in itself. Numerous clock interactions with processes and molecules present in cells further complicate this mechanism. Recently, the anti-aging protein Silencing Information Regulator Two family member, SIRT1, has been linked with the molecular circadian clock. In this study, we investigated the in vitro effect of aminooxyacetic acid on SIRT1 expression in relation to circadian dynamics of Cry1 and Bma11 expressions in serum shocked NIH-3 T3 and HaCaT cells. The study was carried out in the context of

Cry1 and Tef gene polymorphisms are associated with major depressive disorder in the Chinese population.

INTRODUCTION: Accumulating evidences indicate that circadian abnormalities lead to sleep disorder, neurodegenerative diseases and depression. We have reported that the polymorphisms of a clock-related gene, Tef, contributed to the risk of sleep disturbances and depression in the Parkinson disease. The objective of the present study was to examine whether the three clock genes we previously studied are associated with major depressive disorder (MDD) in the Chinese population. METHODS: 105 Subjects with MDD and 485 control subjects participated in this case-control study. Demographics, Mini-men

Development of the rhythmic melatonin secretion in the embryonic chicken pineal gland.

In order to elucidate details on the development of the circadian clock, the effects of light on the in vitro melatonin (MT) release and the presence of mRNAs of several clock genes in the embryonic chicken pineal gland were investigated. Chicken embryos of various developmental stages were exposed to stimuli of light in vitro in dynamic, four day long bioassay (perifusion). MT secretion and mRNA levels of Cry1, Cry2, Clock and Bmal2 clock genes were determined. Our conclusions: (1) environmental illumination modified MT secretion from explanted embryonic pineal glands as early as on the 13th

Red light delays petal discoloration in Brunfelsia acuminata by regulating anthocyanin biosynthetic genes.

Brunfelsia acuminata (Pohl) Benth. exhibits a distinctive post-anthesis transition in flower color from purple to white, which is closely associated with anthocyanin degradation. However, the photobiological mechanisms regulating pigment stability during petal senescence remain poorly understood. In this study, we investigated how different light qualities affect anthocyanin degradation, flavonoid and phenolic metabolism, and the expression of key anthocyanin biosynthetic genes in detached petals and flowers on intact plants. Among nine continuous light-quality treatments, red light most effec

Disrupted riboflavin metabolism impairs circadian rhythm in ovarian granulosa cells: a novel mechanism driving DOR.

Diminished ovarian reserve (DOR) is a major cause of female infertility and a significant indicator of reproductive aging, yet its underlying pathogenesis remains poorly understood. In this study, we initially assessed the expression of core circadian genes in ovarian granulosa cells from DOR patients and in ovaries from cyclophosphamide (Cy)-induced DOR model rats. We observed dysregulated expression of circadian genes in DOR ovaries. Proteomic analysis further revealed a significant decrease in riboflavin kinase (RFK) expression in DOR rat ovaries, concomitant with abnormal elevation of the

Karyopherin Alpha 2-Expressing Pancreatic Duct Glands and Intra-Islet Ducts in Aged Diabetic C414A-Mutant-CRY1 Transgenic Mice.

Our earlier studies demonstrated that cysteine414- (zinc-binding site of mCRY1-) alanine mutant mCRY1 transgenic mice (Tg mice) exhibit diabetes characterized by the reduction of β-cell proliferation and by β-cell dysfunction, presumably caused by senescence-associated secretory phenotype- (SASP-) like characters of islets. Earlier studies also showed that atypical duct-like structures in the pancreas developed age-dependently in Tg mice. Numerous reports have described that karyopherin alpha 2 (KPNA2) is highly expressed in cancers of different kinds. However, details of the expression of KPN

Magnetoreceptive CRY/MagR complexes: linking circadian redox signalling to protein aggregation in Alzheimer's and Parkinson's disease.

BACKGROUND AND PURPOSE: Neurodegenerative disorders such as Alzheimer's disease (AD) and Parkinson's disease (PD) pose an escalating challenge to neuroscience, as disease-modifying therapies remain elusive despite substantial advances in molecular and cellular understanding. These disorders share convergent pathological features, including protein misfolding and aggregation, mitochondrial dysfunction, oxidative stress, impaired proteostasis, and disruption of circadian regulation. Identifying integrative frameworks that connect these processes is therefore essential for advancing conceptual mo

DNA methylation of core clock genes in patients with major depressive disorder: Association with air pollution exposure and disease severity.

BACKGROUND: Major Depressive Disorder (MDD) is a multifactorial disease which could be influenced by exposure to air pollution through disruption of sleep-wake cycles and other circadian-related behaviors. Our study aimed to investigate the interplay between air pollution exposure, DNA methylation of core clock genes involved in circadian rhythms, and MDD severity. METHODS: Four hundred sixteen MDD patients (64 % females) agreed to participate and donated a blood sample to measure DNA methylation of the core clock genes CRY1, PER1, PER2, CLOCK, BMAL1. MDD severity and functioning was assessed

The role of PACAP in the control of circadian expression of clock genes in the chicken pineal gland.

Several features of the molecular circadian oscillator of the chicken pineal gland show homology with those in the mammalian SCN. Studies have shown the effects of PACAP on the mammalian SCN, but its effects on the expression of clock genes in the avian pineal gland have not yet been demonstrated. Clock and Cry1 expression was analyzed in pineal glands of chicken embryos after exposure to PACAP-38 in vitro. PACAP reduced expression of both clock genes within 2h. Ten hours after exposure, mRNA contents exceeded that of the controls. Our results support the hypothesis that the molecular clock ma

Impact of social isolation on circadian rhythmicity and clock genes expression in the prefrontal cortex and hippocampus of mice exposed to simulated rotating shiftwork/jetlag.

Circadian rhythms regulate fundamental physiological and behavioral functions, yet are increasingly disrupted by contemporary environmental stressors such as rotating shift work and social isolation. Although each disruptor independently compromises circadian homeostasis, their interactive effects remain insufficiently characterized. Given the role of social cues as non-photic zeitgebers, we investigated whether social isolation exacerbates behavioral and molecular circadian disruption induced by repeated light-dark phase shifts, and whether social interaction confers resilience. Male AKR mice

Npas4 is activated by melatonin, and drives the clock gene Cry1 in the ovine pars tuberalis.

Seasonal mammals integrate changes in the duration of nocturnal melatonin secretion to drive annual physiologic cycles. Melatonin receptors within the proximal pituitary region, the pars tuberalis (PT), are essential in regulating seasonal neuroendocrine responses. In the ovine PT, melatonin is known to influence acute changes in transcriptional dynamics coupled to the onset (dusk) and offset (dawn) of melatonin secretion, leading to a potential interval-timing mechanism capable of decoding changes in day length (photoperiod). Melatonin offset at dawn is linked to cAMP accumulation, which dire

Loss of hepatic circadian organization in fatty liver syndrome identifies CLOCK as a regulator of lipid homeostasis in laying hens.

Fatty liver syndrome (FLS) is a major metabolic disorder in laying hens, but how hepatic lipid dysregulation is linked to circadian disruption remains unclear. Here, we investigated the role of circadian regulation in FLS pathogenesis and focused on the core clock gene Clock Circadian Regulator (CLOCK). In a high-fat diet-induced FLS model in Lohmann pink layers, affected birds showed hepatomegaly, hepatic lipid accumulation, elevated serum total cholesterol, triglycerides, alanine aminotransferase, and aspartate aminotransferase, increased hepatic pro-inflammatory gene expression, and reduced

Assessment of continuous positive airway pressure effect on the circadian clock signaling pathway in obstructive sleep apnea patients.

Obstructive sleep apnea (OSA) is associated with circadian rhythm dysregulation plausibly through affecting clock genes. The study's purpose was to investigate the effect of one-night continuous positive airway pressure treatment (CPAP treatment) on circadian clock genes: BMAL1, CLOCK, CRY1, and PER1 at mRNA and protein levels. The study included 30 OSA patients, who underwent diagnostic polysomnography (PSG) and next a one-night effective CPAP treatment with PSG monitoring (CPAP). The blood was collected in the evening before and the morning after PSG and CPAP. Protein levels and mRNA express
