Deep space flight imposes higher levels of damage on biological organisms; however, its specific effects on rice remain unclear. To investigate the variations in DNA methylation under deep space flight conditions, this study examined rice seeds carried by Chang'e-5. After 23 days of lunar orbital flight, the samples were planted in an artificial climate chamber and subjected to transcriptome and DNA methylation sequencing during the tillering and heading stages. The methylation patterns in the rice genome exhibited variability in response to lunar orbital stressors. DNA methylation alters the

Quercetin is a flavonoid compound widely found in vegetables, fruits, and medicinal plants. It has carbonyl and phenolic hydroxyl groups in its structure that can easily form complexes with metal ions. In this study, we reacted quercetin in water at 95 °C with five metal trace elements commonly found in plants, namely calcium, magnesium, zinc, iron(III), and copper. Each supernatant was then examined by electrospray ionization-time-of-flight mass spectrometry (ESI-TOF MS). The results showed that quercetin can form complexes with Ca2+, Mg2+, Zn2+, and Cu2+ in ratios of 2:1, 3:1, and 3:2. Inter

Galphimia glauca is a plant that is endemic to Mexico and has been commonly used since pre-Hispanic times to treat various illnesses, including central nervous system disorders and inflammation. The first studies investigating a natural population of G. glauca in Mexico showed that the plant has anxiolytic and sedative activities in mice and humans. The plant's bioactive compounds were isolated and identified, and they belong to a family of nor-secofriedelanes called galphimines. The integration of DNA barcoding and thin-layer chromatography analysis was performed to clarify whether the botani

Marine habitats are Earth's largest aquatic ecosystems, yet little is known about marine organism's genomes. Molecular studies can unravel their genetics print, thus shedding light on specie's adaptation and speciation with precise authentication. However, extracting high molecular weight DNA from marine organisms and subsequent DNA library preparation for whole genome sequencing is challenging. The challenges can be explained by excessive metabolites secretion that co-precipitates with DNA and barricades their sequencing. In this work, we sought to resolve this issue by describing an optimize

Low temperature is an abiotic stress factor that affects plant development and geographic expansion, resulting in significant economic losses in worldwide food production each year. Through evolution, plants have developed intricate adaptation systems, with epigenetic regulation-modifying gene expression without altering DNA sequences-being pivotal in the cold stress response and memory formation. This review provides a systematic overview of the role of the main epigenetic mechanisms, including histone modifications, DNA methylation, non-coding RNA regulation, and chromatin remodeling, in pla

Plants respond to herbivory with the induction of resistance, mediated by distinct phytohormonal signaling pathways and their interactions. Phloem feeders are known to induce plant resistance via the salicylic acid pathway, whereas biting-chewing herbivores induce plant resistance mainly via the jasmonate pathway. Here, we show that a specialist caterpillar (biting-chewing herbivore) and a specialist aphid (phloem feeder) differentially induce resistance against Pieris brassicae caterpillars in Arabidopsis (Arabidopsis thaliana) plants. Caterpillar feeding induces resistance through the jasmon

The plant defensin PDF1.2 has previously been shown to accumulate systemically via a salicylic acid-independent pathway in leaves of Arabidopsis upon challenge by fungal pathogens. To further investigate the signalling and transcriptional processes underlying plant defensin induction, a DNA fragment containing 1184 bp and 1232 bp upstream of the transcriptional and translational start sites, respectively, was cloned by inverse PCR. To test for promoter activity this DNA fragment was linked to the beta-glucuronidase (GUS)-encoding region of the UidA gene as a translational fusion and introduced

Triploid plants are usually sterile or produce non-viable seeds, but may produce fruits. This study examined the morphological and reproductive characteristics of triploid Passiflora cincinnata Masters, with a focus on fruit and seed production. Triploid plants were obtained through in vitro cultivation of the endosperm via somatic embryogenesis, whereas diploid plants were raised through seed germination. Diploid and triploid plants were confirmed by flow cytometry and cytogenetic analyses. Cross-pollination and self-pollination yielded, on average, 18 fruits per plant in the cytotype diploid

DNA replication stress poses a severe threat to genome stability. It is well-known that SOG1 plays a crucial role in DNA replication stress response in plants. Although ATM-mediated phosphorylation and CRL4PRL1-mediated polyubiquitination of SOG1 play important roles in regulating DNA damage response (DDR), the posttranslational modifications of SOG1 during the replication stress response are little known. Here we show that, during replication stress, the activity of the GSK3-like kinase BIN2 is activated by an unknown mechanism independent of ATR. Subsequently, BIN2 phosphorylates SOG1 at T24

Antimicrobial resistance (AMR) is becoming an increasing global concern and the anaerobic digestion (AD) process represents a potential transmission route when digestates are used as fertilizing agents. AMR contaminants, e.g. antibiotic-resistant bacteria (ARB) and plasmid-mediated antibiotic resistance genes (ARGs) have been found in different substrates and AD systems, but not yet been investigated in plant-based substrates. AMR transfer from soils to vegetable microbiomes has been observed, and thus crop material potentially represents a so far neglected AMR load in agricultural AD processe

Infectious diseases pose one of the most significant threats to the survival of great apes in the wild. The critically endangered mountain gorilla (Gorilla beringei beringei) is at high risk for contracting human pathogens because approximately 60% of the population is habituated to humans to support a thriving ecotourism program. Disease surveillance for human and non-human primate pathogens is important for population health and management of protected primate species. Here, we evaluate discarded plants from mountain gorillas and sympatric golden monkeys (Cercopithecus mitis kandti), as a no

The eukaryotic ATAXIA TELANGIECTASIA AND RAD3-RELATED (ATR) kinase is essential for ensuring genomic integrity under conditions that cause replication defects. It triggers a transient cell cycle arrest and DNA repair by activating the DNA damage response pathway. While ATR kinases appear to be functionally conserved, species-specific phenotypes can be observed across the plant kingdom. To investigate the ancestral roles of the plant ATR, we identified its homolog in the model liverwort Marchantia polymorpha, which we named MpATR. Similar to other plant species, MpATR preserves meristematic cel

Envelope glycoprotein E of the dengue virus, which plays a crucial role in its entry into host cells, has an immunogenic domain III (EIII, amino acids 297-394), which is capable of inducing neutralizing antibodies. However, mice immunized with EIII protein without adjuvant elicited low immune responses. To improve low immune responses, a DNA fragment, consisting of cholera toxin B subunit and EIII gene (CTB-EIII), was constructed and introduced into tobacco plant cells (Nicotiana tabacum L. cv. MD609) by Agrobacterium tumefaciens-mediated transformation methods. The integration and transcripti

BACKGROUND: Coal burning provides 70% of the energy for China's industry and power, but releases large quantities of polycyclic aromatic hydrocarbons (PAHs) and other pollutants. PAHs are reproductive and developmental toxicants, mutagens, and carcinogens.
OBJECTIVE: We evaluated the benefit to neurobehavioral development from the closure of a coal-fired power plant that was the major local source of ambient PAHs.
METHODS: The research was conducted in Tongliang, Chongqing, China, where a coal-fired power plant operated seasonally before it was shut down in May 2004. Two identical prospectiv

Cryopreservation is important for the conservation of plant species. Oxidative stress, amongst the variety of stresses induced during the different steps of cryopreservation, can cause shifts in antioxidant contents that can lead to reduced post-cryopreservation survival and regrowth. A link between cryo-induced ascorbate deficiency and oxidative-induced DNA damage is not proven but could potentially contribute to poor regeneration of cryopreserved shoot tips. Accordingly, this study investigated cryo-tolerance (survival and regeneration of shoot tips), ascorbate and glutathione antioxidant co

Plant pollen and spores are promising forensic indicators for geographic sourcing because their DNA is persistent and their community composition is shaped by local vegetation and environmental conditions. However, reference datasets designed specifically for forensic palynological applications remain limited. Here, we evaluated the feasibility of environmental DNA (eDNA) metabarcoding for geographic sourcing of palynological trace evidence across urban functional zones in Xi'an, China. Airborne palynomorphs were collected from nine city-wide sites representing seven functional zones in spring

BACKGROUND: Viruses have notable effects on agroecosystems, wherein they can adversely affect plant health and cause problems (e.g., increased biosecurity risks and economic losses). However, our knowledge of their diversity and interactions with specific host plants in ecosystems remains limited. To enhance our understanding of the roles that viruses play in agroecosystems, comprehensive analyses of the viromes of a wide range of plants are essential. High-throughput sequencing (HTS) techniques are useful for conducting impartial and unbiased investigations of plant viromes, ultimately formin

The study of plant DNA polymerases lags far behind that concerning their animal or yeast counterpart. In this work we describe the first extensive purification to apparent homogeneity, as well as a detailed biochemical and immunological characterization, of a low molecular weight DNA polymerase (DNA polymerase CI) purified from wheat embryos. The monomeric enzyme is a basic protein having a molecular weight of 52 kDa. Polyclonal antibodies raised in rabbits against DNA polymerase CI did not inhibit animal DNA polymerases alpha and beta or wheat DNA polymerase A, whereas wheat DNA polymerases C

Baicalin is an anti-HIV drug purified from the traditional Chinese medicinal plant Scutellaria Baicalensis Georgi. Baicalin has proven to be electroactive at pyrolytic graphite (PG) electrodes. We thus studied its interaction with DNA via the electrochemical approach. We observed that the peak currents corresponding to the baicalin reduction-oxidation (redox) reaction significantly decrease upon the addition of DNA. With complementary ultraviolet/visible (UV/Vis) spectroscopic evidence, we suggest that baicalin binds to DNA through intercalation. This feature has enabled baicalin to discrimina

γ-terpinene, α-terpinene, p-cymene, and myrcene are monoterpenes found in many essential oils extracted from a variety of plants and spices. Myrcene also occurs naturally in plants such as hops, cannabis, lemongrass, and verbena and is used as a flavoring agent in food and beverage manufacturing. In this research, the biological efficacy of γ-terpinene, α-terpinene, p-cymene, and myrcene was studied in human cell lines (HeLa, SH-SY5Y, and HDFa). Cytotoxicity, cell proliferation, cell migration, and morphology assays were performed to obtain detailed information on the anticancer properties. Ou
