Chewing lice (Phthiraptera) on a wild Golden eagle (Aquila chrysaetos) and a zoo-kept Eurasian griffon vulture (Gyps fulvus) in Tyrol, Austria.

Chewing lice (Phthiraptera) are obligate and permanent ectoparasites commonly found on birds. The life cycle of these insects is completed on the body of the host and therefore many are host specific. This is the first report of chewing lice on a wild Golden eagle (Aquila chrysaetos) and a zoo-kept Eurasian griffon vulture (Gyps fulvus) in Tyrol, Austria. Three different species of chewing lice were identified: Craspedorrhynchus aquilinus was found on Aquila chrysaetos and Colpocephalum turbinatum and Falcolipeurus quadripustulatus were found on Gyps fulvus. The lice were identified morphologi

Diversity and seasonality of ectoparasite burden on two species of Madagascar fruit bat, Eidolon dupreanum and Rousettus madagascariensis.

BACKGROUND: Bats are important reservoir hosts for a variety of pathogens, some of which are transmitted by ectoparasite vectors including mites, fleas, lice, ticks, and bat flies (families Nycteribiidae and Streblidae). All these ectoparasite taxa are known to parasitize two endemic fruit bats of Madagascar, Eidolon dupreanum and Rousettus madagascariensis. We aimed to describe the diversity of ectoparasite infestation for both bat species through morphological observation and DNA barcoding and elucidate ecological and climatic correlates of seasonal nycteribiid parasitism of these hosts. Development of a SNP barcode to genotype Babesia microti infections.

Babesia microti is tick-borne disease that is an emerging threat to public health due to increasing prevalence and expanding geographic range. Detection and constant surveillance of babesiosis is imperative for predicting pathogen expansion. Leveraging our whole genome sequence (WGS) analyses of B. microti, we developed a single nucleotide polymorphism (SNP)-based high resolution melt (HRM) surveillance tool. We developed our HRM assay using available sequence data and identified 775 SNPs. From these candidate SNPs, we developed a 32-SNP barcode that is robust and differentiates geographically

DNA barcoding of five Japanese encephalitis mosquito vectors (Culex fuscocephala, Culex gelidus, Culex tritaeniorhynchus, Culex pseudovishnui and Culex vishnui).

Culex mosquitoes can act as vectors of several important diseases, including Japanese encephalitis, West Nile virus, St. Louis encephalitis and equine encephalitis. Besides the neurological sequelae caused in humans, Japanese encephalitis can lead to abortion in sows and encephalitis in horses. Effective vector control and early diagnosis, along with continuous serosurveillance in animals, are crucial to fight this arboviral disease. However, the success of vector control operations is linked with the fast and reliable identification of targeted species, and knowledge about their biology and e

Advances and challenges in barcoding pathogenic and environmental Leptospira.

Leptospirosis is a zoonotic bacterial disease of global importance. A large spectrum of asymptomatic animal hosts can carry the infection and contribute to the burden of human disease. Environmental sources of human contamination also point to the importance of a hydrotelluric reservoir. Leptospirosis can be caused by as many as 15 different pathogenic or intermediate Leptospira species. However, classification of these bacteria remains complicated through the use of both serological and genetic classification systems that show poor correlation. With the advent of molecular techniques, DNA-bas

Pilot longitudinal mosquito surveillance study in the Danube Delta Biosphere Reserve and the first reports of Anopheles algeriensis Theobald, 1903 and Aedes hungaricus Mihályi, 1955 for Romania.

BACKGROUND: Mosquito-borne viruses (moboviruses) are of growing importance in many countries of Europe. In Romania and especially in the Danube Delta Biosphere Reserve (DDBR), mosquito and mobovirus surveillance are not performed on a regular basis. However, this type of study is crucially needed to evaluate the risk of pathogen transmission, to understand the ecology of emerging moboviruses, or to plan vector control programmes. METHODS: We initiated a longitudinal mosquito surveillance study with carbon dioxide-baited Heavy Duty Encephalitis Vector Survey traps at four sampling sites to ana

Establishment of a two-tiered molecular identification system for Dracaena species - the source plants of dragon's blood.

BACKGROUND: The unresolved taxonomy of Dracaena species, the botanical sources of the prized traditional medicine Dragon's blood, poses significant challenges to resource conservation, material traceability, and quality control. This study aimed to resolve these taxonomic ambiguities by developing and validating a high-resolution, integrated molecular authentication system. Based on 26 original plant samples from the genus Dracaena, along with relevant DNA barcode loci retrieved from NCBI, five previously reported DNA barcodes were evaluated and assessed. Simple sequence repeat (SSR) sequences

Pathology associated with three new Clinostomum metacercariae from Argentina with morphological and DNA barcode identification.

In the Laboratory of Parasites of Fishes, Crustaceans and Mollusks (CEPAVE), we undertook a parasitological study on three species of fish from the Espinal and Esteros del Iberá ecoregions of Argentina. Clinostomid metacercariae were found parasitizing Characidium rachovii, Crenicichla vittata and Gymnogeophagus balzanii. In this study, we analysed the damage that these parasites inflict on their hosts through the evaluation of histological sections. In addition, Clinostomum metacercariae were identified using morphological characters and DNA barcoding. In the pathological analysis, we observe

Bloodmeal metabarcoding reveals host feeding patterns for Aedes aegypti and Culex quinquefasciatus in Jutiapa, Guatemala and Texas, USA.

Mosquito host contact determines arboviral transmission efficiency. Aedes aegypti and Culex quinquefasciatus are important vectors of dengue, Zika, chikungunya, West Nile virus, and other arboviruses, yet their feeding patterns remain poorly characterized in many tropical regions. We used bloodmeal metabarcoding to detect DNA from multiple vertebrate species within individual blood-fed mosquitoes collected from rural Guatemala and south Texas, USA. Mosquitoes were collected using aspiration in Guatemala and BG-Sentinel traps in south Texas. We calculated forage ratios (FR) to assess host utili

Aedes (Ochlerotatus) amateuri Ortega and Zavortink (Diptera: Culicidae) in the Madrean Sky Islands of Arizona, USA: a new country record.

The mosquito community of the Madrean Sky Islands of southeastern Arizona is diverse and distinct with several species that are endemic or at their northern distributional limits. We collected mosquitoes across many of southeastern Arizona's Sky Island mountain ranges. Here, we present new collection records supported by morphological and molecular evidence that indicate the presence of Aedes (Ochlerotatus) amateuri in at least two Sky Islands ranges, the Santa Rita Mountains and the Dragoon Mountains. Aedes amateuri was described in 2019, previously known only from the type locality in Nuevo

Geographic distribution and environmental associations of Psorophora mexicana (Diptera: Culicidae) in the Yucatan Peninsula, Mexico.

Psorophora mexicana (Bellardi) is a rare mosquito species with limited information on its taxonomy, geographic distribution, and ecological preferences. In Mexico, presence records are scarce and largely outdated, particularly in the Yucatan Peninsula. Here, we document the current distribution of Ps. mexicana based on collections conducted between 2017 and 2025 across 14 localities in Campeche, Quintana Roo, and Yucatan states, including the first confirmed record for Quintana Roo. Specimens were identified morphologically and genetically using the mitochondrial region cytochrome c oxidase I

Trypanosomatid detection in Culicoides biting midges (Diptera: Ceratopogonidae) from leishmaniasis-endemic Songkhla Province, Southern Thailand: Microscopy and nanopore metabarcoding reveal parasite diversity.

Leishmaniasis caused by Leishmania (Mundinia) spp. has recently emerged in Southeast Asia, including Thailand. Although molecular studies have detected Leishmania DNA in Culicoides midges, complete parasite development, which proves vector competence, remains undemonstrated in field specimens. We collected 215 parous/gravid (n = 195) and blood-engorged (n = 20) female Culicoides representing at least 10 species from two leishmaniasis foci in Songkhla Province, Southern Thailand. Specimens underwent microscopic dissection, culturing in Schneider's medium, ITS1/SSU rRNA-PCR for Leishmania/trypan

Recurrent and niche-specific functional bacteriome of maize hybrid revealed by integrated metabarcoding and culturomics.

The plant microbiome plays a pivotal role in plant survival in natural habitats by facilitating nutrient acquisition, stress adaptation, and disease suppression, while also offering opportunities to enhance crop productivity and climate resilience. However, the distribution of persistent and culturable bacteriome across maize-associated niches and their functional potential remain poorly resolved. This study integrated metagenomic next-generation sequencing (mNGS-based metabarcoding) and culturomics to characterise the maize-associated bacteriome of bulk soil, rhizoplane, phylloplane, and cob

Precise lineage tracking using molecular barcodes demonstrates fitness trade-offs for ivermectin resistance in nematodes.

A fundamental tenet of evolutionary genetics is that the direction and strength of selection on individual loci vary with the environment. Barcoded evolutionary lineage tracking is a powerful approach for high-throughput measurement of selection within experimental evolution that to date has largely been restricted to studies within microbial systems, largely because the random integration of barcodes within animals is limited by physical and molecular protection of the germline. Here, we use the recently developed TARDIS barcoding system in Caenorhabditis elegans to implement the first random

DARLIN mouse for in vivo lineage tracing at high efficiency and clonal diversity.

Lineage tracing is a powerful tool to study cell history and cell dynamics during tissue development and homeostasis. An increasingly popular approach for lineage tracing is to generate high-frequent mutations at given genomic loci, which can serve as genetic barcodes to label different cell lineages. However, current lineage tracing mouse models suffer from low barcode diversity and limited single-cell lineage coverage. We recently developed the DARLIN mouse model by incorporating three barcoding arrays within defined genomic loci and combining Cas9 and terminal deoxynucleotidyl transferase (

Metabarcoding Reveals a Potentially Undescribed Columnaris-Causing Bacterium in Peracute Skin Disease of Rainbow Trout (Oncorhynchus mykiss, Walbaum).

Columnaris-causing bacteria (CCB) represent a group of four Flavobacterium species, previously classified under Flavobacterium columnaris, causing a threatening condition in salmonid farming characterised by cutaneous and gill lesions, commonly referred to as 'saddleback disease'. A peracute skin disease outbreak with high mortality in rainbow trout (Oncorhynchus mykiss) farms in Northern Italy was investigated. The disease presented with skin discoloration and scale lifting without internal organ abnormalities, leading to a weekly cumulative mortality of up to 80%. The disease was successfull

DNA barcoding shows that Toxocara cati infecting domestic and wild felids is a species complex.

The insufficient attention paid to the importance of Toxocara cati in the epidemiology of toxocariasis has resulted in significant gaps in the understanding of the true zoonotic potential of this parasite. Therefore, further research is necessary to understand its biology and epidemiology. In this article, the hypothesis of possible speciation in the T. cati complex was tested using cox1 sequences from T. cati individuals infecting domestic and wild felids from different parts of the world. Using DNA barcoding, a phylogenetic analysis was performed that grouped T. cati representatives into fiv

Development of a new barcode-based, multiplex-PCR, next-generation-sequencing assay and data processing and analytical pipeline for multiplicity of infection detection of Plasmodium falciparum.

BACKGROUND: Simultaneous infection with multiple malaria parasite strains is common in high transmission areas. Quantifying the number of strains per host, or the multiplicity of infection (MOI), provides additional parasite indices for assessing transmission levels but it is challenging to measure accurately with current tools. This paper presents new laboratory and analytical methods for estimating the MOI of Plasmodium falciparum. METHODS: Based on 24 single nucleotide polymorphisms (SNPs) previously identified as stable, unlinked targets across 12 of the 14 chromosomes within P. falciparu
