A Quantitative Systems Pharmacology Approach to Infer Pathways Involved in Complex Disease Phenotypes.

Designing effective therapeutic strategies for complex diseases such as cancer and neurodegeneration that involve tissue context-specific interactions among multiple gene products presents a major challenge for precision medicine. Safe and selective pharmacological modulation of individual molecular entities associated with a disease often fails to provide efficacy in the clinic. Thus, development of optimized therapeutic strategies for individual patients with complex diseases requires a more comprehensive, systems-level understanding of disease progression. Quantitative systems pharmacology

Computer-focused modification of physician prescribing behavior.

Prescribing data downloaded from the MUMPS-based hospital information system of the Department of Veterans Affairs has been used to target specific interventions aimed at changing physician prescribing patterns. Focused interventions aimed at specific groups of physicians, patients, clinics, and drugs have proven effective in reducing pharmaceutical costs by rationalizing prescribing patterns. The results indicate the potential usefulness of a microcomputer with a commercial database program in manipulating hospital information system data.

Alpha-gal syndrome: potential for a hypersensitivity reaction after the use of dental products: Literature review and case report.

BACKGROUND: Alpha-gal syndrome (AGS) is associated with a potentially severe delayed immunoglobulin E-based hypersensitivity reaction produced via transmission of a salivary oligosaccharide (galactose-⍺-1,3-galactose) predominately from the bite of the lone star tick (Amblyomma americanum). Sensitized people are at an increased risk of experiencing cross-reactivity reactions to numerous foods, pharmaceuticals, and medical and dental products that could result in a spectrum of pathophysiological responses, ranging from gastrointestinal and cutaneous disturbances to anaphylaxis. The authors have

Accelerated Repurposing and Drug Development of Pulmonary Hypertension Therapies for COVID-19 Treatment Using an AI-Integrated Biosimulation Platform.

The COVID-19 pandemic has reached over 100 million worldwide. Due to the multi-targeted nature of the virus, it is clear that drugs providing anti-COVID-19 effects need to be developed at an accelerated rate, and a combinatorial approach may stand to be more successful than a single drug therapy. Among several targets and pathways that are under investigation, the renin-angiotensin system (RAS) and specifically angiotensin-converting enzyme (ACE), and Ca2+-mediated SARS-CoV-2 cellular entry and replication are noteworthy. A combination of ACE inhibitors and calcium channel blockers (CCBs), a c

Estimating the Share of Social Surplus Generated by Pharmaceutical Innovation Accruing to Manufacturers: A Systematic Literature Review.

INTRODUCTION: Pharmaceutical innovation operates within an implicit social contract that seeks to balance incentives for future drug development with equitable access to current therapies. A key metric for evaluating this balance is producer surplus (PS), which represents the share of total social surplus (the sum of consumer surplus [CS] and PS) captured by manufacturers. We explore the determinants and distribution of estimated supply-side surplus share across therapeutic areas, drug classes, and product life cycles. METHODS: We conducted a systematic literature review of empirical studies

Pharmacist-led antimicrobial stewardship: A systematic review of implementation barriers, facilitators, and policy-to-practice gaps in Bangladesh.

BACKGROUND: Antimicrobial resistance (AMR) disproportionately affects low- and middle-income countries (LMICs). Bangladesh has adopted a National Action Plan (2023-2028) yet exemplifies the persistent gap between pharmaceutical policy frameworks and frontline antimicrobial stewardship (AMS) practice. No prior systematic review has synthesised barriers, facilitators, and AMS strategies across all Bangladesh healthcare settings, or positioned findings within a pharmaceutical-policy and pharmacy-practice frame. OBJECTIVES: To synthesise the documented barriers, facilitators, and core and supplem

Auto-Generated Physiological Chain Data for an Ontological Framework for Pharmacology and Mechanism of Action to Determine Suspected Drugs in Cases of Dysuria.

INTRODUCTION: Patients often take several different medications for multiple conditions concurrently. Therefore, when adverse drug events (ADEs) occur, it is necessary to consider the mechanisms responsible. Few approaches consider the mechanisms of ADEs, such as changes in physiological states. We proposed that the ontological framework for pharmacology and mechanism of action (pharmacodynamics) we developed could be used for this approach. However, the existing knowledge base contains little data on physiological chains (PCs). OBJECTIVE: We aimed to investigate a method for automatically ge

Large-scale integration of heterogeneous pharmacogenomic data for identifying drug mechanism of action.

A variety of large-scale pharmacogenomic data, such as perturbation experiments and sensitivity profiles, enable the systematical identification of drug mechanism of actions (MoAs), which is a crucial task in the era of precision medicine. However, integrating these complementary pharmacogenomic datasets is inherently challenging due to the wild heterogeneity, high-dimensionality and noisy nature of these datasets. In this work, we develop Mania, a novel method for the scalable integration of large-scale pharmacogenomic data. Mania first constructs a drug-drug similarity network through integr

Drug dispensings among elderly in the year before colon cancer diagnosis versus matched cancer-free controls.

WHAT IS KNOWN AND OBJECTIVE: The concomitant use of multiple drugs is common among the general population of elderly. The aim of this study was to provide an overview of which drugs are dispensed to elderly in the year before colon cancer diagnosis and to compare this with cancer-free controls. METHODS: Data from the Eindhoven Cancer Registry were linked to the PHARMO Database Network. Patients with colon cancer aged ≥70 years were included and matched with controls on gender, year of birth and postal code. Proportions of cases and controls with ≥1 dispensing of each WHO ATC-2-level drug duri

The effect of formulations and experimental conditions on in vitro human skin permeation-Data from updated EDETOX database.

In vitro methods are commonly used in order to estimate the extent of systemic absorption of chemicals through skin. Due to the wide variability of experimental procedures, types of skin and data analytical methods, the resulting permeation measures varies significantly between laboratories and individuals. Inter-laboratory and inter-individual variations with the in vitro measures of skin permeation lead to unreliable extrapolations to in vivo situations. This investigation aimed at a comprehensive assessment of the available data and development of validated models for in vitro skin flux of

PharmMapper server: a web server for potential drug target identification using pharmacophore mapping approach.

In silico drug target identification, which includes many distinct algorithms for finding disease genes and proteins, is the first step in the drug discovery pipeline. When the 3D structures of the targets are available, the problem of target identification is usually converted to finding the best interaction mode between the potential target candidates and small molecule probes. Pharmacophore, which is the spatial arrangement of features essential for a molecule to interact with a specific target receptor, is an alternative method for achieving this goal apart from molecular docking method. Integrating gene expression and clinical data to identify drug repurposing candidates for hyperlipidemia and hypertension.

Discovering novel uses for existing drugs, through drug repurposing, can reduce the time, costs, and risk of failure associated with new drug development. However, prioritizing drug repurposing candidates for downstream studies remains challenging. Here, we present a high-throughput approach to identify and validate drug repurposing candidates. This approach integrates human gene expression, drug perturbation, and clinical data from publicly available resources. We apply this approach to find drug repurposing candidates for two diseases, hyperlipidemia and hypertension. We screen >21,000 compo

Application of Supervised SOM Algorithms in Predicting the Hepatotoxic Potential of Drugs.

The hepatotoxic potential of drugs is one of the main reasons why a number of drugs never reach the market or have to be withdrawn from the market. Therefore, the evaluation of the hepatotoxic potential of drugs is an important part of the drug development process. The aim of this work was to evaluate the relative abilities of different supervised self-organizing algorithms in classifying the hepatotoxic potential of drugs. Two modifications of standard counter-propagation training algorithms were proposed to achieve good separation of clusters on the self-organizing map. A series of optimizat
