Human autoantibody to defensin: disease association with hyperreactive onchocerciasis (sowda).

Chronic hyperreactive onchodermatitis (sowda) is a severe form of onchocerciasis observed in a subset of individuals infected with the filarial nematode Onchocerca volvulus. SDS-PAGE and immunoblot analyses of O. volvulus adult worm extracts were used to characterize the antigens of the marked antibody response of sowda patients. One 2.5-kD antigen was recognized by sera from all 35(100%) sowda patients that were studied. In comparison, only 7 of 44 (16%) patients with generalized onchocerciasis and 11 of 21 (52%) of exposed individuals with no microfilariae in skin snips and no signs of disea

Differential Expression of Defensins and Other Innate Immune Effectors by Indian Isolates of Chikungunya Virus.

Chikungunya virus (CHIKV) infection has been associated with heightened disease burden and fatality rate, especially post its resurgence in 2006, with presentation of various atypical symptoms. Its re-emergence and the subsequent periodic outbreaks indicate the constant evolution of the virus due to the incorporation of mutations in its genome. This can potentially elicit a differential induction of innate immune response of varying degrees, which has been poorly studied in the case of CHIKV. Leveraging an immunocompetent epithelial model cell line to define the varying internalization and rep

An improved lentiviral system for efficient expression and purification of β-defensins in mammalian cells.

β-Defensins are a family of conserved small cationic antimicrobial peptides with different significant biological functions. The majority of mammalian β-defensins are expressed in epididymis, and many of them are predicted to have post-translational modifications. However, only a few of its members have been well studied due to the limitations of expressing and purifying bioactive proteins with correct post-translational modifications efficiently. Here we developed a novel Fc tagged lentiviral system and Fc tagged prokaryotic expression systems provided new options for β-defensins expression a

Delineation of interfaces on human alpha-defensins critical for human adenovirus and human papillomavirus inhibition.

Human α-defensins are potent anti-microbial peptides with the ability to neutralize bacterial and viral targets. Single alanine mutagenesis has been used to identify determinants of anti-bacterial activity and binding to bacterial proteins such as anthrax lethal factor. Similar analyses of α-defensin interactions with non-enveloped viruses are limited. We used a comprehensive set of human α-defensin 5 (HD5) and human neutrophil peptide 1 (HNP1) alanine scan mutants in a combination of binding and neutralization assays with human adenovirus (AdV) and human papillomavirus (HPV). We have identifi

Full sequence and characterization of two insect defensins: immune peptides from the mosquito Aedes aegypti.

We report the complete amino acid sequence and biological activity of two immune peptides, from the yellow fever mosquito Aedes aegypti, that are induced in response to infection. Both peptides display biological activity against the Gram positive microbe Micrococcus luteus and substantial sequence homology to insect defensins, small heat-stable, antibiotic peptides previously described from several non-vector insects. These mosquito peptides, designated Ae. aegypti defensins A and B, are isoforms. Defensin B is the most abundant antibacterial peptide in this species whereas defensin A is much

Beta-Defensin-2 and Beta-Defensin-3 Reduce Intestinal Damage Caused by Salmonella typhimurium Modulating the Expression of Cytokines and Enhancing the Probiotic Activity of Enterococcus faecium.

The intestinal microbiota is a major factor in human health and disease. This microbial community includes autochthonous (permanent inhabitants) and allochthonous (transient inhabitants) microorganisms that contribute to maintaining the integrity of the intestinal wall, modulating responses to pathogenic noxae and representing a key factor in the maturation of the immune system. If this healthy microbiota is disrupted by antibiotics, chemotherapy, or a change in diet, intestinal colonization by pathogenic bacteria or viruses may occur, leading to disease. To manage substantial microbial exposu

From venom peptides to neurotherapeutics: BmK defensins and short-chain peptides as modulators of ion channels.

Scorpions, having inhabited the Earth long before the emergence of humans, represent an ancient lineage of arthropods. While often regarded with fear due to their potential to induce severe pain or fatal envenomation, scorpion venoms constitute complex cocktails of bioactive molecules known as toxins. Notably, these toxic components have been repurposed in medical research as valuable sources for therapeutic development. In traditional Chinese medicine (TCM), the venom of Buthus martensii Karsch (BmK), commonly referred to as the Chinese scorpion, has been historically employed in the treatmen

Preliminary study on the role of human defensins, interleukins and PCSK9 in early and late preeclampsia.

The lack of reliable methods for preeclampsia (PE) early diagnosis limits the opportunities for timely prevention, diagnosis and treatment. This study aims to identify the alterations of biochemical parameters and the immune system activity to build a panel of markers that can support preeclampsia diagnosis. For this study, we recruited 30 pregnant women: 10 healthy pregnant women (CTR); 10 pregnant women with early preeclampsia (EP); 10 pregnant women with late preeclampsia (LP). We evaluated lipid profile and, by gene expression, we assessed PCSK9, IL-2, IL-6, IL-8, IL-10, TNF-α and TGF-β. M

Biomarker assessment in chronic rhinitis and chronic rhinosinusitis: Endothelin-1, TARC/CCL17, neopterin, and α-defensins.

BACKGROUND: Chronic rhinitis (CR) is characteristically divided into several major clinical phenotypes: allergic rhinitis (AR); nonallergic, noninfectious rhinopathy (NAR); and chronic rhinosinusitis (CRS). CRS has two phenotypic variants: CRS with nasal polyps (CRSwNP) and CRS without nasal polyps (CRSsNP). An area of growing interest is to identify biologic markers that could assess different aspects of CR phenotypes. OBJECTIVE: The aim of the study was to evaluate four CR biomarkers: endothelin-1 (ET-1), thymus and activation-regulated chemokine (CCL17), neopterin, and α-defensins in subje

Evidence that the role of plant defensins in radish defense responses is independent of salicylic acid.

Radish leaves contain two homologous 5-kDa plant defensins which accumulate systemically upon infection by fungal pathogens (F.R.G. Terras et al., 1995, Plant Cell 7: 573-588). Here we report on the molecular cloning of the cDNAs encoding the two pathogen-inducible plant defensin isoforms from radish (Raphanus sativus L.) leaves. Tissue-print and whole-leaf electroblot immunostaining showed that the plant defensin peptides not only accumulate at high levels at or immediately around the infection sites in leaves inoculated with Alternaria brassicicola, but also accumulate in healthy tissue furt

Defensins as natural antimicrobial peptide scaffolds against antimicrobial-resistant pathogens: mechanisms, resistance risks, and translational prospects.

The growing threat of antimicrobial resistance has increased the need for anti-infective approaches beyond conventional single-target antibiotics. Defensins, a conserved family of cysteine-rich antimicrobial peptides, are promising candidates owing to their structural stability, membrane activity, target-specific mechanisms, immunomodulatory functions, and potential synergy with existing antibiotics. This critical narrative review discusses defensins as potential therapeutics against antimicrobial-resistant pathogens, with a focus on structure-activity relationships, bacterial envelope biology

Human β-defensins and their synthetic analogs: Natural defenders and prospective new drugs of oral health.

As a family of cationic host defense peptides, human β-defensins (HBDs) are ubiquitous in the oral cavity and are mainly synthesized primarily by epithelial cells, serving as the primary barrier and aiming to prevent microbial invasion, inflammation, and disease while maintaining physiological homeostasis. In recent decades, there has been great interest in their biological functions, structure-activity relationships, mechanisms of action, and therapeutic potential in oral diseases. Meanwhile, researchers are dedicated to improving the properties of HBDs for clinical application. In this revie

Divergent impacts of tocilizumab and colchicine in COVID-19-associated coagulopathy: the role of alpha-defensins.

Patients who are severely affected by coronavirus disease 2019 (COVID-19) may develop a delayed onset 'cytokine storm', which includes an increase in interleukin-6 (IL-6). This may be followed by a pro-thrombotic state and increased D-dimers. It was anticipated that tocilizumab (TCZ), an anti-IL-6 receptor monoclonal antibody, would mitigate inflammation and coagulation in patients with COVID-19. However, clinical trials with TCZ have recorded an increase in D-dimer levels. In contrast to TCZ, colchicine reduced D-dimer levels in patients with COVID-19. To understand how the two anti-inflammat

Molecular Identification and Antibacterial Activity Analysis of Blue Fox (Vulpes lagopus) β-Defensins 108 and 122.

The blue fox (Vulpes lagopus), a fur-bearing animal, is an important component of the breeding industry in China. Semen quality is a key factor for the reproductive process and the breeding effectiveness of the farmed blue fox. However, bacterial contamination in semen samples utilized in artificial fertilization is very common. The β-defensins, a class of important antimicrobial peptides in mammals, could protect the reproductive system of male animals from bacterial invasion, maintain the stability of the genital tract microenvironment and improve semen quality. In this study, molecular clon
