Dentine dysplasia type 1 is a rare and complex dental anomaly. Our aim was to conduct a morphometric assessment of a dentinal dysplasia type 1c (DD1c) caries-free mandibular second molar, extracted due to symptomatic apical periodontitis. Controls consisted of five intact mandibular second molars. Micro-computed tomography analysis showed that the DD1c volume % for enamel, dentine/cementum and pulp chamber fell in the 0.36th, 99.97th and 0.09th percentiles of the control teeth (P < 0.01). It also revealed an extremely complicated root canal system in the DD1c tooth with a varying degree of den

OBJECTIVES: The effects of protein deficiency and sucrose on formation and mineralization of dentine and dental caries in the molars of young rats were investigated.
MATERIALS AND METHODS: Two groups of weaning Wistar rats received raw soy flour to induce protein deficiency with sucrose or starch as the carbohydrate source; the other two groups received skim milk powder as the protein source with sucrose or starch as the carbohydrate source. At the onset, tetracycline was injected to mark the dentine formed at that moment. After 6 weeks, lower molars were sectioned sagittally, and the areas o

Inadequate dentin bonding persists as a major cause of secondary caries and restoration failure, accounting for >60% of global dental restorative costs. Prevailing adhesion theory attributes this to challenges in displacing water from the demineralized dentin matrix (DDM) to infiltrate hydrophobic monomers. However, techniques such as ethanol-wet bonding, which eliminate water, yield only partial improvement, suggesting limitations in this water-centric view. Using chromatography equivalence modeling and Nile red molecular tracing, we identified a solvent-independent redistribution effect exer

The retreatment of failed adhesive restorations consumes more than 60% of clinical resources. Patient factors-such as caries risk, oral hygiene, diet, and occlusal stress-contribute to failure primarily. However, the contribution of the degradation of the adhesive-dentin interface to the failure cannot be ignored and involves 3 interrelated challenges: (1) highly hydrated demineralized dentin matrix (DDM) hindering adhesive infiltration, particularly in the partially demineralized zone; (2) residual hydroxyapatite (HAP) prone to dissolution; and (3) degradation of unprotected collagen by acid-

OBJECTIVES: This study investigated the nanomechanical properties, microstructure, and composition of dentinogenesis imperfecta type II (DGI-II) peritubular dentin (PTD) and intertubular dentin (ITD) and examined the correlations between them.
MATERIALS AND METHODS: Six samples from each of the normal and DGI-II groups were prepared by cutting the midcoronal dentin perpendicular to the dentin tubules. The number and morphology of the dentin tubules were then observed by scanning electron microscopy (SEM). Hydroxyapatite (HAP) was detected using high-resolution atomic force microscopy (HR-AFM)

MicroRNA (miR)-200c enhances osteogenesis, modulates inflammation, and participates in dentin development. This study was to investigate the beneficial potential of miR-200c in vital pulp therapy (VPT) by mitigating pulpitis and promoting dentin regeneration. We explored the miR-200c variations in inflamed pulp tissues from patients with symptomatic irreversible pulpitis and primary human dental pulp-derived cells (DPCs) challenged with P.g. lipopolysaccharide (Pg-LPS). We further assessed the functions of overexpression of miR-200c on odontogenic differentiation, pulpal inflammation, and dent

Dentin Sialoprotein (DSP) and phosphophoryn (PP) are two most dominant non-collagenous proteins in dentin, which are the cleavage products of the DSPP (dentin sialophosphoprotein) precursor protein. The absence of the DSPP gene in DSPP knock-out (KO) mice results in characteristics that are consistent with dentinogenesis imperfecta type III in humans. Symptoms include thin dentin, bigger pulp chamber with frequent pulp exposure as well as abnormal epithelial-mesenchymal interactions, and the appearance of chondrocyte-like cells in dental pulp. To better understand how DSPP influences tooth dev

Human dentin consists of a primary layer produced during tooth formation in early childhood and a second layer which first forms upon tooth eruption and continues throughout life, termed secondary dentin (SD). The effect of attrition on SD formation was considered to be confined to the area subjacent to attrition facets. However, due to a lack of three-dimensional methodologies to demonstrate the structure of the SD, this association could not be determined. Therefore, in the current study, we aimed to explore the thickening pattern of the SD in relation to the amount of occlusal and interprox

Remineralization of dentin during dental caries is of considerable clinical interest. Dentin matrix protein 1 (DMP1) is a non-collagenous calcium-binding protein that plays a critical role in biomineralization. In the present study, we tested if peptides derived from DMP1 can be used for dentin remineralization. Peptide pA (pA, MW = 1.726 kDa) and peptide pB (pB, MW = 2.185), containing common collagen-binding domains and unique calcium-binding domains, were synthesized by solid-phase chemistry. An extreme caries lesion scenario was created by collagenase digestion, and the biomineral-nucleati

BACKGROUND: The prevalence of root caries is increasing with greater life expectancy and number of retained teeth. Therefore, new preventive strategies should be developed to reduce the prevalence of root caries. The aim of this study was to investigate the effects of fluoridated milk on the remineralization of root dentin and to compare these effects to those of sodium fluoride (NaF) application without milk.
METHODS: Thirty extracted human molars were divided into 6 groups, and the root cementum was removed from each tooth. The dentin surface was demineralized and then incubated with one of

The interfacial properties of a new calcium-silicate-based coronal restorative material (Biodentine™) and a glass-ionomer cement (GIC) with dentin have been studied by confocal laser scanning microscopy (CLSM), scanning electron microscopy (SEM), micro-Raman spectroscopy, and two-photon auto-fluorescence and second-harmonic-generation (SHG) imaging. Results indicate the formation of tag-like structures alongside an interfacial layer called the "mineral infiltration zone", where the alkaline caustic effect of the calcium silicate cement's hydration products degrades the collagenous component of

Previous research, in which the bonding effectiveness of the 3 self-etch monomers HAEPA, EAEPA, and MAEPA was determined, showed that MAEPA was most effective. In this study, the molecular interactions of these monomers with hydroxyapatite and dentin were investigated by combining x-ray diffraction, infrared spectroscopy, and scanning electron microscopy. We tested the null hypothesis that the bonding performance of these monomers does not correlate to the formation of monomer-calcium salts and to hydrolytic stability of these monomer-calcium complexes. Monomer/ethanol/water solutions were pre

Fluoride (F) has been a useful instrument in caries prevention. However, only limited data exist on the effect of its long-term use on dentin mineralization patterns and microhardness. The objective of this study was to evaluate the influence of tooth F concentration ([F]) and dental fluorosis (DF) severity on dentin microhardness and mineralization. We collected 137 teeth in Montreal and Toronto, Canada, and Fortaleza, Brazil, where optimum or suboptimum levels of water F were 0.2 ppm, 1 ppm, and 0.7 ppm, respectively. Teeth were analyzed for DF severity, dentin [F], enamel [F], dentin microh

Dentin hypersensitivity is not a new disease, more than a hundred years ago Gysi, later Brännström and many others were dealing with the problems of its symptoms, pathomechanism, differential diagnosis and therapy. The growing interest today maybe attributed to improving oral health, and to the presence of more teeth in older age, as a consequence of decreasing caries prevalence. The exposure of dentine surfaces due to gingival recession, erosion and abrasion may cause serious complaints, occurs in about 40% of the adult population, however, few of them turn to the dentist. Therefore, practisi

An analysis of accumulated experimental results, the mechanism of intratubular crystal deposition in the transparent layer and remineralization of the matrix of carious dentin has led to a more rounded concept and understanding of dentin caries relevant to clinical practice. The transparent layer of carious dentin is not sclerotic but is part of the demineralized softened dentin. The crystal precipitation in the tubules is an intermediate product in a slow demineralization process. Physiological remineralization occurs in matrix of the inner carious dentin which has crossbanded collagen fibers

The incremental lines of von Ebner frequently have been described as ultradian markers of dentin calcification. To determine the relationship between these lines and the quantity of dentin formed, reference was made to an in vivo marker of calcification, tetracycline. These markers were produced by injecting four juvenile monkeys periodically over a period of 175 days. These animals had been subjects in the heat stress study and were exposed to a series of heat stresses and cortisone injections. At the end of the study, undecalcified thin sections of premolars and second molars were prepared b

AIM: The immediate dentin sealing (IDS) approach involves sealing the dentin immediately after tooth preparation and before impression taking. This in vitro study aimed to investigate the effect of saliva contamination at various time intervals on the micro-shear bond strength (µSBS) of dentin sealed using a three-step etch-and-rinse adhesive.
MATERIALS AND METHODS: The occlusal coronal third of 40 intact third molars was removed, and IDS was applied using the three-step adhesive OptiBond FL. The bonded samples were divided into four groups and exposed to non-stimulating natural saliva for 0,
