Transplacental infection with adeno-associated virus type 1 in mice.

Adeno-associated type 1 parvovirus (AAV) was detected in the kidneys and lungs of fetuses and newborns, when pregnant mice were injected subcutaneously with AAV type 1 and murine adenovirus as a helper virus. These findings clearly indicate that transplacental infection with AAV in rodents has been achieved.

Alpha-synuclein overexpression reduces neural activity within a basal ganglia vocal nucleus in a zebra finch model.

Changes in vocal pitch, loudness, and timing are prevalent in Parkinson's Disease (PD) and a target for early intervention and treatment. The neural mechanisms underlying these impairments are not understood, motivating work in animal models. The adult male zebra finch songbird is uniquely poised for these studies given vocally-dedicated brain nuclei and a quantifiable output (birdsong). Our prior publication revealed that injection of an adeno-associated virus (AAV5) expressing the human (h) alpha-synuclein (hSNCA, a-syn) gene into basal ganglia vocal nucleus Area X results in elevated insolu

Trivalent ions kinetic-gating for producing high-concentration and shelf-stable plasmid DNA/PEI particles.

Scalable, cost-effective manufacturing remains a major barrier to the clinical translation of viral vector-mediated gene therapies. The widely used transfection method for producing adeno-associated virus (AAVs) and lentivirus uses plasmid DNA (pDNA)/polyethyleneimine (PEI) particles loaded with multiple plasmids; however, these particles must be prepared at low concentrations and used immediately, limiting scalability and reproducibility. Here we show a kinetic-gating strategy in which transient binding of trivalent citrate ions slows complexation, modulating charge-neutralization kinetics an

Rapid neuronal labeling and functional imaging in the developing mouse brain with AAV-PHP.eB.

Neuronal labeling in the developing brain is essential for studying the formation and maturation of neural circuits. Among labeling techniques, adeno-associated virus (AAV)-based gene delivery offers high efficiency and versatility. However, whether AAV can effectively label neurons in the developing brain has not been fully determined. Here, we demonstrate that administration of AAV-PHP.eB in neonatal mice results in neuronal transduction as early as 3 days after injection. By titrating the AAVs, we achieve sparse neuronal labeling and facilitate high-resolution reconstruction of dendritic mo

AAV capsid-based nanocarriers encapsulating inorganic nanoparticles for targeted delivery.

Current nanoparticle (NP) delivery systems often suffer from off-target accumulation and limited intrinsic targeting capabilities, restricting their biomedical applications. Here, we present a strategy to assemble adeno-associated virus (AAV) capsid proteins around inorganic nanoparticles to generate viral protein nanocarriers (VPNCs) with preserved targeting functionality. AAV capsid protein VP3 is recombinantly expressed in Escherichia coli (E. coli), purified under denaturing conditions, and refolded via a controlled dialysis process. Direct NP addition results in heterogeneous protein coro

Evaluation and Optimization of Different Digestion Strategies for In-Depth Proteomic Characterization of Residual Host Cell Proteins in rAAV-Based Gene Therapy Products.

Recombinant adeno-associated virus vectors (rAAVs) are the most important vectors for in vivo gene therapies, yet their safety relies on low levels of residual host cell proteins (HCPs). While mass spectrometry-based proteomics enables sensitive and untargeted HCP profiling, it faces challenges with matrix interferences from purification buffers and the high dynamic range between abundant viral capsids and trace HCPs. Although sample preparation methods that address these challenges are well-established for antibody products, their adaptation to rAAV purification stages and products remains la

Regulatory Perspective for Biologics License Application of Recombinant Adeno-Associated Virus Products in China.

Recombinant adeno-associated virus (rAAV) vectors have emerged as a leading viral vector for in vivo gene therapy because they offer significant advantages over other viral vectors, such as stable physicochemical properties, low pathogenicity, low integration risk, and long-term expression of the transgene. In recent years, the number of rAAV products that have received approval for commercial marketing and clinical trials has grown rapidly, bringing hope for the treatment of refractory and rare diseases. However, rAAV products are highly innovative and complex, and the manufacturing processes

Suppressing microtubule detyrosination augments adeno-associated virus 2 endosomal escape and gene delivery.

Adeno-associated virus (AAV) is a widely used vector for gene delivery, yet the host intracellular trafficking barriers often limit its therapeutic efficacy. Here, we identify microtubule detyrosination - a microtubule post-translational modification - as a key regulator of AAV2 endo-lysosomal processing. Using super-resolution microscopy (SIM/STORM), we show that upon AAV2 endocytosis, the host upregulates microtubule detyrosination via the GSK3β-CLASP2 signaling axis. Single-particle tracking of recombinant virus reveals that detyrosinated microtubules form a physical and functional barrier,

Channelrhodopsin variants for high-rate optogenetic neurostimulation at low light intensities.

Optogenetics allows versatile control of excitable cell networks, which advances basic science research and drives the development of future medical applications. Fast-closing channelrhodopsins (ChRs) are required for high temporal fidelity of neurostimulation, but their short channel open times require sufficient plasma membrane expression and high light intensity, challenging clinical translation. Here, we addressed the need of high-rate neurostimulation by engineering optimized blue-light-sensitive ChR variants. In particular, we report on the ChR2 variant f-ChR2 TC enabling high frequency

Gene therapy in Duchenne muscular dystrophy.

Duchenne muscular dystrophy (DMD) is a severe X-linked myopathy caused by mutations in the DMD gene, resulting in the absence of functional dystrophin. Gene therapy seems to represent a rational therapeutic strategy, aiming to restore dystrophin expression through delivery of engineered microdystrophin constructs using adeno-associated virus (AAV) vectors. Preclinical studies in murine and canine models demonstrated robust dystrophin restoration, histological improvement, and functional benefit, supporting the transition to clinical trials. Over the past decade, five AAV-microdystrophin progra

Viral Circuit Tracing in Biomedical Pain Research: Methods, Protocols, and Applications.

Viral circuit tracing has transformed pain research by providing detailed insights into the neural circuits underlying pain processing and modulation. This chapter offers a comprehensive overview of the methods, protocols, and applications of viral tracing techniques in pain research, focusing on adeno-associated virus (AAV), rabies virus, and herpes simplex virus (HSV) as key tools. These viral tracers facilitate the mapping of pain-related circuits, such as the periaqueductal gray (PAG)-dorsal raphe (DR) pathway and amygdala-centered circuits, which are pivotal for understanding both the sen

Adeno-associated virus-based gene therapy for hemophilia A and B: a systematic review and meta-analysis.

Adeno-associated virus (AAV)-based gene therapy is an emerging treatment for hemophilia A (HA) and hemophilia B (HB). In this systematic review and meta-analysis, we searched for studies of adult males with severe or moderately severe HA or HB who received AAV-based gene therapy. Annualized bleeding rate (ABR), annualized infusion rate (AIR), total factor use, factor levels, and adverse events (AEs) were extracted. Eight HA trials representing 7 gene therapies and 211 patients and 12 HB trials representing 9 gene therapies and 184 patients were included. For HA, gene therapy resulted in an ann

Neuropathological Findings in Nonclinical Species Following Administration of Adeno-Associated Virus (AAV)-Based Gene Therapy Vectors.

Adeno-associated virus (AAV) gene therapy vectors are an accepted platform for treating severe neurological diseases. Test article (TA)-related and procedure-related neuropathological effects following administration of AAV-based vectors are observed in the central nervous system (CNS) and peripheral nervous system (PNS). Leukocyte accumulation (mononuclear cell infiltration > inflammation) may occur in brain, spinal cord, spinal nerve roots (SNRs), sensory and autonomic ganglia, and rarely nerves. Leukocyte accumulation may be associated with neuron necrosis (sensory ganglia > CNS) and/or gli

Brain-targeted delivery of neuroprotective survival gene minimizing hematopoietic cell contamination: implications for Parkinson's disease treatment.

BACKGROUND: Neurodegenerative diseases, including Parkinson's disease, Amyotropic Lateral Sclerosis (ALS) and Alzheimer's disease, present significant challenges for therapeutic development due to drug delivery restrictions and toxicity concerns. Prevailing strategies often employ adeno-associated viral (AAV) vectors to deliver neuroprotective survival genes directly into the central nervous system (CNS). However, these methods have been limited by triggering immunogenic responses and risk of tumorigenicity, resulting from overexpression of survival genes in peripheral blood mononuclear cells

Extracellular vesicles enhance pulmonary transduction of stably associated adeno-associated virus following intratracheal administration.

Adeno-associated virus (AAV) vector has shown multiple clinical breakthroughs, but its clinical implementation in inhaled gene therapy remains elusive due to difficulty in transducing lung airway cells. We demonstrate here AAV serotype 6 (AAV6) associated with extracellular vesicles (EVs) and secreted from vector-producing HEK-293 cells during vector preparation (EVAAV6) as a safe and highly efficacious gene delivery platform for inhaled gene therapy applications. Specifically, we discovered that EVAAV6 provided markedly enhanced reporter transgene expression in mucus-covered air-liquid interf

Recombinant adeno-associated virus production evaluation in Chinese hamster ovary cells.

The gene therapy field has advanced in recent years with five recombinant adeno-associated virus (rAAV) based products winning Food and Drug Administration (FDA) approval. As the number of therapeutic applications and overall production demands for rAAV increase, it is valuable to evaluate rAAV production in different production cells. Chinese hamster ovary (CHO) cells have been a robust host for biomolecule manufacturing for more than 35 years. However, there is no report to our knowledge describing the use of CHO cells for rAAV production. In this study, we examined the ability of CHO cells

First-in-Patient Dose Prediction for Adeno-Associated Virus-Mediated Hemophilia Gene Therapy Using Allometric Scaling.

In this study, the author compared the performance of two allometric scaling approaches and body-weight-based dose conversion approach for first-in-patient (FIP) dose prediction for adeno-associated virus (AAV)-mediated hemophilia gene therapy using preclinical and clinical efficacy data of nine AAV vectors. In general, body-weight-based direct conversion of effective doses in monkeys or dogs was more likely to underestimate FIP dose but worked for one bioengineered vector with a high transduction efficiency specifically in humans. In contrast, allometric scaling between gene efficiency factor
