A stage-resolved, proteome-supported co-expression atlas reveals coordinated expression remodeling at host entry in Haemonchus contortus.

Haemonchus contortus shifts from free-living larval stages on pasture to parasitic, blood-feeding stages in the host abomasum, but the coordinated gene programs that underlie this transition remain incompletely resolved. We aimed to construct a stage-resolved developmental co-expression atlas of H. contortus using public RNA-seq data and to integrate it with a 2025 developmental proteome atlas. We assembled 42 RNA-seq libraries spanning ten life-cycle stages from egg to adult and built a signed weighted gene co-expression network on 10,485 genes, which resolved 18 co-expression modules whose e

Cellular and molecular basis of tooth eruption.

OBJECTIVES: Tooth eruption requires the presence of a dental follicle (DF), alveolar bone resorption for an eruption pathway, and alveolar bone formation at the base of the bony crypt. The objectives of our investigations have been to determine how the DF regulates both the osteoclastogenesis and osteogenesis needed for eruption. MATERIAL AND METHODS: Multiple experimental methods have been employed. RESULTS: The DF regulates osteoclastogenesis and osteogenesis by regulating the expression of critical genes in both a chronological and spatial fashion. In the rat 1st mandibular molar there is

Efficient Generation of Fetal Hepatic Stellate Cells From hiPSC.

Human induced pluripotent stem cell (hiPSC)-derived liver organoids have emerged as valuable models for studying human liver development. However, existing organoid systems often lack developmentally matched cell populations, particularly fetal hepatic stellate cells (HSCs), limiting their ability to recapitulate key developmental processes. Current approaches for generating HSCs rely on primary cells, immortalized cell lines, or hiPSC differentiation methods that frequently produce activated HSC-like cells and often require cell sorting. Here, we describe an efficient protocol for generating

Deciphering postnatal limb development at single-cell resolution.

The early postnatal limb developmental progression bridges embryonic and mature stages and mirrors the pathological remodeling of articular cartilage. However, compared with multitudinous research on embryonic limb development, the early postnatal stage seems relatively unnoticed. Here, a systematic work to portray the postnatal limb developmental landscape was carried out by characterization of 19,952 single cells from murine hindlimbs at 4 postnatal stages using single-cell RNA sequencing technique. By delineation of cell heterogeneity, the candidate progenitor sub-clusters marked by Cd34 an

Recent advances in Schistosoma mekongi ecology, transcriptomics and proteomics of relevance to snail control.

Mekong schistosomiasis caused by Schistosoma mekongi is a public health problem that occurs along the border between southern Laos and northern Cambodia. Given its restricted distribution and low prevalence, eventual eradication via an effective control program can be expected to be successful. To achieve this goal detailed knowledge of its basic biology, molecular biology, biochemistry, and pathology is urgently required. In this regard, recent studies on transcriptome analysis of adult male and female S. mekongi worms, and proteome analysis of developmental stages have been reported and are

Homology and the evolution of vocal folds in the novel avian voice box.

The origin of novel traits, those that are not direct modifications of a pre-existing ancestral structure, remains a fundamental problem in evolutionary biology. For example, little is known about the evolutionary and developmental origins of the novel avian vocal organ, the syrinx. Located at the tracheobronchial junction, the syrinx is responsible for avian vocalization, but it is unclear whether avian vocal folds are homologous to the laryngeal vocal folds in other tetrapods or convergently evolved. Here, we identify a core developmental program involved in avian vocal fold formation and in

The people behind the papers - Laura Aviñó-Esteban and James Sharpe.

It can be challenging to understand the dynamics of gene expression patterns when embryos develop inside a uterus, making it impossible to image changes to gene expression in real time. A new paper in Development presents a pipeline where images of mouse embryos at discrete developmental timepoints can be used to reconstruct a continuous description of gene expression changes over time. To learn more about the story behind the paper, we caught up with first author Laura Aviñó-Esteban and corresponding author James Sharpe, Head of EMBL Barcelona, Spain.

From embryos to condensates: A developmental biologist's journey.

I am the Huntington Sheldon Professor of Medical Discovery in the Department of Molecular Biology and Genetics in the School of Medicine at the Johns Hopkins University, where I have been running a lab for 30 years. Our research focusses on the molecular control of embryonic polarity and germline development, with an emphasis on asymmetric cell division and biomolecular condensates. We have uncovered mechanisms that localize proteins and RNAs in the cytoplasm by controlling protein diffusion and RNA condensation. My lab has also characterized a repressive program that launches the germline by

The people behind the papers - Ramya Ranganathan and Andrea Streit.

The transcription factor Six1 and its co-factor, Eya1, regulate inner ear development in vertebrates, and loss of Six1 or Eya1 causes developmental differences. In their study, Andrea Streit and colleagues find new gene targets of Six1 that overlap with known human deafness loci in the early inner ear. To know more about their work, we spoke to the first author, Ramya Ranganathan, and the corresponding author, Andrea Streit, Professor of Developmental Neurobiology and Head of the Centre for Craniofacial & Regenerative Biology at King's College London, UK.

The human fetal adrenal cortex and the window of sexual differentiation.

Understanding normal development is fundamental to appreciating postnatal morphology, physiology and, in some instances, pathophysiology. Developmental biology tends to interrogate models in nonprimate species, for instance the mouse, where genetic manipulation gives privileged insight into the function of particular genes. Some human developmental processes, as occur in the adrenal gland, are not faithfully reproduced in these rodent models, yet have an impact on the pathophysiology and treatment of endocrine disorders, such as congenital adrenal hyperplasia. In this setting, in vitro researc

Conserved features and evolutionary shifts of the EDA signaling pathway involved in vertebrate skin appendage development.

It is widely accepted that evolutionary changes in conserved developmental signaling pathways play an important role in morphological evolution. However, few in silico studies were interested in tracking such changes in a signaling pathway. The Ectodysplasin (EDA) pathway provides an opportunity to fill this gap because it is involved in vertebrate skin appendage development such as scales, teeth, hair, and feathers that take an obvious part in the adaptation of species to their environment. We benefited from the large amount of genomic data now available to explore the evolution of the upstre

Developmental programming by prenatal sounds: insights into possible mechanisms.

In recent years, the impact of prenatal sound on development, notably for programming individual phenotypes for postnatal conditions, has increasingly been revealed. However, the mechanisms through which sound affects physiology and development remain mostly unexplored. Here, I gather evidence from neurobiology, developmental biology, cellular biology and bioacoustics to identify the most plausible modes of action of sound on developing embryos. First, revealing often-unsuspected plasticity, I discuss how prenatal sound may shape auditory system development and determine individuals' later cap

Effects and phenotypic consequences of transient thyrotoxicosis and hypothyroidism at different stages of zebrafish Danio rerio (Teleostei; Cyprinidae) skeleton development.

The effects and consequences of changes in thyroid hormones (THs) level are among the actively studied topics in teleost developmental and evolutionary biology. In most of the experimental models used, the altered hormonal status (either hypo- or hyperthyroidism) is a stable characteristic of the developing organism, and the observed phenotypic outcomes are the cumulative consequences of multiple TH-induced developmental changes. Meanwhile, the influence of the transient fluctuations of TH content on skeleton development has been much less studied. Here, we present experimental data on the dev

Activins in reproductive biology and beyond.

BACKGROUND: Activins are members of the pleiotrophic family of the transforming growth factor-beta (TGF-β) superfamily of cytokines, initially isolated for their capacity to induce the release of FSH from pituitary extracts. Subsequent research has demonstrated that activins are involved in multiple biological functions including the control of inflammation, fibrosis, developmental biology and tumourigenesis. This review summarizes the current knowledge on the roles of activin in reproductive and developmental biology. It also discusses interesting advances in the field of modulating the bioac

Color patterns as a model for connecting developmental mechanisms to phenotypic evolution.

Vertebrate color patterns are experimentally accessible and ecologically relevant traits, making them excellent models for studying how diversity arises during development and evolution. Advances in developmental biology, pattern quantification, and broad phylogenetic sampling now offer an opportunity to investigate how pattern development and evolution influence each other. To facilitate this crosstalk, we propose a conceptual framework connecting pattern development with its diversification through two key developmental principles: self-organization and instruction. This integrative framewor

Sildenafil in bronchopulmonary dysplasia-associated pulmonary hypertension: developmental and translational insights.

Bronchopulmonary dysplasia-associated pulmonary hypertension (BPD-PH) is increasingly viewed as a developmental pulmonary vascular disorder rather than a purely reactive vasoconstrictive complication of chronic lung disease. It involves impaired angiogenesis, reduced pulmonary vascular surface area, alveolar simplification, and progressive right ventricle-pulmonary artery uncoupling. Sildenafil, a phosphodiesterase type 5 inhibitor, is widely used because it enhances nitric oxide-cyclic guanosine monophosphate signaling and may reduce pulmonary vascular tone. However, neonatal-specific evidenc

From germline immortality to somatic rejuvenation: Unlocking the ovarian blueprint for longevity.

Aging is typically framed as a one-way, irreversible accumulation of molecular damage in cells and tissues, leading to progressive functional decline. Yet mammalian reproduction, and particularly female reproduction, reveals a striking exception to this rule. Despite residing within an aging organism and within a fast-aging ovarian tissue environment, oocytes give rise to embryos that begin life with restored developmental potential and youthful molecular organization. By reframing ovarian biology as a model for rejuvenation rather than solely as a site of reproductive decline, this Essay prop

Optimization of adenoviral gene transfer in human pluripotent stem cells.

Human pluripotent stem cells, such as embryonic stem cells and induced pluripotent stem cells, have the potential to differentiate into a wide variety of cells in vitro and have applications in basic developmental biology research and regenerative medicine. To understand the process of differentiation from pluripotent stem cells to functional cells, it is necessary to efficiently and safely transfer and express exogenous genes. We attempted to optimize the efficient transfer of genes into pluripotent stem cells using adenoviral vectors. Comparative study of the activities of three representati
