Identification by whole-exome sequencing of new single-nucleotide polymorphisms associated with molar-incisor hypomineralisation among the Lebanese population.

OBJECTIVE: Molar-incisor hypomineralization (MIH) is a developmental qualitative enamel defect, causing a worldwide challenging dental problem. The etiology of this defect remains unclear. Here we identify by whole-exome sequencing (WES) new single-nucleotide polymorphisms (SNPs) in genes expressed during enamel mineralization and in those modulating prenatal, natal and postnatal risk factors among the Lebanese MIH children: immune system and xenobiotic detoxification. DESIGN: Dental examination for MIH was performed based on the MIH index for diagnostic criteria. Saliva samples were collecte

Developmental Abnormalities of Teeth in Children With Nephrotic Syndrome.

INTRODUCTION: Treatment with glucocorticoids in children with nephrotic syndrome can be the cause of developmental disorders of the masticatory organ and bone or teeth abnormalities. The aim was to assess the frequency and type of dental abnormalities and the correlation of their occurrence with a dosage of glucocorticoids and treatment time in children with idiopathic nephrotic syndrome. METHODS: The study group consisted of 31 patients aged 5 to 17 diagnosed with idiopathic steroid-sensitive nephrotic syndrome and 33 overall healthy children. The studies included clinical evaluation of dent

Craniofacial and dental characteristics of three Japanese individuals with genetically diagnosed SATB2-associated syndrome.

Craniofacial defects are one of the most frequent phenotypes in syndromic diseases. More than 30% of syndromic diseases are associated with craniofacial defects, which are important for the precise diagnosis of systemic diseases. Special AT-rich sequence-binding protein 2 (SATB2)-associated syndrome (SAS) is a rare syndromic disease associated with a wide variety of phenotypes, including intellectual disability and craniofacial defects. Among them, dental anomalies are the most frequently observed phenotype and thus becomes an important diagnostic criterion for SAS. In this report, we demonstr

Regional odontodysplasia: morphological, ultrastructural, and immunohistochemical features of the affected teeth, connective tissue, and odontogenic remnants.

PURPOSE: Regional odontodysplasia (RO) is a rare developmental odontogenic defect. The affected teeth are described as "ghost teeth," due to malformation of dentin, enamel, and cementum. The purpose of this study was to describe the ultrastructural characteristics of the affected teeth as well as the immunohistochemical features of the soft tissue associated with 2 cases of regional odontodysplasia. METHODS: Two cases diagnosed as RO were included on the study. After diagnosis, affected teeth and associated soft tissue were surgically removed from both patients and processed for light and sca

Exposure to Per- and Polyfluoroalkyl Substances (PFAS) Causes Dental Developmental Anomalies: Underrecognized Risk of Fluoride Bioaccumulation.

Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals that persist in the environment and have potential health risks. Among them, 8:2 fluorotelomer alcohol (FTOH) undergoes microbial biotransformation in the environment into perfluorooctanoic acid (PFOA) and releases fluoride. While PFOA's toxic effects have been well-studied, the impact of PFOA (and its precursor 8:2 FTOH) on dental health is largely unknown. Moreover, it is not established whether, and to what extent, defluorination and fluoride bioaccumulation occur during the in vivo 8:2 FTOH metabolism. This study is the fir

Proton microprobe determination of fluorine profiles in the enamel and dentine of erupting incisors from sheep given low and high daily doses of fluoride.

Developmental defects in incisors were induced by daily oral ingestion of sodium fluoride solutions. Teeth extracted at eruption from sheep that had been subjected to four different fluoride regimens--0.2 or 0.5 mg F/kg body weight daily for 6 months, 2 or 6 mg F/kg body weight daily for 21 days--were analysed for fluorine by gamma emission using a proton microprobe. Calcium and zinc profiles were also measured using proton-induced X-ray emission. Diffuse opacities, similar in appearance to mild human fluorosis, were produced by the first two regimens, whereas the last two produced hypoplastic

Diagnostic Indices for Epidemiological Assessment of Molar Incisor Hypomineralization: A Systematic Review.

Molar incisor hypomineralization (MIH) is a qualitative developmental defect of enamel and one of the most prevalent oral health conditions in childhood. Over the last two decades, increasing attention has been given to its prevalence and etiology; however, marked heterogeneity in diagnostic criteria and indices has limited the comparability of epidemiological findings. This systematic review aimed to identify and describe the diagnostic indices used for the epidemiological assessment of MIH in the general pediatric population. A systematic literature search was conducted in PubMed, Scopus, an

Nonsurgical management of a dilacerated maxillary lateral incisor with type III dens invaginatus: a case report.

Type III dens invaginatus is a developmental anomaly characterized by an enamel-lined channel that originates on the coronal surface and passes apically through part or all of the root and exits into the periodontal ligament. In this case report, a 13-yr-old male had a Type III dens that exited at the midroot level of tooth #7. At that same level, the root dilacerated severely to the mesial, and a periradicular radiolucency was present on the distal. A 12-mm periodontal defect was present on tooth #6 and a sinus tract was present. All maxillary anterior teeth responded normally to pulp vitalit

Ghost teeth: Regional odontodysplasia of maxillary first molar associated with eruption disorders in a 10-year-old girl.

Regional odontodysplasia (RO) is an uncommon, developmental anomaly of the dental hard tissues that affects ectodermal and mesodermal dental components with characteristic clinical and radiographic findings. Clinically, RO affects a particular segment in either or both dentitions in the maxilla or mandible or both jaws. Radiographic features have consistently demonstrated thin and defective layers of enamel and dentine, resulting in a faint, fuzzy outline, creating a ghost-like appearance. The RO etiology is uncertain; numerous factors have been suggested and considered as local trauma, irradi

Impaired vitamin D signaling reveals novel targets in tooth and alveolar bone.

Teeth are established as targets for vitamin D metabolites but have retained less attention than the well-defined bone cell metabolism. Here we report that VDRgem mice, which bear a point-mutated vitamin D receptor (VDR) unresponsive to the endogenous bioactive vitamin D, are more severely affected and have a greater severity of tooth and alveolar bone defects than VDR-null mice. Transcriptional profiling of postnatal day 5 VDRgem and VDR-null mutant incisors revealed vitamin D ligand signaling targets that promote mineralization, induce alveolar bone growth, and assure dentin integrity. [An analysis of the prevalence of isolated dental anomalies and of those associated with hereditary syndromes: a model for evaluating the genetic control of the dentition characteristics].

Tooth ontogenesis, morphogenesis, and eruption are under a multifactorial (genetic, epigenetic, environmental) control. The aim of this study has been a genetic appraisal of tooth developmental characteristics by means of the prevalence and patterns of association of tooth anomalies in a large sample of hereditary syndromes. MATERIALS AND METHODS. A sample of 169 hereditary syndromes exhibiting tooth abnormalities was collected from the literature: original articles were controlled in order to achieve uniformity of judgement about the definition of the dental defects. Tooth anomalies were clas

Focal odontodysgenesis of the maxillary second premolars in a child.

A few months before her sixth birthday, a Caucasian girl suffered progressively worse episodes of painful, nontender, erythematous swellings of the palate lingual to apparently healthy primary second molars. Neither of the developing maxillary second premolars was discerned clearly in radiographs, but, upon surgical exploration, incomplete crowns were found in both follicles. All the other premolars appeared to be developing normally. Histologic, histochemical, and chemical analyses showed that the enamel was hypocalcified, apparently as a result of hypomaturation; the dentin was essentially n
