OBJECTIVE: Mitochondrial dysfunction is one of the earliest pathological events observed in amyotrophic lateral sclerosis (ALS). The aim of this study is to evaluate the therapeutic efficacy of 2,4-dinitrophenol (DNP), a mild mitochondrial uncoupler, in an ALS mouse model to provide preclinical proof-of-concept evidence of using DNP as a potential therapeutic drug for ALS.

The distribution of transferrin-donated iron between the stroma and cytosol compartments of rabbit reticulocytes was studied. In cells with inhibited haem synthesis iron accumulates in the stroma fraction. The biosynthesis of enterotoxin A by replicating and nonreplicating cells was investigated. Unlike enterotoxin B, a secondary metabolite, enterotoxin A secretion resembled that of a primary metabolite by being secreted during the exponential phase of growth. The amount of toxin produced per unit of growth was not influenced by NaCl, NaNO(2), or NaNO(3). Several surfactants increased toxin secretion. Toxin secretion by nonreplicating cells was inhibited by chloramphenicol and 2, 4-dinitrophenol but not by streptomycin or penicillin G. The optimal pH for enterotoxin A production was 6.5 to 7.0.

Ca(2+) uptake was studied in short-term experiments using 5-day-old excised maize roots. This tissue readily absorbs Ca(2+), and inhibition by dinitrophenol and low temperature shows that the process is metabolically mediated. Female sheep were used to determine the regions of the gastrointestinal tract able to absorb Cu. Copper was absorbed from the abomasum, small intestine and colon, but not from the rumen-reticulum or caecum. In vitro studies indicated that Cu uptake was linearly related to the Cu concentration. We have investigated the effects of different treatments that increase cyclic AMP levels on the in vitro synthesis and release of catecholamines in the rabbit carotid body.
