Extracellular freezing challenges insects in several ways, including mechanical damage from ice crystals, exposure to stressful levels of cold and cellular dehydration. While stress-response mechanisms activated during and immediately after freezing are well studied, less is known about how freeze-tolerant organisms recover from freezing in the longer term and whether this recovery carries energetic costs. Here, we tracked changes in gene expression and energy stores over the course of 15 days following extracellular freezing using the world's southernmost insect, Belgica antarctica (Diptera:

E-20-monooxygenase (E20MO) is an enzymatic product of the shade (shd) locus (cytochrome p450, E20MO). Initially discovered in Drosophila, E20MO facilitates the conversion of ecdysone (E) into 20-hydroxyecdysone (20E) and is crucial for oogenesis. Prior research has implicated 20E in growth, development, and insecticide resistance. However, little attention has been given to the association between the E20MO gene and DENV2 infection. The transcriptome of Ae. aegypti cells (Aag2 cells) infected with DENV2 revealed the presence of the E20MO gene. The subsequent quantification of E20MO gene expres

Ecdysone receptor (EcR) and three insect chitinases (OfChtI, OfChtII, and OfChi-h) are considered as attractive targets for the development of novel insect growth regulators (IGRs) since they are closely related to the insect molting. In this study, to develop potent multi-target IGRs, a series of hexacyclic pyrazol-3-amide derivatives were rationally designed by utilizing the scaffold hopping strategy with the previously reported compound 6j (N-(4-bromobenzyl)-2-phenyl-4,5,6,7-tetrahydro-2H-indazole-5-carboxamide) as a lead compound. The bioassay results indicated that most of the target comp

A mucin-like glycoprotein, peritrophin-55 was isolated and purified from the peritrophic matrix of Lucilia cuprina larvae. When injected into sheep, peritrophin-55 induced an immune response that inhibited larval growth by 51-66% when larvae subsequently fed on sera from the vaccinated sheep. The protein may have potential use as an immunogen probably accompanying other antigens to protect sheep from the cutaneous myiasis caused by these larvae. Peritrophin-55 was uniformly distributed throughout the peritrophic matrix where it probably lubricates the surface of the peritrophic matrix and prot

Mosquitoes impose a significant epidemiological burden on public health through the transmission of malaria parasites and arboviruses. Vector competence, the ability of a mosquito to acquire, maintain, and transmit pathogens, is tightly linked to immune regulation, but immune phenotypes in mosquitoes cannot be interpreted only as pathogen-triggered activation of canonical pathways. Following blood feeding, female mosquitoes undergo extensive physiological remodeling, including endocrine reprogramming, microbiota shifts, oxidative and barrier stress, digestive and metabolic activation, and coor

Optimal feeding strategies for the red claw crayfish, Cherax quadricarinatus, remain to be established. In this study, C. quadricarinatus was fed frozen housefly larva meal (Musca domestica [MD]) at different frequencies to evaluate effects on growth performance, antioxidant capacity, digestive enzyme activity, immune genes, and intestinal microbiota structure. Four feeding protocols were established: control (C) fed basic feed, experimental group 1 (E1) fed basic feed for 2 days and then compound feed (50% MD + 50% basic feed) for 1 day, experimental group 2 (E2) fed basic feed for 1 day and

20-Hydroxyecdysone (20E) is the active form of the steroid hormone ecdysone and is responsible for regulating insect development, molting, and metamorphosis. Infection with the entomopathogenic Beauveria bassiana enhances innate immunity and reduces 20E titers in Lymantria dispar. To further explore the role of the 20E signaling pathway in antifungal immunity, we combined exogenous hormone assays, proteomic profiling, and RNAi-mediated knockdown. In this study, exogenous hormone assays revealed that 20E accelerated the death of B. bassiana-infected larvae, whereas juvenile hormone (JH) markedl

In adult gonads, disruption of somatic sexual identity leads to defective gametogenesis and infertility. However, the underlying mechanisms by which somatic signals regulate germline cells to achieve proper gametogenesis remain unclear. In our previous study, we introduced the chinmoSex Transformation (chinmoST ) mutant Drosophila testis phenotype as a valuable model for investigating the mechanisms underlying sex maintenance. In chinmoST testes, depletion of the Janus Kinase-Signal Transducer and Activator of Transcription downstream effector Chinmo from somatic cyst stem cells (CySCs) femini

MicroRNAs (miRNAs) are small noncoding RNAs that play critical roles in controlling posttranscriptional gene regulation and have a profound effect on mosquito reproduction and metabolism. Juvenile hormone (JH) is critical for achieving reproductive competence in the main vector of human arboviral diseases, Aedes aegypti We report a JH-mediated mechanism governing miRNA expression. Using a transcription factor screen with multiple primary miRNA (pri-miRNA) promoters, we identified that the Ecdysone-induced protein E75 (E75) isoform (E75-RD) induced miRNA gene promoter activity. The use of insect larvae for plastic waste biodegradation has attracted increasing attention as a potential strategy to mitigate environmental plastic pollution. However, the toxicological consequences of ingesting plastics recovered from the environment, particularly those acting as vectors for sorbed contaminants, remain poorly understood. In this study, we investigated the endocrine and neurotoxic effects of field-recovered (environmental) polystyrene (ePS) in the plastivorous insect Zophobas morio. Larvae were fed virgin PS (vPS) or PS collected from a marine coastal area known to be impac

The honeybee (Apis mellifera) is a highly social insect whose caste differentiation is regulated by epigenetic mechanisms, representing a classic example of phenotypic plasticity in social insects. Although the importance of transposable elements (TEs) in epigenetic research is well recognized, their specific role in honeybee caste differentiation has not been fully explored. This study reveals a novel regulatory mechanism where the microRNA (miRNA) ame-mir-3721-3p, derived from ApME (Apis miniature inverted-repeat TEs), suppresses DNA methyltransferase gene DNMT3, promoting queen-like develop

Growth and maturation are coordinated processes in all animals. Integration of internal cues, such as signalling pathways, with external cues, such as nutritional status, is paramount for an orderly progression of development and growth. In Drosophila, this involves insulin and steroid signalling, but the underlying mechanisms and their coordination are incompletely understood. We show that bioactive 20-hydroxyecdysone production by the enzyme Shade in the fat body is a nutrient-dependent process. We demonstrate that under fed conditions, Shade plays a role in growth control. We identify the t

In crustaceans, vitellogenin (Vg) synthesis is regulated by complex hormonal and molecular networks. This study investigates the role of the ecdysone receptor (PmEcR) in regulating Vg expression and identifies downstream effectors in Penaeus monodon. Silencing of PmEcR using RNA interference resulted in a two-fold increase in Vg expression and a significant rise in the gonadosomatic index (GSI), suggesting PmEcR acts as a suppressor of vitellogenesis. Through a suppression subtractive hybridization (SSH) technique, an uncharacterized LOC 113805388 gene, was identified as being downregulated up

INTRODUCTION: Control of the mosquito Aedes albopictus is confounded by its behavior due to females preferring to oviposition in small natural and artificial containers that are often difficult to remove or treat with insecticides. Autodissemination strategies utilizing highly potent insect growth regulators (IGRs) have emerged as promising tools for the control of this container-inhabiting species. The intended goal of autodissemination approaches is to use mosquitoes to self-deliver an IGR to these cryptic oviposition locations. Previous studies have focused on the efficacy of these approach

The ovary in mammals has developed specialized mechanisms for protection against pathogen infections; however, the understanding of the innate immune system in the ovary of crustaceans is still limited. To elucidate the ovary's defense mechanisms in response to viral challenges, we subjected oriental river prawns (Macrobrachium nipponense) to poly I:C, a double-stranded RNA analog that emulates viral dsRNA, and analyzed the ovary's transcriptome profiles. Concurrently, RNA-seq analysis was performed on the hepatopancreas, a well-recognized immune-related tissue, following poly I:C challenge to

Obesity is a global health concern associated with various metabolic disorders including insulin resistance and adipose tissue inflammation characterized by adipose tissue macrophage (ATM) infiltration. In this study, we present a novel Drosophila model to investigate the mechanisms underlying ATM infiltration and its association with obesity-related pathologies. Furthermore, we demonstrate the therapeutic potential of attenuating Eiger/TNFα signaling to ameliorate insulin resistance and ATM. To study ATM infiltration and its consequences, we established a novel Drosophila model (OBL) that mim

UNLABELLED: As in vertebrates, the insect steroid hormones, especially 20-hydroxyecdysone (20E), initiate and regulate sexual behavior by acting on the central nervous system. This 20E action is, in part, triggered by transcriptional events mediated through the binding of 20E to a heterodimer comprising the ecdysone receptor (EcR) and ultraspiracle (USP). However, to date, our knowledge about this genomic steroid pathway remains incomplete. In moths, males detect female sex pheromones, eliciting stereotyped sexual behavior. In Agrotis ipsilon males, the behavioral response and the neuronal sen

BACKGROUND: The insect hemolymph (blood-equivalent fluid), composed of a large number of hemocytes (blood cells) and a variety of soluble immune effectors, is hostile for pathogens including fungi. In order to survive in the insect hemocoel (body cavity), the entomopathogenic fungus (EPF) has evolved two classical coping strategies, namely evasion and suppression of the host immune reactions. However, it remains unclear whether EPF has other ways of coping with host immunity.
RESULTS: In this study, we demonstrated that Metarhizium rileyi (an EPF) infection by injection of blastospores into t

Numerous studies have demonstrated the vital roles of gut microbes in the health, immunity, nutrient metabolism, and behavior of adult worker honeybees. However, a few studies have been conducted on gut microbiota associated with the larval stage of honeybees. In the present study, we explored the role of a gut bacterium in larval development and larval-pupal transition in the Asian honeybee, Apis cerana. First, our examination of gut microbial profiling showed that Bombella apis, a larvae-associated bacterium, was the most dominant bacterium colonized in the fifth instar larvae. Second, we de

OBJECTIVE: Terminal glycosylations such as fucosylation and especially sialylation are crucial in maturation of proteins for their particular function. In cells and tissues where their function is under hormonal control, often terminal processes including glycosylations are tightly regulated by hormones. Therefore, it is not surprising that two key enzyme families involved in these steps, fucosyltransferases and sialyltransferases, are directly controlled by steroid hormones.
METHODS: Gene structure and protein sequences have been analyzed by set of Unix-based complex sequence analysis progra
